Bridging the gap in Parkinson's disease research: from general pathophysiology to advanced 3d cell culture models.

Parkinson’s disease (PD) is the second most common neurodegenerative disorder, having a substantial negative impact on the quality of life of patients with its wide range of motor and non-motor symptoms. PD has a complex etiology involving aging, genetic, and environmental factors. The hallmark of the pathology in PD is the accumulation of misfolded α-synuclein, a presynaptic vesicle-associated protein, in dopaminergic neurons, causing neuronal death in the substantia nigra pars compacta (SNpc) and other brain regions. This review focuses on the main pathophysiological mechanisms of PD and in

A Three-Dimensional Lung Cell Model to Leptospira Virulence Investigations.

Leptospirosis is a worldwide zoonosis and a serious public health threat in tropical and subtropical areas. The etiologic agents of leptospirosis are pathogenic spirochetes from the genus Leptospira. In severe cases, patients develop a pulmonary hemorrhage that is associated with high fatality rates. Several animal models were established for leptospirosis studies, such as rodents, dogs, and monkeys. Although useful to study the relationship among Leptospira and its hosts, the animal models still exhibit economic and ethical limitation reasons and do not fully represent the human infection. As

Protocol for real-time measurement of mitochondrial bioenergetics in 3D-cultured brain tumor stem cells using the Resipher system.

Aberrant mitochondrial function can lead to severe human diseases, including neurodegenerative diseases and cancer. Here, we describe a cell-based protocol for measuring different mitochondrial respiratory parameters using the high-resolution real-time Resipher system. We optimized this protocol on brain tumor stem cells cultured in three-dimensional spheroids. We provide essential optimization steps for cell seeding density, mitochondrial respiration modulator concentrations, running the assay, and data analysis. For complete details on the use and execution of this protocol, please refer to

Advances in 3D culture systems for maintaining prostate tissue architecture and functional ex vivo prostate organ culture for biomedical applications.

Model systems are inevitable for biomedical research, and advanced prostate three dimensional (3D) models help bridge the gap between in vitro and in vivo systems. Prostate diseases are biologically complex and regulated by hormone signaling, stromal-epithelial interactions, immunological infiltration, and glandular microenvironmental factors. Traditional two-dimensional (2D) cell culture techniques and prostatic cell lines exhibit limitations for translational research, as they reflect varied phenotypic and genotypic characteristics compared to native prostate tissue. Recent advancements in p

A novel 3D cell culture model to study the human small intestinal immune landscape.

Several subsets of mononuclear phagocytes and DCs (MDC) populate the small intestine (SI), and these cells reportedly exert specialized functions in anti-microbial immunity and tolerance. Given the specialized phenotype of these cells, differing from other MDC family members, including their putative circulating blood precursors, local intestinal factors play key instructive roles in their differentiation. We designed an SI cell culture model composed of three intestinal epithelial cell (IEC) types, including absorptive enterocytes (E cells), antigen delivering microfold (M) cells, and mucus-p

Development of a low cell density model for examining astrocyte reaction to 3D suspension using AVIFJ.

Alternating viscous and inertial force jetting (AVIFJ) was used to print single-cells and position small numbers of cells precisely. The effect of nozzle size on single-cell encapsulation and droplet spacing ability was tested using astrocytes. Astrocytes were patterned in Matrigel and examined for 7 d after printing to assess viability and their response to being suspended in 3D at low cell density. The results showed AVIFJ had a single-cell printing efficiency around 30% using nozzles with outer diameters (ODs) of 63μm and 96μm at a 300 000 cells ml-1concentration, and 30% for 143µm and 195µ

Stem cell culture and differentiation in 3-D scaffolds.

Conventional two-dimensional (2-D) cultivation are easy to utilize for human pluripotent stem (hPS) cell cultivation in standard techniques and are important for analysis or development of the signal pathways to keep pluripotent state of hPS cells cultivated on 2-D cell culture materials. However, the most efficient protocol to prepare hPS cells is the cell culture in a three dimensional (3-D) cultivation unit because huge numbers of hPS cells should be utilized in clinical treatment. Some 3-D cultivation strategies for hPS cells are considered: (a) microencapsulated cell cultivation in suspen

A Novel Serum-Free 2D and 3D Culture System Comprising Two Small Molecules for Culturing Mouse Lacrimal Gland Epithelial Cells.

Lacrimal gland (LG) dysfunction is a major cause of aqueous-deficient dry eye disease, and cell-based and tissue engineering therapies show significant potential for this condition. However, the limited availability of sufficient seed cells cultured under serum-free conditions has hindered their widespread application. In this study, we developed a novel serum-free culture system using two small molecules, Y27632 and SB431542 (2C), to efficiently expand LG epithelial cells (LGECs). For the 2D primary culture of LGECs, LGs were isolated from 6-8 weeks old mice and enzymatically digested using D

Wax-printing-free fabrication of paper-supported 3D cancer cell culture.

Three-dimensional (3D) in vitro tumor models are critical for studying transport-limited drug efficacy in solid tumors; however, many existing platforms are technically complex and remain difficult to access. Stacked paper-based tumor models ("cells-in-gel-in-paper", CiGiP) address this challenge by enabling formation of diffusion-limited microenvironments while allowing direct access to cells from distinct tissue depths. Nevertheless, current CiGiP implementations rely on wax or hydrophobic barrier patterning of paper, which has become increasingly inaccessible. Here, we present a wax-printin

Magnetic Bead-Guided Assembly of 3D Primary Human Islet Cells in Decellularized Pancreatic Scaffolds.

Background: Three-dimensional (3D) cell cultures are increasingly recognized as effective models for studying diseases and developing cell therapies. In the endocrine pancreas field, organoids/spheroids derived from human islet cells enable advances in diabetes research, drug screening, and tissue engineering. While various 3D culture methods exist, approaches such as magnetic bead-assisted aggregation remain underexplored for endocrine pancreatic cells. Additionally, the use of biological scaffolds, especially those derived from decellularized pancreatic extracellular matrix, provides a biomi

3D printed gelatin methacryloyl hydrogels for perfusion culture of human trabecular meshwork cells and glaucoma studies.

Glaucoma, a progressive eye disease leading to irreversible blindness, currently affects over 70 million people globally. Elevated intraocular pressure (IOP) is implicated in its development. IOP is carefully regulated by the trabecular meshwork (TM). However, studying TM behavior has been limited to traditional tissue culture studies or costly ex vivo cultures of animal and donor eyes. Developing novel functional TM models could enhance cell/tissue behavior understanding and aid therapeutic development for glaucoma. In this study, we 3D printed a simplified and reproducible model of the human

Establishment of a patient-derived 3D in vitro meningioma model in xeno-free hydrogel for clinical applications.

BACKGROUND: Meningiomas exhibit a complex biology that, despite notable successes in preclinical studies, contributes to the failures of pharmaceutical clinical trials. Animal models using patient tumor cells closely mimic in vivo conditions but are labor-intensive, costly, and unsuitable for high-throughput pharmaceutical testing. In comparison, monolayer cell models (two-dimensional, 2D) are cost-efficient but lack primary tumor cell-cell interactions, potentially overestimating treatment effects. Three-dimensional (3D) models offer an alternative through more precise mimicking of tumor morp

Shape, shrink, spheroid: a DIY high-throughput spheroid generation device.

3D spheroids, which closely replicate three-dimensional cell-cell and cell-extracellular matrix interactions, offer superior predictive capabilities compared to conventional 2D monolayer cultures, positioning them as forward-looking platforms in drug testing, cancer biology, and regenerative medicine. However, high-throughput generation of uniform sized spheroids is still a technological challenge. In one hand, the use of conventional ultra-low attachment (ULA) multiwell plates for this purpose is labor intensive and complex. On the other hand, the use of microfabricated facilities demands cut

3D human cell culture models in periodontal research: A scoping review.

OBJECTIVE(S): Despite the increasing use of 3D in vitro models in periodontal research, there is currently no comprehensive synthesis explicitly mapping how effectively these models translate structural achievements into functional biological fidelity. Thus, this scoping review aims to map the existing literature while critically evaluating the biological complexity and translational value of current human 3D in vitro periodontal models. DESIGN: PubMed, Cochrane Library, and Web of Science databases were searched to identify relevant original studies detailing the development and application

Two- and Three-Dimensional In Vitro Models of Parkinson's and Alzheimer's Diseases: State-of-the-Art and Applications.

In vitro models play a pivotal role in advancing our understanding of neurodegenerative diseases (NDs) such as Parkinson's and Alzheimer's disease (PD and AD). Traditionally, 2D cell cultures have been instrumental in elucidating the cellular mechanisms underlying these diseases. Cultured cells derived from patients or animal models provide valuable insights into the pathological processes at the cellular level. However, they often lack the native tissue environment complexity, limiting their ability to fully recapitulate their features. In contrast, 3D models offer a more physiologically rele

3D Spheroid and Organoid Models to Study Neuroinfection of RNA Viruses.

Three-dimensional culture models of the brain enable the study of neuroinfection in the context of a complex interconnected cell matrix. Depending on the differentiation status of the neural cells, two models exist: 3D spheroids also called neurospheres and cerebral organoids. Here, we describe the preparation of 3D spheroids and cerebral organoids and give an outlook on their usage to study Rift Valley fever virus and other neurotropic viruses.

Modeling 3D Glioblastoma: A review of spheroid culture techniques and standardization challenges.

Glioblastoma (GB) is among the most aggressive and treatment-resistant cancers, highlighting the urgent need for physiologically relevant preclinical models. Traditional two-dimensional (2D) cultures fall short in replicating the complex microenvironment of GB, whereas three-dimensional (3D) culture models offer improved structural, biochemical, and functional resemblance to in vivo tumors. However, the lack of a standardized protocol for GB spheroid formation leads to significant inter-study variability and limited reproducibility. This review summarizes the key variables affecting spheroid g

Temporal reassignment and correspondence evaluation with quality control for time-course imaging of 3D cell culture.

Longitudinal imaging of 3D cell cultures like tumor organoids and spheroids offers crucial insights into cancer progression and treatment. However, spatial displacement during time-course imaging, caused by matrix detachment or experimental artifacts, can confound analyses. We present TRACE-QC, an application of the Procrustes technique to evaluate data integrity and rectify mislabeling in longitudinal imaging of 3D cell culture. Our algorithm integrates permutation-based optimization with Procrustes analysis. By using X and Y coordinates of images, it accurately reorders, matches, and aligns
