Antibodies represent indispensable tools in the armamentarium against infectious diseases, with widespread application in prophylactic, therapeutic, and diagnostic settings. Conventional mammalian immunoglobulin G (IgG) antibodies have been extensively utilized in clinical and research contexts; however, their utility is sometimes constrained by intrinsic limitations such as thermal instability, susceptibility to proteolytic degradation, limited mucosal efficacy, and the high costs associated with mammalian expression systems. These challenges have driven increasing interest in alternative ant

This study investigated the distribution and genetic diversity of Entamoeba in 217 faecal samples from mammals, birds, rodents, reptiles, amphibians, fish, and invertebrates sampled across Algeria. By next-generation sequencing (NGS)-based metabarcoding of SSU rDNA, an overall positivity rate of 13 % (n = 29/217) was observed, with positive cases limited to five host families: Bovidae, Camelidae, Equidae, Cervidae, and Caviidae, all of which are herbivores. From the 34 established cluster consensus sequences, we identified one species: Entamoeba bovis, two ribosomal lineages (RL1 and RL8), thr

Trypanosoma brucei is an extracellular protozoan that causes neglected tropical diseases in both humans and livestock. The parasite has a bipartite life cycle involving an insect vector and a mammalian host. Within the latter, it mainly thrives as a blood-borne parasite that relies on glycolysis to support its energy metabolism. It is for this reason that trypanosomal glycolytic enzymes have been investigated as potential targets for the development of trypanosome-killing drugs. Recent work from our lab has shown that they are also interesting biomarkers for the detection of active trypanosome

This review describes the first steps necessary to apply any reproductive biotechnology in South American camelids (SAC) semen or sperm: sample collection, evaluation and handling. In camelids, the length and position adopted for mating and the site of semen deposition have conditioned semen collection methods. The advantages and disadvantages of available collection methods are summarized. The two main drawbacks for applying assisted reproductive techniques in SAC: sperm concentration and rheological characteristics are discussed. Techniques currently available to reliably evaluate diverse sp

Salmonella-related foodborne infections are commonly caused by the serovars of S. Typhimurium, which can be detected using antibody-based immunoassays. The monovalent variable domain of the camelid heavy chain antibody (VHH) performs excellently in constructing multivalent VHH variants, which generally exhibit higher affinities with antigens and consequently enhance the assay sensitivity. In this study, the divalent variants of VHHs (diVHHs) targeting S. Typhimurium were generated by encoding the monovalent VHH genes assembled in tandem with a flexible linker peptide (G4S)2. Soluble diVHHs wer

Characterizing antimicrobial use in animal populations is critical for purposes of antimicrobial stewardship. While dose-based metrics such as the animal daily dose (ADD) are typically used for such purposes, duration of therapy is emerging as a critical and more intuitive metric. In theory, the number of ADDs should approximate the number of days of therapy (DOTs), but no studies have examined whether this is the case. The objective of this study was to compare antimicrobial ADDs with antimicrobial DOTs in three populations: canine patients, large animal hospital patients, and dairy herds. In

Nanobodies, single-domain-based VHHs, are antibody fragments derived from heavy-chain-only IgG antibodies found in the Camelidae family that provide marked advantages compared to traditional monoclonal antibodies. Their small molecular size, simple structure, and high antigen-binding affinity facilitate the precise modulation of immune pathways associated with autoimmune pathophysiology. This review integrates ongoing advancements in experimental and translational research on nanobody therapeutics, encompassing multiple autoimmune conditions, including systemic lupus erythematosus, rheumatoid

Hottentotta saulcyi, medically important scorpion species, causes some of harmful toxic exposure in Iran. Administrated, conventional antivenom-based immunotherapy is still limited and hardly meet ideal characteristic of effective treatment for scorpion envenomation. In this study we aimed to develop a neutralizing agent directed against scorpion venom based on VHH, variable domain of the Camelidae heavy chain antibody or Nanobody. This promising biomolecule is well-established as an advantageous tool for therapeutic purposes due to its small size, stability, monomeric performance and less imm

South American camelids (SAC), including llamas (Lama glama) and alpacas (Vicugna pacos), exhibit distinctive reproductive features, such as induced ovulation and highly viscous seminal plasma (SP), that complicate conventional semen collection and objective sperm evaluation. Consequently, spermatozoa recovered from the cauda epididymis or through surgical vas deferens deviation (VDD) have become valuable alternatives for research and germplasm preservation. These sperm sources also provide a unique opportunity to investigate sperm physiology in the absence of SP. This review critically compar

Plasmodium sporozoites (SPZs) are formed in the Anopheles mosquito midgut from where they travel to the salivary glands and subsequently to the mammalian liver after deposition into the skin. The SPZ's main surface antigen, the circumsporozoite protein (CSP), plays a pivotal role in SPZ biology and constitutes the immunodominant target for host antibodies. In this study, we raised single-domain antibodies (sdAbs) against CSP from P. falciparum (PfCSP) by immunizing two alpacas with recombinant versions of the antigen. We found that all identified sdAbs specifically target PfCSP's globular [For

Emerging coronavirus-associated acute respiratory diseases have posed major global health and economic disruption, highlighting the ongoing risk of zoonotic spillover. Cross-species transmission is largely determined by the interaction between viral receptor-binding domains (RBDs) and host cellular receptors. This study utilizes Porcine Respiratory Coronavirus (PRCoV) as a model to investigate host adaptation via the Aminopeptidase N (APN) receptor. Using molecular docking and structural analysis, we evaluated the docking-estimated binding affinity between the PRCoV RBD and APN proteins from 1

Three-dimensional comparative descriptions of the ossa cranii across representatives of the Camelidae, Giraffidae, and Bovidae remain limited despite their relevance to comparative veterinary anatomy and clinical practice. Given the distinct structural variations and functional adaptations of these families, a descriptive comparison of ossa cranii morphology is needed to advance anatomical knowledge and establish species-specific reference data. This study compared the principal ossa cranii of camel, giraffe, cattle, sheep, and goat to identify conserved anatomical features and species-specifi

Alpacas belong to the Camelidae family. Antibodies produced through alpaca immunization are called nanoantibodies. Compared to traditional antibodies, nanoantibodies have several characteristics, including smaller molecular weight, stable structure, high homology with human antibodies, and suitability for prokaryotic expression. Malignant melanoma (MM) is a severe and aggressive form of cancer that affects both humans and animals. It commonly arises in the mucous membranes of the skin, nose, mouth, anus, and digestive tract, as well as in the choroid of the eyes. Multiple factors contribute to

Nanobodies are the smallest fragments of naturally occurring single-domain antibodies that have evolved to be fully functional in the absence of a light chain. Conventional antibodies are glycoproteins comprising two heavy and two light chains. Surprisingly, all members of the Camelidae family possess a fraction of antibodies devoid of both light chains and the first constant domain. These types of antibodies are known as heavy-chain antibody (HcAb) nanobodies. There are three subclasses of IgG in dromedaries, namely IgG1, IgG2, and IgG3 of which IgG2 and IgG3 are of the HcAb type. These heavy

BACKGROUND: The diagnosis of active Toxocara canis infections in humans is challenging. Larval stages of T. canis do not replicate in human tissues and disease may result from infection with a single T. canis larva. Recently, we developed a nanobody-based electrochemical magnetosensor assay with superior sensitivity to detect T. canis excretory-secretory (TES) antigens. Here, we evaluate the performance of the assay in children from an Ecuadorian birth cohort that followed children to five years of age.
METHODS: Samples were selected based on the presence of peripheral blood eosinophilia and

CD38 is a multifunctional transmembrane glycoprotein found in multiple tissues and overexpressed in many cancer cells, notably in hematological malignancies such as leukemia and multiple myeloma (MM). Therefore, targeting CD38 remains an attractive strategy for cancer treatment in hematological malignancies as well as in solid tumors. It plays a critical role in the progression of these diseases through its ADP-ribosyl cyclase and cADPR-hydrolase activities. Its importance has led to the development of various anti-CD38 monoclonal antibodies (mAbs), including daratumumab and isatuximab, approv

BACKGROUND: Antibody-based therapies targeting CD38 are currently used as single agents as well as in combination regimens for multiple myeloma, a malignant plasma cell disorder. In this study, we aimed to develop anti-CD38 single-domain antibodies (sdAbs) that can be used to trace CD38+ tumour cells and subsequently used for targeted radionuclide therapy. SdAbs are derived from Camelidae heavy-chain antibodies and have emerged as promising theranostic agents due to their favourable pharmacological properties.
METHODS: Four different anti-CD38 sdAbs were produced, and their binding affinities

The recent COVID-19 pandemic has demonstrated again the global threat posed by emerging zoonotic coronaviruses. During the past two decades alone, humans have experienced the emergence of several coronaviruses, such as SARS-CoV in 2003, MERS-CoV in 2012, and SARS-CoV-2 in 2019. To date, MERS-CoV has been detected in 27 countries, with a case fatality ratio of approximately 34.5%. Similar to other coronaviruses, MERS-CoV presumably originated from bats; however, the main reservoir and primary source of human infections are dromedary camels. Other species within the Camelidae family, such as Bac

Antibody treatment is currently the only available countermeasure for botulism, a fatal illness caused by flaccid paralysis of muscles due to botulinum neurotoxin (BoNT) intoxication. Among the seven major serotypes of BoNT/A-G, BoNT/A poses the most serious threat to humans because of its high potency and long duration of action. Prior to entering neurons and blocking neurotransmitter release, BoNT/A recognizes motoneurons via a dual-receptor binding process in which it engages both the neuron surface polysialoganglioside (PSG) and synaptic vesicle glycoprotein 2 (SV2). Previously, we identif
