The use of biofilters to improve indoor air quality: the removal of toluene, TCE, and formaldehyde.

A biofilter composed of a scrubber, a hydroponic planting system, and an aquatic system with green plants as a base maintained air quality within part of a modern office building. The scrubber was composed of five parallel fiberglass modules with external faces of porous lava rock. The face, largely covered with mosses, was wetted by recirculating water. Air was drawn through the scrubber and the immediately adjacent hydroponic region by a dedicated air handling system. The system was challenged for 4 weeks with three common indoor organic pollutants and removed significant amounts of all comp

Functional characterisation of Target of Rapamycin (TOR) signalling in Physcomitrella.

The evolutionary conserved TOR kinase positively controls growth and development of the moss Physcomitrella, development and function of its chloroplasts, its protein synthesis, and cell-cycle progression. Target of Rapamycin (TOR) is a conserved protein kinase and an important signalling hub in eukaryotes. The moss Physcomitrella (Physcomitrium patens) is a model organism for plant physiology, development, and evolution. However, little is known about TOR signalling in non-vascular plants, including Physcomitrella. Here, we report the effects of inhibiting TOR signalling in Physcomitrella. We

Essential mitochondrial activity in Physcomitrium patens relies on complementary cytochrome and alternative oxidase pathways.

Mitochondrial respiration catalyses the transfer of electrons from NADH to oxygen through the activity of five multiprotein complexes. In plants, mitochondria also possess additional alternative electron transport pathways, including the alternative oxidase (AOX) pathway, which transfers electrons from ubiquinol to O2, bypassing the cytochrome-dependent electron transport chain. This study investigates the functional role of AOX during plant evolution by analysing Physcomitrium patens plants that lack or overexpress AOX. In the moss P. patens, AOX exhibits a remarkably high electron transport

Effects of copper accumulation on growth and development of Scopelophila cataractae grown in vitro.

Scopelophila cataractae was cultured in vitro for 16 weeks to assess the contrasting effects of Cu on growth and reproduction, as well as gametophore stage. To induce buds and gametophores of S. cataractae, ten treatments (tr 1 to tr 10) of culture media were prepared using a combination of mineral salts, sugar, vitamin B complex, CuSO4, and exogenous hormones. Highest numbers of gametophores and buds were formed in media containing 500 µM CuSO4 in co-application with auxin and cytokinin, as shown in the modest Cu treatments (tr 6 and tr 7, 26 per cushion and 255 per 25 mm2, respectively). A 5

Agarose/κ-carrageenan-based hydrogel film enriched with natural plant extracts for the treatment of cutaneous wounds.

Hydrogels for complex and chronic wound dressings must be conformable, absorb and retain wound exudates and maintain hydration. They can incorporate and release bioactive molecules that can accelerate the healing process. Wound dressings have to be in contact with the wound and epidermis, even for long periods, without causing adverse effects. Hydrogel dressing formulations based on biopolymers derived from terrestrial or marine flora can be relatively inexpensive and well tolerated. In the present article hydrogel films composed by agarose (1.0 wt%), κ-carrageenan at three different concentra

First contact with greater gravity: Moss plants adapted via enhanced photosynthesis mediated by AP2/ERF transcription factors.

The emergence of land plants required adaptations to altered water availability and increased effective gravity. Bryophytes underwent major changes in physiology, anatomy, and growth during their emergence onto land. However, the link between gravity, photosynthesis, and genetic control remains unclear. Here, we show that leaf carbon dioxide diffusion enhanced photosynthesis in the model moss Physcomitrium patens under increased gravity (6 and 10 times Earth's gravity), driven by increases in plant (gametophore) number and chloroplast size. RNA sequencing analysis showed that 10 gravity up-reg

Moss-produced human complement factor H with modified glycans has an extended half-life and improved biological activity.

Most drugs that target the complement system are designed to inhibit the complement pathway at either the proximal or terminal levels. The use of a natural complement regulator such as factor H (FH) could provide a superior treatment option by restoring the balance of an overactive complement system while preserving its normal physiological functions. Until now, the systemic treatment of complement-associated disorders with FH has been deemed unfeasible, primarily due to high production costs, risks related to FH purified from donors' blood, and the challenging expression of recombinant FH in

Physcomitrella patens DNA methyltransferase 2 is required for recovery from salt and osmotic stress.

DNA methyltransferase 2 (DNMT2) unlike other members of the cytosine DNA methyltransferase gene family has dual substrate specificity and it methylates cytosines in both the DNA and transfer RNA (tRNA). Its role in plants, however, has remained obscure to date. In this study, we demonstrate that DNMT2 from Physcomitrella patens accumulates in a temporal manner under salt and osmotic stress showing maximum accumulation during recovery, i.e. 24 h after plants are transferred to normal growth medium. Therefore, to study its role in stress tolerance, we generated PpDNMT2 targeted knockout plants (

Thallium in terrestrial environments--occurrence and effects.

In this investigation thallium contents in soil and plants in the EuroRegion Neisse were analysed and the distribution determined. The median top-soil content is 0.5 mg/kg in the area investigated. In this low-contaminated area the moss Pleurozium schreberi contains 0.04-0.13 microg/g and the moss Polytrichumformosum between 0.01 and 0.05 microg/g Tl. The effects of thallium on man and the terrestrial environment were examined. In an epidemiological study a significant positive correlation was found between the thallium content of the two mosses and the incidence of diseases of the circulatory

Calcium gradient in plant cells with polarized growth in simulated microgravity.

Plant cells characterized by apical growth, for example, root hairs and apical cells of moss protonema, are a convenient model to address the problem of gravity response mechanisms including initiation of cell polarity. The fluorescent calcium probe, chlorotetracycline, allowed us to display the calcium distribution gradient in these cells. Irradiation by red light led to a sharp decrease in the Ca2+ ion activity in cells. During clinostatting in darkness the pattern of calcium influx and distribution changes inconsiderably as compared with control; in root hairs calcium is detected mainly in

Flavodiiron Proteins Increase Plant Biomass Under Fluctuating Light and Enhance Chilling Tolerance in Tobacco.

The introduction of flavodiiron (FLV) proteins in angiosperms can modulate photosynthesis and photoprotection under fluctuating light conditions. However, the potential of FLV in crop improvement and chilling tolerance remains poorly understood. To address this, transgenic tobacco plants expressing the FLVA and FLVB genes from Physcomitrium patens were used to examine the influence of FLV on plant biomass and chilling tolerance. The results indicated that the steady-state CO₂ assimilation rate (AN) was not altered by the introduction of FLV. However, during shade-sun transitions, FLV lines exh

Lichen secondary metabolites affect growth of Physcomitrella patens by allelopathy.

Lichen secondary metabolites can function as allelochemicals and affect the development and growth of neighboring bryophytes, fungi, vascular plants, microorganisms, and even other lichens. Lichen overgrowth on bryophytes is frequently observed in nature even though mosses grow faster than lichens, but there is still little information on the interactions between lichens and bryophytes. In the present study, we used extracts from six lichen thalli containing secondary metabolites like usnic acid, protocetraric acid, atranorin, lecanoric acid, nortistic acid, and thamnolic acid. Moss-derived human complement factor H modulates retinal immune response and attenuates retinal degeneration.

BACKGROUND: AMD is a multifactorial progressive disease of the central retina that leads to severe vision loss among the elderly. Genome-wide association studies in AMD patients and preclinical data have identified a dysregulated complement system and aberrant microglia responses in the pathogenesis of AMD. Specifically, a genetic variant in the complement factor H (CFH) gene, an important inhibitor of the alternative complement pathway, confers the strongest risk for AMD. Here, we investigated whether moss-derived recombinant human CFH proteins, termed CPV-101 and CPV-104, can modulate microg

Formaldehyde detoxification by the moss Racomitrium japonicum.

As a pervasive indoor air pollutant, formaldehyde (FA) poses significant environmental and health risks. This study investigates the molecular and physiological mechanisms underlying the FA remediation potential of the moss Racomitrium japonicum. The moss employs a dual defense strategy: activation of antioxidant systems (SOD/POD/CAT enzymes and AsA-GSH cycle) under moderate FA stress, and metabolic detoxification via upregulation of key genes (FDH, formate dehydrogenase, and MS, malate synthase) that channel FA into the Calvin cycle and glyoxylate pathway. High FA concentrations induced oxida

Autophagy is required for gamete differentiation in the moss Physcomitrella patens.

Autophagy, a major catabolic process in eukaryotes, was initially related to cell tolerance to nutrient depletion. In plants autophagy has also been widely related to tolerance to biotic and abiotic stresses (through the induction or repression of programmed cell death, PCD) as well as to promotion of developmentally regulated PCD, starch degradation or caloric restriction important for life span. Much less is known regarding its role in plant cell differentiation. Here we show that macroautophagy, the autophagy pathway driven by engulfment of cytoplasmic components by autophagosomes and its s

Microfilament distribution in protonemata of the moss Ceratodon.

Microfilaments were visualized in dark-grown protonemata of the moss Ceratodon to assess their possible role in tip growth and gravitropism. The relative effectiveness of rhodamine phalloidin (with or without m-maleimidobenzoyl-N-hydroxysuccinimide ester (MBS)) and of immunofluorescence (using the C4 antibody) was evaluated for actin localization in the same cell type. Using immunofluorescence, microfilaments were primarily in an axial orientation within the apical cell. However, a more complex network of microfilaments was observed using rhodamine phalloidin after MBS pretreatment, especially

Railroad derived nitrogen and heavy metal pollution does not affect nitrogen fixation associated with mosses and lichens at a tundra site in Northern Sweden.

Traffic derived nitrogen (N) and heavy metal pollution is a well-known phenomenon, but little explored in otherwise pristine ecosystems such as subarctic tundra. Here, the main source of N input to the ecosystem is via N2 fixation by moss- and lichen-associated bacteria. While inhibitory effects of N deposition on moss-associated N2 fixation have been reported, we still lack understanding of the effects of traffic derived N and heavy metal deposition on this ecosystem function in an otherwise pristine setting. To test this, we established a distance gradient (0-1280 m) away from a metal pollut

Moss-derived recombinant Factor H, CPV-104, effectively antagonizes alternative pathway C3/C5 convertases stabilization by NeFs from patients with primary C3 glomerulopathy.

BACKGROUND: C3 glomerulopathy (C3G) is a rare kidney disease caused by uncontrolled activation of the complement alternative pathway (AP), frequently driven by nephritic factors (NeFs) that stabilize the AP C3 and C5 convertases. NeFs are detected in up to 80% of patients and exhibit heterogeneous stabilizing activity, contributing to variable degrees of complement dysregulation. Current complement-targeted therapies reduce complement activation but may compromise immune surveillance by blocking key components of the pathway. Supplementation with the physiological AP regulator factor H (FH) re

Involvement of microRNA in copper deficiency-induced repression of chloroplastic CuZn-superoxide dismutase genes in the moss Physcomitrella patens.

Superoxide dismutases (SODs) are metallo-enzymes that catalyze the dismutation of superoxide radicals. In Arabidopsis thaliana, the expression of CuZn-SOD in both the chloroplast and cytosol was reported to be down-regulated by microRNA398 (miR398) during growth on low copper. The moss Physcomitrella patens contains chloroplastic and cytosolic CuZn-SOD genes, but lacks miR398. From analysis of P. patens microRNA, miR1073 was predicted to target CuZn-SOD mRNAs. We noticed that two chloroplastic CuZn-SOD genes contain the miR1073 target sequence in the 3' untranslated region; however, the cytoso
