Spatial metabolomics reveals brassinosteroid-mediated compartmentalization of saikosaponin biosynthesis in Bupleurum chinense DC. under abiotic stress.

Although the biosynthesis of active ingredients in medicinal plants is co-regulated by diverse environmental stimuli, the endocrine mechanism by which brassinosteroids (BRs) coordinate physiological resilience with the in situ spatial distribution of saikosaponins in Bupleurum chinense DC. under combined drought and salinity remains largely elusive. We hypothesized that exogenous brassinosteroids (BRs) application alleviates oxidative impairment provoked by single and combined osmotic/ionic stresses while orchestrating the tissue-level topological routing of saikosaponin biosynthesis. Here, B.

Effects of drought-re-watering-drought on the photosynthesis physiology and secondary metabolite production of Bupleurum chinense DC.

Drastic changes in soil water content can activate the short-term high expression of key enzyme-encoding genes involved in secondary metabolite synthesis thereby increasing the content of secondary metabolites. Bupleurum chinense DC. is a traditional medicinal herb that is famous for its abundant saikosaponins. In the current study, the effects of drought-re-watering-drought on the photosynthesis physiology and biosynthesis of saikosaponins were investigated in 1-year-old B. chinense. The results showed that alterations in soil moisture altered the photosynthesis physiological process of B. ch

Hyperspectral geographical origin identification of Bupleurum via spatial-spectral features based on adaptive weighted quaternion zernike moments.

Bupleurum, a vital herb in traditional Chinese medicine (TCM), possesses complex chemical compositions heavily dependent on variety and geographical origin. Misidentification often leads to unstable therapeutic efficacy and safety risks. Addressing challenges in market confusion and traceability, this study proposes a rapid, non-destructive identification framework using visible-near infrared (Vis-NIR) hyperspectral imaging. A comprehensive dataset comprising 3670 samples from 17 batches, 7 origins, and 2 varieties was established, with hyperspectral images acquired separately from the cross-s

Bupleurum falcatum L. root extract suppresses norepinephrine-induced M2 macrophage polarization by regulating IL-4/STAT6 axis.

Chronic psychological stress promotes cancer progression by activating tumor-associated macrophages (TAMs), particularly through polarization toward the pro-tumorigenic M2 phenotype. However, therapeutic strategies targeting stress-induced macrophage polarization remain limited. This study investigated whether the ethanolic extract of Bupleurum falcatum L. root (EBF) can modulate chronic stress-driven TAM polarization. To simulate a stress-associated microenvironment, norepinephrine (NE) was used to treat 4 T1 breast cancer cells. Conditioned media from NE-treated cells (NE CM) significantly u

Metabolite Profiles Provide Insights into Underlying Mechanism in Bupleurum (Apiaceae) in Response to Three Levels of Phosphorus Fertilization.

Phosphorus (P) deficiency affects plant yield and quality, yet at the same time, excessive phosphorus application does not necessarily promote the growth of plants. How to maintain a balance between biomass accumulation and phosphorus application is a problem. Therefore, the purpose of this research was to explore the relationship between yield and quality of Bupleurum and phosphorus fertilization, based on three phosphorus fertilization levels (20 kg∙ha-1; 10 kg∙ha-1; and 0 kg∙ha-1). We adopted gas chromatography-mass spectrometry to assess the response of primary metabolites of different pla

Saikosaponin D from Bupleurum targets SaeR to disrupt biofilm formation and pathogenic programs for the treatment of MRSA pneumonia.

Staphylococcus aureus pneumonia, particularly methicillin-resistant Staphylococcus aureus (MRSA), remains a major clinical challenge due to biofilm formation, complex regulatory programs, and rapid acquisition of antimicrobial resistance (AMR). The SaeRS two-component system is a central regulatory node controlling multiple infection-related pathways, making its response regulator SaeR an attractive therapeutic target. Targeting such upstream regulators provides an alternative strategy to conventional bactericidal approaches. Through a network pharmacology-guided screen, we identified saikosap

Polysaccharides from Bupleurum chinense impact the recruitment and migration of neutrophils by blocking fMLP chemoattractant receptor-mediated functions.

Although neutrophils play important roles in host defense, sometimes they contribute to inflammation-related tissue injuries. Thus, restriction of their recruitment to the inflammatory sites is a promising strategy in the amelioration of inflammatory disease. The previous studies reported the anti-inflammatory effects of Bupleurum chinense, but the active ingredients and possible mechanism are still unclear. Here, we isolated water-soluble polysaccharides (BCPs) from B. chinense, to evaluate its anti-inflammatory effects and possible mechanism. The present results showed that BCPs significantl
