Single-cell encapsulation of mesenchymal stromal cells via ECM-mimetic supramolecular hydrogels enhances therapeutic efficacy.

Mesenchymal stromal cells (MSCs) hold great promise for tissue regeneration due to their potent paracrine effects. However, the absence of extracellular matrix (ECM) support following transplantation significantly compromises their survival and therapeutic efficacy. To address this, we developed a single-cell encapsulation strategy using an ECM-mimetic supramolecular hydrogel system based on host-guest chemistry. In this approach, cholesterol-polyethylene glycol-adamantane is inserted into the MSC membrane via hydrophobic interactions, enabling the subsequent formation of a uniform hydrogel co

Encapsulation cell therapy for mucopolysaccharidosis type VII using genetically engineered immortalized human amniotic epithelial cells.

Mucopolysaccharidosis type VII (MPSVII) is a lysosomal storage disease resulted from a deficiency of the enzyme beta-glucuronidase (GUSB), which is necessary for degradation of glycosaminoglycans (GAGs). The deficiency of GUSB causes progressive accumulation of GAGs and subsequent lysosomal distension in multiple tissues, including the central nervous system (CNS). In murine experiments, bone marrow transplant, enzyme replacement, viral vectors, and genetically modified cells were successfully used for correction of the visceral accumulation of GAGs, but little improvement was seen in the brai

Growth of tumour cell lines in polymer capsules: ultrastructure of encapsulated PC12 cells.

Recent studies indicate that polymer-encapsulated PC12 cells release sufficient amounts of dopamine to significantly alter behavioural paradigms in animals with unilateral lesions of dopaminergic midbrain neurons. Because cell fine structure provides a useful measure for assessment of storage function, exocytosis, metabolism, cell activity and cell viability, we examined the ultrastructure of PC12 cells grown in semi-permeable polymer capsules maintained in vitro or implanted into the forebrain of rats or guinea pigs. Encapsulated PC12 cells remained viable and continued to divide for the enti

Sickle cell anemia and severe infections due to encapsulated bacteria.

Overwhelming infections caused by encapsulated bacteria are an important cause of morbidity and death in children with sickle cell anemia. The most important contributing factors to this increased susceptibility to infections are an opsonophagocytic defect due to an abnormality of the alternate pathway of complement activation, a state of functional hyposplenia, and a lack of specific circulating antibodies as a developmental phenomenon. If the inordinately high, early mortality rate associated with sickle cell anemia is to be prevented, early diagnosis of affected infants is crucial. Prophyla

Vesicular Transport of Encapsulated microRNA between Glial and Neuronal Cells.

Exosomes (EXs) and extracellular microvesicles (EMVs) represent a diverse assortment of plasma membrane-derived nanovesicles, 30-1000 nm in diameter, released by all cell lineages of the central nervous system (CNS). They are examples of a very active and dynamic form of extracellular communication and the conveyance of biological information transfer essential to maintain homeostatic neurological functions and contain complex molecular cargoes representative of the cytoplasm of their cells of origin. These molecular cargoes include various mixtures of proteins, lipids, proteolipids, cytokines

Encapsulated cell therapy for neurodegenerative diseases: from promise to product.

Delivering therapeutic molecules, including trophic factor proteins, across the blood brain barrier to the brain parenchyma to treat chronic neurodegenerative diseases remains one of the great challenges in biology. To be effective, delivery needs to occur in a long-term and stable manner at sufficient quantities directly to the target region in a manner that is selective but yet covers enough of the target site to be efficacious. One promising approach uses cellular implants that produce and deliver therapeutic molecules directly to the brain region of interest. Implanted cells can be precise

Engineering Immune-Protective Stem Cell-Derived β-Cell Grafts for Type 1 Diabetes: Encapsulation Technologies and the Emerging Graft Ecosystem.

BACKGROUND: Stem cell-derived β-cell replacement has emerged as a promising strategy for the treatment of type 1 diabetes (T1D), potentially addressing the limitations of exogenous insulin therapy and donor-dependent islet transplantation. However, despite substantial progress in stem cell differentiation and transplantation technologies, long-term graft survival remains constrained by immune rejection, recurrent autoimmunity, fibrosis, hypoxia, and inadequate vascularization. Encapsulation technologies have therefore become central to the development of clinically viable bioartificial pancrea

Transparent Phase Change Materials for Solar-Cell Front-Encapsulation Combining High Transmittance and Passive Thermal Management.

Solid-solid phase change materials (SS-PCMs) are promising candidates for solar-cell thermal management because they can buffer heat accumulation through latent heat storage. However, their practical application is still hindered by poor optical transmittance and insufficient mechanical stability. In this work, we present a robust material design strategy to fabricate highly transparent and morphologically stable phase change films via the synergistic crosslinking polymerization of octadecyl acrylate with tailored diacrylate crosslinkers. Measured at 20 °C (below the transition point), the opt

Poly(ethylene oxide)/Gelatin-Based Biphasic Photocrosslinkable Hydrogels of Tunable Morphology for Hepatic Progenitor Cell Encapsulation.

Macroporous hydrogels have great potential for biomedical applications. Liquid or gel-like pores were created in a photopolymerizable hydrogel by forming water-in-water emulsions upon mixing aqueous solutions of gelatin and a poly(ethylene oxide) (PEO)-based triblock copolymer. The copolymer constituted the continuous matrix, which dominated the mechanical properties of the hydrogel once photopolymerized. The gelatin constituted the dispersed phase, which created macropores in the hydrogel. The microstructures of the porous hydrogel were determined by the volume fraction of the gelatin phase.

Review of Advanced Hydrogel-Based Cell Encapsulation Systems for Insulin Delivery in Type 1 Diabetes Mellitus.

: Type 1 Diabetes Mellitus (T1DM) is characterized by the autoimmune destruction of β-cells in the pancreatic islets. In this regard, islet transplantation aims for the replacement of the damaged β-cells through minimally invasive surgical procedures, thereby being the most suitable strategy to cure T1DM. Unfortunately, this procedure still has limitations for its widespread clinical application, including the need for long-term immunosuppression, the lack of pancreas donors and the loss of a large percentage of islets after transplantation. To overcome the aforementioned issues, islets can be

Mast Cells Selectively Deliver Extracellular Vesicle-Encapsulated mRNA to Colorectal Cancer Cells.

Mast cells (MCs), a type of granulocytic immune cell, exert contrasting effects on tumorigenesis. The anti- or pro-tumorigenic activity of MCs depends on the cancer type, tumor microenvironment, and MC localization within the tumor. Consequently, their role remains controversial and poorly understood across multiple cancer types, including colorectal cancer (CRC). Most proposed mechanisms underlying MC activity in CRC have focused on MC secretion of biological factors. In this study, we demonstrated that MCs transfer extracellular vesicles containing mRNAs and proteins to CRC cells. This proce

Cell-Based Therapies for Type 1 Diabetes: A Narrative Review of Stem Cell-Derived Islets, Encapsulation Strategies, and Recent Clinical Trials.

Type 1 diabetes (T1D) is an autoimmune disease characterized by the destruction of insulin-producing pancreatic β-cells, necessitating lifelong insulin therapy. While islet transplantation has demonstrated efficacy in restoring glycemic control, donor scarcity and the requirement for chronic immunosuppression limit its widespread application. Recent advances in stem cell biology, bioengineering, and immunomodulation have catalyzed a paradigm shift toward stem cell-derived β-cell replacement therapies. This narrative review presents recent clinical trials and technological innovations in cell-b

Preparation of Decellularized Tissue as Dual Cell Carrier Systems: A Step Towards Facilitating Re-epithelization and Cell Encapsulation for Tracheal Reconstruction.

Surgical treatment of tracheal diseases, trauma, and congenital stenosis has shown success through tracheal reconstruction coupled with palliative care. However, challenges in surgical-based tracheal repairs have prompted the exploration of alternative approaches for tracheal replacement. Tissue-based treatments, involving the cultivation of patient cells on a network of extracellular matrix (ECM) from donor tissue, hold promise for restoring tracheal structure and function without eliciting an immune reaction. In this study, we utilized decellularized canine tracheas as tissue models to devel

Ghrelin Receptor Blockade Restricts Hepatic Echinococcus granulosus Infection by Suppressing Host Cell Proliferation and Enhancing Fibrotic Encapsulation.

INTRODUCTION: Cystic echinococcosis (CE) is a parasitic liver disease caused by Echinococcus granulosus (E.g) infection, characterized by hepatic fibrosis and liver necrosis. Ghrelin and its receptor GHSR have been proven to regulate the progression of hepatic E.g infection, and GHSR knockout inhibits infection progression. Nevertheless, the specific regulatory mechanism remains poorly understood. METHODS: In this study, mice with hepatic E.g infection were intraperitoneally injected with ghrelin protein and the GHSR antagonist [D-Lys3]-GHRP-6 for 90 days, and the related molecular and pathol

Genipin-crosslinked double PLL membranes overcome the strength-diffusion trade-off in cell encapsulation without compromising biocompatibility.

Cell microencapsulation technologies allow non-autologous implantation of therapeutic cells for sustained drug delivery purposes. The perm-selective membrane of these systems provides resistance to rupture, stablishes the upper molecular weight limit in bidirectional diffusion of molecules, and affects biocompatibility. Thus, despite being a decisive factor to succeed in terms of biosafety and therapeutic efficacy, little progress has been made in its optimization so far. Here we show that, compared to other usually used coating designs, genipin-crosslinked double poly-L-lysine (GDP) membranes

Comparing Single Cell Versus Pellet Encapsulation of Mesenchymal Stem Cells in Three-Dimensional Hydrogels for Cartilage Regeneration.

While the gold standard for inducing mesenchymal stem cell (MSC) chondrogenesis utilizes pellet culture, most tissue engineering strategies for cartilage regeneration encapsulate MSCs as single cells, partially due to the technical challenge to homogeneously encapsulate cell pellets in three-dimensional (3D) hydrogels. It remains unclear whether encapsulating MSCs as single cell suspension or cell aggregates in 3D hydrogels would enhance MSC-based cartilage formation. In this study, we determined that the optimal size of MSC micropellets (μPellets) that can be homogeneously encapsulated in hyd

Bioprinting of GelMA/PEGDA Hybrid Bioinks for SH-SY5Y Cell Encapsulation: Role of Molecular Weight and Concentration.

Current clinical interventions for large peripheral nerve gap injuries are limited. However, hydrogels that are cell-instructive, mechanically compliant, and have an appropriate biodegradation profile for nerve guidance conduit applications are limited. In this study, a photocrosslinkable gelatin methacryloyl (GelMA) and polyethylene glycol diacrylate (PEGDA) hybrid bioink is developed. Encapsulation of primary salivary gland cells in enzymatically degradable poly(ethylene glycol) hydrogels promotes acinar cell characteristics.

UNLABELLED: Radiation therapy for head and neck cancers leads to permanent xerostomia due to the loss of secretory acinar cells in the salivary glands. Regenerative treatments utilizing primary submandibular gland (SMG) cells show modest improvements in salivary secretory function, but there is limited evidence of salivary gland regeneration. We have recently shown that poly(ethylene glycol) (PEG) hydrogels can support the survival and proliferation of SMG cells as multicellular spheres in vitro. To further develop this approach for cell-based salivary gland regeneration, we have investigated

Non-Stoichiometric Click Polymerization for Size-Controlled and Mild Encapsulation of Cells.

Artificial cell shells can achieve protection, identification, measurement, and manipulation of cells, but most cell encapsulation methods fail to control shell thickness, directly limiting the shell function in practical applications. The in-situ polymerization method has the potential to control the encapsulation shell but lacks chemical driving forces to enable shell thickness from nanoscale to micrometer-scale. The micrometer-scale shell can enhance the functionality in cell encapsulation and extend its applications to broader fields. In this work, we utilized the efficient and mild thiol-
