Mast cells are important elements of the body response to foreign antigens, being those represented either by small molecules (allergic response) or harbored by foreign microorganisms (response to parasite infection). These cells derive from hematopoietic stem/progenitor cells present in the marrow. However, in contrast with most of the other hematopoietic lineages, mast cells do not differentiate in the marrow but in highly vascularized extramedullary sites, such as the skin or the gut. Mast cell differentiation in the marrow is activated as part of the body response to parasites. We will rev

The authors describe a rare tumor with overlapping features of true histiocytic sarcoma and interdigitating dendritic cell sarcoma occurring in infancy. A 3-month-old boy presented with hepatosplenomegaly and abdominal distention. He presented a diagnostic challenge at initial presentation despite extensive investigations. On follow-up at age 16 months he had progressive splenomegaly, thrombocytopenia, and worsening coagulopathy. A diagnosis of histiocytic sarcoma with interdigitating dendritic cell differentiation was established after an open biopsy of liver and spleen. A repeat liver biopsy

Since the heterogeneous development of individual follicles in a given ovary cannot be accounted for by changes in circulating gonadotropin levels, local modulatory factors play an important role in the paracrine control of follicular development. The important paracrine role of ovarian steroids has been well established. Oestrogen is important in the augmentation of gonadotropin action. High local concentration of oestrogens enhances the gonadotropin stimulation of aromatase activity, resulting in further increases in oestrogen production. The elevated local oestrogens in the follicular fluid

Intestinal stem and progenitor cells balance proliferation and lineage commitment to maintain epithelial homeostasis and adapt to environmental challenges. Tuft cells are a rare chemosensory lineage that rapidly expands in response to the cytokine IL-13 during helminth infection, yet how epithelial progenitor states influence this response remains incompletely understood. Using genetic mouse models, intestinal organoids, and single-cell approaches, we identify the Ras guanine nucleotide exchange factor RasGRP1 as an epithelial-intrinsic promoter of efficient tuft cell differentiation. Rasgrp1

Germinal centers (GCs) are specialized microenvironments in which B cells undergo affinity maturation through iterative cycles of somatic hypermutation, proliferation and selection, generating antibodies with improved antigen-binding properties. Although GC B cells do not themselves secrete antibody, they differentiate into memory B cells and plasma cells that provide long-term humoral immunity. GC selection has traditionally been viewed as a process that preferentially expands B cells expressing the highest-affinity B cell receptors. However, recent studies have revealed that physiological GC

Esophageal squamous cell carcinoma with ductal differentiation (ESCC-DD) is exceedingly rare. This article presents a 61-year-old man with progressive dysphagia. Imaging studies and endoscopy were suggestive of malignancy. Pathological examination of the biopsy specimen diagnosed squamous cell carcinoma with focal biphasic glandular structures. The patient received chemotherapy with albumin-bound paclitaxel plus cisplatin, combined with immunotherapy using sintilimab injection, alongside supportive care to manage treatment-related adverse effects. One month later, his dysphagia had significant

Mechanical cues have emerged as critical regulators of stem cell fate, acting alongside biochemical signals to govern differentiation and functional maturation. In native tissues, stem cells reside within dynamic and mechanically heterogeneous microenvironments, where physical factors such as matrix stiffness, viscoelasticity, topography, and externally applied forces play essential roles in directing cellular behavior. Recent advances in mechanically active biomaterials have enabled precise control over the mechanical properties of stem cell microenvironments, providing powerful platforms to

Dedicator of Cytokinesis 8 (DOCK8), a member of the DOCK family of guanine nucleotide exchange factors, serves as a key factor in managing the Rho GTPases activity, notably Cdc42 and Rac1, which control key cellular processes, including migration, adhesion, and phagocytosis. Mutations in the DOCK8 gene cause autosomal recessive hyper-IgE syndrome (AR-HIES), a primary immunodeficiency disorder characterized by elevated serum IgE levels, eczema, recurrent infections, and defective immune responses. In this study, a CRISPR/Cas9-generated Dock81281&1282 TG deletion mutation mouse model was constru

Testicular sex cord-stromal tumors are clonal neoplasms, with the majority being of Leydig cell followed by Sertoli cell origins. In Leydig cell tumors, adipocytic differentiation has been previously reported as a possible distinguishing feature, which has not been reported in other sex cord-stromal tumors. Herein, we report a case of a 48-year-old man who presented with an incidentally discovered 1.1 cm testicular mass, for which he underwent partial orchiectomy. Microscopically, the tumor showed features consistent with sex cord-stromal tumor with strong and diffuse nuclear and cytoplasmic r

Protective immunity against pathogens or cancer is mediated by the activation and clonal expansion of antigen-specific naive T cells into effector T cells. To sustain their rapid proliferation and effector functions, naive T cells switch their quiescent metabolism to an anabolic metabolism through increased levels of aerobic glycolysis, but also through mitochondrial metabolism and oxidative phosphorylation, generating energy and signalling molecules1-3. However, how that metabolic rewiring drives and defines the differentiation of T cells remains unclear. Here we show that proliferating effec

Early placenta development involves cytotrophoblast differentiation into extravillous trophoblast (EVT) and syncytiotrophoblast (STB). Defective trophoblast development and function may result in severe pregnancy complications, including fetal growth restriction and pre-eclampsia. The incidence of these complications is increased in pregnancies of fetuses affected by Rubinstein-Taybi syndrome, a developmental disorder predominantly caused by heterozygous mutations in CREB-binding protein (CREBBP) or E1A-binding protein p300 (EP300). Although the acetyltransferases CREBBP and EP300 are paralogs

Memory CD8 T cells are a critical component of protective immunity, and inducing effective memory T-cell responses is a major goal of vaccines against chronic infections and tumours. Considerable effort has gone into designing vaccine regimens that will increase the magnitude of the memory response, but there has been minimal emphasis on developing strategies to improve the functional qualities of memory T cells. Here we show that mTOR (mammalian target of rapamycin, also known as FRAP1) is a major regulator of memory CD8 T-cell differentiation, and in contrast to what we expected, the immunos

Both in vitro and in vivo studies have implicated the c-Myb transcription factor in vascular smooth muscle cell (SMC) proliferation and hematopoiesis. However, its role in differentiation and maturation of contractile, as opposed to proliferating, SMCs has not been investigated. Here we demonstrate that c-myb(-/-) embryonic stem cells (ESCs) are incapable of producing embryoid bodies (EBs) with spontaneously contracting SMCs but can differentiate into contracting cardiomyocytes unimpaired. Quantitative real-time RT-PCR revealed that whereas mesodermal differentiation was unaffected, myocardin,

BACKGROUND: Fumaric acid esters (FAE) are effective against psoriasis vulgaris and monomethylfumarate (MMF) is believed to be the most bioactive metabolite of this medication. Earlier we found that the beneficial effects of FAE medication are accompanied by a downregulation of type 1 cytokine production by T-helper (Th) lymphocytes, which are important as they maintain a type 1 cytokine [interferon (IFN)-gamma, interleukin (IL)-2] environment in the skin lesions of psoriasis vulgaris patients and once maximal beneficial effects are obtained type 2 cytokine production is also decreased. The main topic of this article is B cell development and differentiation, with a special focus on the mechanisms and molecules that regulate the expression of humoral immunity. Molecular epidemiological analysis was performed on the genes responsible for the X-linked agammaglobulinemia (XLA) phenotype of the majority of Italian patients and their distinct mutations were characterized. Mutations in Bruton's tyrosine kinase (BTK), a member of Tec Family of protein tyrosine kinases, have been found to be mainly responsible for XLA disease. The exact function of BTK in signal transduction is not y

Eleven pineal parenchymal tumors were studied using various antibodies specific to the central nervous system and cell-proliferation-related antigen MIB-1 in order to examine the divergent types of cell differentiation and also evaluate prognosis. Electron microscopy was also performed. All tumors were immunohistochemically positive to chromogranin A and alpha B crystallin, and were also highly positive to retinal S protein. Pineocytoma cells contained microtubules, intermediate filaments, glial bundles, clear-centered vesicles and synaptic apparati. Pineoblastoma cells also had microtubules a

Cholera toxin (CT) is a powerful oral immunogen and adjuvant that elicits strong IgG and IgA antibody responses. In our study we investigated whether this property of CT was associated with an effect on B cell isotype differentiation. Initially, we determined the effect of CT on normal LPS-induced Peyer's patch B cells and found that whereas CT is strongly inhibitory of IgM production, it increases by approximately three-fold the number and frequency of IgG- and IgA-producing cells. Subsequently, using cell sorting technology, we demonstrated that CT acts on membrane (m)IgM+, mIgG/mIgA- B cell

The nonparallel relationships between development of classes of B lymphocytes and of serum immunoglobulins during ontogeny and in some patients imply: 1) that B-cell differentiation is discontinuous and 2) that class-specific-controls for terminal B-lymphocyte differentiation exist. Three separate defects in B-cell differentiation in vivo have been identified using pokeweed mitogen stimulation of lymphocytes. B lymphocytes may fail to develop, may develop but lack the capacity to differentiate further, or may be normal in both respects. The latter result suggests involvement of other cell type

BACKGROUND: In vitro modeling is a powerful approach to investigate the pathomechanisms driving human congenital conditions. Here, we use human embryonic stem cells (hESCs) to model Nager and Rodriguez syndromes, two craniofacial conditions characterized by hypoplastic neural crest-derived craniofacial bones, caused by pathogenic variants of SF3B4, a core component of the spliceosome.
RESULTS: We observed that siRNA-mediated knockdown of SF3B4 interferes with the production of hESC-derived neural crest cells, as seen by a marked reduction in neural crest gene expression. This phenotype is ass
