Prognostic accuracy of 70 individual frailty biomarkers in predicting mortality in the Canadian Longitudinal Study on Aging.

The frailty index (FI) uses a deficit accumulation approach to derive a single, comprehensive, and replicable indicator of age-related health status. Yet, many researchers continue to seek a single "frailty biomarker" to facilitate clinical screening. We investigated the prognostic accuracy of 70 individual biomarkers in predicting mortality, comparing each with a composite FI. A total of 29,341 individuals from the comprehensive cohort of the Canadian Longitudinal Study on Aging were included (mean, 59.4 ± 9.9 years; 50.3% female). Twenty-three blood-based biomarkers and 47 test-based biomark

The Link Between Life Satisfaction and Outcome-wide Health: Evidence from the Canadian Longitudinal Study on Aging.

A promising avenue for improving physical health and reducing health risks in older adulthood is intervening on transdiagnostic protective factors-those that form the mechanisms of several diseases. A potential candidate for a transdiagnostic protective factor is subjective well-being (SWB). Despite the accumulating evidence favoring SWB as a protective factor, individual studies tend to neglect the cumulative effect of publication bias, explicitly specifying a causal model, and the possibility of alternative causal models that guide the selection of confounders. Using three waves of longitudi

Antiseizure Medications and Cardiovascular Events in Older People With Epilepsy in the Canadian Longitudinal Study on Aging.

IMPORTANCE: How epilepsy may promote cardiovascular disease remains poorly understood. OBJECTIVE: To estimate the odds of new-onset cardiovascular events (CVEs) over 6 years in older people with vs without epilepsy, exploring how enzyme-inducing antiseizure medications (EIASMs) and traditional cardiovascular risk factors mediate these odds. DESIGN, SETTING, AND PARTICIPANTS: This was a prospective cohort study using the comprehensive cohort of the Canadian Longitudinal Study on Aging (CLSA), with 6 years of follow-up (2015-2021, analysis performed in December 2023). The CLSA is an ongoing, n

Characterizing Older Adults' Travel Behaviour and Unmet Needs: Findings from the Canadian Longitudinal Study on Aging (CLSA).

This study provides researchers, practitioners, and policy makers with a profile of older adults' travel behaviour and the older adult population that reports unmet travel needs. In addition, we quantified associations between reporting an unmet travel need and measures of health and social connectedness. Data came from the second follow-up survey of the Canadian Longitudinal Study on Aging, collected from 2018 to 2021 (n = 14,167). Nine in ten (90.2%) older adults aged 65 years and older indicated that driving is the main way they get around. Older adults with an unmet travel need were more l

Protocol for validating an algorithm to identify neurocognitive disorders in Canadian Longitudinal Study on Aging participants: an observational study.

INTRODUCTION: In population-based research, disease ascertainment algorithms can be as accurate as, and less costly than, performing supplementary clinical examinations on selected participants to confirm a diagnosis of a neurocognitive disorder (NCD), but they require cohort-specific validation. To optimise the use of the Canadian Longitudinal Study on Aging (CLSA) to understand the epidemiology and burden of NCDs, the CLSA Memory Study will validate an NCD ascertainment algorithm to identify CLSA participants with these disorders using routinely acquired study data. METHODS AND ANALYSIS: Up

Longitudinal associations between movement and sarcopenia, gait speed, and grip strength differ by frailty level in the Canadian Longitudinal Study on Aging.

BACKGROUND: This study tested the hypotheses that more physical activity and less sitting would be associated with lower sarcopenia incidence, stronger grip strength, and faster gait speed, and these associations would be strengthened among higher frailty levels. METHOD: Participants (N = 10871, 50.7% females, age: 61.3 ± 9.3 years) without sarcopenia at baseline from the Canadian Longitudinal Study on Aging were included. Baseline frailty was determined using a 111-item index and participants were grouped as Low (<0.20, n = 9090) or High Frailty (≥0.20, n = 1781). Baseline activity was deter

The association between adverse childhood experiences and epigenetic age acceleration in the Canadian longitudinal study on aging (CLSA).

Research examining the association between exposure to a wide range of adverse childhood experiences (ACEs) and accelerated biological aging in older adults is limited. The purpose of this study was to examine the association of ACEs, both as a cumulative score and individual forms of adversity, with epigenetic age acceleration assessed using the DNA methylation (DNAm) GrimAge and DNAm PhenoAge epigenetic clocks in middle and older-aged adults. This cross-sectional study analyzed baseline and first follow-up data on 1445 participants aged 45-85 years from the Canadian Longitudinal Study on Agi

Sexual dimorphism in metabolomic and phenotypic spectra of UGT deficiency: findings from the Canadian Longitudinal Study on Aging.

BACKGROUND: Two of the most frequently deleted genes in the human genome are the UDP-glycosyltransferases UGT2B17 and UGT2B28. They encode metabolic enzymes of the glucuronidation pathway that plays a pivotal role in the maintenance of cellular homeostasis for a variety of small molecule metabolites. These deletions may impact health, yet their effects remain poorly understood. We evaluated the impact of UGT deficiency on the plasma metabolome and examined the association between altered metabolites and health outcomes. METHODS: The metabolomic profiles of 4262 proficient gene carriers were c
