Pseudomonas sp. strain JS150 was isolated as a nonencapsulated variant of Pseudomonas sp. strain JS1 that contains the genes for the degradative pathways of a wide range of substituted aromatic compounds. Pseudomonas sp. strain JS150 grew on phenol, ethylbenzene, toluene, benzene, naphthalene, benzoate, p-hydroxybenzoate, salicylate, chlorobenzene, and several 1,4-dihalogenated benzenes. We designed experiments to determine the conditions required for induction of the individual pathways and to determine whether multiple substrates could be biodegraded simultaneously. The increasing environmental burden of biodegradable plastics such as poly (butylene adipate-co-terephthalate) (PBAT) necessitates a detailed understanding of their microbial degradation mechanisms. In this study, Gordonia sp. CN2K utilized PBAT as the sole carbon and energy source, with cell density increasing from 1.26 × 108 to 6.26 × 108 CFU mL⁻1 over 90 days. PBAT degradation, determined by weight loss measurements, reached 52.8% within 45 days. ATR-FTIR analysis indicated ester bond cleavage and polymer backbone disruption, demonstrating chemical modifications in the PBAT polymer. SEM ana

BACKGROUND: Carbazole is a recalcitrant compound with a dioxin-like structure and possesses mutagenic and toxic activities. Bacteria respond to a xenobiotic by recruiting exogenous genes to establish a pathway to degrade the xenobiotic, which is necessary for their adaptation and survival. Usually, this process is mediated by mobile genetic elements such as plasmids, transposons, and insertion sequences.
FINDINGS: The genes encoding the enzymes responsible for the degradation of carbazole to catechol via anthranilate were cloned, sequenced, and characterized from a carbazole-degrading Sphingo

Soil from a pesticide disposal site was used to enrich for microorganisms that degraded the acylanilide herbicide propachlor (2-chloro-N-isopropylacetanilide). After seven transfers of the enrichment, the culture contained about six strains. The highest yield of microbial biomass occurred if just two of these isolates, strains DAK3 and MAB2, were inoculated into a mineral salts medium containing propachlor. When only strain DAK3 was grown on propachlor, a metabolite (2-chloro-N-isopropylacetamide) was released into the medium. Strain MAB2 could grow on this metabolite. The results of morpholog

The decomposition of various aromatic hydrocarbon intermediates was examined using a recombinant oxidative enzyme immobilized on single-walled carbon nanotubes (SWCNTs). Hydroxyquinol 1,2-dioxygenase (CphA-I), which catalyzes ring cleavage of catechol and its analogues, was obtained from Arthrobacter chlorophenolicus A6 via cloning, overexpression, and subsequent purification. This recombinant enzyme was immobilized on SWCNTs by physical adsorption and covalent coupling in the absence and presence of N-hydroxysuccinimide. The immobilization yield was as high as 52.1%, and a high level of enzym

The carbazole-degradative plasmid pCAR1 of Pseudomonas resinovorans strain CA10 has two gene clusters, carAaAaBaBbCAcAdDFE and antABC, which are involved in the conversions of carbazole to anthranilate and anthranilate to catechol, respectively. We proved that the antABC gene cluster, encoding two-component anthranilate 1,2-dioxygenase, constitutes a single transcriptional unit through Northern hybridization and reverse transcription-PCR (RT-PCR) analyses. The transcription start point of antA was mapped at 53 bp upstream point of its translation start point, and the -10 and -35 boxes were hom

Pseudomonas sp. strain MT1 is capable of degrading 4- and 5-chlorosalicylates via 4-chlorocatechol, 3-chloromuconate, and maleylacetate by a novel pathway. 3-Chloromuconate is transformed by muconate cycloisomerase of MT1 into protoanemonin, a dominant reaction product, as previously shown for other muconate cycloisomerases. However, kinetic data indicate that the muconate cycloisomerase of MT1 is specialized for 3-chloromuconate conversion and is not able to form cis-dienelactone. Protoanemonin is obviously a dead-end product of the pathway. A trans-dienelactone hydrolase (trans-DLH) was indu

Pyrocatechase (catechol 1,2-oxidoreductase (decyclizing), EC 1.13.11.1), a ferric ion-containing dioxygenase from Pseudomonas arvilla C-1, catalyzes the intradiol cleavage of catechol with insertion of 2 atoms of molecular oxygen to form cis,cis-muconic acid. The enzyme also catalyzed the oxidation of various catechol derivatives, including 4-methylcatechol, 4-chlorocatechol, 4-formylcatechol (protocatechualdehyde), 4,5-dichlorocatechol, 3,5-dichlorocatechol, 3-methylcatechol, 3-methoxycatechol, and 3-hydroxycatechol (pyrogallol). All of these substrates gave products having an absorption maxi

Catechol (Cat) quantification typically requires benchtop instrumentation, restricting its application for on-site environmental analysis. To address this, we developed a portable image-based biosensing platform that integrates a genetically engineered whole-cell biocatalyst with a Multi-Feature Fusion Vision Transformer (MF-ViT). An Escherichia coli strain expressing 2,3-dihydroxybiphenyl-1,2-dioxygenase (BphC) was constructed to specifically convert Cat into a measurable yellow product. Molecular docking and molecular dynamics simulations provided a structural rationale for the sensor's anal

In this study, the stable consortium composed by Pseudomonas reinekei strain MT1 and Achromobacter xylosoxidans strain MT3 (cell numbers in proportion 9:1) was under investigation to reveal bacterial interactions that take place under severe nutrient-limiting conditions. The analysis of steady states in continuous cultures was carried out at the proteome, metabolic profile, and population dynamic levels. Carbon-limiting studies showed a higher metabolic versatility in the community through upregulation of parallel catabolic enzymes (salicylate 5-hydroxylase and 17-fold on 2-keto-4-pentenoate h

Toluene/o-Xylene Monooxygenase (ToMO) is equipped with a broad spectrum of aromatic substrate specificity (such as BTEX; benzene, toluene, ethylbenzene, and isomers of xylenes). TOMO has can hydroxylate more than a single position of aromatic rings in two consecutive monooxygenation reactions. Catechol 1,2-dioxygenase (C1,2D) is an iron-containing enzyme able to cleave the ring of catechol (the converted product from ToMO) for complete detoxification of BTEX. In this study, cold-active ToMO and C1,2D were produced using newly isolated psychrophilic Pseudomonas S2TR-14 in the minimal salt mediu

Microbial consortia are essential for the degradation of complex hydrocarbon mixtures in coastal environments, yet the functional basis of consortium performance rarely resolved at strain level. In this study, we employed a Plackett-Burman experimental design to rationally assemble and optimize a synthetic consortium for the degradation of medium-grade crude oil in a sand-seawater microcosm. Six strains were selected: Stutzerimonas stutzeri CBC1 and CBC2, S. balearica GOM11, Pseudomonas sp. CBC3, Halomonas litopenaei GOM23 and Enterobacter kobei GOM24. After 45 days, the consortium removed 15.

Phenolic compounds are an extensive group of natural and anthropogenic organic substances of the aromatic series containing one or more hydroxyl groups. The main sources of phenols entering the environment are waste from metallurgy and coke plants, enterprises of the leather, furniture, and pulp and paper industries, as well as wastewater from the production of phenol-formaldehyde resins, adhesives, plastics, and pesticides. Among this group of compounds, phenol is the most common environmental pollutant. One of the cheapest and most effective ways to combat phenol pollution is biological puri

This study investigates the advantages of combined microbial degradation of polycyclic aromatic hydrocarbons (PAHs) in reducing the inhibitory effects of high-concentration eluents commonly used in soil washing. A microbial synergistic strategy was proposed using Arthrobacter sp. SZ-3 and Pseudomonas putida B6-2 as the key bacteria in the presence of Tween 80. The results show that in systems with Tween 80, the SZ-3 strain exhibits a strong capacity to degrade three types of PAH compounds, while the B6-2 strain follows multiple degradation pathways. Mixed bacteria achieved degradation rates 60

Coal chemical wastewater (CCW) can destabilize sludge structure and cause performance fluctuations in biological systems. Aerobic granular sludge (AGS) is structurally compact and resistant to shock loading. In this study, AGS was integrated with iron-carbon microelectrolysis (IC-ME) conditioning and co-substrate regulation for CCW treatment. Together, these strategies alleviated toxic stress and substrate limitation, stabilized the granular microenvironment, and promoted pollutant biotransformation. Two sequencing batch reactors were operated in stages to compare AGS alone with co-substrate a
