Bacteriophages are the most abundant biological entities in the biosphere. Due to their host specificity and ability to kill bacteria rapidly, bacteriophages have many potential healthcare applications, including therapy against antibiotic-resistant bacteria. Infection by flagellotropic bacteriophages requires a properly rotating bacterial flagellar filament. The flagella-dependent phage χ (Chi) infects serovars of the pathogenic enterobacterium Salmonella enterica However, cell surface receptors and proteins involved in other stages of χ infection have not been discovered to date. We screened

Bacteriophages are gaining increasing attention as targeted biological control agents against multidrug-resistant (MDR) foodborne pathogens. However, repeated exposure to bacteriophages can induce the emergence of bacteriophage-resistant mutant (BIM) strains, which may exhibit altered physiological characteristics that impair therapeutic efficacy. This study investigated the phenotypic and molecular adaptive mechanisms of multidrug-resistant Salmonella Typhimurium BIMs that acquired resistance to receptor-specific bacteriophages (STP-1 and STP-2) targeting O-antigen and flagella. The TRAIL (Tumour Necrosis Factor-Related Apoptosis-Inducing Ligand) is a promising candidate for cancer treatment due to its unique ability to selectively induce programmed cell death, or apoptosis, in cancer cells while sparing healthy ones. This selectivity arises from the preferential binding of the TRAIL to death receptors on cancer cells, triggering a cascade of events that lead to their demise. However, significant limitations in using the TRAIL for cancer treatment are the administration of the TRAIL protein that can potentially lead to tissue toxicity (off-target) and the short half-l

The ferrichrome-iron receptor encoded by the fhuA gene of Escherichia coli K-12 is a multifunctional outer membrane receptor required for the binding and uptake of ferrichrome and bacteriophages T5, T1, phi 80, and UC-1 as well as colicin M. To identify domains of the protein which are important for FhuA activities, a library of 31 overlapping deletion mutants in the fhuA gene was generated. Export of FhuA deletion proteins to the outer membrane and receptor functions of the deletion proteins were analyzed. All but three of the deletion mutant FhuA proteins cofractionated with the outer membra

The rising prevalence of multidrug-resistant Klebsiella pneumoniae strains, particularly the emerging high-risk ST16 lineage, highlights the urgent need for alternative therapeutic strategies. A major barrier to effective bacteriophage (phage) therapy against K. pneumoniae is its polysaccharide capsule, which hinders phage access to surface receptors. In this study, we identified and characterized an anti-KL51 capsule depolymerase derived from the anti-ST16 phage PWKp9B. The recombinant DpoK51-9B enzyme was expressed in Escherichia coli, purified, and shown to degrade the KL51 capsule using Pe

Pseudomonas aeruginosa is an opportunistic pathogen that can cause sinus infections and pneumonia in cystic fibrosis (CF) patients. Bacteriophage therapy is being investigated as a treatment for antibiotic-resistant P. aeruginosa infections. Although virulent bacteriophages have shown promise in treating P. aeruginosa infections, the development of bacteriophage-insensitive mutants (BIMs) in the presence of bacteriophages has been described. The aim of this study was to examine the genetic changes associated with the BIM phenotype. Biofilms of three genetically distinct P. aeruginosa strains,

Bacteriophage lambda adsorbs to its Escherichia coli K-12 host by interacting with LamB, a maltose- and maltodextrin-specific porin of the outer membrane. LamB also serves as a receptor for several other bacteriophages. Lambda DNA requires, in addition to LamB, the presence of two bacterial cytoplasmic integral membrane proteins for penetration, namely, the IIC(Man) and IID(Man) proteins of the E. coli mannose transporter, a member of the sugar-specific phosphoenolpyruvate:sugar phosphotransferase system (PTS). The PTS transporters for mannose of E. coli, for fructose of Bacillus subtilis, and

The global rise of antibiotic resistance highlights the urgent need for alternative therapeutics such as bacteriophages (phages). Phages exert selective pressure that can "steer" bacteria toward reduced pathogenicity, increased antibiotic susceptibility, or immune clearance. A receptor-focused steering strategy is especially powerful since many phage receptors are also critical for bacterial fitness and virulence. Effective therapy requires identifying these receptors during characterization and combining phages that target distinct, conserved structures to minimize resistance. We review the c

As the adsorption receptor of bacteriophage tail protein, bacterial lipopolysaccharide (LPS) is a main culprit responsible for nonalcoholic fatty liver disease (NAFLD) caused by high-fat diets. However, few studies have focused on how the interaction between intestinal bacteriophages and bacterial LPS affects the development and progression of NAFLD. Herein, we determined that excessive fat intake significantly increases the levels of endogenous LPS, while the administration of beneficial metabolites of the intestinal microbiota (specifically butyrate) alleviated hepatic injury in rats. The be

With the rise of antibiotic-resistant pathogens, bacterial infections have become an increasingly significant threat. Bacteriophages have the potential to complement the current treatment options. To ensure that phage biocontrol is efficient and long-lasting, the risk of resistance development must be minimized, and phage resistance mechanisms should be thoroughly investigated. In this study, a Tn5 transposon mutagenesis library of the fire blight pathogen Erwinia amylovora was screened to identify multi-phage-resistant mutants. These mutants had transposon insertions in the genes Eamy_RS26425

Nucleotide second messengers are widely used to mediate the physiology and antiphage immunity in bacteria. It was revealed that 2',3'-cAMP/2',3'-cGMP synthetase activity is required for immune response in plants. However, it remains unclear whether 2',3'-cNMPs are involved in the interactions between bacteria and phages. Here, we report that 2',3'-cNMPs, which are synthesized by RnaA and degraded by A1S_2339, A1S_0249, and A1S_2666, play critical roles in the modulation of the physiology of Acinetobacter baumannii. 2',3'-cNMPs specifically bind to the receptor protein CRPAb in A. baumannii and

Yersinia pestis is the etiological agent of plague. Marmota himalayana of the Qinghai-Tibetan plateau is the primary host of flea-borne Y. pestis. This study is the report of isolation of Mu-like bacteriophages of Y. pestis from M. himalayana. The isolation and characterization of four Mu-like phages of Y. pestis were reported, which were named as vB_YpM_3, vB_YpM_5, vB_YpM_6, and vB_YpM_23 according to their morphology. Comparative genome analysis revealed that vB_YpM_3, vB_YpM_5, vB_YpM_6, and vB_YpM_23 are phylogenetically closest to Escherichia coli phages Mu, D108 and Shigella flexneri ph

The rise of antibiotic resistance, coupled with a dwindling antibiotic pipeline, presents a significant threat to public health. Consequently, there is an urgent need for novel therapeutics targeting antibiotic-resistant pathogens. Nisin, a promising peptide antibiotic, exhibits potent bactericidal activity through a mechanism distinct from that of clinically used antibiotics. However, its cationic nature leads to hemolysis and cytotoxicity, which has limited its clinical application. Here, nanodelivery systems have been developed by mimicking the mechanisms bacteriophages use to deliver their

Rhizogenic Agrobacterium (Rhizobium) spp. are causative agents of hairy root disease (HRD), a major threat to hydroponic crop production worldwide. The use of specific bacteriophages is considered a prospective approach to control the development of HRD in greenhouses. A combination of diverse bacteriophages is a key step to overcome potential phage resistance in the pathogen. In this study, a novel lytic bacteriophage, named Miki, was identified and characterized for its antibacterial potential against a rhizogenic Agrobacterium sp. strain circulating in greenhouses in Central Russia. PURPOSE: Klebsiella pneumoniae is a multidrug-resistant (MDR) bacterium that has emerged as a major global public health threat. The decreasing effectiveness of conventional antibiotics has renewed interest in bacteriophage therapy as a promising alternative antibacterial strategy. However, the rapid emergence of phage resistance remains a critical limitation. This study aimed to investigate whether indole-3-acetic acid (IAA) can modulate bacterial quorum sensing (QS) and enhance phage therapy by modulating the QS regulator SdiA.
METHODS: Experiments were performed using the lytic bacteriopha

Liver cancer, mainly hepatocellular carcinoma (HCC), remains a global health burden marked by poor prognosis with limited therapeutic efficacy, and high recurrence rates. HCC remains one of the most lethal malignancies worldwide, with limited therapeutic options and high resistance to conventional treatments. Despite low therapeutic efficacy, molecular heterogeneity, treatment resistance and high recurrence rate, hepatocellular carcinoma (HCC) is still a significant health problem worldwide. These restrictions have stimulated the research of focused methods for delivering therapeutic genetic p

In this study, we describe the isolation and characterization of novel bacteriophage vB_EcoP_Kapi1 (Kapi1) isolated from a strain of commensal Escherichia coli inhabiting the gastrointestinal tract of healthy mice. We show that Kapi1 is a temperate phage integrated into tRNA argW of strain MP1 and describe its genome annotation and structure. Kapi1 shows limited homology to other characterized prophages but is most similar to the seroconverting phages of Shigella flexneri and clusters taxonomically with P22-like phages. The receptor for Kapi1 is the lipopolysaccharide O-antigen, and we further

Campylobacter jejuni continues to be the leading cause of bacterial food-borne illness worldwide, so improvements to current methods used for bacterial detection and disease prevention are needed. We describe here the genome and proteome of C. jejuni bacteriophage NCTC 12673 and the exploitation of its receptor-binding protein for specific bacterial detection. Remarkably, the 135-kb Myoviridae genome of NCTC 12673 differs greatly from any other proteobacterial phage genome described (including C. jejuni phages CP220 and CPt10) and instead shows closest homology to the cyanobacterial T4-related

Phage therapy often fails when bacteria evolve resistance. We argue that phage selection should begin with receptors whose modification imposes predictable costs, turning resistance into reduced virulence, antibiotic resensitization, or other exploitable trade-offs. Receptor-constrained evolutionary traps offer a framework for designing phages that steer-not merely suppress-bacterial evolution effectively.
