Ultrastructural analysis of beetle larva cuticles during infection with the entomopathogenic fungus, Beauveria bassiana.

BACKGROUND: Beauveria bassiana is one of the commercially available entomopathogenic fungi (EPF), and a number of isolates with high virulence and broad host spectrum have been used to control agricultural and forest pests. Although the functional importance of genes in EPFs' pathogenesis have been extensively studied, the precise ultrastructural mechanism of the fungal infection, particularly penetration of the host insect cuticles, is not well understood. RESULTS: In this study, we investigated the morphology and ultrastructure of the larval cuticle of the red flour beetle, Tribolium castan

Effect of fermentation additives on the conidium surface-bond enzymatic activity and virulence of Beauveria bassiana.

A Colombian isolate (Bv062) of Beauveria bassiana, previously selected for its pathogenicity against Diatraea saccharalis, was used to evaluate the effect of fermentation additives on conidium surface-bond enzymatic activity and virulence. Genome analysis of this isolate revealed a diverse repertoire of virulence-related factors involved in host infection, including 13 lipases, 18 chitinases, 21 subtilisin-like proteases, and 19 trypsin-like proteases. Comparative genomics with other B. bassiana isolates also identified novel protein candidates not previously annotated in this species. This ge

The synergistic potential of polyethylene glycol 400 for the control of Hyphantria cunea larvae by Beauveria bassiana.

The efficacy of entomopathogenic fungi as pest control agents is constrained by both their physiological state and external environmental factors. This study identified synergists capable of enhancing the insecticidal activity of Beauveria bassiana (Bb) and investigated the underlying synergistic mechanisms. Our results found that among 6 potential synergists, polyethylene glycol 400 (PEG) and trehalose significantly improved Bb's lethality against Hyphantria cunea larvae, with PEG demonstrating the most pronounced effect. PEG treatment markedly increased Bb spore adhesion and germination rate

Fungal Keratitis Due to Beauveria bassiana in a Contact Lenses Wearer and Review of Published Reports.

Fungal keratitis is a severe ocular infection that primarily affects subjects engaged in outdoor activities. Risk factors include allergic conjunctivitis, previous eye surgery, previous treatment with wide-spectrum antimicrobial agents and corticosteroids and using contact lenses. Corneal infection is usually secondary to trauma involving organic material, which is often the only predisposing factor. Early diagnosis based on clinical examination and microbiological investigation (microscopy, cultures and molecular techniques) is crucial to selecting the appropriate antifungal therapy and preve

Bbssk1, a response regulator required for conidiation, multi-stress tolerance, and virulence of Beauveria bassiana.

Ssk1-type response regulator proteins are the core elements of histidine-to-aspartate systems that mediate fungal stress tolerance, a determinant to the biocontrol potential of fungal entomopathogens. We characterized the functions of Beauveria bassiana Ssk1 (Bbssk1) by analyzing multi-phenotypic changes in ΔBbssk1 and differentially expressed genes in the digital gene expression (DGE) libraries of ΔBbssk1 and wild-type constructed under osmotic stress. The Bbssk1 disruption caused 25 % reductions in conidial yield and virulence to Spodoptera litura larvae and significant defects in tolerances

The hidden habit of the entomopathogenic fungus Beauveria bassiana: first demonstration of vertical plant transmission.

Beauveria bassiana strain 04/01-Tip, obtained from a larva of the opium poppy stem gall wasp Iraella luteipes (Hymenoptera; Cynipidae), endophytically colonizes opium poppy (Papaver somniferum L.) plants and protects them against this pest. The goal of this study was to monitor the dynamics of endophytic colonization of opium poppy by B. bassiana after the fungus was applied to the seed and to ascertain whether the fungus is transmitted vertically via seeds. Using a species-specific nested PCR protocol and DNA extracted from surface-sterilised leaf pieces or seeds of B. bassiana-inoculated opi

P4-ATPase BbCrpa2 detoxifies cyclosporine a via AP2-clathrin-dependent endocytic pathway in Beauveria bassiana.

Cyclosporine A (CsA) exerts potent intracellular toxicity in fungi, yet the mechanisms underlying fungal CsA tolerance remain poorly understood. Here, we identify BbCrpa2, a P4-ATPase in the entomopathogenic fungus Beauveria bassiana, as a key determinant of CsA resistance. Unlike the trans-Golgi network-localized BbCrpa, which sequesters cytosolic CsA, BbCrpa2 functions as a plasma membrane gatekeeper. It captures CsA at the cell surface and mediates its internalization into vesicles through AP2-clathrin-dependent endocytosis. Genetic and functional analyses reveal that BbCrpa2 and BbCrpa act

Ecdysteroid UDP-Glucosyltransferase Expression in Beauveria bassiana Increases Its Pathogenicity against Early Instar Silkworm Larvae.

Beauveria bassiana (B. bassiana) is a broad-spectrum entomopathogenic fungus that can control pests in agriculture and forestry. In this study, encoding ecdysteroid uridine diphosphate glucosyltransferase gene (egt) was successfully screened in B. bassiana on the medium containing 500μg/mL G418 sulfate solution through the protoplast transformation method. This enzyme has the function of 20E (20-hydroxyecdysone) inactivation, thus increasing the mortality of the early instar larvae infected with B. bassiana. In this study, we transformed B. bassiana with the egt gene, which deactivates 20-hydr

Summer-form susceptible, winter-form resistant: differential susceptibility of Cacopsylla chinensis nymphs to Beauveria bassiana.

INTRODUCTION: The pear psylla Cacopsylla chinensis is a destructive pear pest whose nymphs exhibit seasonal polymorphism between summer-form and winter-form morphs. Whether the two morphs differ in susceptibility to Beauveria bassiana and the underlying mechanisms have not been characterized. METHODS: Bioassays at three spore concentrations (2.5×106, 2.5×107, and 2.5×108 spores/mL) were conducted to compare fungal virulence against fourth-instar nymphs of both morphs. Hemocyte counts and transcriptome sequencing were performed at 24 h and 36 h post-infection, respectively. qRT-PCR and enzymat

Host plant-specific volatiles of Beauveria bassiana-colonized plants initiate trophic plant-aphid-predator cascades.

BACKGROUND: Entomopathogenic ascomycetes (EAs) have efficacy in insect pest control through direct contact and indirectly as plant endophytes. As endophytes, they lead indirectly to pest mortality, enhance plant resilience to stresses, promote plant growth and alter plant volatile emissions. These changes can influence plant interactions with beneficial fauna, such as predators, parasitoids and pollinators. However, variation in endophytic colonization across plant species and EA strains raises questions about cultivar-specific responses within the same crop species. Here we evaluated the impa

Application of Beauveria bassiana conidia to spotted lanternfly forewings causes fewer infections than abdominal applications.

Adult spotted lanternflies (Lycorma delicatula) were differentially susceptible to Beauveria bassiana when inoculated with conidia on the distal ventral abdomen versus distal forewings. More adults inoculated on the abdomens died of B. bassiana infections than those inoculated on the wings. Abdominal inoculants also died more quickly than wing inoculants. Due to the large dorsal forewings of these planthoppers, typically covering abdomen and thorax, we suggest that the wings can at least partially protect from dorsal sprays of an infectious Hypocreales; we hypothesize that spraying surfaces on

Beauvericin production by endophytic and epiphytic Beauveria bassiana in peach (Prunus persica) and implications for insect biocontrol.

Beauveria bassiana (Balsamo) Vuillemin is a well-known entomopathogenic fungus that occupies diverse ecological niches, including soilborne, epiphytic, and endophytic habitats. Its capacity to function as an endophyte has received growing interest in potential applications for sustainable pest management, particularly in woody perennial systems where delivery and persistence of biological control agents are challenging. This study investigated endophytic colonization of peach (Prunus persica Batsch) seedlings by B. bassiana and quantified production of the insecticidal secondary metabolite bea

Biochemical and in silico structural properties of a thermo-acid stable β-glucosidase from Beauveria bassiana.

β-glucosidase hydrolyses the glycosidic bonds in cellobiose and cello-oligosaccharides, a critical step in the saccharification for biofuel production. Hence, the aim of this study was to gain insights into the biochemical and structural properties of a β-glucosidase from Beauveria bassiana, an entomopathogenic fungus. The β-glucosidase was purified to homogeneity using salt precipitation, ultrafiltration, and chromatographic techniques, attaining a specific activity of 496 U/mg. The molecular mass of the enzyme was then estimated via SDS-PAGE to be 116 kDa, while its activity pattern was conf

Time-series gene expression patterns and their characteristics of Beauveria bassiana in the process of infecting pest insects.

Beauveria bassiana has been widely used as an important biological control fungus for agricultural and forest pests, and clarifying the interaction mechanism between B. bassiana and its host will help to better exert the efficacy of the mycoinsecticide. Here, we proposed a novel pattern analysis (PA) method for analyzing time-series data and applied it to a transcriptomic data set of B. bassiana infecting Galleria mellonella. We screened out 14 patterns including 868 genes, which had some characteristics that were not inferior to differentially expressed genes (DEGs). Compared with the previou

Functional identification of the gene encoding enoyl-CoA hydratase for siderophore biosynthesis in the entomopathogen Beauveria bassiana.

The entomopathogenic fungus Beauveria bassiana possesses a siderophore biosynthetic pathway that closely resembles the general pathway found in filamentous fungi. Although a peroxisomal enoyl-CoA hydratase (Ech), SidH, has been implicated in the siderophore biosynthesis, its ortholog in B. bassiana appears to be functionally dispensable for siderophore production. In this study, gene function analysis was used to characterize the functional Ech involved in siderophore production among other fourteen ECH-domain proteins. The results indicated these proteins performed different roles in fungal d
