Inhaled predatory bacteria-loaded large porous microspheres to eradicate drug-resistant Pseudomonas aeruginosa from the lung.

The pneumonia caused by antimicrobial-resistant Gram-negative bacteria is an intractable clinical problem due to the lack of effective treatments. Inspired by the unique predatory bacterial ability of Bdellovibrio bacteriovorus, we here developed an inhalable live bacterial formulations, i.e., B. bacteriovorus-loaded poly(lactic-co-glycolic acid) (PLGA) large porous microspheres (BPMs), to eradicate antimicrobial-resistant Pseudomonas aeruginosa from the lung. BPMs serve as a "safe house" of B. bacteriovorus to avoid being phagocytized by macrophages due to their large size; while the continua

Effect of predatory bacteria on the gut bacterial microbiota in rats.

Bdellovibrio bacteriovorus and Micavibrio aeruginosavorus are Gram-negative proteobacteria that are obligate predators of other Gram-negative bacteria and are considered potential alternatives to antibiotics. Most studies focusing on predatory bacteria have been performed in vitro, thus the effect of predatory bacteria on a live host, including the impact on the ecology of the native microbiota, has yet to be fully examined. In this study, intrarectal inoculations of Sprague-Dawley rats with predatory bacteria were performed. Additionally, feces were collected for seven days post-inoculation t

Functional analysis of natural variation in the RNA-binding protein CsrA across the bacterial domain predicts regulatory activity.

Bacteria employ sophisticated post-transcriptional regulatory mechanisms to adapt to environmental changes. Carbon storage regulator A (CsrA), a highly conserved RNA-binding protein, serves as a critical post-transcriptional regulator by typically recognizing GGA-containing hairpin loops in target mRNAs and repressing translation. However, how this conserved regulator evolved diverse species-specific regulatory networks remains unclear. We developed Swarm-seq, a high-throughput platform assessing CsrA homologs across the bacterial domain for regulating flagella-dependent swarming in Bacillus s

Pulmonary lesions reshape swine respiratory microbiota: evidence of bacterial dysbiosis and reduced diversity.

The swine lower respiratory tract (LRT) microbiota has increasingly been investigated, but important knowledge gaps remain, limiting our understanding of microbial communities involved in respiratory health and disease. This longitudinal study evaluated the LRT bacterial composition of pigs with pulmonary lesions (PL+) compatible with Mycoplasma hyopneumoniae infection and without pulmonary lesions (PL-) across production phases from nursery to slaughter. Using 16 S-rRNA gene sequencing of intratracheal mucus and lung fragments, we observed longitudinal shifts in bacterial community compositio

Examining the efficacy of intravenous administration of predatory bacteria in rats.

The proteobacteria Bdellovibrio bacteriovorus and Micavibrio aeruginosavorus are obligate predators of Gram-negative bacteria, and have been proposed to be used to treat multidrug-resistant bacterial infections. The ability of predatory bacteria to reduce bacterial burden in vivo within the lungs of rats has been demonstrated, but it was unknown if predatory bacteria can attenuate systemic bacterial burden administered intravenously. In this study, we first assessed the safety of intravenous inoculation of predatory bacteria in rats. No rat morbidity or adverse histopathology of various organs

Oleanolic Acid Modulates the Gut-Liver Axis to Alleviate High-Fat Diet-Induced Hepatic Lipid Deposition in Nile Tilapia (Oreochromis niloticus).

This study examined the protective mechanisms of oleanolic acid (OA) against high-fat diet (HFD)-induced hepatic steatosis and intestinal dysbiosis in Nile tilapia. Fish were allocated to four groups: normal diet (ND), HFD, and OA-supplemented HFD (50 and 250 mg/kg). After 42 days, physiological, biochemical, and histological assessments demonstrated that OA markedly reduced hepatic lipid accumulation, mitochondrial injury, and intestinal shortening. Transcriptomic analysis revealed that OA alleviated lipid dysregulation by inhibiting de novo lipogenesis and promoting lipid trafficking and β-o

Clearance of Gram-Negative Bacterial Pathogens from the Ocular Surface by Predatory Bacteria.

It was previously demonstrated that predatory bacteria are able to efficiently eliminate Gram-negative pathogens including antibiotic-resistant and biofilm-associated bacteria. In this proof-of-concept study we evaluated whether two species of predatory bacteria, Bdellovibrio bacteriovorus and Micavibrio aeruginosavorus, were able to alter the survival of Gram-negative pathogens on the ocular surface. Clinical keratitis isolates of Pseudomonas aeruginosa (strain PAC) and Serratia marcescens (strain K904) were applied to the ocular surface of NZW rabbits followed by application of predatory bac

Combined Application of Bacterial Predation and Violacein to Kill Polymicrobial Pathogenic Communities.

Violacein is a bisindole antibiotic that is effective against Gram-positive bacteria while the bacterial predator, Bdellovibrio bacteriovorus HD100, predates on Gram-negative strains. In this study, we evaluated the use of both together against multidrug resistant pathogens. The two antibacterial agents did not antagonize the activity of the other. For example, treatment of Staphylococcus aureus with violacein reduced its viability by more than 2,000-fold with or without B. bacteriovorus addition. Likewise, predation of Acinetobacter baumannii reduced the viability of this pathogen by more tha

Analysis of gill and skin microbiota in Larimichthys crocea reveals bacteria associated with cryptocaryoniasis resistance potential.

Cryptocaryoniasis, caused by the ciliate parasite Cryptocaryon irritans, poses a significant threat to the large yellow croaker (Larimichthys crocea) in intensive marine aquaculture. This study explores the interaction between skin and gill microbiota and C. irritans infection, focusing on the role of commensal microbes in disease resistance. Fish were challenged with 100 theronts per gram of body weight, leading to substantial microbial dysbiosis, characterized by decreased alpha diversity and disrupted co-occurrence networks, particularly on the skin. Post-infection, Vibrio abundance signifi

Outer membrane changes enable evolutionary escape from bacterial predation.

Antimicrobial resistance (AMR) is a threat to modern medicine. To combat AMR pathogens, natural predators like bacteriophages and predatory bacteria have gained interest recently. Predatory bacterium Bdellovibrio bacteriovorus is ubiquitous and has a broad prey range. It is particularly potent at killing many AMR Gram-negative bacterial pathogens featured on the WHO priority list. However, it is currently unclear whether prey bacteria can evolve genetically-determined resistance against predation by B. bacteriovorus. Here, we show that the model bacterium Escherichia coli K-12 consistently evo

The role of microbiota in bovine tuberculosis: Characterizing bacterial communities across lungs, gut, and lymph nodes.

Animals with bovine tuberculosis (bTB) develop granulomatous lesions as part of a complex immune response to chronic antigenic stimulus involving innate and adaptive mechanisms; however, the association of the microbiota with the development of and susceptibility to bTB remains poorly understood. This study characterizes the bacterial communities at different body sites involved in the pathophysiology of bTB, highlighting the gut-lung axis and their association with disease. Lungs, gut, thoracic, and mesenteric lymph nodes samples were collected from 31 bovines from a single herd (n = 19 bTB;

Predatory bacteria can intensify lung-injury in a multidrug-resistant Acinetobacter baumannii pneumonia model in rat.

INTRODUCTION: Respiratory tract infection caused by antibiotic-resistant bacteria are one of the most important causes of death worldwide. Therefore, in this study, we investigated the possibility of using predatory bacteria to improve the Acinetobacter baumannii pneumonia model in rat. METHODS: Multidrug-resistant (MDR) A. baumannii clinical strain was used to induce pneumonia. In addition to the sham and predator control group, three treatment groups (n = 5) were studied with colistin, Bdellovibrio bacteriovorus HD100, and combination of predator and antibiotics. Also, the colistin MIC valu

[Ultrastructural basis of interactions between prokaryotes and eukaryotes in different symbiotic models].

This paper reviews the Author's contribution to the knowledge of the ultrastructural basis of the prokaryote-eukaryote interactions in different models assessed by an ultrastructural approach. In agreement with the hypothesis of the origin of eukaryotic cells, which are chimeras of several prokaryotes with different morpho-functional specializations, symbiosis had major consequence for evolution of life. In Arthropods, one of the most successful lifestyles, the presence of endosymbiotic prokaryotes, plays an important role in their metabolism. In some cases, genome integration has occurred in

Impact of a tropical monsoon climate on formaldehyde exposure and microbial contamination in anatomy dissection hall.

Gross anatomy dissection is an essential component of medical and health science education, yet it presents notable occupational hazards, particularly from formaldehyde (FA) exposure and microbial contamination. These risks may be intensified in anatomy dissection halls located in tropical monsoon (Am) climates, where elevated humidity and temperature promote both chemical volatility and microbial persistence. This study assessed the combined effects of such climatic conditions on FA concentrations and microbial ecology within a naturally ventilated dissection hall in southern Thailand. FA lev

Fate and removal of iodinated X-ray contrast media in membrane bioreactor: Microbial dynamics and effects of different operational parameters.

Iodinated X-ray contrast media (ICM) are mainly used in medical sector, and their presence in environmental waters is a cause of concern as they are capable of forming highly toxic iodinated disinfection byproducts. In the present study, the removal mechanisms of the three ICM- iohexol, iopromide, and iopamidol were elucidated in a lab-scale aerobic membrane bioreactor (MBR). At steady-state operation (solids retention time (SRT)- 70 days, organic loading rate (OLR)- 0.80 KgCOD/m3-day, nitrogen loading rate (NLR)- 0.08 KgNH4-N/m3-day, hydraulic retention time (HRT)- 12 h), the average removal

Impacts of an azo food dye tartrazine uptake on intestinal barrier, oxidative stress, inflammatory response and intestinal microbiome in crucian carp (Carassius auratus).

Food dyes, or color additives, are often added into foods, cosmetics and beverages during processing to improve the sensory attributes of the final products. However, the toxicity of tartrazine (TZ), one of the most common azo-dyes, is still unclear, and needs to be ascertained by further study. Hence, in the present study, we aimed to evaluate the effects of TZ consumption on health by using a teleost, crucian carp (Carassius auratus) as the experimental fish. TZ consumption (1.4, 5.5 and 10 mg/kg bwt/day) could cause severe histopathological and cellular alterations in intestine and liver. T

Microbiota-based therapies in oral health and disorders.

The human oral microbiome is a complex, dynamic ecosystem critically involved in maintaining oral health and contributing to systemic well-being. Many bacteria and fungi are involved in oral cavities such as Penicillium, Rhodotorula, Saccharomycetales, Streptococcus, Veillonella, Neisseria, Actinomyces, and Schizophyllum. Disruption of microbial homeostasis, or dysbiosis, underpins a wide spectrum of oral diseases, including dental caries, periodontal disease, endodontic infections, and mucosal conditions. Recent advances in microbiome research have elucidated the mechanisms by which pathogeni
