In a series of three experiments during March-October, 1998, two species of captive-bred poison dart frogs (Dendrobates tinctorius and D. auratus) were exposed to Batrachochytrium dendrobatidis, a recently-described chytridiomycete fungus (chytrid) that was originally isolated from a blue poison dart frog (D. azureus). All frogs exposed to the chytrids developed a fatal skin disease, whereas none of the control frogs developed skin lesions. The most consistent clinical sign in chytrid-exposed frogs was excessive shedding of skin. Gross lesions were subtle, usually affected the legs and ventrum

BACKGROUND: Brain metastases (BM) are a major complication in metastatic breast cancer, especially in HER2-positive and triple-negative subtypes. Despite advances in systemic therapy, BM remains associated with a poor prognosis and often requires radiotherapy. This study evaluates the outcomes of brain irradiation across subtypes and proposes a prognostic scoring system to guide treatment.
METHODS: This retrospective study analyzed data from 348 breast cancer patients with BM to develop a prognostic scoring system for radiotherapy decisions. The data included molecular subtypes, number of bra

As human activity accelerates the spread of pathogens and other invasive species, reconstructing the cryptic early dynamics of undetected invasions has become a central challenge in disease ecology and invasion biology. Here, we leverage museum specimens to reconstruct the invasion of Batrachochytrium dendrobatidis (Bd), a fungal panzootic driving global amphibian biodiversity loss, across historically enigmatic declines in the Brazilian Atlantic Forest. Two hypotheses have been proposed to explain Bd's historical dynamics in this region: long-term enzootic persistence versus recent epizootic

Amphibians globally suffer from emerging infectious diseases like chytridiomycosis caused by the continuously spreading chytrid fungi. One is Batrachochytrium salamandrivorans (Bsal) and its disease ‒ the 'salamander plague' ‒ which is lethal to several caudate taxa. Recently introduced into Western Europe, long distance dispersal of Bsal, likely through human mediation, has been reported. Herein we study if Alpine salamanders (Salamandra atra and S. lanzai) are yet affected by the salamander plague in the wild. Members of the genus Salamandra are highly susceptible to Bsal leading to the leth

Pathogens inflict various costs onto their hosts from sublethal changes in physiology and behavior to intense pathology and mortality. The timing of host immune responses and concomitant changes in behavior may be jointly underpinned by an increase in glucocorticoid hormones. The temporal and causal links between these interrelated responses to infection remain equivocal in many host-pathogen systems. Here, using a fungal pathogen implicated in global amphibian declines, Batrachochytrium dendrobatidis (Bd), we examined sublethal consequences of infection in the Cuban tree frog (Osteopilus sept

In the midst of the current biodiversity crisis, amphibians are severely threatened by emerging diseases such as chytridiomycosis. Characterizing the mechanisms that underlie susceptibility to this disease is fundamental to improve amphibian conservation. Using a comprehensive multi-omics approach, this study investigates the impact of an exposure to the chytrid fungus Batrachochytrium salamandrivorans (Bsal) on the gene expression of Italian crested newts (Triturus carnifex) and on the bacterial symbionts that constitute their skin microbiota. Exposure to Bsal affected multiple components of

Batrachochytrium dendrobatidis continues to cause declines in amphibian populations worldwide, and it remains unclear why skin defenses often fail to control the infection. Although amphibians have a complex, multifunctional immune system, the chytridiomycosis agent seems to have evolved countermeasures that enable it to survive and eventually impair critical skin functions. Previous studies show that B. dendrobatidis cells or cell-free supernatants inhibit lymphocytes by inducing apoptosis, suggesting impaired local cell killing. However, there is little evidence of lymphocyte recruitment to

Environmental change is reshaping wildlife reproduction through increasing temperatures and often leads to the spread of emerging infectious diseases, yet the physiological consequences of managing these stressors remain poorly understood. Amphibians are particularly vulnerable owing to their ectothermy and high susceptibility to chytridiomycosis, caused by Batrachochytrium dendrobatidis (Bd). Here, we examined how Bd infection and thermal treatment interact to influence sperm quality and reproductive investment in male green and golden bell frogs (Ranoidea aurea), a species that has suffered

Chytridiomycosis caused by Batrachochytrium dendrobatidis (Bd) has been documented in greater sirens (Siren lacertina) in the wild and in the pet trade. This study evaluated the use of terbinafine-impregnated implants for chytridiomycosis prophylaxis in greater sirens exposed to Bd. Implants were placed intracoelomically in both control (blank implant, n = 4) and treatment (24.5 mg of terbinafine implant, n = 4) groups. Sirens were exposed to Bd zoospores via 24-h immersion bath at 1 and 2 mon postimplant placement. Blood was collected monthly for plasma terbinafine levels, and skin swabs were

Disruption of habitat connectivity alters host movement patterns and pathogen exposure in wildlife. Changes in exposure dynamics have led to increased research interest in host-associated microbial communities (i.e., microbiomes), particularly in how repeated encounters with pathogens may drive microbial filtering processes that favor the assembly of pathogen-inhibiting microbiomes, a concept known as the adaptive microbiome principle. Understanding how habitat connectivity and pathogen exposure shape adaptive microbiomes remains a key frontier in disease ecology. For widely distributed waterb

Bacteriophages play important roles in shaping microbial community dynamics across diverse environments. In the amphibian skin, most microbiome studies have focused on bacteria and their interactions with the fungus Batrachochytrium dendrobatidis (Bd), leaving other microbial components, including viruses, largely unexplored. Here, we present the first characterization of the viral community in the amphibian skin microbiome, focusing on ecological drivers of bacteriophage diversity and functional potential in the axolotl Ambystoma altamirani. Using public shotgun metagenomes, we found that the

In a survey of blood parasites in Ugandan frogs, 30 (17%) of 180 frogs were infected with at least 1 species of Hepatozoon or Trypanosoma, or with microfilariae. There were significant differences in the prevalence of parasitism among species, with parasitemia detected in only 3 of 9 species. The intensity of blood parasite infection ranged from 1 to 1,080 infected cells per 5,000 cells examined. Seasonal changes in the prevalence and intensity of parasitemia were not observed, nor was there any association between parasitemia and infection with the pandemic fungus Batrachochytrium dendrobatid

The chytrid fungus, Batrachochytrium dendrobatidis, causes chytridiomycosis and is a major contributor to global amphibian declines. Although amphibians have robust immune defenses, clearance of this pathogen is impaired. Because inhibition of host immunity is a common survival strategy of pathogenic fungi, we hypothesized that B. dendrobatidis evades clearance by inhibiting immune functions. We found that B. dendrobatidis cells and supernatants impaired lymphocyte proliferation and induced apoptosis; however, fungal recognition and phagocytosis by macrophages and neutrophils was not impaired.

Amphibians are the most threatened vertebrate group worldwide, with emerging infectious diseases playing an important role in their ongoing declines. Although the amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd) has received considerable attention, viruses of the genus Ranavirus (Rv) remain less explored, particularly in South America. In this study, we report the first detection of Rv in two threatened amphibian species from Brazil. Specifically, we found infected tadpoles of the redbelly toads Melanophryniscus admirabilis (1.4% prevalence) and Melanophryniscus biancae (24% preval

With amphibians still holding the record as the most threatened class of terrestrial vertebrates, their skin microbiota has been shown to play a relevant role in their survival in a fast-changing world. Yet little is known about how abiotic factors associated with different aquatic habitats impact these skin microorganisms. Here we chose the yellow-bellied toad (Bombina variegata), a small anuran that colonizes a wide range of wetland habitats, to investigate how the diversity and composition of both its bacterial and fungal skin communities vary across different habitats and with water charac

While epizootics increasingly affect wildlife, it remains poorly understood how the environment shapes most host-pathogen systems. Here, we employ a three-step framework to study microclimate influence on ectotherm host thermal behaviour, focusing on amphibian chytridiomycosis in fire salamanders (Salamandra salamandra) infected with the fungal pathogen Batrachochytrium salamandrivorans (Bsal). Laboratory trials reveal that innate variation in thermal preference, rather than behavioural fever, can inhibit infection and facilitate salamander recovery under humidity-saturated conditions. Yet, a

Terbinafine is an orally and topically active allylamine antifungal agent with fungistatic and fungicidal action. Its mechanism of action is the competitive inhibition of squalene epoxidase which leads to squalene accumulation and ergosterol depletion in the cell membrane. Clinically relevant pathogens can include but are not limited to: Aspergillus spp., Candida spp., Blastomyces dermatitidis, Histoplasma capsulatum, Emydomyces testavorans, Ophidiomyces ophiodiicola, and Batrachochytrium dendrobatidis. Terbinafine is an antifungal drug that has been used successfully in humans with few advers

The fungal pathogen Batrachochytrium dendrobatidis (Bd) has driven severe amphibian declines across the Neotropics, yet some species persist under enzootic pathogen conditions following epizootics. We investigated whether skin bacterial communities and host mucosome activity contributed to the persistence of seven amphibian species in a once megadiverse Chocó-Andean community that experienced drastic Bd-associated declines. Across dry and wet seasons, we characterized skin bacterial communities using 16S rRNA metabarcoding and assessed mucosome-mediated inhibition of a local Bd lineage using i

Fungal infections in humans, wildlife, and plants are a growing concern because of their devastating effects on human and ecosystem health. In recent years, populations of many amphibian species have declined, and some have become extinct due to chytridiomycosis caused by the fungal pathogen Batrachochytrium dendrobatidis. For some endangered amphibian species, captive colonies are the best intermediate solution towards eventual reintroduction, and effective antifungal treatments are needed to cure chytridiomycosis and limit the spread of this pathogen in such survival assurance colonies. Amphibian species around the world are currently becoming endangered or lost at a rate that outstrips other vertebrates—victims of a combination of habitat loss, climate change and susceptibility to emerging infectious disease (Stuart et al. 2004). One of the most devastating such diseases is caused by the chytrid fungus Batrachochytrium dendrobatidis (Bd), which infects hundreds of amphibian species on multiple continents. While Bd itself has been characterized for some time, we still know little about the mechanisms that make it so deadly. In this issue of Molecular Ecology, Rosenblum et al.
