Nanohydroxyapatite as the key factor in bone regeneration.

Bone tissue is a complex material with remarkable durability, elasticity and resistance to mechanical forces, playing a key role in the musculoskeletal system. Hydroxyapatite, a naturally occurring bone-like mineral, is used in a variety of medical applications due to its compatibility with bone tissue. However, synthetic hydroxyapatite has some limitations, such as fragility and low resistance to mechanical straining. Recent developments in nanotechnology have led to the invention of nanohydroxyapatite, which improves bone regeneration and integration with surrounding tissues. The aim of the

Stem Cells in Bone Repair and Regeneration.

Bones normally function to provide both mechanical and locomotion supports in the body. They are highly specialized connective tissues that are characterized by mineralized extracellular components, which provide both rigidity and strength to bones. Stem cells hold great potentials for both the repair and regeneration of different tissue types, including bone tissues. The future use of stem cell therapy is promising for developing regenerative medicine approaches to treat disorders and diseases in a wide range of tissues such as cartilages and bones. Data have been accumulated recently on the

Gene therapy approaches to regenerating bone.

Bone formation and regeneration therapies continue to require optimization and improvement because many skeletal disorders remain undertreated. Clinical solutions to nonunion fractures and osteoporotic vertebral compression fractures, for example, remain suboptimal and better therapeutic approaches must be created. The widespread use of recombinant human bone morphogenetic proteins (rhBMPs) for spine fusion was recently questioned by a series of reports in a special issue of The Spine Journal, which elucidated the side effects and complications of direct rhBMP treatments. Gene therapy - both d

Current understanding of osteoconduction in bone regeneration.

Bone tissue is osteoconductive. In particular, cancellous bone with its porous and highly interconnected trabecular architecture allows easy ingrowth of surrounding tissues. When placed in an osseous environment, living tissue for the host bed migrates into the cancellous structure, which results in new bone formation and incorporation of that structure. This is the process of osteoconduction. The mineral and collagenous components of bone are osteoconductive. Osteoconduction also is observed in fabricated materials that have porosity similar to that of bone structure. Corallin ceramics, hydro

Inflammation, mesenchymal stem cells and bone regeneration.

Achieving satisfactory reconstruction of bone remains an important goal in orthopedic and dental conditions such as bone trauma, osteoporosis, arthritis, osteonecrosis, and periodontitis. Appropriate temporal and spatial differentiation of mesenchymal stem cells (MSCs) is essential for postnatal bone regeneration. Additionally, an acute inflammatory response is crucial at the onset of bone repair, while an adaptive immune response has important implications during late bone remodeling. Various reports have indicated bidirectional interactions between MSCs and inflammatory cells or molecules. F

[Spontaneous bone regeneration following ceratocystic odontogenic tumor].

A patient presents with a keratocystic odontogenic tumour of the left maxillary sinus. In computed tomography scans, extensive pressure-induced osseous atrophy of the sinus walls is detected. Endoscopic cystectomy of the tumour was performed, with subsequent clinical follow-up. A second computed tomography scan revealed almost complete regeneration of the sinus walls. Where spontaneous regeneration of osseous structures is possible, restraint should be exercised when assessing indications for bony reconstruction during initial conservative surgery such as enucleation.

Bone regeneration after chemotherapy for vault lymphoma.

A 76-year-old woman presented with a mass in the left frontal region of the scalp. On admission, neurological examination found no deficits. CT revealed an osteolytic mass lesion in the left frontal cranial vault. She underwent open biopsy of the subcutaneous lesion. Histological examination identified cells with pleomorphic nuclei and marked nucleoli, and immunohistochemical staining showed these cells were positive for CD20, but negative for CD3. The histological diagnosis was diffuse large B-cell lymphoma. The patient received chemotherapy consisting of rituximab, cyclophosphamide, adriamyc

Proresolving nanomedicines activate bone regeneration in periodontitis.

Therapies to reverse tissue damage from osteolytic inflammatory diseases are limited by the inability of current tissue-engineering procedures to restore lost hard and soft tissues. There is a critical need for new therapeutics in regeneration. In addition to scaffolds, cells, and soluble mediators necessary for tissue engineering, control of endogenous inflammation is an absolute requirement for success. Although significant progress has been made in understanding natural resolution of inflammation pathways to limit uncontrolled inflammation in disease, harnessing the biomimetic properties of

Oxygen-releasing biomaterials for bone tissue regeneration.

Bone defects often exceed the body's innate self-healing capacity, inevitably disrupting local vascular networks and establishing a severe hypoxic microenvironment. This pathological hypoxia compromises cell survival, suppresses osteogenic differentiation, and ultimately dictates regenerative failure, representing a critical bottleneck in bone tissue engineering. Oxygen-releasing biomaterials (ORBs) have emerged as a transformative strategy to reprogram this hostile microenvironment into an oxygenous and pro-osteogenic niche. To advance this rapidly evolving field, this review provides a compr

Macrophage polarization-regulating hydrogels for bone regeneration.

Bone regeneration represents a complex and highly orchestrated process that involves not only osteogenic cells but also the immune system, particularly macrophages. Macrophage polarization plays a pivotal role in bone healing, and hydrogel-based immunomodulatory strategies offer promising approaches to guide this process. However, the design of such hydrogels must incorporate bone-specific requirements, such as mechanical support, biodegradation synchronized with bone remodeling, and spatiotemporal regulation of macrophage phenotypes. This review systematically examines advanced hydrogel syste

Gas-powered extracellular vesicles promote bone regeneration.

The signaling gas hydrogen sulfide (H2S) has recently been implicated in the regulation of bone remodeling and the maintenance of periodontal health. Exploring the underlying mechanisms for this regulation holds promise for the development of new treatment strategies to block bone resorption and stimulate bone regeneration. A recent study by Zhou et al. (Bioactive Materials, 2024) showed that treatment with H2S stimulated changes in the extracellular vesicles (EVs) released by M2 macrophages, enhancing their capacity to promote the osteogenic differentiation of mesenchymal stem cells in vitro.

Biomaterial-induced pathway modulation for bone regeneration.

Embryogenic developmental processes involve a tightly controlled regulation between mechanical forces and biochemical cues such as growth factors, matrix proteins, and cytokines. This interplay remains essential in the mature body, with aberrant pathway signaling leading to abnormalities such as atherosclerosis in the cardiovascular system, inflammation in tendon tissue, or osteoporosis in the bone. The aim of bone regenerative strategies is to develop tools and procedures that will harness the body's own self-repair ability in order to successfully regenerate even very large and complex bone

Guided bone regeneration using K-incision technique.

PURPOSE: The present study describes 3 patients with chronic periodontitis and consequent vertical resorption of the alveolar ridge who were treated using implant-based restoration with guided bone regeneration (GBR). METHODS: After extraction of a periodontally compromised tooth, vertical bone augmentation using a K-incision was performed at the healed, low-level alveolar ridge. RESULTS: The partial-split K-incision enabled soft tissue elongation without any change in buccal vestibular depth, and provided sufficient keratinized gingival tissue during GBR. CONCLUSIONS: Within the limits of

Bone morphogenetic proteins for articular cartilage regeneration.

Degeneration of articular cartilage (AC) tissue is the most common cause of osteoarthritis (OA) and rheumatoid arthritis. Bone morphogenetic proteins (BMPs) play important roles in bone and cartilage formation. This article reviews the experimental and clinical applications of BMPs in cartilage regeneration. Experimental evidence indicates that BMPs play an important role in protection against cartilage damage caused by inflammation or trauma, by binding to different receptor combinations and, consequently, activating different intracellular signaling pathways. Loss of function of BMP-related

Bone regeneration: current concepts and future directions.

Bone regeneration is a complex, well-orchestrated physiological process of bone formation, which can be seen during normal fracture healing, and is involved in continuous remodelling throughout adult life. However, there are complex clinical conditions in which bone regeneration is required in large quantity, such as for skeletal reconstruction of large bone defects created by trauma, infection, tumour resection and skeletal abnormalities, or cases in which the regenerative process is compromised, including avascular necrosis, atrophic non-unions and osteoporosis. Currently, there is a plethor

Bone marrow stem cells and liver regeneration.

Development of new approaches to treat patients with hepatic diseases that can eliminate the need for liver transplantation is imperative. Use of cell therapy as a means of repopulating the liver has several advantages over whole-organ transplantation because it would be less invasive, less immunogenic, and would allow the use, in some instances, of autologous-derived cells. Stem/progenitor cells that would be ideal for liver repopulation would need to have characteristics such as availability and ease of isolation, the ability to be expanded in vitro, ensuring adequate numbers of cells, susce

Osteotropic beta-cyclodextrin for local bone regeneration.

An osteotropic alendronate-beta-cyclodextrin conjugate (ALN-beta-CD) was developed as a bone-targeting delivery system for improved treatment of skeletal diseases. The conjugate shows very strong binding to hydroxyapatite (HA, main component of the skeleton). Its ability in forming molecular inclusion complex with prostaglandin E(1) (PGE(1), a potent bone anabolic agent) was confirmed by phase solubility experiments and differential scanning calorimetry (DSC). In a bilateral rat mandible model, ALN-beta-CD/PGE(1) molecular complex was shown to stimulate strong local bone anabolic reaction. In
