Conventional, organic, and regenerative organic farming differentially shape nutritional and phytochemical profile in beetroot (Beta vulgaris) and its commercial powder products.

Beetroots are increasingly recognized as a functional food; however, the extent to which different farming practices influence their primary nutrients, including minerals, sugars, and protein, and secondary metabolites, including phenolics, betalains, and nitrates, remains poorly understood. To address this gap, we analyzed fresh beetroot (Beta vulgaris) samples collected from a conventional, organic, and Regenerative Organic Certified (ROC™) farm (n = 12 per farming system) in Southwest Oregon, USA, along with beetroot powder from five commercially available brands samples (n = 6 samples per

Out of the blue: family-wide loss of anthocyanin biosynthesis in Cucurbitaceae.

Plant pigmentation secrets are among the oldest interests of plant scientists, with pigments such as chlorophyll, carotenoids, flavonoids and betalains contributing to the diversity of hues in higher plants. Anthocyanins, a class of flavonoids responsible for vibrant shades of pink, red and blue pigmentation, are almost ubiquitous in angiosperms but are replaced by betalains in some families in the order Caryophyllales. We investigated anthocyanin pigmentation in Cucurbitaceae, one of the largest fruit and vegetable families, characterised by white and yellow flowers and red, orange and green

A visible and quantitative pEAQ-RUBY system for analysis of transcription factor-promoter regulation in plants.

Understanding how transcription factors (TFs) regulate target promoters is central to molecular biology. However, widely used reporter-based assays typically require specialized instrumentation, costly substrates, and labor-intensive procedures. The recently developed RUBY reporter gene, which enables visible betalain accumulation, offers a simple and equipment-free alternative for monitoring gene expression. Here, we established a pEAQ-RUBY reporter system for efficient analysis of TF-mediated promoter regulation. We first optimized a quantitative method for betalain extraction in tobacco and

Pre-harvest application of sodium nitroprusside enhances storage root quality in red beet cultivated under normal and drought conditions.

BACKGROUND: The role of nitric oxide (NO) in plant stress tolerance, as well as in increasing post-harvest quality, has been extensively demonstrated in several fruits and vegetable crops; however, the effects of its pre-harvest application on post-harvest quality are still poorly documented. Therefore, the pre-harvest application of NO in red beet (Beta vulgaris subsp. vulgaris) plants cultivated under well-watered and drought conditions was evaluated to assess whether it improves the post-harvest quality of their storage roots. Red beet plants cultivated under well-watered (80% of water hold

Comprehensive fecal metabolomics and gut microbiota study of the protective mechanism of herbal pair Polygonum hydropiper-Coptis chinensis in rats with stress-induced gastric mucosal damage.

Introduction: Stress-related gastric mucosal lesions (SGMLs) are the most common complication in critical care patients. Previous studies have demonstrated that herbal pair (HP), Polygonum hydropiper-Coptis chinensis (HP P-C) has the anti-SGML effect. However, the underlying mechanism of HP P-C against SGML remains elusive. This study aimed to elucidate how HP P-C extracts exert their protective effects on SGML by examining the role of gut microbiota and metabolites. Methods: SD rats were pretreated with different doses of HP P-C extracts for 6 days, followed by inducing SGML with water-immers

A Non-Invasive Pathogen‑Responsive RUBY Reporter Enables Visible Monitoring of PR1 Activation.

We report a non‑invasive, pathogen‑responsive reporter system in Arabidopsis thaliana generated by placing the PR1 (AT2G14610) promoter upstream of the betalain‑producing RUBY cassette. Two independent transgenic lines showed robust, dose‑dependent red pigmentation accumulation in response to flg22 (a flagellin-derived peptide) and salicylic acid (a defense-signaling molecule that induces PR1 ) in both seedling and adult tissues. This reporter provides a simple, low‑cost visual readout of defense activation without the need for destructive sampling or specialized instrumentation. Seeds have be
