Intramuscular and topical treatment of cutaneous leishmaniasis lesions in mice infected with Leishmania amazonensis using coumarin (-) mammea A/BB.

Treatment of cutaneous leishmaniasis remains limited to a few available options. Recent studies showed in vitro antileishmanial activity of (-) mammea A/BB, a coumarin isolated from leaves of Calophyllum brasiliense. Moreover, the dichloromethane crude extract and hexane fraction from this plant demonstrated in vivo activity in mice infected with Leishmania amazonensis. We evaluated the antileishmanial activity of (-) mammea A/BB in the L. amazonensis BALB/c mice model. The animals were given intramuscular and topical treatment with (-) mammea A/BB for 30 consecutive days. The results demonstr

Evaluation of the cutaneous wound healing potential of tamanu oil in wounds induced in rats.

AIMS: Tamanu is a plant oil derived from the fruit and seeds of the Calophyllum inophyllum tree. Although scientific data on tamanu oil are limited, it is recommended worldwide for the treatment of abrasions, burns, diabetic wounds and scars. This study aimed to compare the wound healing efficacy of the topical use of tamanu oil with a reference drug in rats. METHODS: Uniform wounds were induced on the dorsum of 21 rats, randomly divided into three groups. The control group received normal saline; the tamanu group received tamanu oil; and the centella group was treated with Centella asiatica.

A natural coumarin, benjaminin, suppresses proliferation and migration and induces apoptosis in MCF-7 breast cancer cells.

Breast cancer remains the leading cause of cancer-related mortality in women, highlighting the need for selective and effective therapeutics. Conventional chemotherapies are often limited by off-target toxicity, prompting interest in natural products, including coumarins from Calophyllum species. Benjaminin, a novel coumarin, has demonstrated cytotoxicity in multiple cancer models, yet its effects in breast cancer remain unexplored. Here, we evaluated benjaminin in MCF-7 (tumorigenic) and MCF-10A (non-tumorigenic) breast cells. Benjaminin exhibited moderate but highly selective cytotoxicity to

Ethnobotany-guided exploration of plant-derived photosensitizer candidates for photodynamic therapy.

The integration of photosensitization with Indigenous knowledge offers a promising framework for exploring more sustainable therapeutic approaches. This review explores how the ethnobotanical knowledge of the Dayak communities of Borneo (Indonesia), particularly the traditional use of brightly coloured medicinal plants for dermatological treatments, may provide a biologically informed framework for prioritizing plant-derived photosensitizer candidates. The biosynthetic pathways and photochemical potential of key metabolites associated with selected Dayak medicinal taxa (Curcuma longa, Etlinger

Antiproliferative and proapoptotic activity of GUT-70 mediated through potent inhibition of Hsp90 in mantle cell lymphoma.

BACKGROUND: Mantle cell lymphoma (MCL) is an aggressive B-cell lymphoma with poor prognosis, requiring novel anticancer strategies. METHODS: Mantle cell lymphoma cell lines with known p53 status were treated with GUT-70, a tricyclic coumarin derived from Calophyllum brasiliense, and the biological and biochemical consequences of GUT-70 were studied. RESULTS: GUT-70 markedly reduced cell proliferation/viability through G(1) cell cycle arrest and increased apoptosis, with greater sensitivity in mutant (mt)-p53-expressing MCL cells than in wild-type (wt)-p53-bearing cells. Mechanistically, GUT-

Testing the carbon starvation hypothesis using a late-successional neotropical tree species with green photosynthetic stems.

Carbon starvation is one of the proposed mechanisms of drought-induced plant mortality. However, it has not been implicated in drought mortality as much as hydraulic failure. We tested the role of carbon on responses to drought by limiting stem photosynthesis and increasing tissue non-structural carbohydrates (NSC) in saplings of the tropical tree Calophyllum longifolium Willd. (Calophyllaceae). We first artificially increased [NSC] by exposing half of the saplings to 2,000 µmol mol-1 of CO2, while the other half remained at ambient [CO2] for 6 wk. Following CO2 treatments, there were no signi
