Listeria monocytogenes infections: laboratory investigation of an unexpected increase in incidence.

Listeria monocytogenes is a facultative intracellular pathogen causing mainly meningitis and septicemia in immunocompromised hosts. From July 1988 through December 1989, 16 patients at The Mount Sinai Hospital were diagnosed as having listeriosis shortly after admission, 14 within a one-year period (July 1988-July 1989). Because this incidence was almost double the incidence in previous years (< 8 annually), an epidemiologic and microbiologic investigation was undertaken to determine a potential route of acquisition of L. monocytogenes. On the basis of plasmid profile, bacteriocin (enterocin)

Staphylococcosis of turkeys. 6. Development of penicillin resistance in an interfering strain of Staphylococcus epidermidis.

Staphylococcus epidermidis strain 115, used as an interfering agent to help reduce the incidence of staphylococcosis in turkeys, was converted into a penicillin- and chloramphenicol-resistant strain designated 115R. This was accomplished by introducing a plasmid carrying the beta-lactamase (penicillinase) and chloramphenicol-resistance genes into S. epidermidis 115 by electroporation. The resultant strain, 115R, was an efficient producer of beta-lactamase and had marked increased resistance to penicillin and chloramphenicol. A beta-lactamase DNA probe was used to confirm the presence of the be

Plasmids Shaped the Recent Emergence of the Major Nosocomial Pathogen Enterococcus faecium.

Enterococcus faecium is a gut commensal of humans and animals but is also listed on the WHO global priority list of multidrug-resistant pathogens. Many of its antibiotic resistance traits reside on plasmids and have the potential to be disseminated by horizontal gene transfer. Here, we present the first comprehensive population-wide analysis of the pan-plasmidome of a clinically important bacterium, by whole-genome sequence analysis of 1,644 isolates from hospital, commensal, and animal sources of E. faecium Long-read sequencing on a selection of isolates resulted in the completion of 305 plas

Unnoticed spread of linear VanA-plasmids in vancomycin-variable Enterococcus faecium strains across different regions: a diagnostics challenge.

BACKGROUND: Vancomycin-variable enterococci (VVE) are van-positive Enterococcus faecium strains with a vancomycin-susceptible phenotype that can revert to resistance upon exposure to vancomycin, posing an emerging diagnostic challenge. METHODS: We screened VVE within a large collection of E. faecium from hospitalised patients (n = 319) and healthy human volunteers (n = 60) during 2009-2022. VVE prevalence, antimicrobial resistance, plasmid replicases, virulence factors and bacteriocins were investigated using culture-based methods and WGS. vanA-carrying clusters and plasmids were analysed and

Co-occurrence of resistance and virulence plasmids in a blaNDM-5-positive Escherichia coli ST602 clone across One Health sectors in China.

Carbapenem-resistant bacteria circulating across human, animal, and environmental sectors are a growing public health concern. We investigated blaNDM-5-positive Escherichia coli ST602 from an integrated duck-egg farming ecosystem in China using whole-genome sequencing with plasmid characterization. Among 44 representative carbapenem-resistant E. coli (CREC) isolates selected from a larger collection of 103 CREC isolates, seven belonged to ST602 and formed a genetically related O103:H21 cluster recovered from human, duck, and environmental samples within the same farm ecosystem. To place these

Genomic safety, bacteriocinogenic potential, and probiotic-associated traits of Enterococcus lactis LBM-M3D isolated from the swine gastrointestinal tract.

Swine production is vital for global food security; however, its sustainability is threatened by the overuse of antibiotics, which contributes to the emergence and dissemination of multidrug-resistant pathogens. Probiotics and bacteriocin-producing bacteria are promising strategies to support animal health and reduce antibiotic dependence. In this study, Enterococcus lactis LBM-M3D, isolated from the porcine gastrointestinal tract, was evaluated for in vitro antimicrobial activity, tolerance to simulated gastrointestinal conditions, genomic safety features, and bacteriocinogenic potential. ColV increases the virulence of Escherichia coli K1 strains in animal models of neonatal meningitis and urinary infection.

Isogenic Escherichia coli strains, differing in their expression of K1 antigen and ColV plasmid, were studied for their ability to produce disease. Newborn rats were used to test the ability of these strains to colonize the intestine and to produce bacteremia and meningitis; adult rats were used to test their ability to produce urinary tract infection. Colonization of intestine and bladder by K1+ ColV+ E. coli was associated with rapid induction of bacteremia and higher mortalities compared with colonization with K1+ ColV- strains. These findings suggest that the ColV plasmid could play a role

[Transcription map of a recombinant plasmid carrying a gene for the synthesis of pesticine I and a protein for pesticine I immunity in the plague microbe].

Molecules of the plasmids pBR322 and pRD17 have been compared by electron microscopy technique of R-loops visualization. Comparative location of R-loops on the plasmids has been computerized on minicomputer HP9825A due to the program making possible to define the coordinates of the transcription start and direction. The 5.1 kb fragment coding for pesticin I and immunity protein to pesticin I and cloned in pBR322 vector plasmid is flanked by Bam HI-EcoRI sites. Five promoter regions and direction of transcription were localized on the fragment.

Genomic Relationship Between Brochothrix campestris and Its Phages: A Cross-Replicon and Interspecies Perspective.

Background/Objectives: The bacterium Brochothrix campestris is closely related to Brochothrix thermosphacta, a known food spoilage agent, and Listeria monocytogenes, the causative agent of listeriosis. B. campestris garnered attention several years ago because it produces brochocin-C, a bacteriocin capable of inhibiting the growth of certain pathogens. It has been recently suggested that phages play a significant role in the evolution of B. thermosphacta, similar to the role they play for L. monocytogenes. However, understanding the role of phages in the evolution of B. campestris has been cha

Trans-complementable copy-number mutants of plasmid ColE1.

Plasmid ColE1, like many other small non-conjugative plasmids, is present in multiple copies (about 15 per chromsome equivalent) in Escherichia coli cells. Because of their high copy number, the replication of such plasmids has been described as 'relaxed', even though there is good evidence that it is strictly controlled: ColE1 derivatives have characteristic but different copy numbers and ColE1 copy-number mutants have been characterised. No plasmid-specified protein is essential for the replication of ColE1 and related plasmids, as extensive replication can occur in chloramphenicol-treated c

Listeria monocytogenes lineages: Genomics, evolution, ecology, and phenotypic characteristics.

Listeria monocytogenes consists of at least 4 evolutionary lineages (I, II, III, and IV) with different but overlapping ecological niches. Most L. monocytogenes isolates seem to belong to lineages I and II, which harbor the serotypes more commonly associated with human clinical cases, including serotype 1/2a (lineage II) and serotypes 1/2b and 4b (lineage I). Lineage II strains are common in foods, seem to be widespread in the natural and farm environments, and are also commonly isolated from animal listeriosis cases and sporadic human clinical cases. Most human listeriosis outbreaks are assoc

Genomic exploration of the fermented meat isolate Staphylococcus shinii IMDO-S216 with a focus on competitiveness-enhancing secondary metabolites.

BACKGROUND: Staphylococcus shinii appears as an umbrella species encompassing several strains of Staphylococcus pseudoxylosus and Staphylococcus xylosus. Given its phylogenetic closeness to S. xylosus, S. shinii can be found in similar ecological niches, including the microbiota of fermented meats where the species may contribute to colour and flavour development. In addition to these conventional functionalities, a biopreservation potential based on the production of antagonistic compounds may be available. Such potential, however, remains largely unexplored in contrast to the large body of r

Vancomycin-resistant enterococci isolates from the rumen content of deer.

The total enumeration of enterococci from the rumen content of deer, their species determination, resistance to antibiotics with the predominant importance of vancomycin resistance, and their metabolic properties, were examined. Enterococci (2.0 x 10(2) CFU per ml) encountered from the rumen of deer were phenotypically allotted to the species Enterococcus casseliflavus (26.6%), E. gallinarum (26.6%) and E. faecium (6.6%). The other isolates were not phenotypically specified. Although the resistance to the other antibiotics was recorded, the emergence of vancomycin-resistant enterococci in a hi

A double outbreak of exfoliative toxin-producing strains of Staphylococcus aureus in a maternity unit.

This report describes a double outbreak of staphylococcal scalded skin syndrome (SSSS) in which two distinct tetracycline-resistant strains of Staphylococcus aureus producing different exfoliative toxins were involved. In the first phase the daytime staff of the delivery unit and eczematous skin conditions in midwives were implicated as the probable source. In the second phase a source within a post-natal ward was suggested with local cross-infection. In the final phase both sources were epidemiologically linked to cases of SSSS. Because early discharge was the policy of the unit many cases pr

[The nature of typhimuricin--a substance produced by Salmonella typhimurium LT2 and relation of its synthesis to the cryptic plasmid].

The effect of typhimuricin on Escherichia coli K12 cells and the properties of this substance produced by Salmonella typhimurium LT2 have been studied. The obtained data permit one to conclude that typhimuricin is similar to the aminoacid L-valine or the chemical very similar to it in properties. The synthesis of typhimuricin is not controlled by the cryptic plasmid present in Salmonella typhimurium LT2 cells. The experiments aimed at isolation of enterobacterial strains producing low molecular mass antibiotics--microcines have been done. It was found that the capability to produce macrocines

Identification, characterization, and recombinant expression of epidermicin NI01, a novel unmodified bacteriocin produced by Staphylococcus epidermidis that displays potent activity against Staphylococci.

We describe the discovery, purification, characterization, and expression of an antimicrobial peptide, epidermicin NI01, which is an unmodified bacteriocin produced by Staphylococcus epidermidis strain 224. It is a highly cationic, hydrophobic, plasmid-encoded peptide that exhibits potent antimicrobial activity toward a wide range of pathogenic Gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA), enterococci, and biofilm-forming S. epidermidis strains. Purification of the peptide was achieved using a combination of hydrophobic interaction, cation exchange, and h

Colicin U, a novel colicin produced by Shigella boydii.

A novel colicin, designated colicin U, was found in two Shigella boydii strains of serovars 1 and 8. Colicin U was active against bacterial strains of the genera Escherichia and Shigella. Plasmid pColU (7.3 kb) of the colicinogenic strain S. boydii M592 (serovar 8) was sequenced, and three colicin genes were identified. The colicin U activity gene, cua, encodes a protein of 619 amino acids (Mr, 66,289); the immunity gene, cui, encodes a protein of 174 amino acids (Mr, 20,688); and the lytic protein gene, cul, encodes a polypeptide of 45 amino acids (Mr, 4,672). Virulence profiles and other biological characters in water isolated Aeromonas hydrophila.

Thirty water isolates of A. hydrophila were tested for potential virulence profiles, antibiotic resistance and Bacteriocin-Like Substances (BLS) production. Cytotoxic activity was present in all strains tested, 87% were hemolytic and 70% adhesive. Lysine decarboxylase reactions (LDC) positivity was correlated with virulence factors: 100% versus cytotoxicity, 84% versus adherence, 76% versus hemolytic activity. The correlation was also present in the LDC-negative strains. Hemolytic and cytotoxic activities were frequently associated: high cytotoxicity, corresponding to high hemolytic activity a

Vancomycin-resistant Enterococcus faecium: A current perspective on resilience, adaptation, and the urgent need for novel strategies.

Vancomycin-resistant Enterococcus faecium (VREfm) has become a critical opportunistic pathogen, urgently requiring new antimicrobial strategies due to its rising prevalence and significant impact on patient safety and healthcare costs. VREfm continues to evolve through mutations and the acquisition of new genes via horizontal gene transfer, contributing to resistance against several last-resort antibiotics. Although primarily hospital-associated, VREfm are also detected in the community, food chain, livestock, and environmental sources like wastewater, indicating diverse transmission pathways
