Bifidobacterium bifidum FB3-12 attenuates ovalbumin-induced allergic diseases by enhancing intestinal barrier function and regulating gut microbiota.

The escalating environmental changes have exacerbated the physiological suffering and economic burden caused by allergic diseases worldwide. Bifidobacterium bifidum (B. bifidum), due to its positive regulatory effects on the gut microenvironment, is considered a promising strategy for the prevention and treatment of allergies. Our study utilized the ovalbumin (OVA)-induced allergy mouse model to explore the anti-allergic potential of the self-screened strain B. bifidum FB3-12 (FB3-12) via intestinal intervention. The results demonstrated that a three-week FB3-12 treatment effectively suppresse…

Bifidobacterium bifidum Alleviate Intestinal Barrier Damage by Regulating the Intestinal Flora and Colonic Mucin O-Glycan Structural Patterns.

Bifidobacterium bifidum can alleviate intestinal barrier damage, but its reliance on mucin O-glycan modifications remains unclear. A dextran sodium sulfate-induced colitis mouse model was established to evaluate the therapeutic potential of B. bifidum Z29-2, C5, and their mixture. The results demonstrated that B. bifidum alleviated intestinal injury by preventing colon shortening and goblet cell depletion, as well as reducing levels of inflammatory cytokines. Furthermore, B. bifidum maintained the structural integrity of gut microbiota by enriching beneficial bacteria like Faecalibaculum and O…

Ratio-Dependent Regulation of Butyrate Production by 2'-Fucosyllactose and Fructooligosaccharides in a Bifidobacterium bifidum-Faecalibacterium prausnitzii Co-Culture.

An appropriate 2'-fucosyllactose (2'-FL) and fructooligosaccharides (FOS) mixing ratio may enhance butyrate production through substrate complementarity and metabolite-mediated interactions; however, the substrate balance required for this effect remains unclear. This study investigated how different 2'-FL/FOS ratios affect butyrate production in a two-species co-culture of Bifidobacterium bifidum and Faecalibacterium prausnitzii. 2'-FL and FOS were mixed at different weight ratios and added to monocultures and two-species co-cultures. The bacterial growth, relative abundance, and organic acid…

In-Silico Rational Design of Proline-Containing Peptides (PCPs) for Efficient Utilization by Bifidobacterium bifidum and Identification of Their Key Transported Sequences.

Industrial production of Bifidobacterium bifidum (B. bifidum) is constrained by its poor use of conventional nitrogen sources. Previous studies have demonstrated that supplementation with proline-containing peptides (PCPs) enhances both growth density and freeze-drying survival rate of B. bifidum. Thus, we developed an in silico-guided strategy to prepare proline-containing peptide-rich hydrolyzates tailored to its nitrogen preference. Casein, glutenin, and whey protein hydrolyzate were selected as substrates, and six enzymatic hydrolysis routes were designed using simulated digestion and mole…

Sprayable dynamic Schiff-base hydrogel engineers an oxygen-limited microenvironment for viable Bifidobacterium bifidum delivery and accelerated oral ulcer healing.

Oral ulcers affect over 25% of the global population, substantially impairing quality of life. Current therapeutic approaches are limited by challenges such as drug resistance and treatment dependency. Although probiotics have shown promise in mucosal repair, local delivery of anaerobic strains such as Bifidobacterium bifidum remains challenging because oxygen exposure rapidly reduces bacterial viability. Here, we report a sprayable, dynamic Schiff-base gelatin/cellulose hydrogel capable of in situ gelation and microenvironment engineering for the viable delivery of Bifidobacterium bifidum. Th…
