Dynamics of in vitro development of azole resistance in Candida tropicalis.

OBJECTIVES: Increasing incidence of azole resistance in Candida tropicalis, especially to fluconazole, has been seen in Asian countries including India. Limited knowledge is available on the molecular mechanisms associated with the development of azole resistance in C. tropicalis. The present study examined the dynamics of in vitro azole resistance in C. tropicalis after prolonged treatment with fluconazole. METHODS: Nine fluconazole-susceptible isolates of C. tropicalis were used in this study. Fluconazole resistance was induced experimentally in C. tropicalis isolates. The stability of indu

Role of protein phosphomannosylation in the Candida tropicalis-macrophage interaction.

Candida tropicalis is an opportunistic fungal pathogen responsible for mucosal and systemic infections. The cell wall is the initial contact point between a fungal cell and the host immune system, and mannoproteins are important components that play key roles when interacting with host cells. In Candida albicans, mannans are modified by mannosyl-phosphate moieties, named phosphomannans, which can work as molecular scaffolds to synthesize β1,2-mannooligosaccharides, and MNN4 is a positive regulator of the phosphomannosylation pathway. Here, we showed that C. tropicalis also displays phosphomann

Protein kinase A governs growth and virulence in Candida tropicalis.

Candida tropicalis is one of the most important human fungal pathogens causing superficial infections in locations such as the oral mucosa and genital tract, as well as systemic infections with high mortality. In its sister species Candida albicans, the cyclic AMP/protein kinase A (cAMP/PKA) pathway regulates fungal adhesion and dimorphism, both of which correlate closely with virulence. CaTpk1 and CaTpk2, the catalytic subunits of PKA, not only share redundant functions in hyphal growth, adhesion, and biofilm formation, but also have distinct roles in stress responses and pathogenesis, respec

Efficacy of posaconazole in a murine disseminated infection by Candida tropicalis.

We evaluated the efficacy of posaconazole against Candida tropicalis in a systemic infection model with immunosuppressed mice. Posaconazole at 50 mg/kg of body weight/day prolonged the survival of mice and reduced the fungal tissue burden of mice infected with any of the five strains tested, with the exception of one strain that had a high MIC against this drug. Our results demonstrate the efficacy of posaconazole in the treatment of invasive murine infection caused by C. tropicalis.

Presumed Candida tropicalis endogenous endophthalmitis with multifocal systemic Candida albicans abscesses: a case report.

BACKGROUND: Endophthalmitis is a kind of infectious diseases that seriously threatens vision; Endogenous fungal endophthalmitis (EFE) accounts for 2% to 8% of all endophthalmitis, and is rare and often misdiagnosed in clinical practice. Different strains of Candida may have organ orientation differences, and mixed infections have also been reported in literature. CASE REPORT: A 72 year old male patient was admitted to the hospital due to fever for 5 months and decreased vision in both eyes. The patient's left vitreous fluid underwent metagenomic next-generation sequencing (mNGS), which reveal

Contribution of N-Linked Mannosylation Pathway to Candida parapsilosis and Candida tropicalis Biofilm Formation.

BACKGROUND: Mycoses are a growing threat to human health, and systemic candidiasis caused by Candida parapsilosis and Candida tropicalis is frequent in immunocompromised patients. Biofilm formation is a virulence factor found in these organisms, as sessile cells adhere to surfaces, the stratification and production of extracellular matrix provides protection and resistance to antifungal drugs. Previous evidence indicated that the N-linked mannosylation pathway is relevant to C. albicans biofilms, but its contribution to other species remains unknown. METHODS: C. parapsilosis and C. tropicalis

Brief exposure to antimycotics reduces the extracellular phospholipase activity of Candida albicans and Candida tropicalis.

BACKGROUND: Although the phospholipase activity is considered a potential virulence determinant of the pathogenic Candida species, the effect of antimycotics on this attribute is not known. Hence we evaluated the phospholipase activity in 10 isolates each of Candida albicans and Candida tropicalis, after their exposure to antifungals. METHODS: The impact of antimycotics on phospholipase activity was also assessed after exposure of the isolates to sub-minimum inhibitory concentrations of nystatin, amphotericin B and fluconazole. RESULTS: All Candida isolates investigated exhibited phospholipa

Candida Tropicalis Biofilm Formation Under Secondary Bile Salt Sodium Deoxycholate.

Candida species are components of the normal intestinal microbiota and are under constant exposure to bacterial metabolites, including secondary bile salts. Secondary bile salts are produced by commensal bacteria in the intestine and not only affect lipid absorption through emulsification but also have great effects on other microorganisms. Here, we examined the effect of a secondary bile salt, sodium deoxycholate (NaDCA), on the formation of biofilms by Candida tropicalis. In contrast to C. albicans, C. tropicalis tended to maintain its absolute biofilm biomass and surface hydrophobicity in t

Biological production and downstream separation of xylitol using Candida tropicalis.

Xylitol is one of the platform chemicals identified by the U.S. Department of Energy as a promising candidate for production from biomass. It is an important chemical building block with applications in both the food and pharmaceutical industries. Xylitol is a pentose sugar alcohol with low caloric value, anti-cariogenic properties, and some ability to inhibit microbial growth. The objective of this work is to identify new routes for production of xylitol from lignocellulosic biomass with a focus on post-fermentation downstream processing and separation. Fermentation (via C. tropicalis) condit

Candida tropicalis Brain Abscess Diagnosed by Metagenomic Next-generation Sequencing.

We report the case of a 59-year-old HIV-negative male diagnosed with a brain abscess following vasculitis-associated stroke. The causative pathogen remains unidentified using conventional methods including culture and rapid multiplex PCR. Craniotomy and biopsy were performed to establish a definitive diagnosis, and metagenomic next-generation sequencing (mNGS) of the abscess tissue identified Candida tropicalis as the causative pathogen. This case highlights the utility of mNGS in identifying pathogens in culture-negative CNS infections, even when conventional methods fail to detect the causat

Spontaneous Remission of Acute Lymphoblastic Leukemia Following Candida tropicalis Fungemia.

Spontaneous remission (SR) in acute lymphoblastic leukemia (ALL) is a poorly understood phenomenon that has been sporadically reported in medical literature for over a century, and the molecular and immunologic mechanisms of remission pose interesting clinical questions. Furthermore, the often-transient nature of these remissions poses a challenge to physicians in formulating an approach to treatment. We report on a rare case of Candida tropicalis sepsis in a three-year-old female with high-risk ALL who received less than two months of treatment prior to sepsis and subsequent SR.

Characteristics of Extracellular Vesicles Released by the Pathogenic Yeast-Like Fungi Candida glabrata, Candida parapsilosis and Candida tropicalis.

Candida spp. yeast-like fungi are opportunistic pathogens in humans and have been recently found to release extracellular vesicles (EVs) that are involved in many vital biological processes in fungal cells. These include communication between microorganisms and host-pathogen interactions during infection. The production of EVs and their content have been significantly characterized in the most common candidal species Candida albicans, including the identification of numerous virulence factors and cytoplasmic proteins in the EV cargo. We have here conducted the isolation and proteomic character

Candida tropicalis Fungal Keratitis: A Case Report and Literature Review.

Fungal keratitis, or keratomycosis, is an infection of the cornea caused by fungi. Although it is less frequently implicated in ocular infections than bacterial keratitis, its prognosis remains more guarded. However, the fungi involved include a variety of rare fungal species. Fungal keratitis caused by C. tropicalis has been reported only rarely in the literature. We report the first case of Candida tropicalis corneal abscess diagnosed in the Parasitology-Mycology Department of the Hassan II University Hospital in Fez: a 66-year-old patient with corneal dystrophy was admitted to the Ophthalmo

Novel antifungal activity of oligostyrylbenzenes compounds on Candida tropicalis biofilms.

UNLABELLED: As sessile cells of fungal biofilms are at least 500-fold more resistant to antifungal drugs than their planktonic counterparts, there is a requirement for new antifungal agents. Olygostyrylbenzenes (OSBs) are the first generation of poly(phenylene)vinylene dendrimers with a gram-positive antibacterial activity. Thus, this study aimed to investigate the antifungal activity of four OSBs (1, 2, 3, and 4) on planktonic cells and biofilms of Candida tropicalis. The minimum inhibitory concentration (MIC) for the planktonic population and the sessile minimum inhibitory concentrations (SM

Molecular epidemiology of Candida tropicalis isolated from urogenital tract infections.

Candida tropicalis is a common human pathogenic yeast, and its molecular typing is important for studying the population structure and epidemiology of this opportunistic yeast, such as epidemic genotype, population dynamics, nosocomial infection, and drug resistance surveillance. In this study, the antifungal susceptibility test and multilocus sequence typing (MLST) analysis were carried out on C. tropicalis from central China. Among 64 urogenital isolates, 45 diploid sequence types (DST) were found, of which 20 DSTs (44.4%) were new to the central database. The goeBURST analysis showed that C

Polymorphism analysis of virulence-related genes among Candida tropicalis isolates.

BACKGROUND: Adhesion, biofilm formation, yeast-hyphal transition, secretion of enzymes, and hemolytic activity are all considered important factors in Candida tropicalis infection. However, DNA sequence data for this pathogen are limited. In this study, the polymorphism and heterogeneity of genes agglutinin-like sequences (ALS)2, Lipase (LIP)1, LIP4, and secretory aspartyl proteinase tropicalis (SAPT)1-4 as well as the relationship between phenotype and genotype were analyzed. METHODS: This study started in August 2013, and ended in July 2017. The complete length of ALS2, LIP1, LIP4, and SAPT

Candida tropicalis-associated bilateral renal papillary necrosis and emphysematous pyelonephritis.

Although the kidney is often involved in disseminated and localized candidiasis, bilateral emphysematous pyelonephritis (EPN) is infrequently reported. Renal papillary necrosis (RPN) caused by fungi is also rare. We describe a patient with bilateral RPN and EPN caused by Candida tropicalis, who suffered from recurrent hematuria, flank pain, acute fulminant renal failure, and obstruction by a sloughed papilla. He was treated successfully with antifungal therapy and percutaneous nephrostomy (PCN). This is the first case report of C. tropicalis-associated EPN and RPN.

Oral candida lesions and Candida tropicalis: Potential prognostic markers in end-of-life cancer patients.

AIMS: Oral candidosis is common in patients with end-of-life cancer; however, its prognosis is unclear. We aimed to assess oral candidosis and Candida species as prognostic indicators in palliative care for these patients. METHODS: We consecutively included palliative care patients, assessed for candidosis via microbiological techniques, and classified into three groups by the extent of oral lesions. The association between oral candidosis and overall survival was assessed using a Cox proportional hazards model adjusted by performance status (PS). RESULTS: We studied 142 patients (median age

Fluconazole resistance among Candida species with special emphasis on ERG11 gene mutations among Candida tropicalis.

INTRODUCTION: Candidiasis, an opportunistic fungal infection, is increasingly caused by non-albicans Candida species that show reduced fluconazole susceptibility, mainly due to ERG11 overexpression. This study aimed to identify Candida species, determine fluconazole resistance using VITEK 2 and disc diffusion methods, and detect ERG11 gene mutations in Candida tropicalis. METHODOLOGY: A total of 410 clinical samples were included in this laboratory-based prospective study conducted at a tertiary care hospital in Mysuru. Fluconazole-resistant Candida species were identified using the Vitek-2 s
