Effect of bombesin on the development of the neonatal rabbit gastrointestinal tract.

The effect of bombesin on the postnatal development of the gastrointestinal tract was examined in New Zealand white rabbits. Bombesin (1.25, 12.5, 30 micrograms/kg body weight) or vehicle was administered intraperitoneally to suckling rabbits for 13 days starting on day 4 of life. The animals were killed at day 17. There was no significant effect of bombesin at doses of 1.25 or 12.5 micrograms/kg in any region of the gut studied. Bombesin administered at 30 micrograms/kg induced a widespread trophic effect in the gastrointestinal tract characterized by significant increases in the wet weight o

Serum gastrin responses to bombesin and food in patients with hypergastrinemia.

Serum gastrin responses to bombesin and to food were studied in patients with hypergastrinemia. Nine unoperated patients with hypergastrinemia of antral origin (seven achlorhydria by two idiopathic G-cell hyperfunction) had significantly higher serum gastrin responses to bombesin (P less than 0.001) and to food (P less than 0.001) than eight unoperated patients with Zollinger-Ellison syndrome. Three gastrectomized Zollinger-Ellison patients showed large serum gastrin responses to bombesin, and two of them had large postprandial increases in serum gastrin. Serum gastrin in a single patient with

Effects of pancreatic duct ligation on pancreatic response to bombesin.

To examine mechanisms that might be related to biliary pancreatitis, we examined the effects of pancreatic duct ligation (PDL) with pancreatic stimulation in vivo. PDL alone caused no increase in pancreatic levels of trypsinogen activation peptide (TAP), trypsin, or chymotrypsin and did not initiate pancreatitis. Although bombesin caused zymogen activation within the pancreas, the increases were slight and it did not cause pancreatitis. However, the combination of PDL with bombesin resulted in prominent increases in pancreatic TAP, trypsin, chymotrypsin, and the appearance of TAP in acinar cel

Endogenous amylin contributes to the anorectic effects of cholecystokinin and bombesin.

Previous studies indicated that amylin contributes to the anorectic effects of cholecystokinin (CCK) and bombesin (BBS), possibly by enhancing the release of pancreatic amylin or by modulating their anorectic actions within the central nervous system (CNS). To elucidate the interaction between amylin and CCK or BBS, respectively, we investigated the influence of an IP injection of CCK or BBS on feeding in amylin-deficient mice (IAPP(-/-)). The anorectic effects of CCK and BBS were nearly abolished in IAPP(-/-) mice compared to wildtype (WT) mice (e.g. 20 microg/kg CCK, 1-h food intake: WT/NaCl

In vitro response of rat gastrointestinal segments to cholecystokinin and bombesin.

In previous studies of the rat gastrointestinal (GI) tract, we have demonstrated specific binding of cholecystokinin (CCK) to the pylorus and of bombesin (BN) to the gastric fundus, gastric antrum, duodenum, and ileum. We now present the results of an investigation of the in vitro response of the same regions of the rat GI tract to CCK-8 (the active octapeptide of CCK) and BN. Sections of rat fundus, antrum, pylorus, duodenum, and ileum were suspended in a Tyrode buffer and attached to an isometric pressure transducer in a longitudinal orientation. Dose-response curves to CCK-8 and BN were gen

Effect of bombesin on lower esophageal sphincter pressure in humans.

Intravenous infusion of bombesin in humans results in increased serum gastrin levels and increased lower esophageal sphincter pressure. To differentiate a gastrin- from a nongastrin-mediated mechanism, the effect of intravenous bombesin infusion on lower esophageal sphincter pressure was studied in 5 healthy subjects and in 5 antrectomized patients. Lower esophageal sphincter pressure was recorded by three continuously perfused catheters with side-openings located 5 mm apart. Blood samples were taken at regular intervals during the study periods to assay gastrin and pancreatic polypeptide. Man

A novel bombesin-like peptide from skin of Rana shuchinae.

Bombesin and its receptors have been demonstrated to be involved in a larger array of physiological and pathophysiological conditions including memory and fear behavior, lung development and injury, and tumor growth. A bombesin-like peptide named bombesin-RS was purified and characterized from the skin secretions of the frog, Rana shuchinae. Its amino acid sequence (pETSFMAPSWALGHLM-NH2) was determined by auto Edman degradation and mass spectrometry. The cDNA encoding bombesin-RS precursor was cloned from skin cDNA library of this frog. The precursor is composed of 67 amino acid residues inclu

Intracerebroventricular infusion of bombesin modulates GIP secretion in conscious dogs.

UNLABELLED: Glucose-dependent insulinotropic polypeptide (GIP) is an incretin with important role in glucose homeostasis and energy conservation. Thus far, the neural mechanisms involved in the regulation of GIP secretion, have not yet been fully elucidated. The aim of this study was to evaluate a possible effect of intracerebroventricular administration of Bombesin in the regulation of GIP secretion. METHODS: Thirty-two adult dogs were used in this study. In group 1 the animals received a bolus icv infusion of 200 ng bombesin or an equivalent amount of artificial cerebrospinal fluid (aCSF).

Central neuromodulation of gastric acid secretion by bombesin-like peptides.

The amphibian skin tetrapeptide bombesin shows potent action in reducing gastric acid secretion by intracerebral ventricular (ICV) administration in rats. In order to establish a relationship between this action and the amino acid composition of the bombesin-like peptides, most of the natural bombesin-like peptides and some synthetic analogues were tested on their ability to reduce gastric acid secretion by ICV administration. The amphibian peptides bombesin, its [Tyr4]-bombesin analogue, alytesin, ranatensin and litorin, and the mammalian peptide GRP significantly reduced gastric acid output

Bombesin stimulates insulin secretion by a pancreatic islet cell line.

The amphibian tetradecapeptide, bombesin (BBS) has been shown to stimulate insulin secretion both in vivo and by pancreatic islet cells in vitro. To determine whether BBS can act directly on pancreatic beta cells, we examined its effects on insulin secretion by HIT-T15 cells (HIT cells), a clonal islet cell line. Addition of 100 nM BBS to HIT cells stimulated insulin release 25-fold within 30 sec. The rapid stimulatory effect of BBS on insulin release was short-lived: the secretory rate returned to basal levels after 90 min of BBS treatment. The decrease in the rate of insulin release in the c

Evaluation of radiolabelled bombesin analogues for receptor-targeted scintigraphy and radiotherapy.

The 14-aminoacid peptide bombesin (BN) has a high affinity for the gastrin-releasing peptide receptor which is expressed by a variety of tumours. Thus, radiometal-labelled DTPA-BN derivatives are potentially useful radioligands for receptor-targeted scintigraphy and radiotherapy of BN receptor-expressing tumours. A number of such DTPA-BN analogues, [DTPA-D-Tyr6]BN(6-13)NHEt (Et=ethyl), [DTPA-Tyr5,D-Phe6]BN(5-13)NHEt, [DTPA-D-Phe6,Leu13psiPhe14]BN(6-14), [DTPA-Tyr5,D-Phe6,Leu13psiPhe14]BN(5-14), [DTPA-Pro1,Tyr4]BN and [DTPA-Pro1,Tyr4,Nle14]BN, were synthesized and studied for their binding char

Caudal hindbrain participation in the suppression of feeding by central and peripheral bombesin.

We have previously demonstrated that bombesin (BBS) infused into the fourth cerebral ventricle in rats suppressed feeding at doses 10-1,000 times less than those required after lateral ventricular administration. These results suggest that centrally administered BBS acts in the caudal hindbrain to suppress food intake. To investigate the participation of caudal hindbrain structures in fourth-ventricular BBS-induced suppression of feeding, we made lesions that destroyed the following: 1) area postrema (AP) and 2) and medial nucleus of the solitary tract (NST). Additionally, we examined the role

Intrathecal injection of bombesin inhibits gastric acid secretion in the rat.

Bombesin (100-500 ng) injected intrathecally (T9-10) inhibited gastric acid secretion stimulated by pentagastrin and the GABAB agonist baclofen in urethane-anesthetized rats and basal gastric acid secretion in conscious, pylorus-ligated rats. Peptide action was dose-related, occurred within 30 min, and lasted for greater than 1 h. Bombesin-induced inhibition of pentagastrin-stimulated gastric acid secretion was not altered by cervical cord transection. Intravenous infusion of the monoclonal bombesin antibody 2A11 abolished intravenous bombesin (10 micrograms/kg.h)-induced 33% inhibition of gas

Neuromedin B receptor in esophagus: evidence for subtypes of bombesin receptors.

To identify receptors for bombesin-related peptides in the rat esophagus, we measured binding of 125I-Bolton-Hunter neuromedin B (125I-BH-neuromedin B) and 125I-[Tyr4]bombesin to tissue sections from the rat esophagus and compared the results with those for rat pancreas. Esophagus bound both tracers, whereas pancreas bound only 125I-[Tyr4]bombesin. In each tissue binding was saturable, dependent on pH, on time, and on temperature, reversible, and specific. Autoradiography demonstrated binding of both tracers only to the muscularis mucosae of the esophagus and binding of 125I-[Tyr4]bombesin dif

Effect of bombesin and gastrin-releasing peptide on canine sphincter of Oddi.

This study reports the effects of bombesin and gastrin-releasing peptide (GRP) on the canine sphincter of Oddi using a method that allows repeated cannulation of the biliary sphincter in the unanesthetized animal through a Thomas cannula placed opposite the biliary papilla. Immediately after intravenous administration of bombesin or GRP, phasic sphincter contractions disappeared, basal sphincter pressures fell, and common bile duct pressures rose. Because bombesin releases cholecystokinin (CCK) and CCK resulted in a similar pattern to that of bombesin, the bombesin effect on the sphincter of O

Localization of specific binding sites for bombesin in the canine gastrointestinal tract.

The goal of these studies was to determine the tissue and cell types possessing specific binding sites for bombesin/gastrin-releasing peptide in the canine gastrointestinal tract. Monoiodinated, biologically active (Tyr-4)-bombesin 14 (100 pM) was applied to sections of canine gut and localized using quantitative autoradiography. The highest density of bombesin/gastrin-releasing peptide specific binding sites occurred over endocrine cells in the antral mucosa. Specific binding sites were also found on the circular muscle layer of the gastric fundus, gastric antrum, and ileum, on longitudinal m

Bombesin receptor subtype 3 as a potential target for obesity and diabetes.

INTRODUCTION: Diabetes mellitus and obesity are important health issues; increasing in prevalence, both in the USA and globally. There are only limited pharmacological treatments, and although bariatric surgery is effective, new effective pharmacologic treatments would be of great value. This review covers one area of increasing interest that could yield new novel treatments of obesity/diabetes mellitus. It involves recognition of the central role the G-protein-coupled receptor, bombesin receptor subtype 3 (BRS-3) plays in energy/glucose metabolism. AREAS COVERED: Since the initial observatio
