Potential roles of gut microbiota in metal mixture and bone mineral density and osteoporosis risk association: an epidemiologic study in Wuhan.

Few studies have focused on the effects of multiple metal mixtures on bone health and the underlying mechanisms related to alterations in the gut microbiota. This study aimed to examine the potential roles of gut microbiota alterations in metal mixtures and their association with osteoporosis traits. Adults aged ≥ 55 years were recruited from two community healthcare centers in Wuhan City during 2016-2019. The plasma concentrations of six metals (zinc, iron, selenium, lead, cadmium, and arsenic) were measured using an inductively coupled plasma mass spectrometer. The k-means clustering method

Influence of programmed death ligand 1 (PD-L1) knockout on gut microbiota in experimental autoimmune uveitis.

PURPOSE: Programmed death ligand 1 (PD-L1) is a potential target for autoimmune disease therapies. The gut microbiota plays a critical role in autoimmunity, and may influence therapeutic outcomes of immune therapies in cancer. However, the relationship between PD-L1 and gut microbiota in autoimmune conditions remains unclear. This study aims to investigate the effect of PD-L1 knockout on gut microbiota in an experimental autoimmune uveitis (EAU) model. METHODS: EAU was induced via immunization with interphotoreceptor retinoid-binding protein peptide 651-670 (IRBP651-670) in either wild type (

In vitro antibacterial activity of zinc oxide on a broad range of reference strains of intestinal origin.

Minimal inhibitory concentrations (MIC) and growth behaviour in ZnO supplemented media were determined by the broth micro dilution method against 75 reference strains. No clear clustering according to bacterial group was observed, but 10 of 11 Lactobacillaceae strains showed high zinc resistance (≥520 μg mL(-1)). Enterobacterial strains showed high (6/11) to medium resistance (5/11). The Clostridia and Bacteroidaceae strains exhibited a diverse range of MIC. The results of this study show that zinc resistance of commensal intestinal bacteria cannot be grouped according to their taxonomic origi

Gastrointestinal flora of cotton rats.

The gastrointestinal (GI) flora of cotton rats was examined. No lactobacilli were detected in any part of the GI tract. Anaerobes, including Peptococcaceae, Bacteroidaceae, bifidobacteria and eubacteria, were the predominant bacteria in the stomach, small intestine, caecum and faeces. Aerobes and facultative anaerobes, including Enterobacteriaceae and streptococci, were detected at low numbers and very low frequency of occurrence in all parts of the GI tract. Sixty-one isolates of bifidobacteria were recovered from the stomach, small intestine, caecum and faeces of cotton rats. They were ident

[Promotion of Porphyromonas gingivalis to viral disease].

Chronic periodontitis is one of the most common oral diseases in humans, the main recognized pathogenic bac-terium of which is the Porphyromonas gingivalis. Various types of viruses have been detected in periodontal disease in situ, and the joint action of viral and bacterial pathogens infection mechanism are complicated. Porphyromonas gingivalis has the characteristics resulting from the interaction with a variety of bacterium viruses, which may be the reason for chronic perio-dontitis being a protracted disease associated with a variety of systemic diseases. In this paper, we reviewed the re

Non-oral Prevotella stepping into the spotlight.

Species now affiliated to genus Prevotella have been known for decades as an integral part of human oral cavity microbiota. They were frequently isolated from patients with periodontitis or from dental root canals but also from healthy subjects. With the exception of Prevotella intermedia, they were considered opportunistic pathogens, as they were isolated also from various bacterial abscesses from the head, neck, breast, skin and various other body sites. Consequently, Prevotella were not in the focus of research activities. On the other hand, the four species found in the rumen never caused

Serious infection with Leptotrichia buccalis. Report of a case and review of the literature.

Leptotrichia buccalis is an obligately anaerobic, gram-negative rod classified as a member of the family Bacteroidaceae. Organisms found in this family (Bacteroides and Fusobacteria) are frequent causes of severe infection in human beings. L. buccalis is a constituent of normal oral flora. Leptotrichia has rarely been recovered from mixed infections primarily of the head and neck. It has not previously been reported as a cause of serious infection in man. We report a case of cavitary pneumonia and septicemia caused by L. buccalis. The colonial morphology and gram stain of L. buccalis are chara

Black-pigmented gram-negative anaerobes in endodontic infections.

Necrotic dental root canal infections are polymicrobial infections dominated by anaerobic bacteria. The number of different species in one canal is usually low, approx. 4-7 species. The species isolated most frequently belong to the genera Prevotella, Porphyromonas, Fusobacterium, Peptostreptococcus, Eubacterium and Streptococcus. The frequency of isolation of black-pigmented Gram-negative anaerobes in endodontic infections varies from 25% to > 50%. Pr. intermedia is the most commonly found pigmented species, followed by Pr. denticola and two Porphyromonas species, P. gingivalis and P. endodon

Black-pigmented gram-negative anaerobes in genito-urinary tract and pelvic infections.

Black-pigmented Gram-negative anaerobes are part of the normal vaginal flora and contribute to a range of superficial and deep genital infections. Prevotella melaninogenica is found in moderate numbers (10(4-6) cfu/g) in healthy women; the numbers and detection rates increase in anaerobic vaginosis (where it may be a significant contributor to changed microbial metabolic activity that gives the signs and symptoms of this condition) and in other vaginal infective conditions. Pr. melaninogenica is also part of the mixed flora in deep pelvic infections: endometritis, post-partum and post-abortal

Deep metagenomic characterization of the gut virome in pregnant women with preeclampsia.

Preeclampsia (PE), a pregnancy-specific syndrome, has been associated with the gut bacteriome. Here, to investigate the impact of the gut virome on the development of PE, we identified over 8,000 nonredundant viruses from the fecal metagenomes of 40 early-onset PE and 37 healthy pregnant women and profiled their abundances. Comparison and correlation analysis showed that PE-enriched viruses frequently connected to Blautia species enriched in PE. By contrast, bacteria linked to PE-depleted viruses were often the Bacteroidaceae members such as Bacteroides spp., Phocaeicola spp., Parabacteroides

Integrated analysis of human-mouse gut microbiota in RSV infection based on machine learning.

BACKGROUND: Respiratory syncytial virus (RSV) is a leading cause of lower respiratory tract infections in children, but effective treatment options remain limited. The gut-lung axis, which highlights the role of gut microbiota in regulating respiratory immunity, provides new opportunities for developing probiotic-based therapies. However, existing studies on RSV-associated gut microbiota are often small-scale and lack systematic integration. To address this gap, we conducted a comprehensive machine learning-based analysis of gut microbiota data from RSV-infected children and mice, integrating

[Oral infection and rheumatic diseases].

Periodontitis (PD) is one of the most common infectious diseases of dental attachment. From epidemiological studies there is known association of periodontitis with chronic diseases as for example diabetes mellitus, cardiovacular diseases, atherosclerosis or Crohn disease. In last decade there is an increasing evidence for association of rheumatoid arthritis (RA) and periodontitis also. RA and PD have some common genetic, environmental and immunopathological characters. Important aspect of reciprocal relationship is also ability to citrullination, which is innate to one of most important oral

Feasibility and therapeutic strategies of vaccines against Porphyromonas gingivalis.

Periodontitis is a chronic infectious disease that is highly prevalent worldwide and is characterized by inflammation of the gums, and loss of connective tissue and bone support. The Gram-negative anerobic bacterium Porphyromonas gingivalis is generally accepted as the main etiological agent for chronic periodontitis. The objective of this paper is to elucidate the feasibility of achieving protection against periodontitis though immunization against P. gingivalis. Until now, animal studies have showed no complete protection against P. gingivalis. However, current knowledge about P. gingivalis

HTP-1 exhibited significant immunoregulatory activities on CTX-induced immunosuppressed mice in a dose-depend manner by alleviating jejunum injury and improving the levels of immune organ indexes, cytokines and immunoglobulins. Moreover, HTP-1 supplementation boosted the content of SCFAs, altered the intestinalmicrobiota composition, and raised the abundances of beneficial bacteria M

Spatial organization of intestinal microbiota in the mouse ascending colon.

Complex microbial populations are organized in relation to their environment. In the intestine, the inner lining (mucosa) is a potential focal point for such organization. The proximal murine colon contains mucosal folds that are known to be associated with morphologically distinct microbes. To identify these microbes, we used the technique of laser capture microdissection (LCM) to sample microbes associated with these folds (interfold region) and within the central lumen (digesta region). Using 16S rRNA gene tag pyrosequencing, we found that microbes in the interfold region were highly enrich

Acute myocardial infarction induces sex-specific, time-dependent remodeling of the gut microbiome and intestinal immune compartment in retired breeder C57BL/6N mice.

INTRODUCTION: The gut microbiome influences cardiovascular health through metabolite production and immune modulation. Gut microbial dynamics and cardiovascular outcomes are also shaped by biological sex. However, sex-specific responses to myocardial infarction (MI) that involve the gut microbiome and intestinal milieu remain poorly defined, particularly in older hosts. Here, we characterize gut microbiome structure and function alongside physiological and immune responses to MI across multiple tissues in aging male and female mice. METHODS: MI was induced by permanent LAD ligation and confir

Gut microbiota composition and tumor immune features in meningioma patients.

UNLABELLED: Meningiomas are prevalent intracranial tumors with poorly understood extraneural drivers. While the gut-brain axis influences neuro-oncogenesis, meningioma-specific gut microbiome alterations and their clinical implications remain uncharacterized. This study integrated 16S rRNA sequencing of fecal samples from 15 treatment-naïve WHO grade I meningioma patients (MPs) and 15 healthy controls (HCs) with immunohistochemical profiling of tumor immune infiltrates (MPO⁺ neutrophils, CD68⁺ macrophages, CD3⁺ T cells). Compared with HCs, MPs exhibited significantly reduced alpha diversity (S
