In Vitro and In Vivo Models for Drug Transport Across the Blood-Testis Barrier.

The blood-testis barrier (BTB) is a selectively permeable membrane barrier formed by adjacent Sertoli cells (SCs) in the seminiferous tubules of the testes that develops intercellular junctional complexes to protect developing germ cells from external pressures. However, due to this inherent defense mechanism, the seminiferous tubule lumen can act as a pharmacological sanctuary site for latent viruses (e.g., Ebola, Zika) and cancers (e.g., leukemia). Therefore, it is critical to identify and evaluate BTB carrier-mediated drug delivery pathways to successfully treat these viruses and cancers. M

[Role of autophagy in cadmium-induced damage to the blood-testis barrier in mice].

OBJECTIVE: To investigate the role of autophagy in cadmium chloride (CdCl2)-induced damage to the blood-testis barrier (BTB) in mice. METHODS: Twenty four-week-old male C57BL/6 mice were randomly divided into four groups and intraperitoneally injected with CdCl2 at 0 mg/kg/d (the control), 0.5 mg/kg/d (low-dose), 1.0 mg/kg/d (medium-dose) and 2.0 mg/kg/d (high-dose) respectively for 28 consecutive days. Then the morphological changes of the testis tissue was observed by HE staining, the integrity of BTB measured with the biotracer, and the expressions of the BTB components ZO-1 and N-Cadherin

GATA4 Regulates Blood-Testis Barrier Function and Lactate Metabolism in Mouse Sertoli Cells.

Conditional deletion of Gata4 in Sertoli cells (SCs) of adult mice has been shown to increase permeability of the blood-testis barrier (BTB) and disrupt spermatogenesis. To gain insight into the molecular underpinnings of these phenotypic abnormalities, we assessed the impact of Gata4 gene silencing in cell culture models. Microarray hybridization identified genes dysregulated by siRNA-mediated inhibition of Gata4 in TM4 cells, an immortalized mouse SC line. Differentially expressed genes were validated by quantitative RT-PCR analysis of primary cultures of Gata4(flox/flox) mouse SCs that had

Spermatogenesis and blood-testis barrier in rats after long-term Vitamin A deprivation.

It has been established that experimental avitaminosis A in rats results in a 'Sertoli cell-only situation' after about 10 weeks, and that replacing the vitamin immediately reinitiates spermatogenesis. The present study deals with testicular recovery after prolonged deprivation (up to 19 weeks). The Sertoli cell-only situation reached under this condition was thought to be refractory to Vitamin A replacement. However, spermatogenesis did reinitiate about 11 weeks after vitamin restoration, although in an atypical manner. The blood-testis barrier, normally assembled when spermatocytes reaches t

Signaling pathways regulating blood-tissue barriers - Lesson from the testis.

Signaling pathways that regulate blood-tissue barriers are important for studying the biology of various blood-tissue barriers. This information, if deciphered and better understood, will provide better therapeutic management of diseases particularly in organs that are sealed by the corresponding blood-tissue barriers from systemic circulation, such as the brain and the testis. These barriers block the access of antibiotics and/or chemotherapeutical agents across the corresponding barriers. Studies in the last decade using the blood-testis barrier (BTB) in rats have demonstrated the presence o

Human interleukin-1 alpha crosses the blood-testis barriers of the mouse.

Interleukin-1 alpha (IL-1 alpha) has been shown to have direct effects on the gonads, affecting steroidal secretion, DNA synthesis by spermatogonia, and the immune function of the testes. It is unclear, however, how IL-1 alpha exerts these effects because the testis is partitioned into basal and adluminal compartments by both a vascular and a Sertoli cell barrier. The authors used a highly sensitive method to quantify the unidirectional flux rates (Ki) into the testis of technetium pertechnetate-labeled human albumin (T-alb), a compound that does not readily cross the vascular barrier, and hum

Permeability of selected drugs and chemicals across the blood-testis barrier of the rat.

Physiological studies by Setchell and others have described the existence of a blood-testis barrier (BTB) surrounding the seminiferous tubules of the mammalian testis. These studies were initiated to better define the role of the BTB with regard to the penetration of exogenous chemicals to male germ cells. The rete testis was cannulated in rats and fluid was collected. Test chemicals or drugs were usually administered by continuous i.v. infusion. Permeability of nonelectrolytes of various molecular sizes, acidic compounds with varying partition coefficients and pKalpha values, such as salicyli

Experimental varicocele does not affect the blood-testis barrier, epididymal electrolyte concentrations, or testicular blood gas concentrations.

It has been previously shown that 30-day experimental left varicocele (ELV) in adult rats produces a bilateral increase in testicular blood flow and temperature, as well as a concomitant decrease in epididymal sperm count and motility. In the present study, adult male rats with induced ELV were subjected to a variety of studies to determine the mechanism by which unilateral ELV causes a bilateral testicular response. The results demonstrate that ELV does not alter the blood-testis barrier (BTB) to 3H-inulin (MW 5000), it being largely excluded from entry into the tubule lumen in both control a

Chronic sleep loss disrupts blood-testis and blood-epididymis barriers, and reduces male fertility.

Sleep loss increases blood-brain barrier permeability. As the blood-brain barrier and the blood-tissue barriers in the reproductive tract (blood-testis and blood-epididymis barriers) share common characteristics, we hypothesized that sleep restriction may also modify their barrier function. Previous reports showed that sleep loss decreased sperm viability and progressive fast mobility, which may be a consequence of altered blood-testis and blood-epididymis barrier. Therefore, we quantified changes in blood-testis and blood-epididymis barrier after sleep loss and related them to male fertility.

Penetration of some compounds through blood-brain and blood-testis barriers in chronically hypertensive rats.

Acute hypertension impairs the function of biological barriers, e.g. that of blood-brain barrier. We tested the hypothesis that chronic hypertension might influence the penetration of compounds through blood-brain and blood-testis barriers. With this in mind, the penetration of relatively small radioactive compounds (14C-2-methyl-4-chlorophenoxyacetic acid = 14C-MPA, 14C-sucrose and 14C-antipyrine, 5 microCi/kg intravenously) into the brain, liquor space and testis was compared in male normotensive (Wistar and chronically hypertensive SH rats (11-22 months old). In chronic hypertension after 1

Ferroptosis inhibitor liproxstatin-1 alleviates triptolide-induced blood testis-barrier disruption and testicular injury.

Triptolide (TP), the principal bioactive component of Tripterygium wilfordii (T. wilfordii), exhibits potent pharmacological activities; however, its clinical application is limited by severe male reproductive toxicity, and effective interventions to mitigate reproductive toxicity remain limited. Although the relationship between ferroptosis and TP-induced male reproductive damage has garnered attention, it is still not fully understood whether intervening in ferroptosis can serve as a viable approach to ameliorate TP-induced testicular injury. Thus, this study aimed to characterize TP-induced

In Vivo Tracking Deciphers the Traversal of Ebola Virus across the Blood-Testis Barrier.

Ebola virus (EBOV) can persist in semen to mediate sexual transmission and drive epidemic resurgence. However, the mechanism by which EBOV breaches the blood-testis barrier (BTB) to invade the male reproductive system remains poorly understood. Here, we developed a quantum dot (QD)-based virus-barrier visualization system (QD-VBVS) integrating QD-encapsulated EBOV-like particles (EBO-VLP-QDs) with two-photon microscopy to enable real-time, high-resolution visualization of viral-BTB interactions in live mice. Single-particle tracking uncovered a four-stage BTB traversal mode, with ∼2.48% of EBO

2,2',4,4'-Tetrabromodiphenyl ether exposure disrupts blood-testis barrier integrity through CMA-mediated ferroptosis.

2,2',4,4'-Tetrabromodiphenyl ether (PBDE-47), being the most prevalent congener of polybrominated diphenyl ethers (PBDEs), has been found to accumulate greatly in the environment and induce spermatogenesis dysfunction. However, the specific underlying factors and mechanisms have not been elucidated. Herein, male Sprague-Dawley (SD) rats were exposed to corn oil, 10 mg/kg body weight (bw) PBDE-47 or 20 mg/kg bw PBDE-47 by gavage for 30 days. PBDE-47 exposure led to blood-testis barrier (BTB) integrity disruption and aberrant spermatogenesis. Given that Sertoli cells are the main toxicant target

Nickel induces blood-testis barrier damage through ROS-mediated p38 MAPK pathways in mice.

Nickel (Ni) is an essential common environmental contaminant, it is hazardous to male reproduction, but the precise mechanisms are still unknown. Blood-testis barrier (BTB), an important testicular structure consisting of connections between sertoli cells, is the target of reproductive toxicity caused by many environmental toxins. In this study, ultrastructure observation and BTB integrity assay results indicated that NiCl2 induced BTB damage. Meanwhile, BTB-related proteins including the tight junction (TJ), adhesion junction (AJ) and the gap junction (GJ) protein expression in mouse testes a

Continual maintenance of the blood-testis barrier during spermatogenesis: the intermediate compartment theory revisited.

Tight junctions occur between the lateral processes of neighboring Sertoli cells that divide the seminiferous epithelium into two compartments: basal and adluminal compartments. These tight junctions constitute the blood-testis barrier (BTB). The established theory that the BTB must open when spermatocytes translocate from the basal compartment to the adluminal compartment is marked by one contradiction, that is, normal spermatogenesis occurs in the testis because the BTB is expected to constantly seclude the adluminal compartment from the basal compartment in order to protect haploid germ cel

Irisin-treated microbiota restore blood-testis barrier integrity and spermatogenesis in chronically stressed rats.

Chronic psychological stress impairs male fertility by disrupting spermatogenesis and the blood-testis barrier (BTB), which is essential for testicular function. Irisin, a myokine/adipokine involved in metabolic regulation, protects against testicular dysfunction and may beneficially modulate gut microbiota, highlighting its potential in the gut-testis axis. This study investigated the effect of fecal microbiota transplantation (FMT) from irisin-treated rats in restoring BTB integrity in a chronic unpredictable stress (CUS) rat model. Male Sprague-Dawley rats were randomized into four groups:

Curcumin ameliorates aging-induced blood-testis barrier disruption by regulating AMPK/mTOR mediated autophagy.

The blood-testis barrier (BTB) is composed of tight junctions (TJ) between adjacent Sertoli cells (SCs) and is crucial for sperm growth and development. Aging-induced TJ impairment is closely related to testicular dysfunction. Curcumin, a natural compound, has been widely demonstrated to have a wide range of pharmacological activities, but its regulatory effects on tight junction damage in the testis remain unclear. We here explored the effect of curcumin on TJ function and its underlying molecular mechanism by using D-galactose (D-gal)-induced mouse testis and mouse testicular SCs (TM4) aging

Cross-talk between Vimentin and autophagy regulates blood-testis barrier disruption induced by cadmium.

The blood-testis barrier (BTB) plays a vital role in mammalian spermatogenesis by separating the seminiferous epithelium into an adluminal and a basal compartment. Cadmium (Cd) is a toxic heavy metal that is widely present in the environment. We observed that Cd can induce BTB disruption, leading to apoptosis of testicular cells. However, the molecular mechanisms contributing to BTB injury induced by Cd have not yet been fully clarified. Vimentin (Vim) is an important desmosome-like junction protein that mediates robust adhesion in the BTB. In this study, we investigated how Vim responds to Cd

Effect of Metformin on Sertoli Cell Fatty Acid Metabolism and Blood-Testis Barrier Formation.

Sertoli cells (SCs) are essential to maintaining germ cell development. Metformin, the main pharmacologic treatment for pediatric type 2 diabetes, is administered to children during SC maturation. The present study aimed to analyze whether metformin affects SC energy metabolism and blood-testis barrier (BTB) integrity. Primary SC cultures were used for the in vitro studies. In vivo effects were studied in Sprague-Dawley rats treated with 200 mg/kg metformin from Pnd14 to Pnd30. Metformin decreased fatty acid oxidation and increased 3-hydroxybutyrate production in vitro. Moreover, it decreased
