Transcriptome analysis of the bursa of Fabricius and thymus of laying ducks reveals immune gene expression changes underlying the impacts of stocking densities.

1. The thymus and bursa of Fabricius are important immune organs in poultry as they play essential roles in sustaining the normal immune function to maintain health. The following trial investigated whether the stocking density affected gene expressions in immune organs.2. Jinding ducklings were raised in either low or high density (4 or 8 birds/m2) conditions from four to 14 weeks of age, and were then slaughtered and tissues removed. Samples were subjected to high-throughput sequencing to sequence RNA extraction. After filtering calculations with R software, a total of 508 (thymus) and 1,356

Marek's disease virus-1 unique gene LORF1 is involved in viral replication and MDV-1/Md5-induced atrophy of the bursa of Fabricius.

Marek's disease virus (MDV), an alphaherpesvirus, causes severe immunosuppression and T cell lymphomas in chickens, known as Marek's disease (MD), an economically important poultry disease primarily controlled by vaccination. Importantly, it also serves as a comparative model for studying herpesvirus-induced tumor formation in humans. MDV encodes more than 100 genes, most of which have unknown functions. MDV LORF1 is unique to serotype I MDV (MDV-1), lacking homologs in other herpesviruses, and has not been explored yet. To this end, an infectious bacterial artificial chromosome (BAC) harborin

Appearance of T cells in the bursa of Fabricius and cecal tonsils during the acute phase of infectious bursal disease virus infection in chickens.

Groups of 3-wk-old specific-pathogen-free chickens were inoculated intraocularly with two virulent and one vaccine strain of infectious bursal disease virus (IBDV). During the acute phase of infection, the bursa of Fabricius and cecal tonsil were examined by immunohistochemistry for IBDV antigen and T cell. With all three viruses examined, the infection resulted in the appearance of viral antigen in the bursa accompanied by the presence of CD3+ cells. The CD3+ cells were predominantly TCR2+ and appeared at the site where viral antigens were present. The CD3+ cells continued to persist in the b

Transcriptome analysis identifies CCR7 and cell adhesion molecules as mediators of B cell migration to the bursa of Fabricius during chicken embryonic development.

The development of functional B lymphocytes during chicken embryogenesis relies on a series of tightly regulated processes. Precursor B cells migrate from the spleen via the blood to the bursa of Fabricius, where they colonize the bursal follicles to undergo further maturation and differentiation. To better understand the molecular mechanisms underlying early B cell migration in the chicken embryo, transcriptome analysis of B cells isolated from the spleen, blood, and bursa at embryonic days (ED) 12, ED14, and ED16 was performed. These findings suggest that sphingosine-1-phosphate (S1P) and it

The miR-29a/c-3p/HMCCR pathway is a potential mechanism by which chicken bursa of Fabricius responds to stress affecting immune response.

The bursa of Fabricius (BF), a central immune organ of birds, plays a critical role in immune regulation. Cholesterol metabolism is an important pathway for regulating lymphocyte proliferation and activation. However, the mechanisms by which cholesterol metabolism influences immune regulation in avian BF remain unclear. In this study, dexamethasone (Dex) and Newcastle disease virus (NDV) attenuated vaccine were used to simulate stress and induce immune response in chickens, respectively. The dynamic expression patterns and interrelationships of B cell receptor signaling subunit β (CD79B), sphi

Hydrogen sulfide inhalation-induced immune damage is involved in oxidative stress, inflammation, apoptosis and the Th1/Th2 imbalance in broiler bursa of Fabricius.

Hydrogen sulfide (H2S) is widely accepted to be a signaling molecule that exhibits some potentially beneficial therapeutic effects at physiological concentrations. At elevated levels, H2S is highly toxic and has a negative effect on human health and animal welfare. Studies have shown that H2S exposure induces an immune function in mice, but there are few studies of the effect of continuous H2S exposure on immune organs in poultry. In this study, one-day-old broilers were selected and exposed to 4 or 20 ppm of H2S gas for 14, 28 and 42 days of age. After exposure, the bursa of Fabricius (BF) wa

Lead exposure induced inflammation in bursa of Fabricius of Japanese quail (C. japonica) via NF-κB pathway activation and Wnt/β-catenin signaling inhibition.

Bursa of Fabricius (BF), one of primary lymphoid organ, is unique to birds. Meanwhile, lead (Pb) is well known for its high toxicology to birds. Therefore, this study aimed to examine the chronic toxic effects of lead exposure on BF in Japanese quails (C. japonica) and the underlying mechanism of lead immunotoxicity. One-week old male quails were exposed to 0 ppm, 50 ppm, 500 ppm and 1000 ppm Pb concentrations by drinking water for four weeks. The results showed that Pb accumulation in BF increased in a dose dependent way. The growth and development of BF was retarded in 500 ppm and 1000 ppm P

Regulation of H2S-induced necroptosis and inflammation in broiler bursa of Fabricius by the miR-15b-5p/TGFBR3 axis and the involvement of oxidative stress in this process.

Hydrogen sulfide (H2S) is an air pollutant, having toxic effects on immune system. Necroptosis has been discussed as a new form of cell death and plays an important role in inflammation. To investigate the mechanism of H2S-induced immune injury, and the role of microRNAs (miRNAs) in this process, based on the results of high-throughput sequencing, we selected the most significantly changed miR-15b-5p for subsequent experiments. We further predicted and determined the targeting relationship between miR-15b-5p and TGFBR3 in HD11 through miRDB, Targetscan and dual-luciferase, and found that miR-1

Integrated analysis of miRNA and mRNA expression profiles in the bursa of Fabricius of specific pathogen-free chickens infected with avian reticuloendotheliosis virus strain SNV.

Reticuloendotheliosis virus (REV) is a gamma retrovirus that can cause immunosuppression, dwarf syndrome and acute reticulocytoma in poultry. The molecular mechanism by which REV infection leads to immunosuppression and tumorigenesis is poorly understood. In this study, we elucidated the regulatory network of miRNA-mRNA and the major signaling pathways involved in REV-SNV infection. Therefore, we used the spleen necrosis virus (SNV) model of REV to inoculate one-day-old specific pathogen-free (SPF) chickens and then performed global miRNA and mRNA expression profiling by conducting high-throug

The role of the bursa of Fabricius in the immune response to vaccinal antigens and the development of immune tolerance in chicks (Gallus domesticus) vaccinated at a very young age.

Vaccination is recognized to be the most cost-effective means of preventing, controlling, and even eradicating infectious diseases. Conventional poultry are vaccinated through various routes including eye/nose drops, drinking water, vent brush, or injection. Efficient vaccination is an essential part of any good poultry management. The bursa of Fabricius is intimately connected to the cloaca and the intestinal system. It is well-known as a primary lymphoid organ in the chicken and a major channel through which environmental antigens stimulate the immune system. In this study we tested whether d

Single fluff-spray application of mother hen uropygial secretion analogue positively influences bursa of Fabricius development and the heterophil-to-lymphocyte ratio in ROSS 308 chicks.

Stress is an important cause of illness and mortality in chick production. Stressors such as manipulation, absence of maternal care, transport, and housing can lead to welfare issues, immunodepression, and decreased productivity. The mother hen uropygial secretion analogue (MHUSA), a synthetic analog of a maternal semiochemical secretion, has been proven to protect chicks and broilers against stress, significantly reducing the heterophil-to-lymphocyte ratio. The aim of the present study was to test the effects of the MHUSA on chicks' stress when single-sprayed on their fluff at the age of 1 d.

[24,25-Dihydroxyvitamin D3 modulates the effect of 1alpha-hydroxyvitamin D3 on B lymphocyte maturation in chickens].

Patterns of calcium metabolism and maturation of lymphocytes in chicken Fabricius bursa were studied in presence of various doses of I alpha-hydroxy vitamin D3 and 24,25-dihydroxy vitamin D3 administered separately and at various combinations. 24,25(OH2)D3 was distinctly less effective in the antirickets activity as compared with I alpha (OH)D3. A decrease in mass of Fabricius bursa and inhibition of lymphocytes maturation within early steps were observed in the birds with rickets. A toxic dose of I alpha (OH)D3 caused pseudohypertrophy of Fabricius bursa. Small doses of these drugs administer

Expression characteristics of miR-222b-5p/MAPK10 in major immune organs of SPF chickens infected with avian reticuloendotheliosis virus strain SNV (REV-SNV).

Reticuloendotheliosis virus (REV) is a retrovirus in poultry that can atrophy immune organs and cause immunosuppression and tumor diseases. Our previous results revealed that, in the spleen of SPF chickens infected with REV-SNV, gga-miR-222b-5p expression was upregulated at 7, 14 and 21 dpi, and MAPK10 expression was downregulated, both of which were negatively correlated, with a targeted relationship between the two at 28, 35 and 42 dpi. To understand and analyze the expression patterns of MAPK10 and gga-miR-222b-5p in infected chickens at different times and in different immune organs, qRT-P

Examination of macroscopic and microscopic lesions in IBDV-infected organs and molecular characterization of IBDV VP1 gene fragments obtained from commercial broiler farms in Indonesia.

BACKGROUND AND AIM: Infectious bursal disease (IBD) is an infectious immunosuppressive disease that affects young chickens. Instead of strict biosecurity practices, vaccination is used to control IBD. However, the disease has not been effectively managed. Variations in the observed clinical symptoms lead to confounding diagnoses. The study aimed to obtain pathological lesion data from chickens suspected of IBD virus (IBDV) infection by gross pathology, confirm IBDV infection through molecular diagnostics, and genotype the VP1 gene fragments of circulating IBDV in the field.
