Artificial cell vesicle-mediated delivery of Catharanthus roseus (L.) G. Don-derived vinca alkaloids for enhanced antitumor efficacy.

Vinca alkaloids, a class of naturally derived antimitotic agents isolated from Catharanthus roseus, have long been established as potent chemotherapeutic drugs for the treatment of various malignancies. Despite their clinical efficacy, the therapeutic utility of vinca alkaloids, such as vincristine, vinblastine, and vinorelbine, is frequently constrained by systemic toxicity, poor bioavailability, and the emergence of multidrug resistance. In this study, we developed a universal strategy to construct cell membrane-derived vesicles for the encapsulation of vinca alkaloids, thereby enhancing the

Effects of stress factors, bioregulators, and synthetic precursors on indole alkaloid production in compact callus clusters cultures of Catharanthus roseus.

Compact callus cluster (CCC) cultures established from Catharanthus roseus consist of cohesive callus aggregates displaying certain levels of cellular or tissue differentiation. CCC cultures synthesize about two-fold more indole alkaloids than normal dispersed-cell cultures. Our studies here show that additions of KCl, mannitol, and a variety of synthetic precursors and bioregulators to the CCC cultures markedly improved indole alkaloid production and release of these alkaloids into the medium. Treatment with 250 mM mannitol and 4 g/l KCl yielded 42.3 mg l(-1) and 33.6 mg l(-1)of ajmalicine, r

Cerium dioxide nanoparticles protect Catharanthus roseus against arsenic (As) toxicity by enhancing oxidative tolerance, modulating gene expression and minimizing As uptake.

Arsenic (As) is a toxic metalloid, and its increasing contamination in soil has become an important public health concern. Nevertheless, cerium dioxide nanoparticles (CeO2 NPs) have shown potential for phytostabilization of metal polluted soil. Currently during pot trial, As toxicity (As30, As70 mg kg-1) decreased growth, photosynthetic pigments, gas exchange attributes, membrane stability indices (MSIs) and relative water contents (RWCs) of Catharanthus roseus. Moreover, As stress obviously increased, malondialdehyde (MDA), As, and hydrogen peroxide (H2O2) levels in C. roseus plants. Converse

Crosstalk between melatonin and nitric oxide restrains Cadmium-induced oxidative stress and enhances vinblastine biosynthesis in Catharanthus roseus (L) G Don.

Nitric oxide functions downstream of the melatonin in adjusting Cd-induced osmotic and oxidative stresses, upregulating the transcription of D4H and DAT genes, and increasing total alkaloid and vincristine contents. A few studies have investigated the relationship between melatonin (MT) and nitric oxide (NO) in regulating defensive responses. However, it is still unclear how MT and NO interact to regulate the biosynthesis of alkaloids and vincristine in leaves of Catharanthus roseus (L.) G. Don under Cd stress. Therefore, this context was explored in the present study. Results showed that Cd t

Cryo-derived plants through embryogenesis showed same levels of vinblastine and vincristine (anticancer) in Catharanthus roseus and had normal genome size.

Cryopreservation of rare plant materials is an important approach for preserving germplasms and is a good added concept to tissue banking. The preservation of embryogenic cell suspensions is even more valuable as the tissues facilitate in producing millions of embryos, plantlets and generates transgenics en masse. Catharanthus roseus is a medicinally important plant that produces a variety of anticancerous phytocompounds and needs conservation of alkaloid producing cell lines. In this study, embryogenic tissue banking has been attempted in C. roseus by the two-step cryopreservation method comb

Red-Blue LED Light and Plasma-Activated Water Enhance Vindoline and Catharanthine Yields in Catharanthus roseus Grown in a Closed Plant Production System.

Madagascar periwinkle [Catharanthus roseus (L.) G. Don] produces vindoline and catharanthine, key precursors used in the semi-synthesis of the high-value anticancer alkaloids vinblastine and vincristine. Closed plant production systems (CPPS) allow precise application of environmental stimuli, including light spectrum and plasma-activated water (PAW). In a previous study, we demonstrated the potential of combining artificial light (white and red) and PAW to enhance the production of these alkaloids in periwinkle grown under controlled CPPS conditions. The present study expands this approach by
