Bulinus species on Madagascar: molecular evolution, genetic markers and compatibility with Schistosoma haematobium.

Of the four species of Bulinus found on Madagascar, three species: B. obtusispira, B. liratus and B. bavayi are endemic while the fourth, B. forskalii, is probably a recent introduction from the African mainland. The evolutionary relationships of these species with Bulinus species from Africa were studied by phylogenetic analysis of DNA sequence variation at two mitochondrial loci: cytochrome oxidase subunit I (COI) and large ribosomal subunit (LSU) or 16S. The observed levels of nucleotide divergence within Bulinus were substantial but may underestimate the true levels as there was evidence o

Compatibility of Schistosoma haematobium, S. intercalatum and their hybrids with Bulinus truncatus and B. forskalii.

Schistosoma haematobium and S. intercalatum readily hybridize with each other producing generations of viable hybrid offspring. Experiments were designed to investigate the infectivity and viability of the S. haematobium x S. intercalatum F1 and F2 hybrid larvae in their two intermediate snail hosts compared with the parental species. Analysis of the data obtained suggested that the S. haematobium male x S. intercalatum female F1 hybrid miracidia were more infective to Bulinus truncatus than to B.forskalii, and also more infective to B. truncatus compared with the parental S. haematobium mirac

Observations on the compatibility between Bulinus spp. and Schistosoma haematobium in the Senegal River basin.

Snail-infection experiments were carried out with a number of different species and populations of Bulinus and isolates of Schistosoma haematobium. The parasites came from six localities in the Senegal River basin (SRB), in the Lower Valley (Mbodiene), Middle Valley (Podor, Diatar and Nguidjilone), and Upper Valley (Aroundou and Galladé). Isolates of S. haematobium from the Middle and Upper Valleys all showed some compatibility with laboratory-bred B. truncatus from Mali, but none of these isolates was compatible with laboratory-bred B. truncatus originating from Senegal. Schistosoma haematobi

Molecular detection of Schistosoma haematobium in infected Bulinus truncatus snails associated with immune response.

Schistosomiasis is one of the most common waterborne parasite illnesses, it is a major public health issue in developing countries. The polymerase chain reaction (PCR) technique is used to find Schistosoma haematobium DNA in Bulinus truncatus, which could speed up the discovery of infections before cercariae are shed. DraI-PCR detected S. haematobium infection at different infection intervals with bands at 300 bp in shedding snails 40 days after exposure and even on the first day after B. turancuts snails exposure to miracidia. Transmission electron microscopy showed the structure of sporocyst

First Molecular Identification of Bulinus africanus in Lake Malawi Implicated in Transmitting Schistosoma Parasites.

The freshwater snail genus Bulinus plays a vital role in transmitting parasites of the Schistosoma haematobium group. A hybrid schistosome between S. haematobium and S. mattheei has been recently detected using DNA-based identification methods in school children along the Lake Malawi shoreline in Mangochi District. This finding raised the need for contemporary revaluation of local interactions between schistosomes and snails, with a particular focus on snail species within the Bulinus africanus group. In 2017 and 2018, malacological surveys sampled several freshwater sites in Mangochi District

Population dynamics model for Bulinus globosus, intermediate host for Schistosoma haematobium, in river habitats.

A mathematical model is developed that describes the population dynamics of the freshwater snail Bulinus globosus, an intermediate host of Schistosoma haematobium. The model is developed from field data recording abundance changes, recruitment rates, and mortality rates for adult snails in river habitats in Zimbabwe. Rate of recruitment into the adult population is dependent on temperature, incorporating a time lag to allow for growth to adult size. Mortality rate is also a function of temperature. A temperature-dependent model provides a useful description of changes in abundance during the d

Seasonal abundance of trematode cercariae in Bulinus truncatus in a small focus of schistosomiasis in the Nile Delta.

Bulinus truncatus was most numerous from June to August, and echinostome cercariae, present in 19.3% of snails, were the most prevalent of the 4 types of trematode cercariae found. The echinostomes occurred throughout the year, other cercariae only in certain months. Double infections were uncommon. The findings suggest that echinostome cercariae exercise some biological control of Schistosoma haematobium in the area studied.

Genetic Diversity and Ecological Distribution of Bulinus Populations in Southern Benin Using PCR-RFLP Analysis.

Schistosomiasis remains a major public health concern in Benin. Freshwater snails of the genus Bulinus serve as intermediate hosts for schistosomes, trematode parasites responsible for bilharzia. The urinary form, caused by Schistosoma haematobium, is the most prevalent and is primarily transmitted by Bulinus truncatus and Bulinus globosus, whereas the intestinal form, caused by Schistosoma mansoni, is transmitted by Bulinus forskalii. Effective control strategies require an understanding of the molecular diversity and genetic structure of Bulinus populations. Bulinus samples were collected fr

Development of environmental loop-mediated isothermal amplification (eLAMP) diagnostic tool for Bulinus truncatus field detection.

BACKGROUND: Global changes are reshaping the distribution of vector-borne diseases by spreading vectors to previously non-endemic areas. Since 2013, urogenital schistosomiasis has emerged in Corsica and threatens European countries. Gastropod vectors release schistosome larvae that can infect humans who come into contact with freshwater bodies. Monitoring schistosomiasis host vectors is a prerequisite to understand and subsequently to control this pathogen transmission. Because malacological surveys are time consuming and require special expertise, the use of a simple molecular method is desir

Biological activity of artemisinin-naphthoquine phosphate on Schistosoma haematobium stages and the vector Bulinus truncatus.

BACKGROUND: Schistosoma haematobium infection is a major public health problem in most of Africa and the Middle East and praziquantel remains the only drug used for schistosomiasis control, therefore emergence of drug resistance is unavoidable. The antimalarial artemisinin-naphthoquine phosphate combination (co-ArNp) was recently documented to have promising effects on Schistosoma mansoni and its snail host. METHODS: We conducted this in vitro study to assess the bioactivity of co-ArNp on S. haematobium and its snail vector Bulinus truncatus. RESULTS: Treatment of S. haematobium worms with 1

Interactions between intermediate snail hosts of the genus Bulinus and schistosomes of the Schistosoma haematobium group.

Within each of the four species groups of Bulinus there are species that act as intermediate hosts for one or more of the seven species of schistosomes in the Schistosoma haematobium group, which includes the important human pathogens S. haematobium and S. intercalatum. Bulinus species have an extensive distribution throughout much of Africa and some surrounding islands including Madagascar, parts of the Middle East and the Mediterranean region. Considerable variation in intermediate host specificity can be found and differences in compatibility between snail and parasite can be observed over

Selection of susceptible lines of Biomphalaria alexandrina and Bulinus truncatus snails to S.mansoni and S.haematobium infection.

Selection of progenies from infected B.alexandrina and B.truncatus snails resulted in higher infection rates. This observation was consistent when the source of micardia were from either infected human or hamsters. Infection rate of B.alexandrina increased up to 88.2% while B.truncatus did not. Compatibility between the parasites and their intermediate hosts were also studied.

Effect of schistosome infection on protein, glycogen and glucose contents in Biomphalaria arabica and Bulinus truncatus.

B. arabica and B. truncatus the snail intermediate hosts of intestinal and urinary schistosomiasis in Yemen Arab Republic, were bred in the laboratory and infected with S. mansoni and S. haematobium respectively. Schistosome infection caused a marked decrease in the glycogen and protein contents in the tissue of B. arabica and B. truncatus. The glucose content in the hemolymph of both infected snails was lower than that in the normal snails.

Development of cloned DNA probes for the identification of snail intermediate hosts within the genus Bulinus.

DNA fragments have been isolated from Bulinus cernicus and cloned in Escherichia coli strain JM109 using pUC18 as the vector. Four recombinant plasmids designated pBC123, pBC126, pBC127 and pBC134 were selected for further analysis and were shown by both dot and Southern blot hybridisations to have different specificities to species representing the four species groups of Bulinus. Non-specific hybridisation occurred with pBC126 whereas pBC123 and pBC134 hybridised only to B. cernicus. Partial restriction maps of the inserts revealed sequence differences between pBC123 and pBC134. Intermediate
