Expression and antigenicity of virus-like particles of norovirus and their application for detection of noroviruses in stool samples.

Human noroviruses (NoVs), members of the genus Norovirus in the family Caliciviridae, are the leading agents of nonbacterial acute gastroenteritis worldwide. Human NoVs are currently divided into at least two genogroups, genogroup I (GI) and genogroup II (GII), each of which contains at least 14 and 17 genotypes. To explore the genetic and antigenic relationship among NoVs, we expressed the capsid protein of four genetically distinct NoVs, the GI/3 Kashiwa645 virus, the GII/3 Sanbu809 virus, the GII/5 Ichikawa754 virus, and the GII/7 Osaka10-25 virus in baculovirus expression system. Virus hazards from food, water and other contaminated environments.

Numerous viruses of human or animal origin can spread in the environment and infect people via water and food, mostly through ingestion and occasionally through skin contact. These viruses are released into the environment by various routes including water run-offs and aerosols. Furthermore, zoonotic viruses may infect humans exposed to contaminated surface waters. Foodstuffs of animal origin can be contaminated, and their consumption may cause human infection if the viruses are not inactivated during food processing. Molecular epidemiology and surveillance of environmental samples are necessa

Investigations on SARS-CoV-2 and other coronaviruses in mink farms in France at the end of the first year of COVID-19 pandemic.

UNLABELLED: Soon after the beginning of the COVID-19 pandemic in early 2020, the Betacoronavirus SARS-CoV-2 infection of several mink farms breeding American minks ( Neovison vison ) for fur was detected in several countries of Europe. The risk of a new reservoir formation and of a reverse zoonosis from minks was then a major concern. The aim of this study was to investigate the four French mink farms for the circulation of SARS-CoV-2 at the end of 2020. The investigations took place during the slaughtering period thus facilitating different types of sampling (swabs and blood). In one of the f

Investigation of caliciviruses and astroviruses in Gabonese rodents: A possible influence of national and international trade on the spread of enteric viruses.

Caliciviruses (Caliciviridae) and astroviruses (Astroviridae) are among the leading cause of non-bacterial foodborne disease and gastroenteritis in human. These non-enveloped RNA viruses infect a wide range of vertebrate species including rodents. Rodents are among the most important hosts of infectious diseases globally and are responsible for over 80 zoonotic pathogens that affect humans. Therefore, screening pathogens in rodents will be is necessary to prevent cross-species transmission to prevent zoonotic outbreaks. In the present study, we screened caliciviruses and astroviruses in order

Human caliciviruses and pediatric gastroenteritis.

Surveillance using molecular diagnostic methods developed in the past decade shows that human caliciviruses, particularly the Noroviruses, are the most important cause of pediatric acute gastroenteritis, next to rotavirus. Noroviruses also cause severe gastroenteritis in children, requiring hospitalization. The Sapovirus, a morphologically and genetically distinct genus of calicivirus, cause mainly milder endemic pediatric gastroenteritis. Noroviruses and Sapoviruses are genetically and antigenetically diverse, and multiple genetic types cocirculate in the same community. Human caliciviruses s

Identifying SNP threshold from P2 sequences for investigating norovirus transmission.

Noroviruses are a group of non-enveloped single-stranded positive-sense RNA virus belonging to Caliciviridae family. They can be transmitted by the fecal-oral route from contaminated food and water and cause mainly acute gastroenteritis. Outbreaks of norovirus infections could be difficult to detect and investigate. In this study, we developed a simple threshold detection approach based on variations of the P2 domain of the capsid protein. We obtained sequences from the norovirus hypervariable P2 region using Sanger sequencing, including 582 pairs of epidemiologically-related strains from 35 n

A viral metagenomic survey identifies known and novel mammalian viruses in bats from Saudi Arabia.

Bats are implicated as natural reservoirs for a wide range of zoonotic viruses including SARS and MERS coronaviruses, Ebola, Marburg, Nipah, Hendra, Rabies and other lyssaviruses. Accordingly, many One Health surveillance and viral discovery programs have focused on bats. In this report we present viral metagenomic data from bats collected in the Kingdom of Saudi Arabia [KSA]. Unbiased high throughput sequencing of fecal samples from 72 bat individuals comprising four species; lesser mouse-tailed bat (Rhinopoma hardwickii), Egyptian tomb bat (Taphozous perforatus), straw-colored fruit bat (Eid

Faecal virome of healthy chickens reveals a large diversity of the eukaryote viral community, including novel circular ssDNA viruses.

This study is focused on the identification of the faecal virome of healthy chickens raised in high-density, export-driven poultry farms in Brazil. Following high-throughput sequencing, a total of 7743 de novo-assembled contigs were constructed and compared with known nucleotide/amino acid sequences from the GenBank database. Analyses with blastx revealed that 279 contigs (4 %) were related to sequences of eukaryotic viruses. Viral genome sequences (total or partial) indicative of members of recognized viral families, including Adenoviridae, Caliciviridae, Circoviridae, Parvoviridae, Picobirna

Molecular characteristics of bovine norovirus and nebovirus in Swedish dairy herds.

BACKGROUND: Neonatal enteritis is a major cause of losses in dairy calves and bovine norovirus (BNoV) and nebovirus (NeV) are underdiagnosed contributors to this disease. In this study, we report for the first time molecular characteristics of bovine norovirus (BNoV) and nebovirus (NeV) detected in calves in Swedish dairy herds. 700 samples from preweaned calves with and without diarrhea were analysed. RESULTS: BNoV was more prevalent (19%) than NeV (4.5%), and among BNoV, the GIII.P2 genotype was more frequently detected than the GIII.P1 genotype. These viruses were detected at similar frequ

Two outbreaks of virulent systemic feline calicivirus infection in cats in Germany.

Over the last years, several outbreaks of virulent systemic feline calicivirus (VS-FCV) infection have been described in the USA and several European countries. The paper describes two outbreaks of VS-FCV infection in cats in Germany. Data concerning clinical, laboratory, and histopathological features ofVS-FCV infection were collected from two outbreaks affecting 55 and 4 cats, respectively. Presence of feline calicivirus was confirmed by PCR followed by sequencing of the PCR-products. Clinical signs were variable, including severe upper respiratory tract infection, dyspnoea, oral and footpad

LPS and IL-4 Stimulation Favor Murine Norovirus Infection of B Cells.

Noroviruses are single-stranded positive-polarity RNA viruses classified in the family Caliciviridae. Human noroviruses (HuNoVs) are a leading cause of acute viral gastroenteritis worldwide. Despite their ability to infect various epithelial and nonepithelial cell types, establishing robust in vitro culture systems for HuNoVs remains challenging. As a result, murine norovirus 1 (MNV-1) has become a widely used model for investigating norovirus biology and pathogenesis, due to its ability to replicate efficiently in primary dendritic cells and macrophages. Although different B-cell lines are su

Evaluating Nanotrap Microbiome Particles as A Wastewater Viral Concentration Method.

Wastewater-based surveillance has emerged as a powerful approach to monitoring infectious diseases within a community. Typically, wastewater samples are concentrated before viral analyses to improve sensitivity. Current concentration methods vary in time requirements, costs, and efficiency. Here, we evaluated the concentration efficiency and bias of a novel viral concentration approach, Nanotrap Microbiome Particles (NMP), in wastewater. NMP concentration efficiency was target-specific, with significantly lower concentrations of the bacterial indicator HF183 and viral indicator Carjivirus (for

Advances in human norovirus research: Vaccines, genotype distribution and antiviral strategies.

Norovirus, belonging to the Caliciviridae family, is a non-enveloped, positive-sense single-stranded RNA virus. It is widely acknowledged as a significant etiological agent responsible for non-bacterial acute gastroenteritis and considered a major cause thereof. Norovirus is primarily tranmitted via fecal-oral route, but can also be transmitted via airborne routes. Clinical manifestations often include symptoms associated with acute gastroenteritis, like nausea, vomiting, watery diarrhea, stomach cramps, and others. Due to the specific pathogenic mechanism of the virus, and genomic diversity,

Viruses and Parasites in Swamp Rabbits (Sylvilagus aquaticus): A Baseline Survey to Aid Conservation Efforts.

Technical advancements have enabled the discovery of potential pathogens in an ever-broadening range of wildlife taxa. To further the scope of this body of knowledge and to inform conservation efforts, we examined potential disease agents present in swamp rabbits (Sylvilagus aquaticus) of southeastern Missouri, USA, during winter 2023. This region represents the northernmost portion of the species' range and is characterized by an intermixed landscape of preferred bottomland hardwood forest and agricultural landcover. Concerns about infectious disease threats to swamp rabbits have increased si

Viral gastroenteritis.

There are many viruses infecting the human gut: some are found to cause acute gastroenteritis regularly (although not always) like rotaviruses, enteric adenoviruses, small round structured viruses and astroviruses; others enter the host via the gut and most often cause systemic infection (entero-viruses, parvoviruses); and others are not regularly associated with human disease (reoviruses, viruses of the Coronaviridae family). The human gut can also be infected directly by HIV and, as a consequence of immuno-suppression, by viruses of the Herpesviridae family. Most remarkable during the last o

PEG-8000 precipitation as a rapid method for enrichment and purification of feline calicivirus.

Feline calicivirus (FCV) is widely used as a surrogate model to study calicivirus biology, including human noroviruses, for which robust cell culture systems are limited. The availability of concentrated and purified viral preparations is essential for structural, biochemical, and infectivity studies. Here, we describe and validate a polyethylene glycol (PEG-8000) precipitation method for the enrichment and purification of FCV from infected cell culture supernatants. PEG-8000 precipitation resulted in a more than 2-log increase in viral titers while markedly reducing contaminating cellular pro
