Detection and isolation of Bluetongue virus from commercial vaccine batches.

In this report we describe the detection and identification of Bluetongue virus (BTV) contaminations in commercial vaccines. BTV RNA was detected in vaccine batches of Lumpy skin disease (LSD) and Sheep pox (SP) using quantitative PCR (qPCR) for VP1 and NS3 genes. Both batches were positive for VP1 and NS3 in qPCR. The LSD vaccine-derived sample was positive for VP1 and VP2 in conventional PCR. The SP vaccine-derived sample was examined by amplification of VP1, VP4, VP6, VP7, NS2 and NS3 gene segments in conventional PCR. The SP vaccine-derived sample was further propagated in embryonated chic

Predicting temperature-dependent transmission suitability of bluetongue virus in livestock.

The transmission of vector-borne diseases is governed by complex factors including pathogen characteristics, vector-host interactions, and environmental conditions. Temperature is a major driver for many vector-borne diseases including Bluetongue viral (BTV) disease, a midge-borne febrile disease of ruminants, notably livestock, whose etiology ranges from mild or asymptomatic to rapidly fatal, thus threatening animal agriculture and the economy of affected countries. Using modeling tools, we seek to predict where the transmission can occur based on suitable temperatures for BTV. We fit thermal

Quantifying bluetongue vertical transmission in French cattle from surveillance data.

Bluetongue is a vector-borne disease of ruminants with economic consequences for the livestock industry. Bluetongue virus serotype 8 (BTV-8) caused a massive outbreak in Europe in 2006/2009 and re-emerged in France in 2015. Given the unprecedented epidemiological features of this serotype in cattle, the importance of secondary routes of transmission was reconsidered and transplacental transmission of BTV-8 was demonstrated in naturally and experimentally infected cattle. Here we used surveillance data from the on-going outbreak to quantify BTV-8 vertical transmission in French cattle. We used

Bluetongue outbreaks: Looking for effective control strategies against Culicoides vectors.

Several arthropod-borne diseases are now rising with increasing impact and risks for public health, due to environmental changes and resistance to pesticides currently marketed. In addition to community surveillance programs and a careful management of herds, a next-generation of effective products is urgently needed to control the spread of these diseases, with special reference to arboviral ones. Natural product research can afford alternative solutions. Recently, a re-emerging of bluetongue disease is ongoing in Italy. Bluetongue is a viral disease that affects ruminants and is spread throu

[Vaccination against two vector-borne diseases: bluetongue and West Nile].

In 1999 and 2006, two viral diseases emerged massively and unexpectedly in the United States (West Nile disease) and northern Europe (bluetongue disease). Control of infectious diseases transmitted by insect vectors is based on a variety of approaches (including sanitary measures), but primarily on vaccination. Vaccination is more efficient and less expensive than monitoring of insect vectors. The dynamics and epidemiology of two arboviral diseases (West Nile and bluetongue) are described, together with the different vaccines and vaccination methods.

Experimentally induced infection with bluetongue virus serotype 11 in cows.

The consequences of inoculation of bluetongue virus (BTV) serotype 11 into 16 susceptible cows either at the time of breeding or at specified stages of pregnancy were studied. The cows were free of BTV or epizootic hemorrhagic disease virus, and none had antibodies to BTV before virus inoculation. A group of 4 cows was mated naturally to a bull reported to shed BTV-11 (CO75B300 strain) in the semen. The bull was suspected of infecting cows at mating with BTV-11, which subsequently transplacentally infected the developing fetuses and induced persistently infected and congenitally malformed prog

Sero-epidemiology and molecular detection of Bluetongue virus in Indian ruminants.

Bluetongue (BT) is a non‑contagious arthropod‑borne viral disease of domestic and wild ruminants. It is endemic to India and clinical outbreaks of disease have been reported mainly in sheep, although BT is often asymptomatic in other ruminant species. In the present serological survey, a total of 576 serum samples, comprising of 416 cattle and 160 sheep, covering different agro‑climatic zones of Rajasthan, Uttar Pradesh, and Karnataka states, were screened for the presence of Bluetongue virus (BTV) specific antibodies using competitive enzyme‑linked immunosorbent assay (c‑ELISA). Overall 73.08

Detection in and circulation of Bluetongue virus among domestic ruminants in Madagascar.

So far, no published data was available concerning the circulation of Bluetongue virus (BTV) in Madagascar. During a survey on Rift Valley Fever, we were able to detect a virus belonging to BTV. Therefore, we conducted a study aiming at characterizing molecularly the BTV isolated and assess the importance of circulation of BTV in Madagascar. A total of 4393 sera from ruminants selected randomly by stratification and sampled in 30 districts of Madagascar were tested for BTV. Moreover, 175 cattle were followed during 11 months. Phylogenetic analyses were performed from virus isolated from unfed

A possible overwintering mechanism for bluetongue virus in the absence of the insect vector.

Bluetongue virus (BTV) and several other Orbivirus species are transmitted between mammalian hosts via bites from adults of certain species of Culicoides midges. However, BTV can survive for 9-12 months (typically during the winter), in the absence of adult vectors, with no detectable cases of viraemia, disease or seroconversion in the host. The survival of the virus from one 'vector season' to the next is called 'overwintering' but the mechanism involved is not fully understood. It is demonstrated that BTV can persistently infect ovine gammadelta T-cells in vitro, a process that may also occu

Detection and serotyping of bluetongue viruses in Khyber Pakhtunkhwa Province, Pakistan.

This study investigated the presence of bluetongue virus (BTV) RNA and its serotypes in clinically healthy ruminants in Pakistan. A total of 673 blood samples were randomly collected from apparently healthy sheep, goats, buffaloes and cattle across various localities of the Khyber Pakhtunkhwa, Pakistan, between 2019 and 2021. BTV RNA detection was performed using pan-BTV reverse-transcriptase polymerase chain reaction (RT-PCR). Positive samples were further analyzed using serotype-specific real-time RT-PCR assays and nCounter NanoString technology. Overall, 118 samples scored positive for BTV

Bluetongue Virus in Cervids in Piedmont During the 2024-2025 Outbreak.

The mid-end of 2024 witnessed a significant outbreak of bluetongue virus (BTV) in the Piedmont region of Italy, affecting a wide range of species including domestic ruminants and wild cervids. From the beginning of August 2024 to the end of March 2025, 1081 samples were confirmed positive for BTV, of which 1066 were from routinely monitored domestic species (632 from sheep, 405 from cattle, 23 from goats, one from a camel, four from alpacas and one from yak across 687 outbreak sites), while 15 were collected from cervids. This report focuses on the identification of the 15 cases of BTV in red

Nanoparticle-based bio-barcode assay for the detection of bluetongue virus.

The ultrasensitive bio-barcode amplification assay (BCA) technique was developed for the specific detection of the outer-core protein VP7 of bluetongue virus (BTV). The target antigen VP7 was first captured by gold nanoparticles (GNPs) coated with polyclonal antibodies. Magnetic microparticles (MMP) coated with VP7 monoclonal antibody were then added to form a sandwich immuno-complex. After the immuno-complex was formed, signal DNA annealed to DNA strands covalently bound to the GNPs were released by heating and characterized by PCR and real-time fluorescence PCR. A detection limit of 0.1fg/ml

Spatio-temporal machine learning for multi-horizon prediction of bluetongue outbreaks.

Reliable early warning of infectious disease outbreaks remains a major challenge for surveillance systems, particularly for vector-borne pathogens whose transmission depends on interactions among hosts, vectors, and climate-sensitive environmental conditions. Data-driven forecasting offers a promising approach for predicting outbreak risk using surveillance and environmental data. This study develops a logit-weighted ensemble (LWE), a machine-learning framework that predicts outbreak occurrence 1-6 months ahead at the administrative unit-month scale using routinely available outbreak notificat

Limited adaptive mutations but deterministic genome formula variation in bluetongue virus.

Arthropod-borne viruses or arboviruses alternate between vertebrates and arthropods, facing distinct selection pressures in each host. Surprisingly, mechanisms facilitating host alternation remain unstudied. Here, we investigate two phenomena potentially facilitating host alternation in bluetongue virus (BTV): adaptive mutations and genome formula variation (GFV; i.e., differences in segment copy numbers). First, we searched for mutations indicative of host adaptation in cattle, sheep, and midges infected during an epizootic. No mutation was fixed in all samples of a given host. Thus, adaptive

Virological and pathological findings in Bluetongue virus serotype 8 infected sheep.

Twenty-seven sheep of the four most common Swiss breeds and the English breed Poll Dorset were experimentally infected with a northern European field strain of bluetongue virus serotype 8 (BTV-8). Animals of all breeds developed clinical signs, viremia and pathological lesions, demonstrating that BTV-8 is fully capable of replicating and inducing bluetongue disease (BT) in the investigated sheep. Necropsy performed between 10 and 16 days post-infectionem (d.p.i.) revealed BT-typical hemorrhages, effusions, edema, erosions and activation of lymphatic tissues. Hemorrhages on the base of the Arte

Experimental reproduction of bluetongue virus serotype 8 clinical disease in calves.

Cattle are commonly subclinically infected following natural or experimental infection with bluetongue virus (BTV). The introduction of BTV serotype 8 (BTV-8) in Europe has been characterized by the manifestation of clinical signs in infected cattle. In order to study the pathogenesis of BTV-8 in this host, an animal model able to reproduce the clinical manifestations of the disease is required. In this work, two calves were subcutaneously and intravenously injected with a low passage cell-adapted strain of BTV-8. Both calves showed typical bluetongue clinical signs, including pyrexia, ocular

Plasmid DNA-based reverse genetics as a platform for manufacturing of bluetongue vaccine.

Control of bluetongue disease is predominantly through vaccination with licensed inactivated or live-attenuated vaccines (LAVs). Manufacturing of LAVs in endemic countries requires formulation with a high number of serotypes for effective protection. Herein, we evaluated a plasmid DNA-based reverse genetics platform for manufacturing a multivalent vaccine. The synthetic vaccine was characterized by a common BTV1 backbone, with exchange of outer capsid proteins. Recombinant South African vaccine serotypes 1, 5, and 14 were rescued by exchanging the VP2 protein on the backbone. BTV6 rescue was a

Safety and efficacy of a Bluetongue inactivated vaccine (serotypes 1 and 4) in sheep.

A new inactivated vaccine against Bluetongue virus (BTV) serotypes 1 and 4, was developed from field isolates. Safety and efficacy of the vaccine were evaluated in sheep by serological monitoring and virus nucleic acid detection after experimental infection of vaccinated animals. Seroconversion was observed in vaccinated animals at day 14 post vaccination (pv) with neutralizing antibody titer of 1.9 and 1.8 for serotypes 1 and 4, respectively. The titer increase significantly after the booster reaching 2.7 and persist one year >1.5 for both serotypes. After challenge with virulent isolates, vi

From the bluetongue vaccination campaigns in sheep to overimmunization and ovine ASIA syndrome.

The use of vaccines has proven to be very effective in controlling and eradicating infectious diseases, both in veterinary and human medicine; however, vaccines can be also the source of an array of problems caused by procedures such as overimmunization. Bluetongue, an orbiviral disease that affects ruminants, is best controlled by the use of inactivated vaccines. During the last years of the past decade, these vaccines were applied all over Europe to control the spreading of the disease, a goal that was accomplished; however, at the same time, several adverse effects related to the vaccinatio
