Masquerading as pea plants: behavioural and morphological evidence for mimicry of multiple models in an Australian orchid.

BACKGROUND AND AIMS: While there is increasing recognition of Batesian floral mimicry in plants, there are few confirmed cases where mimicry involves more than one model species. Here, we test for pollination by mimicry in Diuris (Orchidaceae), a genus hypothesized to attract pollinators via mimicry of a range of co-occurring pea plants (Faboideae). METHODS: Observations of pollinator behaviour were made for Diuris brumalis using arrays of orchid flowers. An analysis of floral traits in the co-flowering community and spectral reflectance measurements were undertaken to test if Di. brumalis an

New insights to the immunopathology and autoimmune responses in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is an autoimmune liver disease with profound changes in different compartments of the immune system, including those involved in innate, and adaptive immunity. New data from epidemiological studies of PBC have reinforced the thesis that the cause for this relatively uncommon disease is likely to be a combination of both environmental factors and a susceptible genetic predisposition. Recent findings of abnormalities of the innate immune system in PBC suggest that they may serve as links between the environmental factors and the early events in PBC development. Vi

Electrochemical simulation of psychotropic drug metabolism compared to in vivo processes using liquid chromatography and mass spectrometry.

INTRODUCTION: Psychotropic drugs strongly affect the human psyche through their ability to modulate the neurotransmitter activity and to treat mental disorders and diseases. Monitoring of psychotropic drugs in clinical studies is significant. Thus. establishing methodologies for analyzing these drugs and their pharmacologically active metabolites in biological matrices is essential for patients' safety. Therefore, therapeutic drug monitoring (TDM) of these drugs in patients receiving pharmacotherapy in psychiatric hospitals is necessary to avoid medical complications, psychiatric adverse effec

Metabolite mimicry identifies butyrate analogs with select protective functions in the intestinal mucosa.

Microbial-derived short-chain fatty acids regulate a variety of pathways in the healthy colonic mucosa. In particular, butyrate serves as the primary energy source for colonocytes and regulates gene transcription by stabilizing the transcription factor hypoxia-inducible-factors (HIF) and functioning as a histone deacetylase (HDAC) inhibitor. A limitation of butyrate as a therapeutic is its rapid metabolism in differentiated colonocytes. Furthermore, intestinal stem cells (ISCs) respond differently to butyrate, preferentially using glucose for energy procurement. To address these limitations, w

Molecular mimicry: cross-reactive antibodies from patients with immune-mediated neurologic disease inhibit neuronal firing.

Recent data indicate that molecular mimicry may play a role in the pathogenesis of human-T-lymphotropic virus type-1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP), an immune-mediated disease of the central nervous system (CNS). Specifically, HAM/TSP patients developed antibodies that cross-react with heterogeneous nuclear ribonuclear protein A1 (hnRNP A1), an antigen highly expressed in neurons. Antibodies to HTLV-1-tax cross-reacted with hnRNP A1, suggesting molecular mimicry between the two proteins. In support of this hypothesis, HAM/TSP IgG and antibodies to hnRNP A

Gangliosides as targets for autoimmune injury to the nervous system.

Immune responses directed towards gangliosides and their microbial mimics are important mediators of several subtypes of acute post-infectious autoimmune neuropathy, collectively referred to as the Guillain-Barré syndromes. In this diverse group of paralytic syndromes, the immunopathology is in a proportion of cases characterised by anti-ganglioside antibody deposits, accompanied by inflammatory destruction of both axonal and glial components within the PNS. By gaining an understanding of the immunological mechanisms underlying these pathological pathways, it should be possible to select the c

Molecular Mimicry: a Paradigm of Host-Microbe Coevolution Illustrated by Legionella.

Through coevolution with host cells, microorganisms have acquired mechanisms to avoid the detection by the host surveillance system and to use the cell's supplies to establish themselves. Indeed, certain pathogens have evolved proteins that imitate specific eukaryotic cell proteins, allowing them to manipulate host pathways, a phenomenon termed molecular mimicry. Bacterial "eukaryotic-like proteins" are a remarkable example of molecular mimicry. They are defined as proteins that strongly resemble eukaryotic proteins or that carry domains that are predominantly present in eukaryotes and that ar

UAS stealth: target pursuit at constant distance using a bio-inspired motion camouflage guidance law.

The aim of this study is to derive a guidance law by which an unmanned aerial system(s) (UAS) can pursue a moving target at a constant distance, while concealing its own motion. We derive a closed-form solution for the trajectory of the UAS by imposing two key constraints: (1) the shadower moves in such a way as to be perceived as a stationary object by the shadowee, and (2) the distance between the shadower and shadowee is kept constant. Additionally, the theory presented in this paper considers constraints on the maximum achievable speed and acceleration of the shadower. Our theory is tested

Desmoglein-2 as a cancer modulator: friend or foe?

Desmoglein-2 (DSG2) is a calcium-binding single pass transmembrane glycoprotein and a member of the large cadherin family. Until recently, DSG2 was thought to only function as a cell adhesion protein embedded within desmosome junctions designed to enable cells to better tolerate mechanical stress. However, additional roles for DSG2 outside of desmosomes are continuing to emerge, particularly in cancer. Herein, we review the current literature on DSG2 in cancer and detail its impact on biological functions such as cell adhesion, proliferation, migration, invasion, intracellular signaling, extra

A continuous fiber distribution material model for human cervical tissue.

The uterine cervix during pregnancy is the vital mechanical barrier which resists compressive and tensile loads generated from a growing fetus. Premature cervical remodeling and softening is hypothesized to result in the shortening of the cervix, which is known to increase a woman׳s risk of preterm birth. To understand the role of cervical material properties in preventing preterm birth, we derive a cervical material model based on previous mechanical, biochemical and histological experiments conducted on nonpregnant and pregnant human hysterectomy cervical tissue samples. In this study we pre

Immunogenicity of Adalimumab in Bacterial Molecular Mimicry: In Silico Analysis.

BACKGROUND: Adalimumab, a monoclonal antibody targeting tumor necrosis factor α, treats autoimmune diseases but induces antidrug antibodies in 30% to 60% of patients, reducing its efficacy. OBJECTIVE: This study aims to investigate molecular mimicry as a mechanism behind this immunogenicity, where bacterial immunoglobulin domains structurally resemble adalimumab's light chain, triggering immune responses. METHODS: Using PSI-BLASTp (National Center for Biotechnology Information) and PRALINE (Center for Integrative Bioinformatics), there are 40 bacterial antigens homologous to adalimumab, with

HTLV-1 induced molecular mimicry in neurological disease.

As a model for molecular mimicry, we study patients infected with human T-lymphotropic virus type 1 (HTLV-1) who develop a neurological disease called HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP), a disease with important biological similarities to multiple sclerosis (MS) (Khan et al. 2001; Levin et al. 1998, 2002a; Levin and Jacobson 1997). The study of HAM/TSP, a disease associated with a known environmental agent (HTLV-1), allows for the direct comparison of the infecting agent with host antigens. Neurological disease in HAM/TSP patients is associated with immune resp

Investigating the Biomechanics to Mimic Blinking using an Electromagnetic Actuator.

The Bionic Lid Implant for Natural Closure (BLINC) is an active implantable medical device aimed at restoring blinking for people with facial nerve paralysis (FNP), with the aim to restore complete, symmetrical, function. BLINC utilizes electromagnetic force to actuate a sling implanted in the upper eyelid. The horizontal pull translates to the downward movement of the eyelid to mimic a blink. Current work aims to move this project from laboratory to clinic, by gaining a better understanding of the biomechanics required to achieve these goals in humans. This follows a chronic large animal stud

Cytogenetic and Molecular Characterization of Three Mimetic Species of the Genus Alagoasa Bechyné 1955 (Coleoptera: Alticinae) from the Neotropical Region.

Coleoptera is a mega-diverse order, but only about 1% of its species have been analyzed cytogenetically. In this order, the subfamily Alticinae presents many identification problems, mainly due to the occurrence of mimicry. The objective of this work was to cytogenetically characterize 3 very similar species of the genus Alagoasa (A. pantina, A.areata, and A.scissa). We used classical and molecular cytogenetic as well as molecular genetic techniques. All 3 species showed a diploid chromosome number of 2n = 22 (20+X+y), but differences in the morphology of the chromosomes. All had a meiotic for

Caloric restriction mimetics for the treatment of cardiovascular diseases.

Caloric restriction mimetics (CRMs) are emerging as potential therapeutic agents for the treatment of cardiovascular diseases. CRMs include natural and synthetic compounds able to inhibit protein acetyltransferases, to interfere with acetyl coenzyme A biosynthesis, or to activate (de)acetyltransferase proteins. These modifications mimic the effects of caloric restriction, which is associated with the activation of autophagy. Previous evidence demonstrated the ability of CRMs to ameliorate cardiac function and reduce cardiac hypertrophy and maladaptive remodelling in animal models of ageing, me

The deubiquitinase USP7 and E3 ligase TRIM21 regulate vasculogenic mimicry and malignant progression of RMS by balancing SNAI2 homeostasis.

BACKGROUND: Rhabdomyosarcoma (RMS) is a rare malignancy and the most common soft tissue sarcoma in children. Vasculogenic mimicry (VM) is a novel tumor microcirculation model different from traditional tumor angiogenesis, which does not rely on endothelial cells to provide sufficient blood supply for tumor growth. In recent years, VM has been confirmed to be closely associated with tumor progression. However, the ability of RMS to form VM has not yet been reported. METHODS: Immunohistochemistry, RT-qPCR and western blot were used to test the expression level of SNAI2 and its clinical signific

Molecular mimicry: an important virulence strategy employed by Legionella pneumophila to subvert host functions.

It is 32 years since Legionella pneumophila was identified and recognized as a human pathogen, causing the severe form of pneumonia termed Legionnaires' disease, or legionellosis. This bacterium is found in freshwater reservoirs where it replicates in aquatic protozoa and can invade man-made water distribution systems. Although the disease can be treated by antibiotherapy and prevented through surveillance and control measures, reported cases of Legionnaires' disease continue to rise across Europe and outbreaks of major public health significance still occur. Genome sequencing and analyses led

MicroRNA-7 Exerts Antiangiogenic Effect on Colorectal Cancer via ERK Signaling.

BACKGROUND: Recent studies have suggested that microRNA-7 (miR-7) family members may play important roles in human cancer by regulating cell proliferation, apoptosis, migration, and invasion. Therefore, the present study aimed to investigate the clinical significance and biological function of miR-7 in colorectal cancer (CRC). METHODS: Initially, cancer and adjacent tissues were collected from 76 patients with CRC. Then, microvascular density was detected using the Weidner counting method. The functional role of miR-7 in CRC was determined using ectopic expression, knockdown, and reporter ass
