Identification and characterization of two Bacillus anthracis bacteriophages.

Anthrax is an acute infectious zoonotic disease caused by Bacillus anthracis, a bacterium that is considered a potential biological warfare agent. Bacillus bacteriophages shape the composition and evolution of bacterial communities in nature and therefore have important roles in the ecosystem community. B. anthracis phages are not only used in etiological diagnostics but also have promising prospects in clinical therapeutics or for disinfection in anthrax outbreaks. In this study, two temperate B. anthracis phages, vB_BanS_A16R1 (A16R1) and vB_BanS_A16R4 (A16R4), were isolated and showed sipho

Inactivation of Lactobacillus Bacteriophages by Dual Chemical Treatments.

Phage contamination is one of the significant problems in the food fermentation industry, which eventually causes economic losses to the industry. Here, we investigated the viability of Lactobacillus plantarum phage P1 and P2 using various biocides treatments (ethanol, isopropanol, sodium hypochlorite and peracetic acid). Results indicated that phage P1 and P2 could be completely inactivated by treatment with 75% ethanol for 5 min, followed by 400 ppm of sodium hypochlorite treatment for 5 min. Phage P2 could be completely inactivated in the reverse sequence, while 800 ppm of sodium hypochlori

Bacteriophages for the Targeted Control of Foodborne Pathogens.

Foodborne illness is exacerbated by novel and emerging pathotypes, persistent contamination, antimicrobial resistance, an ever-changing environment, and the complexity of food production systems. Sporadic and outbreak events of common foodborne pathogens like Shiga toxigenic E. coli (STEC), Salmonella, Campylobacter, and Listeria monocytogenes are increasingly identified. Methods of controlling human infections linked with food products are essential to improve food safety and public health and to avoid economic losses associated with contaminated food product recalls and litigations. Filamentous bacteriophages, natural nanoparticles, for viral vaccine strategies.

Filamentous bacteriophages are natural nanoparticles formed by the self-assembly of structural proteins that have the capability of replication and infection. They are used as a highly efficient vaccine platform to enhance immunogenicity and effectively stimulate the innate and adaptive immune response. Compared with traditional vaccines, phage-based vaccines offer thermodynamic stability, biocompatibility, homogeneity, high carrying capacity, self-assembly, scalability, and low toxicity. This review summarizes recent research on phage-based vaccines in virus prevention. In addition, the expre

Bacteriophages Isolated From Turkeys Infecting Diverse Salmonella Serovars.

Salmonella is one of the leading causes of foodborne illnesses worldwide. The rapid emergence of multidrug-resistant Salmonella strains has increased global concern for salmonellosis. Recent studies have shown that bacteriophages (phages) are novel and the most promising antibacterial agents for biocontrol in foods because phages specifically kill target bacteria without affecting other bacteria, do not alter organoleptic properties or nutritional quality of foods, and are safe and environmentally friendly. Due to the vast variation in Salmonella serotypes, large numbers of different and highl

Potential of Therapeutic Bacteriophages in Nosocomial Infection Management.

Nosocomial infections (NIs) are hospital-acquired infections which pose a high healthcare burden worldwide. The impact of NIs is further aggravated by the global spread of antimicrobial resistance (AMR). Conventional treatment and disinfection agents are often insufficient to catch up with the increasing AMR and tolerance of the pathogenic bacteria. This has resulted in a need for alternative approaches and raised new interest in therapeutic bacteriophages (phages). In contrast to the limited clinical options available against AMR bacteria, the extreme abundance and biodiversity of phages in n

Bacteriophages as model organisms for virus emergence research.

Viruses fully emerge by gaining the ability to sustainably infect new host populations. When the hosts are humans, emerging viruses can present major public health issues, as exemplified by the AIDS pandemic. Therefore, heuristic approaches to identify nascent diseases before they become pandemic would be valuable. Unfortunately, the current patient-based and epidemiological approaches are ill-suited in this regard because they are largely responsive and not predictive. Alternative approaches based on virus evolutionary ecology might have greater potential to predict virus emergence. However,

Impact of Pseudomonas aeruginosa biofilm exopolysaccharide composition on bacteriophage and bacteriophage-antibiotic combination activity.

Multidrug-resistant Pseudomonas aeruginosa is a leading cause of hospital-acquired infections, including medical device infections, partially due to the organism's ability to produce biofilm on prosthetic material. Increased antibiotic tolerance of bacteria within biofilm, along with the increasing prevalence of infections caused by multidrug-resistant P. aeruginosa, including extensively drug-resistant phenotypes, results in scenarios where conventional antibiotics may fail to effectively treat these infections. Bacteriophage (phage) therapy is a promising alternative to conventional antimicr

Temperate bacteriophage induced in Pseudomonas aeruginosa biofilms can modulate bacteriophage and antibiotic resistance.

Given the high resistance levels in Gram-negative bacteria, phage therapy is receiving increasing attention. In Germany, a clinical study is already underway evaluate a phage cocktail for treating Pseudomonas aeruginosa in cystic fibrosis (CF) patients. In this context, we investigated the prevalence of PF1-like prophages in P. aeruginosa isolates from a local CF cohort, their ability to undergo lytic conversion during biofilm formation, and the resulting impact on the resistance profile of the P. aeruginosa population. Consistent with other studies, prophage Pf4 was the most prevalent in this

Bacteriophages as Immune Modulators: Current Insights and Future Directions.

Bacteriophages are by far the most abundant biological entities on Earth. A phage is a virus that targets bacterial hosts via the lysogenic cycle or lytic cycle, with the latter serving the principal mechanism in personalized phage therapy for treating bacterial infections. Although phage therapy has been routinely used in some countries for more than half a century ago, the rise of modern antibiotic production and its widespread adoption diminished interest. The emergence of increasingly severe antimicrobial resistance has prompted health care to explore nonantibiotic treatment options, inclu

Bacteriophages as vaccine platforms: Opportunities and challenges in translation.

Bacteriophages (phages) have recently received increased interest as versatile candidates for vaccine development. Their inherent characteristics, such as ease of genetic manipulation, high-density antigen display, intrinsic immunostimulatory properties, demonstrated human safety, and scalability in bacterial hosts, make them attractive as next-generation vaccine platforms. Additionally, their cost-effective production, stability, and existing regulatory approval for food and compassionate phage therapy provide a strong foundation for further development of phage-based vaccines. LYTIC CAPABILITY OF BACTERIOPHAGES (FAMILY MYOVIRIDAE) ON BURKHOLDERIA PSEUDOMALLEI.

Burkholderia pseudomallei, a gram-negative bacillus found in soil and water, is the causative agent of melioidosis. It can produce a biofilm, which increases resistance to antibacterial agents. Bacteriophages (phages) have been suggested as alternative antibacterial agents. In this study, the ability of six phages (family Myoviridae) to lyse B. pseudomallei isolates was examined using a microplate phage virulence assay. The six phages were more efficient in lysing soil than clinical B. pseudomallei isolates. Phage ST79 had the highest lytic capability, independent of inoculating phage quantity

Inactivation of Lactobacillus delbrueckii bacteriophages by heat and biocides.

The effect of several biocides and thermal treatments on the viability of four Lactobacillus delbrueckii phages was investigated. Time to achieve 99% inactivation of phages at 63 and 72 degrees C in three suspension media (Tris Magnesium Gelatin (TMG) buffer, Man Rogosa Sharpe (MRS) broth and reconstituted nonfat dry skim milk (RSM)) was calculated. Thermal resistance depended on the phage considered, but a marked heat-resistance was exhibited by one phage (Ib(3)) since its high titre suspensions were completely inactivated only after 45 min at 72 degrees C or 15 min at 90 degrees C. [Bacteriophage Therapy in Orthopaedics and Trauma].

Bacteriophage therapy has re-emerged as a potential adjunctive strategy in the management of orthopedic and trauma-related infections, particularly fracture-related infection (FRI) and periprosthetic joint infection (PJI). These conditions are associated with biofilm formation, repeated surgery, prolonged antimicrobial therapy, impaired functional outcomes, and increasing rates of multidrug-resistant pathogens. The therapeutic rationale of bacteriophages is based on their high specificity for target bacteria, their ability to replicate at the site of infection, and their potential activity aga

Assembly of the tail of bacteriophage T4.

The protein products of at least 21 phage genes are needed for the formation of the tail of bacteriophage T4. Cells infected with amber mutants defective in these genes are blocked in the assembly process. By characterizing the intermediate structures and unassembled proteins accumulating in mutant-infected cells, we have been able to delineate most of the gene-controlled steps in tail assembly. Both the organized structures and unassembled proteins serve as precursors for in vitro tail assembly. We review here studies on the initiation, polymerization, and termination of the tail tube and con

Treatment and prophylaxis of cholera with bacteriophage.

The treatment of cholera with bacteriophage has, in the past, been unsuccessful, owing to the fact that the phages prepared by the usual method (multiplication on cholera vibrios in broth) proved incapable of parasitizing cholera vibrios in vivo. In this paper, the author presents the results of therapeutic and prophylactic trials of a phage preparation prepared by a different method-namely, cultivation on cholera vibrios through alternate passages in the small intestine of guinea-pigs and in bile. This preparation was used effectively for the treatment of cholera patients and for the control
