The role and mechanisms of gram-negative bacterial outer membrane vesicles in inflammatory diseases.

Outer membrane vesicles (OMVs) are spherical, bilayered, and nanosized membrane vesicles that are secreted from gram-negative bacteria. OMVs play a pivotal role in delivering lipopolysaccharide, proteins and other virulence factors to target cells. Multiple studies have found that OMVs participate in various inflammatory diseases, including periodontal disease, gastrointestinal inflammation, pulmonary inflammation and sepsis, by triggering pattern recognition receptors, activating inflammasomes and inducing mitochondrial dysfunction. OMVs also affect inflammation in distant organs or tissues v

Bacterial outer membrane vesicles in colorectal cancer: interdomain communication hubs in pathogenesis and immunotherapy.

The complex interaction between the intestinal microbiota and host mucosal immunity plays a defining role in colorectal cancer (CRC) development and therapeutic outcomes. Recently, bacterial outer membrane vesicles (OMVs) - nano-sized, lipid-bilayered extracellular particulates released by both commensal and pathogenic microorganisms - have emerged as critical long-range signalling vehicles within the gut. This review provides a comprehensive synthesis of the dual functionalities of OMVs in CRC pathogenesis and oncology. Mechanistically, pathogenic OMVs cross compromised mucosal barriers to dr

Assessment of periodontitis vaccine using three different bacterial outer membrane vesicles in canine model.

UNLABELLED: Canines frequently develop periodontitis, which is similar and relevant to immunopathology and microbiology of human periodontitis. The aim of this study was to investigate whether bacterial outer membrane vesicle (OMV)-based periodontal vaccines induced humoral immune response in canines from a human vaccine development perspective. Porphyromonas gingivalis (Pg) and Treponema denticola (Td), two major periodontal pathobionts, were chosen as vaccine targets. Intranasal (IN) immunization with Pg OMVs and Td OMVs strongly elicited humoral immune responses against the two respective s

Nanobody Engineered and Photosensitiser Loaded Bacterial Outer Membrane Vesicles Potentiate Antitumour Immunity and Immunotherapy.

Bacterial outer membrane vesicles (OMVs) are promising as antitumour agents, but their clinical application is limited by toxicity concerns and unclear mechanisms. We engineered OMVs with cadherin 17 (CDH17) tumour-targeting nanobodies, enhancing tumour selectivity and efficacy while reducing adverse effects. These engineered OMVs function as natural stimulator of interferon genes (STING) agonists, activating the cyclic GMP-AMP synthase (cGAS)-STING pathway in cancer cells and tumour-associated macrophages (TAMs). Loading engineered OMVs with photoimmunotherapy photosensitisers further enhance

Disrupting Gram-Negative Bacterial Outer Membrane Biosynthesis through Inhibition of the Lipopolysaccharide Transporter MsbA.

There is a critical need for new antibacterial strategies to counter the growing problem of antibiotic resistance. In Gram-negative bacteria, the outer membrane (OM) provides a protective barrier against antibiotics and other environmental insults. The outer leaflet of the outer membrane is primarily composed of lipopolysaccharide (LPS). Outer membrane biogenesis presents many potentially compelling drug targets as this pathway is absent in higher eukaryotes. Most proteins involved in LPS biosynthesis and transport are essential; however, few compounds have been identified that inhibit these p

Cysteine deleted protegrin-1 (CDP-1): anti-bacterial activity, outer-membrane disruption and selectivity.

BACKGROUND: Protegin-1 (PG-1: RGGRLCYCRRRFCVCVGR-amide) assumes a rigid β-hairpin like structure that is stabilized by two disulfide bridges between Cys6-Cys15 and Cys8-Cys13. Previous studies, employing linear analogs of PG-1, with Cys to Ala mutations or modified Cys, have demonstrated that the disulfide bridges are critical for the broad spectrum and salt resistant antimicrobial activity of PG-1. METHODS: In order to understand structural and functional roles of disulfide bonds in protegrins, we have synthesized a Cys deleted variant of PG-1 or CDP-1, RGGRLYRRRFVVGR-amide, and two of its a

Bacterial outer membrane vesicles as intrinsically immunogenic and highly modifiable nanocarriers for precision tumor therapy.

Bacterial outer membrane vesicles (OMVs) were previously considered merely as waste products of bacterial metabolism. But its inherent stability, immunogenicity, ability to be highly regulated, and targeting potential are turning them into a platform with revolutionary potential in tumor drug delivery. Tumor remains a major challenge in medicine. Even though traditional therapies are effective to some extent, they are often accompanied by intolerable side effects. Furthermore, conventional treatments have low targeting efficiency, and tumor cells almost always develop resistance to them. OMVs

Platelet backpacking nanoparticles based on bacterial outer membrane vesicles enhanced photothermal-immune anti-tumor therapy.

Bacterial outer membrane vesicles (OMVs), produced by Gram-negative bacteria, retain the immunostimulatory capacity of parental bacteria. OMVs have been recognized as potent natural immune adjuvants and drug delivery vehicles. Photothermal therapy that triggers immunogenic cell death further stimulates the immune system by releasing damage-associated molecular patterns. This therapeutic effect can be synergized with OMVs to achieve enhanced anti-tumor outcomes. We also observed that tumors can recruit platelets. Leveraging the phenomenon, we have innovatively employed platelets as "couriers" t

Magnetically-targetable outer-membrane vesicles for sonodynamic eradication of antibiotic-tolerant bacteria in bacterial meningitis.

Treatment of acute bacterial meningitis is difficult due to the impermeability of the blood-brain barrier, greatly limiting the antibiotic concentrations that can be achieved in the brain. Escherichia coli grown in presence of iron-oxide magnetic nanoparticles secrete large amounts of magnetic outer-membrane vesicles (OMVs) in order to remove excess Fe from their cytoplasm. OMVs are fully biomimetic nanocarriers, but can be inflammatory. Here, non-inflammatory magnetic OMVs were prepared from an E. coli strain in which the synthesis of inflammatory lipid A acyltransferase was inhibited using C

Mesenchymal stromal cell-derived nanovesicles ameliorate bacterial outer membrane vesicle-induced sepsis via IL-10.

BACKGROUND: Sepsis remains a source of high mortality in hospitalized patients despite proper antibiotic approaches. Encouragingly, mesenchymal stromal cells (MSCs) and their produced extracellular vesicles (EVs) have been shown to elicit anti-inflammatory effects in multiple inflammatory conditions including sepsis. However, EVs are generally released from mammalian cells in relatively low amounts, and high-yield isolation of EVs is still challenging due to a complicated procedure. To get over these limitations, vesicles very similar to EVs can be produced by serial extrusions of cells, after

Lipopolysaccharide confinement in the bacterial outer membrane is governed by interactions within the conserved Lipid A anchor.

The outer membrane of Gram-negative bacteria is an asymmetric bilayer in which lipopolysaccharide (LPS), the principal component of the outer leaflet, promotes tight packing and ordering of the membrane components that are essential for the barrier and load-bearing functions of this membrane. Lipopolysaccharide mobility is known to be restricted in the outer membrane, but this confinement and the underlying biophysical interactions responsible remain to be fully characterized. Here, we apply a bio-orthogonal strategy for in situ site-specific fluorescent labeling of LPS. Using fluorescence mic

Bacterial outer membrane proteins modulate extracellular Dscam isoforms and Dorsal-mediated immunity in Eriocheir sinensis.

The Down syndrome cell adhesion molecule (Dscam) is a highly diverse immune receptor produced through extensive alternative splicing, but whether specific bacterial surface ligands can guide stimulus-specific Dscam splicing and subsequent immune signaling in crustaceans remains unclear. In this study, we examined the role of the Chinese mitten crab (Eriocheir sinensis) Dscam in hemocyte responses to two outer membrane proteins of Vibrio parahaemolyticus, OmpA0764 and OmpA1186. Bacterial challenge increased Dscam expression in hemocytes. Both recombinant proteins significantly elevated Dscam an

Gonococcal outer membrane vesicle vaccines: bacterial population biology, clinical trials, immune profiling, and vaccine design.

Gonorrhoea is a global health concern exacerbated by rising antimicrobial resistance. Retrospective analyses indicate that outer membrane vesicle (OMV) vaccines derived from Neisseria meningitidis (MeNZB, 4CMenB) may offer partial cross-protection against gonococcal infection. This review outlines the influence of gonococcal population biology on coverage, immune responses elicited by 4CMenB, and emerging strategies that offer the prospect of rationally designed vaccines dedicated to the prevention of gonococcal disease.

Spatiotemporal Targeted Delivery of Biomimetic Bacterial Outer Membrane Nanoparticles for Enhanced Spinal Cord Injury Repair.

Spinal cord injury (SCI) is a complex and dynamic pathological condition characterized by disrupted lipid metabolism and neuroinflammatory responses, posing significant therapeutic challenges. To address these, biomimetic bacterial outer membrane nanoparticles (BM-NPs) are designed by integrating the precise targeting capability of detoxified outer membrane vesicles (dOMVs) with the efficient drug-loading properties of liposomes. BM-NPs exhibit superior targeting efficiency toward peripheral neutrophils and macrophages, enabling spatiotemporal drug delivery via immune cells. An innovative "Tor

The role of bacterial outer membrane vesicles in inflammatory response of acute-on-chronic liver failure.

Acute-on-chronic liver failure (ACLF) is a clinical syndrome that manifests as acute deterioration of liver function due to a series of etiologies and triggers in patients with pre-existing chronic liver diseases. Systemic inflammatory response is the major feature of ACLF. Gut microbiota dysbiosis impairs the intestinal barrier, facilitating the translocation of microorganisms and their metabolites into the liver and thereby exacerbating liver inflammation and disease progression. Recent studies have revealed that bacterial outer membrane vesicles (OMVs) derived from gut microbiota act as key

The structure of the bacterial outer membrane transporter FusA enabled by addition of the native lipid lipopolysaccharide.

Lipopolysaccharide (LPS) is a glycolipid found uniquely in the outer membrane of diderm bacteria, formed of 4-7 acyl chains covalently linked to an extended polysaccharide chain. While a few examples of the interaction between LPS and outer membrane proteins (OMPs) have been structurally characterised, either experimentally or computationally, the precise nature of LPS-OMP interactions and their functional consequences remains unclear. Here, we show that the addition of LPS facilitated cryoEM structure determination of FusA, a 100 kDa TonB-dependent outer membrane transporter from P. atrosepti

Engineering CRISPR System-Based Bacterial Outer Membrane Vesicle Potentiates T Cell Immunity for Enhanced Cancer Immunotherapy.

Immune checkpoint blockade (ICB) therapy has revolutionized cancer treatment but only benefits a subset of patients because of insufficient infiltration and inactivation of effector T cells. Bacterial outer membrane vesicles (OMVs) can activate immunity and deliver therapeutic agents for immunotherapy. However, efficiently targeting and packaging therapeutic molecules into OMVs remains challenging. Here, the engineered E. coli BL21-derived OMVs enable the packaging of multiple genes, resulting in a 7-fold increase in DNA enrichment efficiency and gene silencing in vitro. Moreover, the engineer
