Since 1987, infectious bursal disease virus (IBDV) has circulated and evolved in Vietnam, but little is known about the genotypes present. IBDV samples were collected in 1987, 2001-2006, 2008, 2011, 2015-2019, and 2021 in 18 provinces. We conducted phylogenotyping analysis based on an alignment of 143 VP2-HVR (hypervariable region) sequences from 64 Vietnamese isolates (26 previous and 38 additional sequences and two vaccines, and alignment of 82 VP1 B-marker sequences, including one vaccine and four Vietnamese field strains. The analysis identified three A-genotypes, A1, A3, and A7, and two B

IPNV is a salmonid birnavirus that possesses the ability to establish asymptomatic persistent infections in a number of valuable fish species. The presence of IPNV may interfere with subsequent infection by other viruses. In the present study we show that an IPNV-carrier cell line (EPCIPNV) can induce an antiviral state in fresh EPC by co-cultivating both cell types in three different ways: a "droplet" culture system, a plastic chamber setup, and a transmembrane (Transwell®) system. All three cell co-culture methods were proven useful to study donor/target cell interaction. Naïve EPC cells gro

Many studies have demonstrated that miRNAs play a crucial role in virus-host interactions. This study aimed to investigate the antiviral effects of miR-146a in Epithelioma papulosum cyprini (EPC) cells infected with viral hemorrhagic septicemia virus (VHSV), infectious pancreatic necrosis virus (IPNV), and infectious haematopoietic necrosis virus (IHNV), which are major causes of mortality in various freshwater and marine fish worldwide. Cytopathic effects (CPE) were observed as early as 72 h post-infection in both the control group and the miR-146a inhibitor group. In contrast, CPE formation

Infectious bursal disease virus (IBDV) and fowl adenovirus serotype 4 (FAdV-4) cause infectious bursal disease (IBD) and hydropericardium-hepatitis syndrome, respectively. Recently, studies have reported co-infections of poultry with IBDV and FAdV-4, which is an important problem in the poultry industry. Here, the variant IBDV strain ZD-2018-1 and FAdV-4 isolate HB1501 were used to assess the pathogenicity of co-infection in 1-day-old specific pathogen-free (SPF) chickens. Compared with chickens infected with only FAdV-4, those coinfected with IBDV and FAdV-4 showed enhanced clinical symptoms,

Infectious bursal disease virus (IBDV) is one of the most impactful pathogens of poultry, with disease manifestations ranging from acute forms to subclinical but immunosuppressive infections. This heterogeneity, accompanied by a significant antigenic variability, is sustained by high mutation rates and frequent reassortments between the two genome segments, along with less frequent recombination events. In recent years, the proposal of several classification systems relying on phylogeny contributed to the characterization of several new IBDV genotypes, shedding light on an increasingly diverse

Recently, several attempts have been made to replace egg-based with cell-based vaccines to prevent and control Infectious Bursal Disease Virus (IBDV). This study aimed to evaluate a new fish cell line (M99) for culturing and replicating IBDV. After observing complete cytopathic effects (CPE) on the M99 cell line, virus titers were determined using the TCID50 test, and the presence of the virus was confirmed using an RT-PCR test. Subsequently, 135 broiler chickens (14 days old) were randomly divided into three equal groups for immune response measurements: G1: immunized with a commercial vaccin

Infectious pancreatic necrosis virus (IPNV), belonging to the genus Aquabirnavirus within the family Birnaviridae, causes huge economic loss to the global salmonid industry every year. Recently, outbreaks of disease caused by genogroup I IPNV were found in many rainbow trout (Oncorhynchus mykiss) farms worldwide. An inactivated vaccine was prepared using a genogroup I IPNV isolate with an optimized procedure as incubation with β-propanolactone (BPL) at the final concentration of 0.5% at room temperature for 48 h. The inactivated vaccine was used to immunize rainbow trout, and the protection ef

Infectious bursal disease virus (IBDV) vaccines do not induce sterilizing immunity, and vaccinated birds can become infected with field strains. Vaccine-induced immune selection pressure drives the evolution of antigenic drift variants that accumulate amino acid changes in the hypervariable region (HVR) of the VP2 capsid, which may lead to vaccine failures. However, there is a lack of information regarding how quickly mutations arise, and the relative contribution different residues make to immune escape. To model IBDV antigenic drift in vitro, we serially passaged a classical field strain bel

Infectious bursal disease (IBD) is a highly contagious disease in young chickens worldwide. The major strategy for the prevention and control of IBD virus (IBDV) is vaccination. Therefore, the present study aimed to compare the immunogenicity of four commercially available IBD vaccines on broilers (Ross 308) that were raised in areas with very virulent IBDV infection history. Two commercial broiler farms with four standard poultry houses were selected in Alborz (n=6,250 birds per house) and Khorasan Razavi (n=8,000 birds per house) provinces of Iran. In each farm, the houses were randomly assi

Infectious bursal disease (IBD), also known as Gumboro disease, is a globally well-known disease with a significant socio-economic effect. For control of IBD, several commercial egg- and cell-based vaccines are prepared. The cell-based IBD vaccines are significantly cost-effective; however, it is essential to confirm their safety and efficacy. The main cell line used to product the cell-based IBD vaccines, is a primary chicken embryo fibroblast (CEF). Nevertheless, manipulation of CEF is extremely challenging and time-consuming. This study aimed to characterize a sensitive suspension cell cult

Infectious bursal disease (IBD) is an acute, highly contagious, and immunosuppressive condition in chickens, caused by the infectious bursal disease virus (IBDV). The recent emergence of novel variant IBDV (nVarIBDV) poses a significant threat to the global poultry industry. However, currently available vaccines provide only limited protection against nVarIBDV strains. In this study, a recombinant avian metapneumovirus subtype B (aMPV/B) expressing the nVarIBDV VP2 gene (rLN16A-nVarVP2) was successfully rescued by inserting the gene between the G and L genes of the attenuated aMPV/B strain LN1

Feeding various probiotic lactic acid bacteria, including Enterococcus faecium, can alleviate intestinal inflammation and improve gut health in animals. Recently, postbiotics-non-living preparations derived from microbial cells or their metabolites-have gained attention. However, studies on the effects of these postbiotics on immune markers and changes in the gut microbiota of chickens are limited. In this study, we evaluated the effects of the probiotic strain E. faecium JB00008 on the chicken intestinal tract and characterized immune markers and gut microbiota following viral vaccination. Ch

Nowadays, vaccination is the primary and most effective method for preventing and controlling Infectious bursal disease virus (IBDV). A study examined the potential of a new Gecko cell line (Z1) for cultivating IBDV due to evolutionary similarities between birds and reptiles. After observing complete cytopathic effects (CPE), virus titre was determined using the TCID50 assay, and viral replication was confirmed using an RT-PCR test. In the next step, 150 broiler chickens (15 days old) were randomly divided into three equal groups for immune response measurements: G1: immunized/commercial vacci

At present, stress-induced immunosuppression is still a hidden threat that leads to immunization failure and outbreaks of poultry diseases, and causes huge economic losses to the modern poultry industry. However, the molecular mechanisms of stress-induced immunosuppression affecting viral vaccine immunity are still poorly understood. Here, we identified circAKIRIN2 as a conserved circular transcript in chicken, and explored its expression patterns in different immune states by quantitative real-time PCR (qRT-PCR), then conducted bioinformatics analysis. The results showed that circAKIRIN2 acti

The emerging hepatitis-hydropericardium syndrome (HHS) caused by the novel genotype of fowl adenovirus 4 (FAdV-4) and the infectious bursal disease (IBD) caused by the infectious bursal disease virus (IBDV) are important avian diseases, both cause huge economic losses to the poultry industry. Therefore, it is necessary to develop an efficient and convenient FAdV-4/IBDV bivalent vaccine to prevent the spread of FAdV-4 and IBDV infections. Given that VP2 is the main structural protein and protective antigen of IBDV, we constructed a recombinant FAdV-4 expressing IBDV VP2 in our previous study. I

Outbreaks of infectious bursal disease (IBD), a highly contagious immunosuppressive disease of young chickens, are still reported globally despite vaccination efforts. This study investigated the genetic characteristics of infectious bursal disease virus (IBDV) from 26 reported outbreaks in 2019 in Nigeria. Nucleotide sequences of VP2 hypervariable (hvVP2) region (n=26) and VP1 (n=23) of Nigerian IBDVs were determined. Our results revealed the detection of reassortant strains with segment A related to very virulent IBDV (vvIBDV) having virulence marker (222A, 242I, 256I, 294I and 299S), wherea

Infectious bursal disease (IBD) is an acute, highly contagious, economically important disease of young chickens caused by Avibirnavirus, the infectious bursal disease virus (IBDV). The causative virus is highly resilient in poultry environments and vaccination is the most effective measure for IBDV control. However, the susceptibility of highly attenuated IBDV vaccine strains to neutralization by maternally derived antibodies (MDA) and overwhelming virulence of partly attenuated strains have limited the application of conventional live IBDV vaccines in pre- and posthatch chicks. Nevertheless,

Infectious bursal disease (IBD) remains a potential worldwide threat to the poultry industry despite several vaccination approaches. Because maternally derived antibodies (MDA) constitute a critical problem for IBD vaccination, we examined the efficiency of the intracloacal vaccination approach in breaking through MDA. Experiment 1 determined the ability of the vaccinal strain to multiply in the bursa of Fabricius (BF) in chicks with a high level of MDA. Using real-time polymerase chain reaction, we quantified the strain in the bursae of vaccinated and non-vaccinated chicks. Experiment 2 was p
