Sodium rutin attenuates blood-spinal cord barrier disruption and mitochondrial oxidative stress via SIRT1/β-catenin pathway in distractive spinal cord injury.

BACKGROUND: Distractive spinal cord injury (DSCI) is a severe complication of spinal deformity correction surgery, causing early disruption of the blood-spinal cord barrier (BSCB). However, the molecular mechanisms that translate mechanical distraction stress into vascular endothelial dysfunction remain unclear, and effective pharmacological strategies targeting this process are lacking. PURPOSE: This study aimed to investigate the therapeutic effects of sodium rutin (NaR), a water-soluble derivative of rutin, on BSCB disruption following DSCI, and to elucidate the underlying molecular mechan…

Intrathecal transplantation of bone marrow stromal cells attenuates blood-spinal cord barrier disruption induced by spinal cord ischemia-reperfusion injury in rabbits.

OBJECTIVE: Intrathecal administration of bone marrow stromal cells has been found to produce beneficial effects on ischemia-reperfusion injury to the spinal cord. The blood-spinal cord barrier is critical to maintain spinal cord homeostasis and neurologic function. However, the effects of bone marrow stromal cells on the blood-spinal cord barrier after spinal cord ischemia-reperfusion injury are not well understood. This study investigated the effects and possible mechanisms of bone marrow stromal cells on blood-spinal cord barrier disruption induced by spinal cord ischemia-reperfusion injury.…

Valproic acid attenuates blood-spinal cord barrier disruption by inhibiting matrix metalloprotease-9 activity and improves functional recovery after spinal cord injury.

The disruption of blood-spinal cord barrier (BSCB) after spinal cord injury (SCI) elicits an intensive local inflammation by the infiltration of blood cells such as neutrophils and macrophages, leading to cell death and permanent neurological disability. SCI activates matrix metalloprotease-9 (MMP-9), which is known to induce BSCB disruption. Here, we examined whether valproic acid (VPA), a histone deacetylase inhibitor, would attenuate BSCB disruption by inhibiting MMP-9 activity, leading to improvement of functional outcome after SCI. After moderate spinal cord contusion injury at T9, VPA (3…

Salidroside-Pretreated Small Extracellular Vesicles Derived from Human Umbilical Cord Mesenchymal Stem Cells Promote Blood-Spinal Cord Barrier Repair After Spinal Cord Injury in Mice.

This study investigates the role of salidroside-pretreated small extracellular vesicles (SAL-sEVs) derived from human umbilical cord mesenchymal stem cells (Huc-MSCs) in spinal cord injury (SCI). Results demonstrate that these sEVs repair the damaged blood-spinal cord barrier (BSCB) and facilitate functional recovery. sEVs were collected from Huc-MSCs pretreated with salidroside via ultracentrifugation. sEVs were characterized using transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), and Western blot. The uptake of sEVs by endothelial cells was confirmed through immun…

Blood-brain and blood-spinal cord barrier permeability during the course of experimental allergic encephalomyelitis in the rat.

Experimental allergic encephalomyelitis (EAE) was induced in young male Lewis rats. Blood-brain barrier permeability to radiotracers of different molecular sizes was studied at intervals after induction using a tissue sampling technique. The results were correlated to the clinical picture and to the histological appearance of the central nervous system. Significant increase in blood-brain barrier permeability to small molecules was found to precede clinical symptoms by one day in the lumbar spinal cord and to coincide with the onset of clinical disease in other regions. In all regions, increas

A KETOGENIC DIET PROMOTES BLOOD-SPINAL CORD BARRIER RESEALING TO DECREASE SPINAL CORD IMMUNE CELL INFILTRATION IN A PRECLINICAL MODEL OF MULTIPLE SCLEROSIS.

We previously reported that a medium chain triglyceride and W-3 fatty acid rich ketogenic diet (KD) restores vision and motor functions to mice undergoing experimental autoimmune encephalomyelitis (EAE), a preclinical model of multiple sclerosis (MS). The studies reported here use immune cell profiling to identify the mechanisms underlying this efficacy. We show here that the diet dramatically reduces the infiltration of CD45 HI immune cells into the spinal cords of EAE mice when fed for 2 weeks beginning at symptom onset. This reduction was preceded by an increase of myeloid cells in the bloo

Early blood-spinal cord barrier stabilization preserves peri-lesional tissue and improves recovery after SCI.

Vascular disruption is an early and critical event in spinal cord injury (SCI), but its contribution remains poorly understood. Using novel in vivo two-photon dual-dye imaging, we found simultaneous blood-spinal cord barrier (BSCB) leakage and venous dilation in both the injury epicenter and adjacent transitional segment after cervical SCI in rats. Notably, vascular permeability in the transitional zone preceded axonal and neuronal loss, revealing a therapeutic window. Systemic delivery of ferulic acid-glycol chitosan (FA-GC) nanoparticles, a membrane-sealant, rapidly stabilized the compromise…

Blood-spinal cord barrier disruption contributes to early motor-neuron degeneration in ALS-model mice.

Humans with ALS and transgenic rodents expressing ALS-associated superoxide dismutase (SOD1) mutations develop spontaneous blood-spinal cord barrier (BSCB) breakdown, causing microvascular spinal-cord lesions. The role of BSCB breakdown in ALS disease pathogenesis in humans and mice remains, however, unclear, although chronic blood-brain barrier opening has been shown to facilitate accumulation of toxic blood-derived products in the central nervous system, resulting in secondary neurodegenerative changes. By repairing the BSCB and/or removing the BSCB-derived injurious stimuli, we now identify…

Endothelial HOXB9/SDC4 signaling exacerbates post-ischemic blood-brain and blood-spinal cord barrier disruption by promoting PKCα activation.

BACKGROUND: Ischemic stroke is one of the most deadly chronic diseases, it damages blood-brain barrier (BBB) and blood-spinal cord barrier (BSCB). Syndecan-4 (SDC4) affects BBB integrity and function, but the specific mechanism is unclear. METHODS: The middle cerebral artery occlusion (MCAO) rat model and the oxygen-glucose deprivation/reoxygenation (OGD/R) cell model were conducted. The influences of SDC4 on the integrity and function of BBB were detected by the transmembrane resistance (TEER) assay, immunofluorescence staining and Evans Blue assay. Luxol Fast Blue staining was used to obser…

Exosomal miR-149 from human umbilical cord mesenchymal stem cells attenuates spinal cord injury-induced blood-spinal cord barrier disruption by suppressing the ET-1/PI3K/Akt signaling pathway.

BACKGROUND: Spinal cord injury (SCI) leads to persistent neurological deficits partly by disruption of the blood-spinal cord barrier (BSCB). Small extracellular vesicles (sEVs) from human umbilical cord mesenchymal stem cells (hUC-MSCs) can promote BSCB repair, but their active components remain unclear. This study examined whether miR-149 carried by hUC-MSC-derived sEVs (hUC-MSCs-sEVs) protects the BSCB after SCI by targeting endothelin-1 (ET-1). METHODS: Human brain microvascular endothelial cells (HBMECs) were subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) to model barrier i…

The protective role of nuclear Heme oxygenase-1 in blood-spinal cord barrier after hypoxia in vitro.

Spinal cord injury (SCI), which results in severe lower-limb motor dysfunction, is a neurological disorder that damages the central nervous system. Disruption of the blood-spinal cord barrier (BSCB) results in secondary injury and neuronal apoptosis. Thus, this study aimed to investigate the effect of nuclear heme oxygenase-1 (HO-1) on BSCB after hypoxia in an in vitro model and to identify the potential regulatory mechanism of nuclear HO-1 (nHO-1) in BSCB. In an in vitro hypoxia model, nHO-1 treatment attenuated BSCB permeability. In addition, western blot analysis indicated that the expressi…

TIMP1 inhibits Rac1-mediated ROS production to ameliorate blood-spinal cord barrier disruption in amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder characterized by the progressive degeneration of motor neurons, for which therapeutic strategies and pharmacological interventions remain limited. Disruption of the blood-spinal cord barrier (BSCB) has been identified as a significant factor that may exacerbate motor neuron damage. Tissue inhibitor of metalloproteinase-1 (TIMP1), a molecule known for its dual roles in inhibiting matrix metalloproteinase (MMP) activity and exerting cytokine-like effects via receptor interactions, has been demonstrated to ameliorate endothelial …

CXCL13 Повреждает позвоночник крови, поощряя RNF6/Sqstm1-Ubictiination.

Peripheral nerve injury alters blood-spinal cord barrier functional and molecular integrity through a selective inflammatory pathway.

Peripheral nerve lesion triggers alterations in the spinal microenvironment that contribute to the pathogenesis of neuropathic pain. While neurons and glia have been implicated in these functional changes, it remains largely underexplored whether the blood-spinal cord barrier (BSCB) is also involved. The BSCB is an important component in the CNS homeostasis, and compromised BSCB has been associated with different pathologies affecting the spinal cord. Here, we demonstrated that a remote injury on the peripheral nerve in rats triggered a leakage of the BSCB, which was independent of spinal micr…

BACKGROUND: Valeriana jatamansi Jones, a globally utilized medicinal plant, exhibits favorable pharmacological effects against depression and tumors. Iridoids derived from V. jatamansi (IRFV) promote recovery following spinal cord injury (SCI). Inflammation and disruption of the blood-spinal cord barrier (BSCB) represent key pathological processes in SCI. However, the specific effects of IRFV on neuroinflammation and BSCB integrity remain unexplored. METHODS AND RESULTS: This study aims to elucidate the functional significance and molecular mechanisms by which IRFV modulates neuroinflammation…

Nuclear heme oxygenase-1 enhances blood-spinal cord barrier repair via corticotropin-releasing hormone-mediated tight junction phosphorylation.

OBJECTIVE: The hallmark of secondary spinal cord injury (SCI) is destruction of the blood-spinal cord barrier (BSCB). Nuclear heme oxygenase-1 (NHO-1) is involved in neuroprotection in the central nervous system. Previously, we demonstrated that adenoviral delivery of nuclear-targeted HO-1 (NHO-1) alleviates BSCB disruption, but the molecular mechanisms remain to be clarified. This study was conducted to investigate the specific regulatory mechanisms regulating tight junctions in the BSCB. METHODS: A moderate T10 contusion SCI was induced in adult male Sprague-Dawley rats. Nuclear-targeted HO…

A Novel Retrievable Drug Infusion Stent and Blood-Brain Barrier Portal for Antibody Therapies to the Spinal Cord.

BACKGROUND AND OBJECTIVES: The blood-brain barrier (BBB) is prohibitive to intravascular drug delivery. We hypothesized that isolation of spinal cord perfusion would focus on a higher effective dose while minimizing systemic toxicity and that a histamine portal would increase BBB permeability and facilitate large molecule therapies to the spinal cord. METHODS: A dumbbell-shaped, retrievable drug infusion stentgraft (RDIS) was constructed from nitinol and polytetrafluorethylene. A porcine model under anesthesia underwent percutaneous femoral insertion of the sheathed RDIS and deployment in the
