The rest-activity and body temperature 24 hours cycles, as well as the pattern of spontaneous sleep, were investigated in rats after infection with Trypanosoma brucei brucei. In the infected rats, which were entrained to a 12 hours/12 hours photoperiod, a considerable hypokinesia was detected during the hours of darkness. In most of the infected animals, the body temperature cycle displayed a lower amplitude and an advance of about 3 hours in respect to control rats; in addition, the body temperature rhythm was not significant in some infected rats. The relative proportion of slow wave synchro

Circadian rhythms widely exist in living organisms, and they are regulated by the biological clock. Growing evidence has shown that circadian rhythms are tightly related to the physiological function of the cardiovascular system, including blood pressure, heart rate, metabolism of cardiomyocytes, function of endothelial cells, and vasoconstriction and vasodilation. In addition, disruption of circadian rhythms has been considered as one of the important risk factors for cardiovascular diseases, such as myocardial infarction. This review summarizes the recent research advances in the relationshi

Extensive research defines the impact of advanced maternal age on couples' fecundity and reproductive outcomes, but significantly less research has been focused on understanding the impact of advanced paternal age. Yet it is increasingly common for couples at advanced ages to conceive children. Limited research suggests that the importance of paternal age is significantly less than that of maternal age, but advanced age of the father is implicated in a variety of conditions affecting the offspring. This review examines three aspects of advanced paternal age: the potential problems with concept

Temporal preferences of animals and humans often exhibit inconsistencies, whereby an earlier, smaller reward may be preferred when it occurs immediately but not when it is delayed. Such choices reflect hyperbolic discounting of future rewards, rather than the exponential discounting required for temporal consistency. Simultaneously, however, evidence has emerged that suggests that animals and humans have an internal representation of time that often differs from the calendar time used in detection of temporal inconsistencies. Here, we prove that temporal inconsistencies emerge if fixed duratio

BACKGROUND: Combination antiretroviral therapy has significantly improved survival among people living with HIV. However, many people living with HIV develop cardiovascular, metabolic, neurocognitive, musculoskeletal, and frailty-related conditions earlier or more frequently than HIV-negative individuals of a similar chronological age. DNA methylation-based epigenetic clocks provide a method for estimating biological age and may help explain this increased burden of age-related disease. This short review aims to summarize current evidence on epigenetic age acceleration in people living with HI

Age acceleration, or the difference between biological and chronological age, is a research area of growing interest, particularly in the field of age-related and neurodegenerative disorders, including Alzheimer's disease (AD). First-generation epigenetic clocks that predict chronological age utilising DNA methylation data were the first to derive estimates of age acceleration, which have been associated with several age-related conditions. More recently, second-generation epigenetic clocks that are predictors of specific traits such as age-related health, disease or morbidity and mortality ph

BACKGROUND: Chronic heavy alcohol use is a major risk factor for premature aging and age-related diseases. DNA methylation (DNAm)-based epigenetic clocks are novel tools for predicting biological age. However, the newest configurations, causality-enriched epigenetic clocks, have not been assessed in the context of alcohol consumption and alcohol use disorder (AUD).
METHODS: Epigenetic aging was evaluated in a sample of 615 individuals (372 AUD patients and 243 healthy controls) by applying the GrimAge Version 1 (V1) and Version 2 (V2) clocks alongside three causality-enriched clocks (CausAge,

This paper examines discussions of women's and men's reproductive aging in a series of western European medical texts written in the period 1100-1300. It uses the modern image of the biological clock to explore how far physicians in earlier periods understood reproductive aging to be a process of slow decline before a final age at which fertility ended (menopause for women, or a less defined 'old age' for men), and how far they viewed women's reproductive aging as different from men's. The article argues that, in contrast to modern medical and popular understandings, medieval physicians assume

Circadian disruption represents a global health issue associated with cardiometabolic diseases, sleep disturbances, and mood disorders, driven by a pathophysiological network including clock gene dysregulation and impaired melatonin synthesis. Vitamin D exerts pleiotropic effects on metabolic regulation, immune function, neurotransmission, and possibly circadian synchronization. Emerging evidence suggests that vitamin D and its hydroxyderivatives modulate clock gene expression, influence transcriptional regulators such as retinoic acid receptor-related orphan receptors and REV-ERBs, and intera

Mood disorders, including major depressive disorder, bipolar disorder, and seasonal affective disorder, exhibit significant heterogeneity, with disturbances in biological rhythms playing a central role. These disturbances not only contribute to the onset and progression of mood disorders but also serve as important predictors of relapse ("Chronos syndrome") and treatment response. Circadian disruptions, which are influenced by factors such as seasonality, jet lag, shift work, and childbirth, are hallmarks of mood episodes and are pivotal in transitions between mood states. Longitudinal studies

OBJECTIVE: Pituitary tumours that compress the optic chiasm are associated with long-term alterations in sleep-wake rhythm. This may result from damage to intrinsically photosensitive retinal ganglion cells (ipRGCs) projecting from the retina to the hypothalamic suprachiasmatic nucleus via the optic chiasm to ensure photoentrainment (i.e. synchronisation to the 24-h solar cycle through light). To test this hypothesis, we compared the post-illumination pupil response (PIPR), a direct indicator of ipRGC function, between hypopituitarism patients with and without a history of optic chiasm compres

The liver ranks among the peripheral organs exhibiting the most robust circadian rhythmicity, with glucose homeostasis, lipid metabolism, and bile acid turnover governed by tightly phased diurnal oscillations. Mitochondria execute these programs, their output coordinated with the hepatocyte circadian state. The mitochondrial network undergoes dynamic remodeling across the 24 h cycle, encompassing oscillatory changes in bioenergetics, fusion-fission balance, and quality control. This interplay is bidirectional: core clock components drive rhythmic remodeling via cyclin-dependent kinase 1/mitoge

Sleep disorders such as insomnia, hypersomnolence, nightmares and circadian rhythm disorders play a critical role across all stages of mood disorders, including Chronos syndrome, and are recognized as significant risk factors for their onset, relapse, and recurrence. Moreover, emerging evidence suggests that the co-occurrence of sleep or circadian disruptions with mood disturbances may represent a more severe and distinct clinical phenotype, likely associated with a poorer prognosis than when each condition occurs independently. Personalized interventions targeting sleep, circadian rhythms, an

INTRODUCTION: Alzheimer's disease (AD) involves progressive cognitive decline. Plasma proteomics data may provide insights into disease risk and mechanisms.
METHODS: Plasma proteomics data (N = 498) by the SomaScan 7k platform from the Indiana AD Research Center was used to calculate proteomics-based Alzheimer's risk scores: organ-specific aging clocks and pseudotime trajectory. The Alzheimer's risk scores were investigated for associations with plasma- and imaging-based biomarkers for AD and diagnosis and evaluated for classification performance for diagnosis and biomarker positivity status.

In the last decade, the in-vitro fertilization (IVF) sector has witnessed a shift from so-called 'reactive IVF' to a new model of proactive fertility care. Whereas IVF was traditionally developed to treat people who found they were unable to conceive, the indication for IVF has broadened significantly to include a much wider group of potential patients through a new focus on proactive treatment of future (in)fertilities. This shift combines a number of new trends pertaining to preservation, prediction, private equity and platformization, all of which have gained influence in contemporary assis

Non-alcoholic fatty liver disease (NAFLD) is the most frequent hepatic pathology in the Western world and may evolve into steatohepatitis (NASH), increasing the risk of cirrhosis, portal hypertension and hepatocellular carcinoma. NAFLD derives from the accumulation of hepatic fat due to discrepant free fatty acid metabolism. Other factors contributing to this are deranged nutrients and bile acids fluxes as well as alterations in nuclear receptors, hormones, and intermediary metabolites, which impact on signaling pathways involved in metabolism and inflammation. Autophagy and host gut-microbiot

Second generation antipsychotics (SGA) are associated with adverse cardiometabolic side effects contributing to premature mortality in patients. While mechanisms mediating these cardiometabolic side effects remain poorly understood, three independent studies recently demonstrated that melatonin was protective against cardiometabolic risk in SGA-treated patients. As one of the main target areas of circulating melatonin in the brain is the suprachiasmatic nucleus (SCN), we hypothesized that the SCN is involved in SGA-induced early cardiovascular effects in Wistar rats. We evaluated the acute eff

OBJECTIVE: To study the effects of biological clock protein on circadian disorders in hypoxic-ischemic brain damage (HIBD) by examining levels of CLOCK and BMAL1 proteins in the pineal gland of neonatal rats.
METHODS: Seventy-two 7-day-old Sprague-Dawley (SD) rats were randomly divided into sham-operated and HIBD groups. HIBD model was prepared according to the modified Levine method. Western blot analysis was used to measure the levels of CLOCK and BMAL1 in the pineal gland at 0, 2, 12, 24, 36 and 48 hours after operation.
RESULTS: Both CLOCK and BMAL levels in the pineal gland increased si
