Impact of in vitro fertilization by refrigerated versus frozen buffalo semen on developmental competence of buffalo embryos.

The objective of this study was to evaluate the fertility of buffalo semen for in vitro embryo production (IVEP) by comparing the effectiveness of refrigerated versus frozen semen. Three OPU sessions were held at 30-day intervals. For oocyte fertilization three buffalo bulls were used, one per session. At each OPU-IVEP session, one ejaculate was collected and divided into two equal aliquots. Each aliquot was either refrigerated at 5ºC/24 hours or frozen. A TRIS extender containing 10% low density lipoproteins, 0.5% lecithin and 10 mM acetylcysteine was used adding 7% glycerol for freezing. Spe

Fixed-time artificial insemination technology in buffaloes: a review.

Buffalo occupies a leading position as a major livestock commodity and is the primary milk-producing animal in many countries like Italy, China, India, Pakistan, Bangladesh, and Nepal. Buffalo farming emphasizes the significance of effective reproductive strategies. Among effective reproductive strategy, artificial insemination has a significant influence on herd's genetic progress. Nonetheless, buffaloes exhibit unique reproductive behavior, which complicates the insemination process. These animals demonstrate inconsistent periods (ranging from 6-48 h) of mounting acceptance. Therefore, timed

Accidental monensin poisoning in buffaloes in Minas Gerais, Brazil.

The manuscript reports accidental water buffalo (Bubalus bubalis) poisoning by monensin, a food supplement commonly used to feed cattle, and indicate some relevant aspects related to buffalo breeding. Most of the buffalo intoxication reports are accidental and occur in areas where buffalo breeding is not a traditional activity or in new buffalo breed without expert assistance. The present communication reports accidental buffalo poisoning due to monensin consumption at one corral that usually breed bovine to slaughter, a situation not reported before in Brazil. Monensin poisoning hypothesis wa

Leucoderma in Buffaloes (Bubalus bubalis) in the Amazon Biome.

Leucoderma is a condition that affects the skin and hair of animals, causing depigmentation and acromotrichia. In buffaloes, this condition results in significant economic losses for the production chain due to its impact on the leather trade. This study aimed to investigate the epidemiological and clinicopathological aspects of leucoderma in buffaloes in the Amazon biome and describe the prophylactic treatment to control the disease. The study included 40 buffaloes, 16 males and 24 females, aged between 1 and 10 years, and were of the Murrah, Jafarabadi, Mediterranean, and Murrah × Mediterran

Leptospira interrogans serogroup Pomona strains isolated from river buffaloes.

At present, little is known regarding the prevalence of buffalo leptospirosis worldwide, especially with respect to which Leptospira strains may infect this animal species. Furthermore, most investigations into this disease in buffaloes have only been performed with serological studies. In Brazil, particularly in the Amazon, buffalo production is growing and is just as important as cattle production, although few studies have been performed on buffalo compared to cattle. Thus, the aim of this study was to isolate and characterise Leptospira strains from river buffaloes raised in the Brazilian

Selection strategies for dairy buffaloes: economic and genetic consequences.

Buffaloes are generally raised in Brazil without milk-recording programs, and thus without genetic evaluations of any of their traits. This study evaluated the economic impacts of three different selection strategies on buffalo populations and the evolution of genetic trends, genetic variances and inbreeding coefficients resulting from each of them. The selection strategies used were as follows: (i) random selection; (ii) phenotypic selection; and (iii) progeny testing (PT). As the numbers of herds enrolled in milk-recording programs increased, phenotypic selection and PT strategies increased

Prevalence of Sarcocystis spp. in water buffaloes in Vietnam.

Muscles from heart, tongue, oesophagus, neck and abdomen from 502 adult water buffaloes (Bubalus bubalis) slaughtered in Ho Chi Minh City, Vietnam, between 1996 and 1997 were examined for Sarcocystis cysts by a combination of ocular and histological examination. Sarcocysts were present in 396 (79%) of the animals and the prevalence increased with age from a 57% infection rate among 2-3 year old animals to 93% among 6-7 year olds. The prevalence was higher in animals originating from the northern part (89%) than in those from the southern part (69%) of the country. Four species of Sarcocystis w

Hereditary suprabasilar acantholytic mechanobullous dermatosis in buffaloes (Bubalus bubalis).

A skin disease characterized by trauma-induced sloughing of haired skin, hooves, and horns is described in four calves from a herd of Murrah buffaloes (Bubalus bubalis) in Brazil. Affected calves were detected shortly after birth by the presence of lesions affecting the distal extremities, the scapular and gluteal regions, and the tip of the tail. On histologic evaluation of affected skin, the lesions were characterized by suprabasilar vesicles and acantholysis affecting the epidermis and outer root sheath of the hair follicle infundibulum. The basal cell layer was intact and appeared as a sin

Early pregnancy diagnosis in buffaloes from plasma progesterone concentration.

Blood samples were collected from 84 buffalo cows 21 days after fixed time artificial insemination following oestrus synchronisation and cloprostenol. Progesterone concentration in plasma was determined by radioimmunoassay. The animals were examined for pregnancy by rectal palpation 60 to 90 days after insemination. Forty-two animals were predicted pregnant on the basis of progesterone concentration (more than 1.0 ng per ml), and 28 (66.7 per cent) of them were subsequently confirmed pregnant by rectal palpation. Thirty-five animals were predicted non-pregnant (progesterone less than 0.7 ng pe

Chronological profiling of early pregnancy transcripts in Murrah buffaloes.

Effective reproductive management in cattle, such as cows and buffaloes, requires early and accurate pregnancy detection. Early identification of pregnancy enables farmers to promptly identify non-pregnant animals for treatment and/or rebreeding, thereby reducing the calving interval. This study aimed to standardize the expression of the CCL8 and CXCL10 genes as markers for early pregnancy detection in Murrah buffaloes. Blood samples were collected on the 16th day post-artificial insemination for gene expression analysis and on days zero, seven, 14, and 21 post-artificial insemination for prog

Primary experimental infection of riverine buffaloes with Fasciola gigantica.

The clinical course of the primary experimental Fasciola gigantica infection was investigated in riverine buffalo calves of the Murrah breed. Nine male calves aged 12-15 months were randomly assigned to two groups of five (Group I) and four (Group II) animals. Each animal in Group I, was orally infected with 1000 metacercariae (mc) of F. gigantica, whereas Group II animals did not receive any infection dose and served as uninfected controls. No clinical signs of fasciolosis were observed until the sixth week post-infection (PI). Group I animals, however, developed recognised symptoms of acute

Cystic echinococcosis in water buffaloes: epidemiological survey and molecular evidence of ovine (G1) and buffalo (G3) strains.

A survey of cystic echinococcosis (CE) in the water buffalo (Bubalus bubalis) of the Italian Mediterranean breed was carried out in Campania, a region of southern Italy. In addition, a molecular study was performed on 48 hydatid cysts coming from 48 water buffaloes in order to determine the Echinococcus granulosus strain(s) present in this host. Out of a total of 722 water buffaloes examined for CE, 76 (10.5%) were found infected. The average number of cysts per buffalo was 4.3 (minimum 1, maximum 45). Seventeen buffaloes had hydatid cysts only in the liver (with an average of 5 cysts/liver),

Detection and distribution of Stephanofilaria sp. in buffalo flies and buffalo fly lesions in north Australian beef cattle.

Buffalo flies (Haematobia irritans exigua) are ectoparasites of major animal health and production concern in north Australian beef herds. Skin lesions associated with buffalo fly infestation, cause hide damage and welfare issues and are manifested as dermatitis or ulcerated areas found most commonly near the medial canthus of the eye, along the lateral and ventral neck and on the abdomen. Buffalo flies can transmit a nematode, Stephanofilaria sp., which has been considered the main aetiological agent for buffalo fly lesions, but the role of nematodes in the development of the lesions has not

Successful cloning of a superior buffalo bull.

Somatic cell nuclear transfer (SCNT) technology provides an opportunity to multiply superior animals that could speed up dissemination of favorable genes into the population. In the present study, we attempted to reproduce a superior breeding bull of Murrah buffalo, the best dairy breed of buffalo, using donor cells that were established from tail-skin biopsy and seminal plasma. We studied several parameters such as cell cycle stages, histone modifications (H3K9ac and H3K27me3) and expression of developmental genes in donor cells to determine their SCNT reprogramming potentials. We successfull

A review of hemorrhagic septicemia in cattle and buffalo.

Hemorrhagic septicemia (HS), an acute, fatal and septicemic disease of cattle and buffaloes caused by Pasteurella multocida, is important in tropical regions of the world, especially in African and Asian countries. The prevalence of disease has been well documented with predominant isolation of P. multocida serotypes B:2 and E:2. Conventional methods of identification such as serotyping, biotyping, antibiogram determination and pathogenicity as well as molecular methods (P. multocida-specific polymerase chain reaction (PCR), a serogroup B-specific PCR assay, multiplex capsular typing system an
