Improvement of spinal muscular atrophy via correction of the SMN2 splicing defect by Brucea javanica (L.) Merr. extract and Bruceine D.

BACKGROUND: Spinal muscular atrophy (SMA) is a rare neuromuscular disease and a leading genetic cause of infant mortality. SMA is caused primarily by the deletion of the survival motor neuron 1 (SMN1) gene, which leaves the duplicate gene SMN2 as the sole source of SMN protein. The splicing defect (exon 7 skipping) of SMN2 leads to an insufficient amount of SMN protein. Therefore, correcting this SMN2 splicing defect is considered to be a promising approach for the treatment of SMA. PURPOSE: This study aimed to identify active compounds and extracts from plant resources to rescue SMA phenotyp

Brusatol induced ferroptosis in osteosarcoma cells by modulating the Keap1/Nrf2/SLC7A11 signaling pathway.

Osteosarcoma (OS) is a rare primary malignant bone tumor. Despite ongoing research efforts, improvements in the five-year survival rate of OS patients remain limited. Therefore, there is an urgent need to identify novel therapeutic agents to improve OS prognosis. Brusatol (Bru), a quassinoid extracted from the seeds of Brucea javanica and Brucea sumatrana, has been shown to possess the potential to inhibit tumor metastasis and proliferation. In recent years, increasing evidence has highlighted the critical role of ferroptosis-a distinct form of regulated cell death-in cancer therapy. However,

Bruceine A Ameliorates Osteoporosis Through Suppression of RACK1-Mediated Osteoclastogenesis.

Bruceine A (BA) is a quassinoid compound derived from Brucea javanica with various pharmacological activities. Our previous work has revealed that BA exhibits anti-inflammatory and glucose-lowering effects in db/db mice, resulting in significant protection against diabetic kidney disease. However, considering the pivotal role of inflammatory signaling pathways during osteoclast differentiation, whether and how BA exerts protection against osteoporosis remains to be explored. In the present work, we have demonstrated that BA exhibits significant protective efficacy against bone loss in ovariect

Bruceine A induces cell growth inhibition and apoptosis through PFKFB4/GSK3β signaling in pancreatic cancer.

Pancreatic cancer is one of the most aggressive cancers with a poor prognosis and 5-year low survival rate. In the present study, we report that bruceine A, a quassinoid found in Brucea javanica (L.) Merr. has a strong antitumor activity against human pancreatic cancer cells both in vitro and in vivo. Human proteome microarray reveals that 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 4 (PFKFB4) is the candidate target of bruceine A and both fluorescence measurement and microscale thermophoresis suggest bruceine A binds to PFKFB4. Bruceine A suppresses glycolysis by inhibiting PFKFB4, l

Oleic acid inhibits the proliferation, invasion, and migration of tongue squamous cell carcinoma cells under high glucose conditions through S100A9-HK2/PKM2-SOD2 signaling pathway.

The incidence of tongue squamous cell carcinoma (TSCC) has been increasing annually. Type 2 diabetes mellitus (T2DM) is one of the risk factors for TSCC, with hyperglycemia being a hallmark of T2DM that promotes the development of oral cancer and results in poorer prognoses for TSCC patients who also have T2DM. Oleic acid (OA), a component of Brucea javanica oil emulsion, exhibits both anti-cancer and anti-diabetic properties. However, the precise regulatory mechanisms of OA on tongue squamous cell carcinoma (TSCC) under high glucose conditions have yet to be fully elucidated. Our findings dem
