This study investigated the genotype classification and pathogenicity of infectious bursal disease virus (IBDV) circulating in vaccinated broiler chicken farms in Egypt. A total of 150 samples were collected from 30 vaccinated commercial broiler chicken farms and pooled into 30 working samples. IBDV was tested using reverse transcriptase polymerase chain reaction (RT-PCR) amplification of the hypervariable region of the viral protein 2 (hvVP2) and the VP1 gene 5' extremity. Both RT-PCR fragments were sequenced from six samples, and then the obtained nucleotide sequences were analyzed. The IBDV

There are two types of secretory cells in the chicken bursa of Fabricius (BF): (a) interfollicular epithelial cells (IFE), and (b) bursal secretory dendritic cells (BSDC) in the medulla of bursal follicles. Both cells produce secretory granules, and the cells are highly susceptible to IBDV vaccination and infection. Before and during embryonic follicular bud formation, an electron-dense, scarlet-acid fuchsin positive substance emerges in the bursal lumen, the role of which is unknown. In IFE cells, IBDV infection may induce rapid granular discharge, and in several cells, peculiar granule forma

Infectious bursal disease (IBD) is an acute, highly contagious, immunosuppressive disease of young chickens that causes considerable economic loss in the poultry industry worldwide. Vaccination with live attenuated vaccines is still the most important method used for the control and prevention of IBD in chickens. Here we present the results of in vitro characterization, as well as efficacy and safety testing of a live, intermediate plus vaccine against IBD based on strain G6. Strain characterization confirmed that G6 strain is an intermediate plus strain, showing a high degree of homology with

This study was conducted to investigate the molecular characterization and pathogenicity of very virulent infectious bursal disease virus (vvIBDV) isolated from naturally infected turkey poults and possible spread to chickens. Thirty samples were collected from turkey poults in the vicinity or in the same backyards with chickens suspected to be infected with IBDV and from live bird markets from different localities in Dakahlia governorate, Egypt. There were no obvious clinical signs in tested turkey poults except dehydration and whitish diarrhoea in some birds with no mortality, and post-morte

BACKGROUND: Infectious Pancreatic Necrosis (IPN) is a highly contagious birnavirus disease of farmed salmonid fish, which often causes high levels of morbidity and mortality. A large host genetic component to resistance has been previously described for Atlantic salmon (Salmo salar L.), which mediates high mortality rates in some families and zero mortality in others. However, the molecular and immunological basis for this resistance is not yet fully known. This manuscript describes a global comparison of the gene expression profiles of resistant and susceptible Atlantic salmon fry following c

Antimicrobial immune defense is evolutionarily preserved in all organisms. Mammals have developed robust, protein-based antiviral defenses, which are under constant investigation. Studies have provided evidences for the various fish immune factors sharing similarity with those of mammals. In this study, we have identified an ortholog of mitochondrial antiviral signaling protein from rock bream, Oplegnathus fasciatus. RbMAVS cDNA possesses an open reading frame (ORF) of 1758 bp coding for a protein of 586 amino acids with molecular mass of approximately 62 kDa and isoelectric point of 4.6. In s

Infectious Bursal Disease (IBD), or Gumboro disease, is an acute, highly contagious viral infection that primarily affects young chickens, causing immunosuppression and mortality. This study aimed to characterize the genetic and pathological features of recent IBD outbreaks in Kashmir, India. A total of 32 suspected outbreaks were investigated from 2021 to 2023 across three districts: Srinagar, Ganderbal, and Budgam. Bursal tissues were collected for both histopathology and molecular detection. RT-PCR targeting the hypervariable region (HVR) of the VP2 gene confirmed IBDV infection in 29 sampl

Since 1987, infectious bursal disease virus (IBDV) has circulated and evolved in Vietnam, but little is known about the genotypes present. IBDV samples were collected in 1987, 2001-2006, 2008, 2011, 2015-2019, and 2021 in 18 provinces. We conducted phylogenotyping analysis based on an alignment of 143 VP2-HVR (hypervariable region) sequences from 64 Vietnamese isolates (26 previous and 38 additional sequences and two vaccines, and alignment of 82 VP1 B-marker sequences, including one vaccine and four Vietnamese field strains. The analysis identified three A-genotypes, A1, A3, and A7, and two B

IPNV is a salmonid birnavirus that possesses the ability to establish asymptomatic persistent infections in a number of valuable fish species. The presence of IPNV may interfere with subsequent infection by other viruses. In the present study we show that an IPNV-carrier cell line (EPCIPNV) can induce an antiviral state in fresh EPC by co-cultivating both cell types in three different ways: a "droplet" culture system, a plastic chamber setup, and a transmembrane (Transwell®) system. All three cell co-culture methods were proven useful to study donor/target cell interaction. Naïve EPC cells gro

Infectious bursal disease virus (IBDV) is one of the most impactful pathogens of poultry, with disease manifestations ranging from acute forms to subclinical but immunosuppressive infections. This heterogeneity, accompanied by a significant antigenic variability, is sustained by high mutation rates and frequent reassortments between the two genome segments, along with less frequent recombination events. In recent years, the proposal of several classification systems relying on phylogeny contributed to the characterization of several new IBDV genotypes, shedding light on an increasingly diverse

Recently, several attempts have been made to replace egg-based with cell-based vaccines to prevent and control Infectious Bursal Disease Virus (IBDV). This study aimed to evaluate a new fish cell line (M99) for culturing and replicating IBDV. After observing complete cytopathic effects (CPE) on the M99 cell line, virus titers were determined using the TCID50 test, and the presence of the virus was confirmed using an RT-PCR test. Subsequently, 135 broiler chickens (14 days old) were randomly divided into three equal groups for immune response measurements: G1: immunized with a commercial vaccin

Infectious bursal disease virus (IBDV) vaccines do not induce sterilizing immunity, and vaccinated birds can become infected with field strains. Vaccine-induced immune selection pressure drives the evolution of antigenic drift variants that accumulate amino acid changes in the hypervariable region (HVR) of the VP2 capsid, which may lead to vaccine failures. However, there is a lack of information regarding how quickly mutations arise, and the relative contribution different residues make to immune escape. To model IBDV antigenic drift in vitro, we serially passaged a classical field strain bel

Infectious bursal disease (IBD) is a highly contagious disease in young chickens worldwide. The major strategy for the prevention and control of IBD virus (IBDV) is vaccination. Therefore, the present study aimed to compare the immunogenicity of four commercially available IBD vaccines on broilers (Ross 308) that were raised in areas with very virulent IBDV infection history. Two commercial broiler farms with four standard poultry houses were selected in Alborz (n=6,250 birds per house) and Khorasan Razavi (n=8,000 birds per house) provinces of Iran. In each farm, the houses were randomly assi

Infectious bursal disease (IBD), also known as Gumboro disease, is a globally well-known disease with a significant socio-economic effect. For control of IBD, several commercial egg- and cell-based vaccines are prepared. The cell-based IBD vaccines are significantly cost-effective; however, it is essential to confirm their safety and efficacy. The main cell line used to product the cell-based IBD vaccines, is a primary chicken embryo fibroblast (CEF). Nevertheless, manipulation of CEF is extremely challenging and time-consuming. This study aimed to characterize a sensitive suspension cell cult

Infectious Bursal Disease Virus (IBDV) (Avibirnavirus genus, Birnaviridae family) is a non-enveloped virus with a double-stranded RNA (dsRNA) genome. IBDV causes a highly contagious and immunosuppressive disease in domestic chickens (Gallus gallus), representing a major threat to the global poultry industry. Apoptotic cell death and exacerbated innate immune responses have been implicated in IBDV pathogenesis. Previous studies from our laboratory demonstrated the crucial role of type I interferon (IFN) in triggering apoptosis in IBDV-infected cell cultures. Genomic IBDV dsRNA is recognized by

Feeding various probiotic lactic acid bacteria, including Enterococcus faecium, can alleviate intestinal inflammation and improve gut health in animals. Recently, postbiotics-non-living preparations derived from microbial cells or their metabolites-have gained attention. However, studies on the effects of these postbiotics on immune markers and changes in the gut microbiota of chickens are limited. In this study, we evaluated the effects of the probiotic strain E. faecium JB00008 on the chicken intestinal tract and characterized immune markers and gut microbiota following viral vaccination. Ch

Nowadays, vaccination is the primary and most effective method for preventing and controlling Infectious bursal disease virus (IBDV). A study examined the potential of a new Gecko cell line (Z1) for cultivating IBDV due to evolutionary similarities between birds and reptiles. After observing complete cytopathic effects (CPE), virus titre was determined using the TCID50 assay, and viral replication was confirmed using an RT-PCR test. In the next step, 150 broiler chickens (15 days old) were randomly divided into three equal groups for immune response measurements: G1: immunized/commercial vacci

At present, stress-induced immunosuppression is still a hidden threat that leads to immunization failure and outbreaks of poultry diseases, and causes huge economic losses to the modern poultry industry. However, the molecular mechanisms of stress-induced immunosuppression affecting viral vaccine immunity are still poorly understood. Here, we identified circAKIRIN2 as a conserved circular transcript in chicken, and explored its expression patterns in different immune states by quantitative real-time PCR (qRT-PCR), then conducted bioinformatics analysis. The results showed that circAKIRIN2 acti

The emerging hepatitis-hydropericardium syndrome (HHS) caused by the novel genotype of fowl adenovirus 4 (FAdV-4) and the infectious bursal disease (IBD) caused by the infectious bursal disease virus (IBDV) are important avian diseases, both cause huge economic losses to the poultry industry. Therefore, it is necessary to develop an efficient and convenient FAdV-4/IBDV bivalent vaccine to prevent the spread of FAdV-4 and IBDV infections. Given that VP2 is the main structural protein and protective antigen of IBDV, we constructed a recombinant FAdV-4 expressing IBDV VP2 in our previous study. I
