Cloning and characterization of a calcium-activated chloride channel in rat uterus.

In a search for genes involved in regulation of uterine contractility, we cloned a novel calcium-activated chloride channel gene, named rat Clca4, from pregnant rat uterus. The gene shares approximately 83% and 70% nucleotide homology with mouse Clca6 and human CLCA4, respectively, and was expressed primarily in rat uterus. The transcripts were upregulated at Gestational Day 22 (prior to parturition), implying a functional involvement in parturition. Western blot analysis showed that rat CLCA4 protein was present in uterus, lung, and heart, but not in any other tissues examined. Confocal micro

Skin and soft tissue necrosis from calcium chloride in a deicer.

Calcium chloride salt is the principle ingredient of many commercially available deicers. Calcium chloride melts snow and ice by its osmotic action. We present a case of skin and soft tissue necrosis associated with the use of a calcium chloride-containing deicer. Although calcium chloride is known to produce soft tissue necrosis if it extravasates during intravenous administration, necrosis and skin sloughing has rarely been described after topical exposure to this salt. Calcium chloride likely produces tissue injury from the heat liberated by mixing calcium chloride with water (exothermic re

CaMKII mediates stimulation of chloride conductance by calcium in T84 cells.

We used the secretory colonic cell line T84 to study the regulatory pathways controlling the Ca-stimulated Cl conductance [GCl(Ca)]. Under whole cell patch clamp, basal (unstimulated) current levels averaged 73 +/- 9 pA/20 pF (n = 93) and increased to 600 +/- 100 pA/20 pF (n = 53; at +100 mV) on exposure to 1-2 microM ionomycin. Bath application of the calmodulin (CaM) antagonists trifluoperazine, calmidazolium, or sphingosine (50 microM) reversibly inhibited GCl(Ca), whereas the protein kinase C antagonists H7 and phloretin (50 microM) were without effect. This suggests that increases in intr

Calcinosis cutis secondary to percutaneous penetration of calcium chloride in dogs.

Accidental exposure of the skin of a dog to a commercial hygroscopic landscaping product (77-80% CaCl2) caused contact-irritant dermatitis within 24 hours. Papules and eroded to ulcerated plaques with hyperproliferative margins developed on various areas of the skin that had come in contact with CaCl2. Histopathologic findings were typical of calcinosis cutis. Reproducible lesions similar to those observed on the referred dog were induced on 5 clinically normal dogs exposed to small amounts of the compound. Histopathologic documentation of the percutaneous penetration of CaCl2, with evidence o

Calcium Chloride-Mediated Transformation of Chlamydia trachomatis and Other Chlamydia spp.

Despite a high level of conservation within its core genome, the Chlamydia genus comprises various species that exhibit diverse pathogenic profiles. Genetic engineering techniques employing shuttle vectors are crucial tools for elucidating the pathogenesis of these obligate intracellular bacteria. In recent years, the introduction of calcium chloride-mediated transformation methods has significantly advanced genetic manipulation techniques for Chlamydia trachomatis. However, the efficiency of transformation using shuttle vectors, as well as plasmid maintenance, varies not only between Chlamydi

Theoretical study of 2D cancer drug nanocarriers based on calcium chloride.

CONTEXT: This study aims to use two-dimensional nanocarriers composed of cisplatin and fluorouracil molecules adsorbed on the surface of calcium chloride crystals. We found that cisplatin molecules release an alkaline ammonia molecule, neutralizing the acidic environment of cancerous tumors through acid-base interactions. This behavior is similar to the enzyme urease, which has shown good results in cancer treatment. The compounds cisplatin/CaCl2(100), fluorouracil/CaCl2(100), and cisplatin/CaCl2(110) exhibited a wavy geometric structure, enhancing their efficiency as promising nanocarriers fo

Calcium/calmodulin-modulated chloride and taurine conductances in cultured rat astrocytes.

Osmotically swollen rat cerebral astrocytes develop an increased anion conductance which can mediate chloride and taurine release. We used whole cell patch clamp to study mechanisms that modulate this conductance. Astrocyte chloride conductance increased within 4 min of exposure to 200 mOsm medium and was 670+/-123% of its initial value after 15 min (mean+/-S.E.M.). This conductance was substantially reduced in 0.1 mM extracellular calcium with 20 mM BAPTA added to the electrode solution and was completely inhibited with calcium-free perfusion solution containing 1 mM EDTA (n=4). The conductan

Rapid reversal of life-threatening diltiazem-induced tetany with calcium chloride.

We describe a patient who developed tetany with sudden respiratory arrest after the infusion of intravenous diltiazem. The administration of calcium chloride rapidly resolved the patient's tetany with prompt recovery of respiratory function, averting the need for more aggressive airway management and ventilatory support. The emergency physician should be aware that life-threatening tetany may accompany the administration of intravenous diltiazem and that calcium chloride may be a rapid and effective remedy.

Seed priming with calcium chloride enhances stress tolerance in rice seedlings.

Calcium is a crucial second messenger in plant cells and contributes to plant resistance against biotic and abiotic stress. Plant defense priming with natural or synthetic compounds leads to quicker and stronger resistance responses. However, whether pretreatment of plant seeds with calcium could improve their resistance to stress remains poorly understood. In this study, we showed that rice seedlings grown from calcium chloride (CaCl2)-pretreated seeds displayed enhanced resistance to the rice blast fungus Magnaporthe oryzae and the rice bacterial pathogen Xanthomonas oryzae pv. Oryzae (Xoo).

Location of release sites and calcium-activated chloride channels relative to calcium channels at the photoreceptor ribbon synapse.

Vesicle release from photoreceptor ribbon synapses is regulated by L-type Ca(2+) channels, which are in turn regulated by Cl(-) moving through calcium-activated chloride [Cl(Ca)] channels. We assessed the proximity of Ca(2+) channels to release sites and Cl(Ca) channels in synaptic terminals of salamander photoreceptors by comparing fast (BAPTA) and slow (EGTA) intracellular Ca(2+) buffers. BAPTA did not fully block synaptic release, indicating some release sites are <100 nm from Ca(2+) channels. Comparing Cl(Ca) currents with predicted Ca(2+) diffusion profiles suggested that Cl(Ca) and Ca(2+

Substitution of dietary calcium chloride for calcium carbonate reduces urinary ph and urinary phosphorus excretion in adult cats.

In a 4x4-wk cross-over study, eight adult cats were given four moist diets containing identical amounts of calcium (13.9 mmol/MJ) but with different ratios of calcium carbonate to calcium chloride, the calcium salts providing half of the total dietary calcium. Increasing amounts of calcium chloride were substituted for equimolar amounts of calcium carbonate. Higher intakes of calcium chloride caused significantly lower pH values in postprandial and 24-h urine samples. The urinary excretion of ammonium and titratable acid rose with increasing calcium chloride intake. The urinary concentrations

Calcium chloride within carbon dioxide absorbents prevents Compound A production from sevoflurane.

Calcium chloride (CaCl2)-containing carbon dioxide (CO2) absorbers are characterized by their ability to produce almost no pentafluoroisopropenyl fluoromethyl ether (Compound A), a potentially nephrotoxic byproduct of sevoflurane, in vitro. However, the precise role of CaCl2 in this process remains unclear. We aimed to clarify the role of CaCl2 in vitro and determine whether CaCl2-containing CO2 absorbents produce Compound A during prolonged surgery under low- and minimal-flow sevoflurane anesthesia using Japanese brand Yabashi Lime-f (YL-f). In vitro, a reaction between 1 L of sevoflurane gas

Calcium-induced chloride secretion is decreased by Resveratrol in ileal porcine tissue.

OBJECTIVE: Chloride (Cl-) secretion is crucial for intestinal fluid secretion. Therefore, effects of the polyphenol Resveratrol (RSV) on Cl- secretion have been investigated. In a previous study, we observed effects of RSV on forskolin-induced Cl- secretion in the porcine jejunum but not the ileum although RSV itself induced a transepithelial ion current that may represent Cl- secretion in the ileum. The aim of this study was to gain further insights regarding the effects of RSV on characteristics of Cl- secretion in the porcine ileum using the Ussing chamber technique (recording of short circ

Direct electrochemical reduction of titanium dioxide to titanium in molten calcium chloride.

Many reactive metals are difficult to prepare in pure form without complicated and expensive procedures. Although titanium has many desirable properties (it is light, strong and corrosion-resistant), its use has been restricted because of its high processing cost. In the current pyrometallurgical process--the Kroll process--the titanium minerals rutile and ilmenite are carbochlorinated to remove oxygen, iron and other impurities, producing a TiCl4 vapour. This is then reduced to titanium metal by magnesium metal; the by-product MgCl2 is removed by vacuum distillation. The prediction that this

Inverse micelle mediated calcium chloride transportation for facile alginate gelation in microdroplets.

Block copolymer fluorosurfactants are frequently utilized to stabilize water-oil interfaces in droplet microfluidics, enabling parallel and compartmentalized biochemical reactions within individual droplets. Surfactants are able to self-assemble into inverse micelles with the concentration exceeding the critical micelle concentration (CMC), which has been identified as the main reason causing cross-contamination among droplets. This study explored the possibility to utilize the inverse micelles for passive cargo delivery from the fluorocarbon oil phase into the aqueous droplet interior, which

[Calcium-activated chloride channels: structure, properties, role in physiological and pathological processes].

Ca2+-activated chloride channels (CaCC) are a class of intracellular calcium activated chloride channels that mediate numerous physiological functions. In 2008, the molecular structure of CaCC was determined. CaCC are formed by the protein known as anoctamine 1 (ANO1 or TMEM16A). CaCC mediates the secretion of Cl- in secretory epithelia, such as the airways, salivary glands, intestines, renal tubules, and sweat glands. The presence of CaCC has also been recognized in the vascular muscles, smooth muscles of the respiratory tract, which control vascular tone and hypersensitivity of the respirato

Glycine-gated chloride channels in neutrophils attenuate calcium influx and superoxide production.

Recently, it was demonstrated that liver injury and TNF-alpha production as a result of endotoxin (lipopolysaccharide, LPS) were attenuated by feeding animals a diet enriched with glycine. This phenomenon was shown to be a result of, at least in part, activation of a chloride channel in Kupffer cells by glycine, which hyperpolarizes the cell membrane and blunts increases in intracellular calcium concentrations ([Ca(2+)](i)) similar to its action in the neuron. It is well known that hepatotoxicity due to LPS has a neutrophil-mediated component and that activation of neutrophils is dependent on

Augmentation of calcium influx by stannous chloride at mouse motor nerve terminals.

Stannous chloride (SnCl2) facilitates the calcium (Ca) entry into the frog motor nerve terminals. To compare the mode of action of SnCl2 on the Ca channels in the mouse with that in the frog, we investigated the effects of SnCl2 on the inward Ca current at the nerve terminals. SnCl2 (0.1 mM) did not change the second positive component of the action potential (an outward potassium (K) current) at the terminal part of the nerve terminal. SnCl2 (0.1 mM) increased the amplitude of the prolonged negative or positive deflection (an inward Ca current) evoked by treatment with 1 mM tetraethylammonium

Interaction between stannous chloride and calcium channel blockers in frog neuromuscular transmission.

We have investigated the interactions between stannous chloride (SnCl2) and calcium (Ca) channel blockers on endplate potentials (e.p.p.) and on miniature endplate potentials (m.e.p.p.) to determine which type of channel (among L-, N-, and T-type) participates in the SnCl2-induced increase in Ca entry into motor nerve terminals. The e.p.p. amplitude augmented by 30 microM SnCl2 was decreased by cumulative addition of 10 microM CdCl2 or 0.5 microM omega-conotoxin but not by 10 microM NiCl2 or 5 microM nicardipine. The SnCl2 (30 microM)-induced rise in m.e.p.p. frequency in high-potassium medium
