Accelerating the discovery of industrial PET-degrading enzymes: Evolving paradigms from bioprospecting to computational discovery.

Polyethylene terephthalate (PET) waste remains a major environmental and resource challenge, and enzymatic depolymerization offers a promising route for recovering terephthalic acid and ethylene glycol under aqueous conditions for closed-loop PET recycling. However, the practical deployment of PET-degrading enzymes is constrained by substrate crystallinity and morphology, limited thermal robustness near the glass transition temperature, biocatalyst manufacturability and reusability, product inhibition and pH-mediated activity loss, and the heterogeneity and contamination of post-consumer feeds

Characterization of mycobacteria isolated from the Brazilian Atlantic Forest: a public health and bioprospection perspective.

The Mycobacterium genus remains highly relevant today due to the rising incidence of tuberculosis and opportunistic infections caused by environmental mycobacteria. While much is known about M. tuberculosis, M. leprae and M. bovis, studies focusing on environmental mycobacteria remain limited. These microorganisms are globally distributed and have been identified in diverse biomes, including the Atlantic Forest. This study aims to provide a characterization of four mycobacterial strains isolated from the Atlantic Forest, assessing their metabolic capabilities and biotechnological potential. We

Bioprospection of anti-inflammatory phytochemicals suggests rutaecarpine and quinine as promising 15-lipoxygenase inhibitors.

15-Lipoxygenase (15-LOX) belongs to the family of nonheme iron containing enzymes that catalyzes the peroxidation of polyunsaturated fatty acids (PUFAs) to generate eicosanoids that play an important role in signaling pathways. The role of 15-LOX has been demonstrated in atherosclerosis as well as other inflammatory diseases. In the present study, drug-like compounds were first screened from a set of anti-inflammatory phytochemicals based on Lipinski's rule of five (ROF) and in silico toxicity filters. Two lead compounds-quinine (QUIN) and rutaecarpine (RUT) were shortlisted by analyzing molec

Declaration of Fermentation: community-embedded wild yeast bioprospecting as a model for place-based CURE design.

Course-based undergraduate research experiences (CUREs) are widely recognized as a high-impact practice in biology education, yet most existing CURE frameworks treat the research organism as an interchangeable teaching prop rather than a genuine scientific contribution. We argue that place-based, community-embedded CUREs-in which students isolate, characterize, and publicly deploy a locally meaningful wild organism-constitute a qualitatively distinct model warranting broader adoption. As proof of concept, we present the Declaration of Fermentation project at Indiana University Bloomington: gra

Bioprospection of Selected Plant Secondary Metabolites as Modulators of the Proteolytic Activity of Plasmodium falciparum Plasmepsin V.

Malaria is a devastating disease, and its management is only achieved through chemotherapy. However, resistance to available medication is still a challenge; therefore, there is an urgent need for the discovery and development of therapeutics with a novel mechanism of action to counter the resistance scourge consistent with the currently available antimalarials. Recently, plasmepsin V was validated as a therapeutic target for the treatment of malaria. The pepsin-like aspartic protease anchored in the endoplasmic reticulum is responsible for the trafficking of parasite-derived proteins to the e

Bioprospecting in potato fields in the Central Andean Highlands: screening of rhizobacteria for plant growth-promoting properties.

The Central Andean Highlands are the center of origin of the potato plant (Solanum tuberosum). Ages of mutualism between potato plants and soil bacteria in this region support the hypothesis that Andean soils harbor interesting plant growth-promoting (PGP) bacteria. Therefore, the aim of this study was to isolate rhizobacteria from Andean ecosystems, and to identify those with PGP properties. A total of 585 bacterial isolates were obtained from eight potato fields in the Andes and they were screened for suppression of Phytophthora infestans and Rhizoctonia solani. Antagonistic mechanisms were

Chemical Ontogeny in Plants: Developmental Dynamics of Specialized Metabolism and Implications for Ecology, Quality Control, and Bioprospecting.

Plant specialized metabolism changes throughout development, but developmental context remains inconsistently incorporated into phytochemical interpretation. This integrative narrative review synthesizes evidence on chemical ontogeny in plants, defined here as the qualitative and quantitative reorganization of specialized metabolism along plant and organ development. We distinguish ontogenetic variation from phenology, seasonality, organ maturation, developmental plasticity, and environmentally induced chemical plasticity, and examine the genetic, hormonal, anatomical, ecological, and analytic

In Silico Bioprospection of Daniellia oliveri-Based Products as Quorum Sensing Modulators of Escherichia coli SdiA.

Escherichia coli is a common pathogen responsible for various gut-related infections, and it utilizes the SdiA-mediated quorum sensing (QS) system to regulate biofilm formation, other virulence factors, and pathogenicity. With rising antibiotic resistance, there is a pressing need to discover alternative QS inhibitors (QSIs) targeting SdiA. This study evaluated 239 phytochemicals from Daniellia oliveri as potential SdiA modulators using in silico techniques. Virtual screening identified four lead compounds (cadala-1(10),3,8-triene, carotenoid K, valencene, and β-sesquiphellandrene), with carot
