Sanitization of Oak Barrels for Wine-A Review.

Oak barrels form an integral part of wine production, especially that of high-quality wines where they are implemented as fermentation and aging vessels. Insufficient cleaning and sanitization of barrels can result in microbial spoilage which may have a detrimental impact on wine quality. To date, no review has been published on the various sanitization methods for wine barrels. The objective of this review is to provide an overview of the sanitization methods used in wineries from conventional techniques like the use of sulfur dioxide and steam to alternative and new approaches using ozone an

Emerging Non-Thermal Technologies as Alternative to SO2 for the Production of Wine.

SO2 is an antioxidant and selective antimicrobial additive, inhibiting the growth of molds in the must during the early stages of wine production, as well as undesirable bacteria and yeasts during fermentation, thus avoiding microbial spoilage during wine production and storage. The addition of SO2 is regulated to a maximum of 150-350 ppm, as this chemical preservative can cause adverse effects in consumers such as allergic reactions. Therefore, the wine industry is interested in finding alternative strategies to reduce SO2 levels, while maintaining wine quality. The use of non-thermal or cold

Changes in microbial communities during the spoilage of palm wine stored at room temperature.

This study was to determine the spoilage microbiota of palm wine stored at room temperature in order to define appropriate strategies for shelf-life extension. For this purpose, during the storage of palm wine samples, yeast, lactic acid bacteria (LAB), and acetic acid bacteria (AAB) counts were performed. Then, species were identified by PCR-RFLP followed by sequencing and the overall bacterial community was determined by NGS (Next Generation Sequencing). The results showed that yeast, LAB and AAB counts decreased in the palm wines collected from producers after 24-48 h, while they remained c

Does Fungal Chitosan Leave Noticeable Traces in Treated Wines?

Background (1): The use of fungal chitosan as an antiseptic in wine appears as a promising alternative to sulfur dioxide for the elimination of Brettanomyces bruxellensis sensitive strains. Nevertheless, its utilization raises the question, "how are the treated wines different from the untreated ones?" Methods (2): Chitosan treatment residues were sought in the oligosaccharide and polysaccharide fractions and among 224 low MW ions (<1800 g·mol-1) in several wines by using liquid chromatography (size exclusion HPLC or LC-MS) and GC-MS. Standard oenological parameters were also examined as well

Microbial communities of artisanal ciders in Slovenia and their role in fermentation.

Traditional cider production relies on spontaneous fermentation driven by complex microbial communities. In this study, we investigated the microbial communities, chemical compositions, and functional interactions underlying artisanal cider production in Carinthia, a subalpine region of northern Slovenia. Samples of spontaneously fermented ciders and swabs from wooden cellar equipment were collected from 11 remote farms using traditional practices to identify potential microbial reservoirs. Yeasts and bacteria were isolated, enumerated, dereplicated, and identified using sequencing of barcodin

Zygosaccharomyces bailii: A Promising Probiotic Candidate for Functional Food Development.

The growing interest in functional foods is accompanied by the need to select new probiotic microorganisms. Artisanal fermented foods serve as excellent sources for identifying probiotics, particularly yeasts. In this work, two strains isolated from kombucha were identified: Zygosaccharomyces bailii (UVI60) and Z. lentus (UVI61). Strain UVI61 did not grow at 37 °C, so the evaluation of probiotic properties was carried out with strain UVI60. Z. bailii UVI60 showed excellent survival (98%) under adverse conditions involving synthetic gastric and duodenal juices. The self-aggregation capacity (97

Kombucha SCOBY as a Fermentation-Derived Biofilm Matrix: Species-Resolved Microbial Communities and Multidimensional In Vitro Bioactivities.

Kombucha fermentation is driven by a Symbiotic Culture of Bacteria and Yeast (SCOBY), a cellulose-rich biofilm that hosts a complex microbial consortium. While most kombucha studies focus on the liquid beverage, the SCOBY pellicle itself remains underexplored, particularly with respect to species-level microbial resolution and its intrinsic biological activities. In this study, a commercial kombucha SCOBY was characterized using full-length 16S rRNA gene and ITS amplicon sequencing based on Oxford Nanopore Technology, enabling species-level taxonomic resolution. In parallel, hydroalcoholic and

The impact of beer-spoiling yeast on flavour development in non-alcoholic beers.

No- and low-alcohol beverages reflect a commercially important and rapidly expanding market. These products represent a unique environment that differs nutritionally and environmentally from standard product types and are generally regarded as being at higher risk of spoilage. Beer-spoiling yeasts represent a particular risk within the brewing chain, typically impacting the sensorial properties of the final beverage. However, little is known about their effects in no- and low-alcohol products. This study aimed to determine the impacts of beer-spoiling yeasts on the flavour profiles of non-alco

Yeast Biosensors for the Safety of Fermented Beverages.

Yeast biosensors represent a promising biotechnological innovation for ensuring the safety and quality of fermented beverages such as beer, wine, and kombucha. These biosensors employ genetically engineered yeast strains to detect specific contaminants, spoilage organisms, or hazardous compounds during fermentation or the final product. By integrating synthetic biology tools, researchers have developed yeast strains that can sense and respond to the presence of heavy metals (e.g., lead or arsenic), mycotoxins, ethanol levels, or unwanted microbial metabolites. When a target compound is detecte

Bioprotection with Saccharomyces cerevisiae: A Promising Strategy.

Bioprotection in winemaking refers to the use of naturally occurring microorganisms-mainly non-Saccharomyces yeasts-to inhibit the growth of spoilage microbes and reduce the need for chemical preservatives like sulfur dioxide (SO2). Numerous studies have demonstrated the benefits of non-Saccharomyces as bioprotectants. However, the use of Saccharomyces cerevisiae as a bioprotectant has been studied very little. Furthermore, it can offer many advantages for the production of sulfite-free wines. To test if S. cerevisiae could be used in bioprotection, we compared the ability of different strains

Beer ethanol and iso-α-acid level affect microbial community establishment and beer chemistry throughout wood maturation of beer.

Sour beers produced by barrel-aging of conventionally fermented beers are becoming increasingly popular. However, as the intricate interactions between the wood, the microbes and the beer are still unclear, wood maturation often leads to inconsistent end products with undesired sensory properties. Previous research on industrial barrel-aging of beer suggests that beer parameters like the ethanol content and bitterness play an important role in the microbial community composition and beer chemistry, but their exact impact still remains to be investigated. In this study, an experimentally tracta

Unraveling yeast diversity in food fermentation using ITS1-2 amplicon-based metabarcoding.

A detailed characterization of the microbial ecosystem involved in the production processes of fermented foods is essential. Although fermented foods are an important part of the human diet and have seen an increasing interest nowadays, some challenges still need to be solved. Specifically, yeast identification through culture-independent methodologies is still limited to the genus level. Unlike bacterial species identifications, long-read sequencing technologies have barely been used for yeast species identification, and, to the best of the authors' knowledge, it has not been validated with m

Metagenomic, organoleptic profiling, and nutritional properties of fermented kombucha tea substituted with recycled substrates.

Kombucha fermentation yields a diverse range of beneficial macro and micronutrients. In our study, we examined the metabolites, antioxidant activity, organoleptic characteristics, and nutritional attributes of traditionally prepared kombucha tea, using black tea and sugar (control) as substrates, and compared them with tea made from tea dust and blackstrap molasses (test). Kombucha tea crafted from functional raw materials exhibited enhanced sensory qualities and improved health-promoting properties. The levels of tannins, flavonoids, and phenols play a crucial role in determining the antioxid
