Neuropharmacological potentials of β-carboline alkaloids for neuropsychiatric disorders.

Neuropsychiatric disorders are diseases of the central nervous system (CNS) which are characterised by complex pathomechanisms that including homeostatic failure, malfunction, atrophy, pathology remodelling and reactivity anomaly of the neuronal system where treatment options remain challenging. β-Carboline (βC) alkaloids are scaffolds of structurally diverse tricyclic pyrido[3,4-b]indole alkaloid with vast occurrence in nature. Their unique structural features which favour interactions with enzymes and protein receptor targets account for their potent neuropharmacological properties. However,

Preparation and antiprotozoal evaluation of promising β-carboline alkaloids.

The synthesis of β-carbolines and their in vitro antiplasmodial and antileishmanial activities were described herein. These molecules have also been studied concerning their in vitro cytotoxicity toward the human cell line THP1, in order to calculate their respective selectivity indexes (SI). Among the 20 tested molecules, four exhibited significant antiplasmodial activity on the W2 multi-resistant Plasmodium falciparum strain (0.7 < IC₅₀ < 1.7 μM), in comparison with two references drugs (chloroquine and doxycycline), and a low cytotoxicity. These β-carbolines were also evaluated concerning t

Elevation of blood beta-carboline alkaloids in essential tremor.

BACKGROUND: beta-Carboline alkaloids are normal body constituents but are also potent tremor-producing chemicals that are naturally present in the food chain. OBJECTIVE: To explore the hypothesis that high concentrations of beta-carboline alkaloids are associated with essential tremor (ET). METHODS: One hundred cases and 100 controls were frequency matched on age, sex, and ethnicity. Blood concentrations of harmane and harmine were quantified by high-performance liquid chromatography, blinded to clinical information. RESULTS: The mean log blood concentration of harmane was higher in cases t

Simple beta-carboline alkaloids as nucleic acids fluorochromes.

Treatment of cell smears (chicken blood, Trypanosoma cruzi epimastigotes, Ehrlich ascites tumor cells and mouse spleen) with simple beta-carboline alkaloids induced a strong bluish white fluorescence emission of condensed chromatin and basophilic cytoplasm under ultraviolet excitation. The compounds used were harmine, harmol, harman, norharman, harmalol and harmaline (25-50 micrograms/ml aqueous solutions for 2-3 min). Spectrofluorometric studies on harmine solutions in vitro (emission peak at 420 nm) showed fluorescence quenching at high concentrations as well as in the presence of DNA. Micro

Effects of harmane and other β-carbolines on apomorphine-induced licking behavior in rat.

Harmane, harmine and norharmane are β-carboline compounds which have been referred to as inverse agonists of benzodiazepine receptors. The effect of these compounds on apomorphine-induced licking behavior was studied in rats. Subcutaneous (s.c.) injection of apomorphine (0.5 mg/kg) induced licking. The licking behavior was counted with a hand counter and recorded for a period of 75 min by direct observation. Intraperitoneal (i.p.) injections of harmane (1.25-5 mg/kg), harmine (2.5-10 mg/kg) and norharmane (1.25-5 mg/kg) significantly reduced the licking behavior. In rats pretreated with reserp

Cytotoxicity of chloral-derived beta-carbolines is not specific towards neuronal nor dopaminergic cells.

beta-Carbolines structurally related to the selective dopaminergic neurotoxin 1-methyl-4- phenylpyridinium (MPP(+)) may contribute to dopaminergic neurodegeneration in Parkinson's disease. The chloral-derived mammalian alkaloid derivative 1-trichloromethyl-1,2,3,4-tetrahydro-beta-carboline (TaClo) is formed endogenously by a Pictet-Spengler condensation from the biogenic amine tryptamine (Ta) and the hypnotic aldehyde chloral (Clo). Here we examine the dopaminergic toxicity of TaClo and related compounds by testing their differential cytotoxicities in dopaminergic SH-SY5Y and non-dopaminergic

Beta-carbolines with agonistic and inverse agonistic properties at benzodiazepine receptors of the rat.

The anxiogenic or anxiolytic properties of five beta-carbolines were assessed in rats trained in two-lever operant chambers to operate one lever to obtain food reward when treated with a training drug, and the other when treated with saline. Two such drug-discrimination tests were used, employing either chlordiazepoxide (CDP) or pentylenetetrazol (PTZ) as training drugs. The benzodiazepine (BZ) agonist, ZK 93 423, substituted for the CDP cue and antagonized the PTZ cue. The inverse agonists DMCM and FG 7142 showed the opposite effects. An antagonist, ZK 93 426, antagonized the CDP cue but did

Vaginal lactobacilli produce anti-inflammatory β-carboline compounds.

The optimal vaginal microbiome is a Lactobacillus-dominant community. Apart from Lactobacillus iners, the presence of Lactobacillus species is associated with reduced vaginal inflammation and reduced levels of pro-inflammatory cytokines. Loss of Lactobacillus-dominance is associated with inflammatory conditions, such as bacterial vaginosis (BV). We have identified that Lactobacillus crispatus, a key vaginal bacterial species, produces a family of β-carboline compounds with anti-inflammatory activity. These compounds suppress nuclear factor κB (NF-κB) and interferon (IFN) signaling downstream o

The halogenated tetrahydro-beta-carboline "TaClo": a progressively-acting neurotoxin.

"TaClo", (1-trichloromethyl-1,2,3,4-tetrahydro-beta-carboline) which is structurally very similar to MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine), increases the sensitivity to apomorphine in aging rats after subchronic daily application (0.2 mg/kg i.p.) for 7 weeks. Nine weeks after the last treatment with TaClo, the running speed of the animals during spontaneous nocturnal activity was significantly diminished, and 9 months after the last injection rats developed a pronounced hypersensitivity to apomorphine (0.4 mg/kg s.c.). TaClo, which can be readily produced from endogenous tryptami

Plasma norharman (beta-carboline) levels are elevated in chronic alcoholics.

Based on the hypothesis that condensation products of neurotransmitters with aldehydes are involved in the pathogenesis of alcoholism, aromatic beta-carbolines (norharman and harman) were measured in the blood plasma of alcoholics and nonalcoholics. The identity of the extracted compounds was confirmed by various elution conditions of the high performance liquid chromatography (HPLC), newly developed radioreceptor assays, and the mass spectrum of norharman. The levels of norharman and harman in nonalcoholics were unchanged after a load with ethanol (1 g/kg body weight). The norharman levels of

Targeting Chemical-induced Hepatocellular Carcinoma: Ameliorative Potential of Natural Compounds with Focus on Beta-carbolines.

INTRODUCTION: Hepatocellular carcinoma (HCC), the predominant form of primary liver malignancy, remains a major global health concern owing to its aggressive progression, limited therapeutic efficacy, and high fatality rate. A significant proportion of HCC arises from chronic exposure to chemical carcinogens, which trigger hepatocarcinogenesis through oxidative stress, DNA damage, and dysregulation of signalling networks. Natural compounds, particularly beta-carboline alkaloids, are emerging as safer, multi-targeted candidates with promising hepatoprotective and anticancer potential. This revi

3-Tetrazolyl-β-carboline derivatives as potential neuroprotective agents.

3-Tetrazolyl-β-carbolines were prepared by the Pictet-Spengler approach using a tryptophan analogue as building block, in which the carboxylic acid was replaced by the bioisosteric tetrazole group. Knowing that β-carbolines are often associated with psychopharmacological effects, the study of the 3-tetrazolyl-β-carbolines as potential neuroprotective agents against Parkinson's disease was investigated. The evaluation of neuroprotective effects against 1-methyl-4-phenylpyridin-1-ium (MPP+)-induced cytotoxicity allowed to identify compounds with relevant neuroprotective activity. One derivative,

Current development of β-carboline derived potential antimalarial scaffolds.

β-Carboline alkaloids are an eminent class of nitrogen-based natural alkaloids and therapeutic molecules which exert various pharmacological activities through diverse mechanisms. A lot of attention has recently been directed towards this moiety in order to develop effective antimalarial drugs. "Malaria", an acute febrile illness caused by diverse Plasmodium parasites, is a continuing and escalating problem that devastates economically less developed countries by significantly increased morbidity and mortality rates. The mounting parasite resistance towards the antimalarial drugs and augmentin

Anxiogenic effects of noreleagnine, a water soluble beta-carboline in rats.

We tested noreleagnine, a beta-carboline which unlike other beta-carbolines remains in aqueous solution at physiological pH, as well as chlordiazepoxide, a traditional benzodiazepine, and their interaction with the benzodiazepine receptor antagonist R0 15-1788 in a modified approach schedule with two levels of conflict. Noreleagnine displayed pro-conflict activity characteristic of beta-carbolines and chlordiazepoxide elicited an anti-conflict effect, both of which were blocked by pretreatment with R0 15-1788. We believe this to be the first clear demonstration that noreleagnine has an anxioge

Potentiation of the propunishment, but not the convulsant action of the beta-carboline DMCM by naltrexone.

The ability of naltrexone (NTX) to potentiate the propunishment and convulsant properties of DMCM, a benzodiazepine receptor inverse agonist, was studied in mice. Doses (0.39 and 1.56 mg/kg) of DMCM which were below the threshold for propunishment effects showed a marked ability to enhance the suppressive effects of punishment on locomotor activity in the presence of naltrexone (0.5 or 2.5 mg/kg IP), higher doses of DMCM and NTX (3.13 mg/kg and 10 mg/kg, respectively) had a depressant effect of their own on both punished and unpunished locomotor activity. DMCM given alone induced clonic convul

Sequential Organic/Metal Relay Catalysis Enabling the Regioselective Synthesis of Functionalized α-Carbolines from Alkynyl Ketimines.

Herein, we report an efficient organo/silver relay catalytic system that enables the sequential Mannich/6-endo-dig cyclization/aromatization cascade of β,γ-alkynyl-α-ketimino esters with indolin-2-imines, which exhibits remarkable substrate generality and affords a broad range of α-carboline derivatives in good to excellent yields. Mechanistic studies confirm a stepwise relay catalytic pathway, and the practical utility of this protocol is verified by late-stage functionalization, gram-scale synthesis, and facile synthetic transformations of the resulting α-carboline products for accessing mor

The strong inhibition of triosephosphate isomerase by the natural beta-carbolines may explain their neurotoxic actions.

The natural beta-carbolines (BC) closely resemble the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in structure. The N-methylated beta-carbolinium ions (BC+) are potent inhibitors of mitochondrial respiration and are nigrostriatal neurotoxins. Utilizing [3H]BC, we have identified several proteins to which BC binds with high affinity (e.g. the chaperone member glucose regulated protein 78, the enzyme carboxylesterase, the cytochrome P450 2E1, the enzyme monoamine oxidase B and a small G-protein of the Rho subfamily). In the present study we isolated a protein from bovine brain

Comparisons between patterns of convulsions induced by two beta-carbolines in 10 inbred strains of mice.

The beta-carbolines, methyl-beta-carboline-3-carboxylate (beta-CCM) and 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM) are known to have pharmacological properties opposite to those of agonistic benzodiazepines. Convulsions induced by these drugs lead to differential patterns, such as clonus, myoclonic or tonic seizures. In 10 different inbred strains of mice we investigated whether the responsiveness to the two drugs was the same, irrespective of the pattern of convulsions. We found the same ranking in the responsiveness of the strains to both drugs in the case of myoclonic seizure
