Impact of sodium caprate dosed as a mini-tablet or suspension on insulin delivery and mucosa histomorphology.

Oral administration of solid dosage forms for delivery of therapeutic peptides is highly desired. Preclinical investigations on co-administration with permeation enhancers (PEs) to enable sufficient oral bioavailabilities are, however, predominantly done using liquid formulations despite the commercial end-goal being a solid dosage form. Given the amounts needed of PE, this will typically result in a compacted tablet with high amounts of the PE of choice. The aim of this study was to compare the pharmacokinetics (PK) and pharmacodynamics (PD) of insulin after co-formulation with a fixed dose o

Localized heat application as a strategy for enhanced oral peptide absorption - A proof-of-concept study.

The oral delivery of peptides remains a major challenge, particularly due to poor permeability across the gastrointestinal mucosa. While chemical permeation enhancers (PEs) are commonly used to improve absorption, recent efforts have explored physical strategies, including mechanical disruption of the gut lining. In this study, we investigated the potential of localized mild heat (≤ 42 °C) as a permeation-enhancing approach, alone and in combination with the chemical PE sodium caprate (C10). Applied to colorectal adenocarcinoma cell monolayers, heat treatment of 2 h moderately increased permea

Fortified anti-proliferative activity of niclosamide for breast cancer treatment: In-vitro and in-vivo assessment.

Breast cancer represents one of the top lethal cancer types among the females worldwide. Several factors manipulate the clinical outcome of the treatment as the stage of the cancer upon detection, genetic and hormonal factors, drug resistance and metastasis. Accordingly, drug's repositioning, enhancing the bioavailability and encapsulation into nanoparticles (NPs) are among the predilected pathways for enhanced therapeutic outcome. Niclosamide (NIC) is an anthelmintic drug and has been repositioned as anticancer agent after revealing its anti-neoplastic activity. Piperine (PIP) was used as foo

Double emulsions enable in situ generation of permeation enhancers for oral delivery of macromolecules.

Oral administration of peptide therapeutics is limited by gastrointestinal degradation and poor epithelial permeability. Here, water-in-oil-in-water (W/O/W) double emulsions produced by microfluidics were designed to co-entrap octreotide and medium-chain triglycerides, enabling digestion-triggered generation of permeation-enhancing medium chain fatty acids. Synthesis parameters were systematically optimized to obtain stable, monodisperse droplets with defined core-shell morphology. The emulsions comprised an inner aqueous phase containing the payload, encapsulated within a Miglyol® 812 N oil p

Breaking the Solubility-Permeability Tradeoff: Surfactant-Mediated Enhancement of Oral Etoposide Absorption.

Developing effective oral formulations for poorly soluble anticancer drugs remains a major pharmaceutical challenge due to the combined limitations of solubility, permeability, and efflux transporter activity. In this work, we investigated the influence of the nonionic surfactants Cremophor EL, Pluronic P-85, and Pluronic F-68 on the solubility and intestinal permeability of the anticancer drug etoposide. All surfactants significantly increased etoposide aqueous solubility. While in vitro permeability across an artificial membrane demonstrated the expected solubility-permeability tradeoff, in

Improving Paclitaxel permeation and anticancer efficacy through Soluplus-based solid dispersion with Piperine.

OBJECTIVE: This study aimed to develop and optimize a Soluplus-based Paclitaxel-Piperine (PTX-PPN) solid dispersion to enhance PTX solubility, intestinal permeation, and anticancer efficacy in metastatic breast cancer cells. SIGNIFICANCE: PTX suffers from extremely poor aqueous solubility and dose-limiting toxicity associated with Cremophor® EL-based formulations. PPN is a well-documented bioenhancer and P-glycoprotein inhibitor that may modulate PTX permeation and cytotoxicity. METHODS: PTX-PPN solid dispersions were prepared via freeze-drying using different PTX-PPN-Soluplus ratios (1:1:1,

New Peceol™/Span™ 60 Niosomes Coated with Chitosan for Candesartan Cilexetil: Perspective Increase in Absolute Bioavailability in Rats.

PURPOSE: Candesartan cilexetil (CC), a prodrug of candesartan (CDT), is a class II BCS drug that suffers from poor oral bioavailability because of low aqueous solubility, P-gp efflux and first-pass metabolism. The absolute bioavailability reported for CC was only 15% and the methods to increase it remain elusive, thus the aim of our work was to prepare new CC-loaded niosomes encompassing, for the first time, glycerol monooleate GMO (Peceol™), as P-gp efflux inhibitor and promoter of lymphatic transport with Span™ 60 as bioenhancer. The prepared niosomes were further coated with chitosan for au

Oxidative stress induced by piperine leads to apoptosis in Candida albicans.

UNLABELLED: Candida albicans is a member of pathogens with potential drug resistance threat that needs novel chemotherapeutic strategies. Considering the multifarious biological activities including bioenhancer activity, anti-Candida potential of piperine was evaluated against planktonic/biofilm and hyphal growth of C. albicans alone or in combination as a synergistic agent with fluconazole. Piperine inhibits planktonic growth at or less than 15 μg/ml, hyphae induction at 5 μg/ml concentration, and exhibits stage-dependent activity against biofilm growth of a fluconazole-resistant strain of C.

Can Medical Interventions Serve as 'Criminal Rehabilitation'?

'Moral bioenhancement' refers to the use of pharmaceuticals and other direct brain interventions to enhance 'moral' traits such as 'empathy,' and alter any 'morally problematic' dispositions, such as 'aggression.' This is believed to result in improved moral responses. In a recent paper, Tom Douglas considers whether medical interventions of this sort could be "provided as part of the criminal justice system's response to the commission of crime, and for the purposes of facilitating rehabilitation (Douglas in Journal of Ethics 18(2): 101-122, 2014)." He suggests that they could "at least in so
