Pasture plots in the south of England were contaminated each month throughout 1975 with pig faeces containing Trichuris suis ova. At regular intervals thereafter, soil samples were taken, the T suis ova extracted and their state of development noted. Depending on the time of year that the plots were contaminated, ova required between 62 and 90 weeks to complete their development to the infective stage. Little or no development occurred during winter. Once the infective stage was reached, the ova survived for at least two years. Samples taken from the plots at various depths demonstrated that T

Life in the highly turbulent surf zone poses a severe challenge to reproduction in free-spawning animals. Not only can breaking waves quickly dilute the gametes shed by spawning organisms, but turbulence-induced shear stresses may limit fertilization and interfere with normal development. A Couette cell was used to re-create some of the effects of turbulent water motion to study effects of environmentally relevant shear stresses on fertilization in the purple sea urchin (Strongylocentrotus purpuratus). Although low shear stresses improved fertilization success (presumably by increasing mixing)

Tissue or cell transplantation has been an extremely valuable technique for studying developmental potential of certain cell population, dissecting cell-environment interaction relationship, identifying stem cells, and many other applications. One key technical requirement for performing transplantation assay is the capability of distinguishing the transplanted donor cells from the endogenous host cells, and tracing the donor cells over time. Zebrafish has emerged as an excellent model organism for performing transplantation assay, thanks to the transparency of embryos during development and e

The zebrafish oocyte differs substantially from the zygote and cleavage-stage embryo with regard to the ease with which it can be microinjected with proteins or reagents that modify subsequent development. The objective of this chapter is to describe methods developed in this and other laboratories for microinjection and calcium imaging in the unfertilized zebrafish egg. Methods of immobilizing the oocyte include a holding chamber and a holding pipette. The holding chamber allows imaging of three or four oocytes simultaneously, while the holding pipette facilitates imaging of localized regions

pbx genes encode homeodomain-containing transcriptional regulators that interact with other proteins to control embryogenesis and tumorigenesis. We present the characterization of a zebrafish pbx CDNA that appears to encode a novel family member, pbx4. pbx4 RNA is maternally deposited and is detected throughout the zebrafish embryo during blastula stages. It becomes excluded from ventroanterior structures at late gastrula stages and is detected within the forming central nervous system during segmentation stages. pbx4 expression overlaps with that of two other homeobox genes, hoxb1b and meis3,

A 4-day embryo-larval zebrafish test, from blastula stage to hatching included, was developed. The observations of embryo developmental were made at different development stages, for which morphological, physiological, and behavioral endpoints were selected and quantified for unexposed and exposed embryos. The sensitivity and the ability of these endpoints to inform about mode of action (MoA) were established in testing three model toxicants with well-known toxic effects (propranolol, malathion, cadmium). Lethal, sublethal (heart rate/edema, spontaneous movements, and hatching rate/time distur

Perfluorooctanoic acid (PFOA) and cadmium (Cd) are persistent environmental pollutants, bioaccumulative, and capable of crossing the placental barrier. There is evidence of developmental toxicity induced by PFOA and Cd, but their effects on early development are still poorly understood. In the present study, chicken embryos were exposed to PFOA, Cd, and a mixture of PFOA + Cd from blastula stage up to 72 h of incubation. Embryo mortality and morphology were assessed. PFOA and Cd exposure increased mortality and induced malformations, mainly in the cephalic region, the neural tube, and the vent

Cutting the blastodisc of nonincubated chick eggs in parallel and perpendicularly to the presumptive axis results in the development of twin embryos in 28% of cases. After tripsin and versen solutions were injected in various concentrations into the blastula cavity, the part of the developed twins was but negligible (5% and 3%, respectively). In all experimental groups embryos with different developmental anomalies were often occurrence, as well as various types of malformation were noted in twins. Since Hans Driesch's pioneering work in 1891, it has been known that animal embryos can develop into complete individuals even when divided. However, the developmental processes and molecular mechanisms enabling this self-organization remain poorly understood. In this study, we revisit Driesch's experiments by examining the development of isolated 2-cell stage blastomeres in the sea urchin, Hemicentrotus pulcherrimus. Contrary to intact embryos, these isolated blastomeres initially form a flat, single layer of dividing cells that eventually round up to be a blastula. Live imaging and knockdown

OBJECTIVE: The aim of this study was to evaluate the efficacy of a non-expensive, easy and fast technique (direct micro swim-up) for sperm preparation in intracytoplasmic sperm injection (ICSI) treatments without the use of centrifuge.
METHODS: We carried out a multicentric study in which a total of 140 ICSI-cycles were included. Sibling oocytes were divided into two groups according to semen preparation procedures: group A, discontinuous gradients (DG) (oocytes n=668), and group B, direct micro swim-up (MSU) (oocytes n=660). We analyzed differences in some key performance indicators.

Cryopreservation of zebrafish embryos remains challenging due to their low tolerance for cryoprotectant exposure and their susceptibility to lethal ice formation. Ice recrystallization inhibitors (IRIs) such as N-2-fluorophenyl-D-gluconamide (2FA) have shown promise in improving post-thaw survival in other systems, yet their biocompatibility in zebrafish embryos has not been established. Here, we evaluated the cytotoxicity and teratogenicity of 2FA during discrete developmental stages and under continuous 72-h exposure. Embryos were submerged in embryo media containing 9 mM or 15 mM 2FA during

Pre-B-cell leukemia homeobox transcription factor 1 (PBX1) is a member of the TALE (three-amino acid loop extension) family and functions as a homeodomain transcription factor (TF). When dimerized with other TALE proteins, it can act as a pioneer factor and provide regulatory sequences via interaction with partners. In vertebrates, PBX1 is expressed during the blastula stage, and its germline variations in humans are interrelated with syndromic anomalies of the kidney, which plays an important role in hematopoiesis and immunity among vertebrates. Herein, we summarize the existing data on PBX1

The peri-implantation period from blastula to gastrula is one of the crucial stages of human embryo and stem cell development. During development, human embryos undergo many crucial events, such as embryonic lineage differentiation and development, structural self-assembly, pluripotency state transition, cell communication between lineages, and crosstalk between the embryo and uterus. Abnormalities in these developmental events will result in implantation failure or pregnancy loss. However, because of ethical and technical limits, the developmental dynamics of human peri-implantation embryos a

Collecting information about biochemical processes occurring inside a single cell or embryo is traditionally done either using fluorescent dyes with microscopy or via microelectrode voltage-clamp techniques. This paper demonstrates that a more direct method - transmission of information using an electronic chip implanted in an embryo - is feasible. A light-activated microtransponder with dimensions 250 μm × 250 μm × 100 μm (a "p-Chip") was implanted into a blastula-stage frog (Xenopus laevis) embryo. To implant the chip, a small slit is made in the blastocoel roof with an electrolytically-shar

The body of a vertebrate arises through a series of inductive interactions in the embryo. Macrocephaly is a distortion of the body in which a disproportionate amount of tissue is devoted to the head. This syndrome occurs in certain hybrids between frog species and appears to be due to an alteration of inductive relationships. Chimeric blastulae between normal and hybrid embryos developed macrocephaly when the marginal zone was derived from the hybrid. In these cases, a large cement gland, characteristic of the hybrid head, was induced to form from normal ectoderm. When hybrid zygotes were irra

The developmental toxicity of N-nitrosodimethylamine (NDMA) and trichloroethylene (TCE) was assessed with Frog Embryo Teratogenesis Assay: Xenopus (FETAX). Late Xenopus laevis blastulae were exposed to NDMA and TCE for 96-h in two separate static-renewal tests with and without the presence of three differently induced exogenous metabolic activation systems (MAS). The MAS consisted of Aroclor 1254-induced (Aroclor 1254 MAS), isoniazid-induced (INH MAS), and a post-isolation mixture (mixed MAS) of Aroclor 1254- and isoniazid-induced rat liver microsomes. Addition of the INH MAS and the mixed MAS

DIS3-like 3'-5' exoribonuclease 2 (DIS3L2), an exoribonuclease, is known to preferentially degrade uridylated RNA substrates, miRNAs, and ncRNAs. Recent reports show that DIS3L2 also plays a key role in cell proliferation and tumor growth. Mutations in DIS3L2 are associated with congenital overgrowth disorders such as Perlman syndrome, yet the developmental functions of DIS3L2 remain unknown. We report the developmental role of dis3l2 in neural crest specification, patterning, and survival in the zebrafish embryo. The dis3l2 morphants exhibited reduced expression of neural crest specifier gene

The activity of ADP-ribosyltransferase in nuclei isolated from sea-urchin embryos was estimated by the incorporation of [adenosine-14C]NAD+ into the acid-insoluble fraction. Hydrolysis of this acid-insoluble product by snake venom phosphodiesterase yielded radioactive 5'-AMP and phosphoribosyl-AMP. The incorporation of [14C]-NAD+ was inhibited by 3-aminobenzamide and nicotinamide, potent inhibitors of ADP-ribosyltransferase. [14C]NAD+ incorporation into the acid-insoluble fraction results from the reaction of ADP-ribosyltransferase. The optimum pH for the enzyme in isolated nuclei was 7.5. The

The dorsal organizer, essential for vertebrate embryonic axis formation, is induced by microtubule-mediated transport of maternal determinants. Maternal Huluwa (Hwa) has been identified as an essential organizer inducer in zebrafish and frogs, functioning at midblastula stages to activate β-catenin signaling in the preorganizer. It remains unknown if maternal Hwa functions at or before fertilization. Here, we report that maternal Hwa protein is critical for organizing the vegetal parallel microtubule array immediately after fertilization in zebrafish. Hwa protein and mRNA are enriched at the v
