Drug-loaded Fe3O4/lignin nanoparticles to treat bacterial infections.

Lignin is a biopolymer employed for various biomedical applications. Nevertheless, the drug loading and release mechanism of Fe3O4 nanoparticles with surface functionalization by lignin has yet to be described. Hence, this study functionalizes Fe3O4 nanoparticles surface with lignin (Fe3O4/Lig) for ciprofloxacin delivery and examines its adsorption-release mechanisms. The presence of lignin and ciprofloxacin on Fe3O4 nanoparticles were verified using FT-IR that shows distinct peaks for each functional group of lignin and ciprofloxacin. The study has proved selective adsorption of ciprofloxacin

Bacterial loads of Ureaplasma urealyticum contribute to development of urethritis in men.

Ureaplasma urealyticum could be a pathogen of non-gonococcal urethritis (NGU) in men. However, ureaplasma is often detected in men without NGU, and the proportion of cases possibly attributable to this pathogen is still undefined. We attempted to determine the bacterial loads of U. urealyticum significantly associated with NGU. The 16S rRNA genes of U. urealyticum were quantified by a real-time polymerase chain reaction-based assay in first-void urine (FVU) from 26 asymptomatic and 25 symptomatic men positive for U. urealyticum. The leucocyte counts in first-void urine (FVU) were determined as

Transposable element loads in a bacterial symbiont of weevils are extremely variable.

Not only are transposable elements profuse in the bacterial endosymbiont of maize weevils, but we found that their quantities also vary approximately 10-fold among individual weevils. Because multicopy elements can facilitate homologous recombination, this insertion sequence (IS) load variability suggests that these essentially asexual bacteria may exhibit substantial intraspecific genomic variation.

Modification of Silver-Loaded Biodegradable Polymer Nanoparticles with Bacterial Membrane Vesicles for Treating Intracellular Bacterial Infections.

Salmonella enterica serovar Typhimurium (S. Typhimurium) is an intracellular pathogen capable of surviving and replicating within macrophages, which causes foodborne diseases such as gastroenteritis. To develop a strategy against intracellular bacteria in macrophages, we designed silver-loaded biodegradable polymer nanoparticles functionalized with S. Typhimurium membrane vesicles (MVs). Silver nanoparticles (Ag NPs) were initially encapsulated within biodegradable poly(lactic-co-glycolic) nanoparticles (Ag-P NPs), which were then surface-modified with polyethyleneimine to form Ag-PP NPs. Thes

Reduced bacterial load in stallion semen by modified single layer centrifugation or sperm washing.

The presence of bacteria poses a significant challenge to the quality of stallion semen used in artificial insemination. The bacterial content of insemination doses arises from various sources, such as the healthy stallion, environment, and collection equipment, and is implicated in fertility problems as well as reduced sperm quality during storage. The conventional approach of adding antibiotics to semen extenders raises concerns about antimicrobial resistance and potential negative effects on sperm characteristics, and may not be effective in inhibiting all bacteria. The objective of this st

Men and Women Repeatedly Infected With Chlamydia trachomatis Have a Lower Urogenital Bacterial Load.

We assessed whether patients repeatedly infected with Chlamydia trachomatis (CT) have a lower urogenital or anorectal CT load. A CT-positive retest was independently associated with higher vaginal and higher urine Cq values (P<0.01). Partial immunity could play a role in repeat urogenital infections, potentially not in anorectal infections.

Impact of bacterial load on pharmacodynamics and susceptibility breakpoints for tigecycline and Klebsiella pneumoniae.

OBJECTIVES: In the absence of other therapeutic options, tigecycline is used to treat bloodstream infections and pneumonia caused by carbapenemase-producing Klebsiella pneumoniae (CP-Kp). In this study, the standard and high tigecycline dosing regimens were simulated and tested against different inocula of CP-Kp isolates in an in vitro pharmacokinetic (PK)/pharmacodynamic (PD) model. METHODS: Four susceptible isolates (EUCAST MICs of 0.125-1 mg/L) and two intermediately susceptible CP-Kp clinical isolates (MICs of 2 mg/L) were tested at three different inocula (107, 105 and 103 cfu/mL), simul

Evaluating the effect of virgin coconut oil pulling on viral load, bacterial load and inflammatory mediator levels in chronic periodontitis - A clinical study.

BACKGROUND AND OBJECTIVE: Periodontitis and dental caries are among the most prevalent oral diseases, with chronic periodontitis being a multifactorial, infectious condition that leads to inflammation in the supporting structures of the teeth, progressive attachment loss, and bone resorption. Chronic periodontitis is driven by a consortium of pathogenic microorganisms. This study aimed to evaluate the efficacy of virgin coconut oil (VCO) pulling in reducing the microbial load and inflammatory mediators responsible for chronic periodontitis, in comparison to chlorhexidine (CHX) mouthwash and di

Estimating bacterial load in S. aureus and E. coli bacteremia using bacterial growth graph from the continuous monitoring blood culture system.

BACKGROUND: We examined whether the time to positivity (TTP) and growth and detection plot graph (GDPG) created by the automated blood culture system can be used to determine the bacterial load in bacteremic patients and its potential association correlation with disease severity. METHODS: Known bacterial inocula were injected into the blood culture bottles. The GDPGs for the specific inocula were downloaded and plotted. A cohort of 30 consecutive clinical cultures positive for S. aureus and E. coli was identified. Bacterial load was determined by comparing the GDPG with the "standard" curves

Histological progression and bacterial load dynamics of Renibacterium salmoninarum in Chinook salmon Oncorhynchus tshawytscha.

Renibacterium salmoninarum, the cause of bacterial kidney disease (BKD), severely impacts salmonid populations. Much of our understanding of the BKD pathology in salmonids comes from evaluating fatal infections in wild populations or spawning Pacific Northwest salmonids. Our study investigated the histological progression and bacterial load dynamics of R. salmoninarum infection in Chinook salmon following intraperitoneal injection to enhance understanding of the disease's dynamics. Seventy presmolt salmon were injected with the ATCC-33209 isolate of R. salmoninarum and monitored over 10 wk. Hi

Exhaustion of CD4+ T cells is associated with bacterial load in active tuberculosis patients.

BACKGROUND: Immunity against TB largely relies on CD4+ T cells. The degree of T cell exhaustion is inversely associated with mycobacterial burden and can be partially reversed upon effective anti-tuberculosis therapy. However, the relationship between PD-1 expression and bacterial load remains incompletely defined. In this study, we sought to characterize PD-1 expression across distinct lymphocyte subsets in a well-characterized cohort of TB patients with quantified bacterial burden. METHODS: Suspected cases were prospectively enrolled in Beijing Chest Hospital between January and December 20

Point-of-care fluorescence imaging reveals extent of bacterial load in diabetic foot ulcers.

Elevated levels of bacteria, including biofilm, increase the risk of chronic wound infection and inhibit healing. Addressing asymptomatic high bacterial loads is challenged by a lack of clinical terminology and diagnostic tools. This post-hoc multicenter clinical trial analysis of 138 diabetic foot ulcers investigates fluorescence (FL)-imaging role in detecting biofilm-encased and planktonic bacteria in wounds at high loads. The sensitivity and specificity of clinical assessment and FL-imaging were compared across bacterial loads of concern (104 -109  CFU/g). Quantitative tissue culture confir

Febrile Children with Pneumonia Have Higher Nasopharyngeal Bacterial Load Than Other Children with Fever.

Febrile episodes are common in children and the most frequent reason for attending emergency services. Although most infections have a benign and self-limiting course, severe and sometimes life-threatening infections occur. This prospective study describes a cohort of children presenting to a single-centre pediatric emergency department (ED) with suspected invasive bacterial infection, and explores the relationships between nasopharyngeal microbes and outcomes. All children attending the ED who had a blood culture taken were offered to participate over a two-year period. In addition to convent

MOTS-c peptide increases survival and decreases bacterial load in mice infected with MRSA.

Sepsis is a life-threatening disease characterized by uncontrolled inflammatory responses upon pathogen infections, especially for the antibiotic-resistant strains, such as Methicillin-resistant S. aureus (MRSA). Here we demonstrated that a Mitochondria-derived peptide (MOTS-c) could significantly improve the survival rate and decrease bacteria loads in MRSA-challenged mice, accompanied with declined levels of pro-inflammatory cytokines, such as TNF-α, IL-6 and IL-1β, but with increased level of anti-inflammatory cytokine IL-10. Moreover this peptide enhanced bactericidal capacity of macrophag

Roflumilast Increases Bacterial Load and Dissemination in a Model of Pseudomononas Aeruginosa Airway Infection.

Exacerbations present a major clinical problem in many patients suffering from chronic obstructive pulmonary disease (COPD). Roflumilast, an inhibitor of phosphodiesterase 4, has shown beneficial effects in several clinical trials and is currently widely used to prevent exacerbations in severe COPD. Roflumilast has anti-inflammatory properties that may interfere with potentially important host defense functions, including cytotoxic properties of neutrophils at sites of inflammation. Since chronic bacterial infection is prevalent in severe COPD, Pseudomonas aeruginosa being a major pathogen, we
