Goatpox virus (GTPV) belongs to the genus Capripoxvirus associated with characteristic clinical lesions in fully susceptible breeds of sheep and goats. To date, there is no report of outbreaks of GTPV infection in any wild animals. This study reports the outbreak of GTPV infection in wild Red Serow (Capricornis rubidus.) in Mizoram, India. A total of 113 wild Serow carcasses were recovered from seven districts of Mizoram between May 2015 to October 2016. A postmortem revealed presumptive pox-like lesions. Clinical specimens (lung, skin, and trachea) were examined for the aetiological agents. G

Lumpy skin disease virus (LSDV), a Capripoxvirus, is of economic importance in the cattle industry and is controlled by vaccination. A comparison was made of the host response to the two LSDV vaccines Neethling and Herbivac LS, with reference to the well-studied Orthopoxvirus, modified vaccinia Ankara (MVA), in a mouse model. Because the vaccines differ at the superoxide dismutase homologue (SOD) gene locus, recombinant SOD knock-out and knock-in nLSDV vaccines were constructed and all four vaccines were tested for the induction and inhibition of apoptosis. The SOD homologue was associated bot

Sheeppox virus (SPPV) is a major livestock pathogen causing economic hardship through reduced production and death of vulnerable sheep, with written descriptions of sheeppox-like disease recorded since antiquity. We report 21 novel ancient SPPV genomes spanning the Eurasian steppe Bronze Age (∼1700 BCE) to the Early Modern period in Western Europe, including multiple genomes obtained from medieval parchment. We estimate that major capripoxvirus lineages diverged ∼11,500 to 3700 years ago, overlapping known translocations and bio-cultural developments in sheep. Our dataset supports SPPV divergi

Lumpy skin disease caused by Capripoxvirus is at the moment the most important threat to European cattle industry. The only way for successful control of disease is fast and efficient diagnosis and vaccination. According to EU legislation, vaccination against LDS can be conducted only after confirmation of the disease. Croatia has a special position regarding LSD-in 2016, for the first-time vaccination of the entire cattle population was conducted without an index case. The presence of vaccine viral particles was detected in milk, skin nodules, blood and nasal swabs in seven from total of eigh

Sheep pox (SP) is a contagious viral disease affecting sheep, characterized by fever, respiratory distress, hypogalactia, and skin lesions. In response to a series of outbreaks of pox-like lesions with morbidity (75%) and mortality (37%) rates among sheep in the Upper East Region of Ghana, nasal samples were obtained from affected sheep for diagnosis and characterization. The DNA extracted from these samples was tested using quantitative PCR (qPCR). Positive samples were subjected to further analysis for poxvirus marker genes using conventional PCR. Positive amplicons were sequenced, and phylo

Lumpy skin disease (LSD) caused by the Capripoxvirus LSD virus which infects cattle, leading to a serious disease characterized by fever and the eruption of skin nodules all over the surface of the body. Our understanding of the pathogenesis of this disease is still incomplete, particularly the immunopathological alterations occurring in the skin nodules of infected animals. Therefore, we collected skin nodules from naturally infected cattle with different forms of the disease, both in the early stage of clinical infection and after disease progression. The skin samples were examined both hist

Lumpy skin disease virus (LSDV), a double-stranded DNA virus from the Capripoxvirus genus, primarily affects Bos indicus, Bos taurus breeds, and water buffalo. Arthropod vectors, including mosquitoes and biting flies, are the main LSDV transmitters. Although LSDV is not zoonotic, this study unexpectedly detected LSDV reads in the upper respiratory tract microbiome of humans from rural and urban areas in Maharashtra, India. Nasopharyngeal and oropharyngeal swab samples collected for SARS-CoV-2 surveillance underwent whole-genome metagenomics sequencing, revealing LSDV reads in 25% of samples. S

The aim of this work was to evaluate the immunogenicity and neutralizing activity of sheep pox virus (SPPV; genus Capripoxvirus, family Poxviridae) structural proteins as candidate subunit vaccines to control sheep pox disease. SPPV structural proteins were identified by sequence homology with proteins of vaccinia virus (VACV) strain Copenhagen. Four SPPV proteins (SPPV-ORF 060, SPPV-ORF 095, SPPV-ORF 117, and SPPV-ORF 122), orthologs of immunodominant L1, A4, A27, and A33 VACV proteins, respectively, were produced in Escherichia coli. Western blot analysis revealed the antigenic and immunogen

Sheeppox virus (SPPV), goatpox virus (GTPV), and lumpy skin disease virus (LSDV) are the three members of the genus Capripoxvirus within the Poxviridae family and are the etiologic agents of sheeppox (SPP), goatpox (GTP), and lumpy skin disease (LSD), respectively. LSD, GTP, and SPP are endemic in Africa and Asia, causing severe disease outbreaks with significant economic losses in livestock. Incursions of SPP and LSD have occurred in Europe. Vaccination with live attenuated homologous and heterologous viruses are routinely implemented to control these diseases. Using the gold standard virus n

INTRODUCTION: Lumpy Skin Disease (LSD), caused by a Capripoxvirus, has caused severe outbreaks among cattle in India. The disease status and vaccine response of Mithun (Bos frontalis), a semi-domesticated bovine species of major socio-economic importance in Northeast India, remain poorly understood. Because sero-epidemiological findings have indicated LSDV exposure in Mithun, preventive interventions are needed.
METHODS: We conducted a 12-month longitudinal field study in Nagaland to assess the safety and vaccine-induced immune response of a heterologous goatpox vaccine in seronegative Mithun

In this study, the duration of immunity following a single-dose vaccination using an attenuated live goatpox vaccine (GTPV/Uttarkashi/1978 strain) was evaluated in goatpox-seronegative goats for 52 months. Long-term immunity was evaluated by clinical protection upon virulent virus challenge and serum neutralization assay applied to serum samples. The rise in the level of GTPV-specific antibodies was found to reach a maximum at 21 days post-vaccination, and these antibodies were maintained for 1 to 2 years after immunization, with a steady decline. Upon virulent virus challenge at 12, 24, 42, a

An outbreak of sheeppox was investigated in a cluster of villages situated in Western Himalayan ranges of a Northern Indian state. Non-migratory sheep (n = 80) of native breeds namely Gaddi and Rampur Bushair were infected and 15 have died. The outbreak started after a few animals contracted the disease during the summer grazing period at the highland pastures from migrating flocks of sheep. This initial outbreak resulted in a further spreading of the disease into the valley. Clinical examination revealed varying degree of cutaneous papular lesions and respiratory distress. Upon necropsy, visc

Lumpy skin disease virus (LSDV), a member of the capripoxvirus genus, induces lumpy skin disease (LSD) in cattle and buffalo populations. The initial documentation of LSD dates back to 1929 in Zambia, with subsequent rapid dissemination within the cattle community in Africa and Asia. Described as the "smallpox" equivalent in cattle, LSD manifests through clinical features such as fever, extensive cutaneous nodules, body wasting, lymph node enlargement, and skin edema. Isolation of LSDV constitutes an important step in evaluating virulence and implementing disease control measures. Skin samples

BACKGROUND: Direct whole genome sequencing of Capripox virus genomes from diagnostic samples is not always straightforward. Low viral content in a sample, biased sequencing and subsequent assembly and mapping methods may all influence the outcome. METHODS: In this study we have tested and compared six next generation sequencing approaches on a homogenized skin sample from a bull infected with LSDV. We compared enrichment vs. non-enrichment strategies, different library preparation methods, short read Illumina sequencing with long read sequencing methods. RESULTS: We found that methods that use

Lumpy skin disease (LSD) is a serious emerging infectious disease in cattle caused by a virus of the family Poxviridae. According to the Department of Animal Health, LSD first occurred in Vietnam at the end of October 2020 in Cao Bang and Lang Son provinces. So far, the disease has infected over 63,000 animals, resulting in 9170 deaths occurring in 32 different provinces in northern and central Vietnam. In this study, skin samples from lumpy skin disease virus (LSDV)-infected cattle from the northern provinces of Vietnam displaying clinical symptoms including fever (> 40 °C), runny nose, drool

The present study aimed to evaluate the cell-mediated and the humoral immune response to Romanian sheep pox vaccine in pregnant cows (n = 12) vaccinated at different times of gestation period and the duration of maternal immunity in calves born to these cows. Evaluation of cellular immunity revealed an increase in lymphocytic proliferation that peaked at 10th day post vaccination (dpv) then gradually decreased. Capripoxvirus-specific antibodies were detected by SNT and ELISA in sera collected from vaccinated dams and also in calves born to these cows. In cows, the antibody titers persisted abo

BACKGROUND: Goatpox is a highly contagious disease caused by the Capripoxvirus, primarily affecting sheep and goats. Breeds like the Saanen are particularly vulnerable, especially in enzootic areas, and face risks not only from the disease itself but also from adverse reactions to live attenuated vaccines. This study compares inactivated and attenuated vaccines to find the safest and most effective vaccination strategy for Saanen goats against the Goatpox Virus (GTPV). In this study, 375 pure-breed Saanen goats were strategically divided into four groups to explore the most effective vaccinati

Rational design of capripoxvirus-based vaccine vectors can be achieved by knockout of immunomodulatory genes. In this study, the effect of knockout of the immunomodulatory genes LSDV005, LSDV008 and LSDV066 on the replication of Lumpy skin disease virus in cell cultures and the immune response to an integrated foreign antigen were assessed. The knockout of genes was performed by homologous recombination under conditions of temporary dominant selection. It was found that single knockout of the LSDV005 gene and the LSDV008 gene did not affect the replicative activity of recombinant viruses in vi

The goat pox chick embryo-attenuated virus (GTPV) has been developed as an effective vaccine that can elicit protective immune responses. It possesses a large genome and a robust ability to express exogenous genes. Thus, this virus is an ideal vector for recombinant live vaccines for infectious diseases in ruminant animals. In this study, we identified a novel bidirectional promoter region of GTPV through screening named PbVV(±). PbVV(±) is located between ETF-l and VITF-3, which are transcribed in opposite directions. A new recombinant goat pox virus (rGTPV) was constructed, in which duplicat
