Differential biotin labelling of the cell envelope proteins in lipopolysaccharidic diderm bacteria: Exploring the proteosurfaceome of Escherichia coli using sulfo-NHS-SS-biotin and sulfo-NHS-PEG4-bismannose-SS-biotin.

UNLABELLED: Surface proteins are the major factor for the interaction between bacteria and its environment, playing an important role in infection, colonisation, virulence and adaptation. However, the study of surface proteins has proven difficult mainly due to their hydrophobicity and/or relatively low abundance compared with cytoplasmic proteins. To overcome these issues new proteomic strategies have been developed, such as cell-surface protein labelling using biotinylation reagents. Sulfo-NHS-SS-biotin is the most commonly used reagent to investigate the proteins expressed at the cell surfa

Epilepsy in biotinidase deficiency after biotin treatment.

Patients with severe biotinidase deficiency (BD), if untreated, may exhibit seizures, psychomotor delay, deafness, ataxia, visual pathology, conjunctivitis, alopecia, and dermatitis. Clinical features normally appear within the first months of life, between two and five. Seizures are one of the most common symptoms in these patients (55%), usually presented as generalized tonic-clonic, and improving within 24 h of biotin treatment. Treatment delay has been associated with irreversible neurological damage, mental retardation, ataxia, paraparesis, deafness, and epilepsy exceptionally. We report t

111In-DTPA-Biotin uptake by Staphylococcus aureus.

The potential of indium-111 labelled diethylenetriaminepentaacetic acid α,ω-bis(biocytinamide) (In-DTPA-Biotin) as a specific tracer in nuclear medicine imaging of vertebral osteomyelitis has been shown in a large series of consecutive patients. Biocytin is known to serve as a biotin source for a number of different microorganisms and quantitative studies on staphylococci indicated that on a molar basis biocytin seemed to have an activity equal to that of biotin. In this study, we evaluated the possibility of an illicit transport of In-DTPA-Biotin in cultures of Staphylococcus aureus on contin

Biotin deficiency facilitates kindling hyperexcitability in rats.

Biotin-deficient conditions are frequently associated with epileptic disorders. Biotin deficiency may be caused by long-term treatment with anticonvulsants or excessive ingestion of avidin. Absence of biotinidase activity can also lead to biotin deficiency, and is characterized by developmental delay as well as neurological and dermatological abnormalities. Because seizures are one of the most frequent signs of the latter, biotin-deficient conditions could conceivably facilitate convulsive disorders. To test this hypothesis, we investigated the occurrence of a latent kindling hyperexcitability

[Pharmacologic effects of biotin on epidermal cells].

Biotin deficiency in animals causes pathological changes of the skin and its appendages including, for example, exfoliative dermatitis, depigmentation, and alopecia. The hooves of biotin-deficient swine are weak, brittle, and often necrotic. These changes disappear after dietary biotin supplementation. Biotin supplementation also noticeably improves the hoof quality of horses, cattle and swine having no apparent biotin deficiency. In order to elucidate the molecular basis of these effects, the influence of biotin on cytokeratin expression in a keratinocyte cell line (Ha-CaT) was investigated u

Intestinal distribution and absorption of biotin in the chicken.

The initial distribution and absorption of biotin in chickens was studied in vivo from either changes in the composition of intestinal contents or analysis of blood draining the gut. The progressive decline in biotin concentration and flow along the small intestine demonstrated absorption throughout this organ. Quantitatively the duodenum was the most significant site of biotin absorption when crystalline biotin was included in the diet and when dietary ingredients contained highly available biotin. In contrast, when diets of low biotin availability were given the distal segments of the small

High-dose biotin therapy leading to false biochemical endocrine profiles: validation of a simple method to overcome biotin interference.

BACKGROUND: High-dose biotin therapy is beneficial in progressive multiple sclerosis (MS) and is expected to be adopted by a large number of patients. Biotin therapy leads to analytical interference in many immunoassays that utilize streptavidin-biotin capture techniques, yielding skewed results that can mimic various endocrine disorders. We aimed at exploring this interference, to be able to remove biotin and avoid misleading results. METHODS: We measured free triiodothyronine (fT3), free thyroxine (fT4), thyroid-stimulating hormone (TSH), parathyroid homrone (PTH), 25-hydroxyvitamin D (25OH

NeutrAvidin Functionalization of CdSe/CdS Quantum Nanorods and Quantification of Biotin Binding Sites using Biotin-4-Fluorescein Fluorescence Quenching.

We developed methods to solubilize, coat, and functionalize with NeutrAvidin elongated semiconductor nanocrystals (quantum nanorods, QRs) for use in single molecule polarized fluorescence microscopy. Three different ligands were compared with regard to efficacy for attaching NeutrAvidin using the "zero-length cross-linker" 1-ethyl-3-[3-(dimethylamino)propyl]carbodiimide (EDC). Biotin-4-fluorescene (B4F), a fluorophore that is quenched when bound to avidin proteins, was used to quantify biotin binding activity of the NeutrAvidin coated QRs and biotin binding activity of commercially available s

Bypass of the canonical biotin synthesis pathway in yeast.

Biotin serves as an essential cofactor for several carboxylation enzymes that catalyze reactions vital for cell survival and proliferation. While bacterial biotin synthesis pathways are well characterized, the mechanisms in eukaryotic organisms remain poorly understood. In Saccharomyces cerevisiae, Bio1 catalyzes the initial and rate-limiting step in the yeast biosynthetic pathway. Previous workers demonstrated that the Bio1 ortholog from the related yeast, Cyberlindnera fabianii, functionally substituted for S. cerevisiae Bio1 and greatly increased growth in the absence of biotin. However, we

Biotin Deficiency in Telogen Effluvium: Fact or Fiction?

BACKGROUND: Biotin is popular in hair loss treatment supplements, although the frequency of its deficiency in patients with hair loss has not been established. OBJECTIVES: This work sought to assess the serum levels of biotin in patients with telogen effluvium. METHODS: This case-control study included 60 patients with telogen effluvium and 20 control subjects. Subjects who were on biotin therapy three months prior to the study and those reporting other causes of hair loss were excluded. The scalp of each patient was clinically and dermoscopically examined. Serum biotin levels were measured us

Biotin for the treatment of nail disease: what is the evidence?

AIM: To describe the pharmacology, mechanism of action, and clinical reports using biotin to treat nail conditions. METHODS: A review of articles indexed for MEDLINE on PubMed using keywords 'biotin' and 'nail' was performed and applicable articles were selected for review. RESULTS: Clinical trials have shown an improvement in firmness, hardness, and thickness of brittle nails with oral biotin. There are some case reports and series demonstrating that oral biotin may improve triangular worn down nails, trachyonychia, and habit tic nail deformity. CONCLUSIONS: Oral biotin has been used to tr

Scintigraphic imaging of vertebral osteomyelitis with 111in-biotin.

STUDY DESIGN: Early diagnosis of vertebral infection (hematogenous or postsurgical) is necessary to choose a correct therapy and to minimize dramatic complications. All patients suspected to have vertebral infection underwent radiologic imaging and In-Biotin scintigraphy. OBJECTIVE: Biotin is a growth factor used by many bacteria. The aim of our study is to use In-Biotin to diagnose vertebral infections. SUMMARY OF BACKGROUND DATA: Magnetic resonance imaging, even if endowed with fairly good sensitivity and specificity, shows some limitations in the study of the onset of pathology and in pos

Biotin in metabolism and its relationship to human disease.

Biotin, a water-soluble vitamin, is used as cofactor of enzymes involved in carboxylation reactions. In humans, there are five biotin-dependent carboxylases: propionyl-CoA carboxylase; methylcrotonyl-CoA carboxylase; pyruvate carboxylase, and two forms of acetyl-CoA carboxylase. These enzymes catalyze key reactions in gluconeogenesis, fatty acid metabolism, and amino acid catabolism; thus, biotin plays an essential role in maintaining metabolic homeostasis. In recent years, biotin has been associated with several diseases in humans. Some are related to enzyme deficiencies involved in biotin me

Avidin-biotin system for delivery of diagnostic agents.

Radiolabeled monoclonal antibodies specific for tumor-associated antigens are used for the diagnosis and therapy of malignant tumors. However, the blood clearance of antibodies is slow and tumor-to-nontumor ratios of radioactivity in the current system are not high. To increase tumor-to-nontumor ratios, the concept of pretargeting has been proposed. The strategy is based on the separate administration of the monoclonal antibody and the radiolabel. Because avidin and streptavidin bind to biotin selectively with extremely high affinity, the avidin-biotin system has been applied for pretargeting.

Biotin deficiency per se is teratogenic in mice.

We examined whether maternal biotin deficiency would potentiate the latent teratogenicity of relatively low doses of vitamin A in mice. The incidence and the type of gross congenital malformations (cleft palate, micrognathia, and micromelia) induced by biotin deficiency were similar among the groups given three different concentrations of vitamin A (4000, 12,000 and 60,000 IU) in the diet. Also, the type of these malformations was different from those (exencephaly, cleft palate and macroglossia) induced by a known teratogenic dose of vitamin A (1,200,000 IU). We conclude that in mice concentra

Rett syndrome revisited: a patient with biotin dependency.

A patient with Rett syndrome (cerebral atrophy associated with hyperammonemia) was studied. Primary defects of urea cycle enzymes were excluded as causes of the disorder. The analysis of urinary organic acids showed a moderate increase of lactate, methylcitrate, tiglyglycine and 3-hydroxisovalerate, indicating an abnormality of multiple carboxylases. Biotin supplementation reversed the urinary abnormalities. In fibroblasts grown with a low biotin medium propionylCoA and 3-methylcrotonylCoA carboxylase activities were reduced. Holocarboxylase synthetase activity was normal (Vmax and Km). Surpri
