5-Fluorouracil causes leukocytes attraction in the peritoneal cavity by activating autophagy and HMGB1 release in colon carcinoma cells.

Signals released by leukocytes contribute to tumor growth and influence the efficacy of antineoplastic treatments. The outcome of peritoneal carcinomatosis treatments is unsatisfactory, possibly because chemotherapy activates events that have in the long-term deleterious effects. In this study we offer evidence that 5-fluorouracile (5-FU), besides provoking apoptosis of MC38 colon carcinoma cells, induces a striking attraction of leukocytes both in an orthotopic model of colon carcinomatosis in vivo and in monocyte-migration assays in vitro. Leukocyte attraction depends on the presence of High

Decidual Inflammation Drives Chemokine-Mediated Immune Infiltration Contributing to Term Labor.

Infiltration of maternal peripheral leukocytes into the uterine tissues is a critical event occurring before, during, and after term labor (TL). In this article, we investigate the contribution of uterine smooth muscle (myometrium) and pregnant endometrium (decidua) to the inflammatory process during human TL. We hypothesize that labor-related physiological inflammation is orchestrated by uterine-secreted cytokines, which dually activate the uterine vascular endothelium and maternal leukocytes to promote their adhesion and infiltration into the uterus. Using Luminex and ELISA assays, we examin

Epstein-Barr herpesvirus infection: inhibition by immunologically induced mediators with interferon-like properties.

When sensitized leukocytes were re-exposed to EBV antigen or to tuberculin-purified protein derivatives, they produced lymphokines, including the migration inhibition factor, which inhibited the migration of guinea-pig peritoneal exudate cells, and other lymphokines with the characteristics of interferon, which inhibited EBV-induced transformation and EBV superinfection of target cells. Unsensitized leucocytes from sero-negative adults and from neonates did not produce lymphokines when challenged with the antigens. This indicates that cell-mediated immunity and its associated soluble mediators

Conjugated linoleic acid targets β2 integrin expression to suppress monocyte adhesion.

Chronic recruitment of monocytes and their subsequent migration through the activated endothelium contribute to atherosclerotic plaque development. Integrin-mediated leukocyte adhesion is central to this process. Conjugated linoleic acid (CLA) has the unique property of inducing regression of pre-established murine atherosclerosis via modulation of monocyte/macrophage function. Understanding the mechanisms through which CLA mediates its atheroprotective effect may help to identify novel pathways that limit or reverse atherosclerosis. In this study, we identified a novel mechanism through which

Oral Cladribine Impairs Intermediate, but Not Conventional, Monocyte Transmigration in Multiple Sclerosis Patients across a Model Blood-Brain Barrier.

Multiple sclerosis (MS) is a disease in which the immune system damages components of the central nervous system (CNS), leading to the destruction of myelin and the formation of demyelinating plaques. This often occurs in episodic "attacks" precipitated by the transmigration of leukocytes across the blood-brain barrier (BBB), and repeated episodes of demyelination lead to substantial losses of axons within and removed from plaques, ultimately leading to progressive neurological dysfunction. Within leukocyte populations, macrophages and T and B lymphocytes are the predominant effectors. Among c

Lanostane-type triterpenoids from ethnomedicinal fungi inhibit endothelial cell proliferation.

ETHNOPHARMACOLOGICAL RELEVANCE: Ethnomedicinal fungi have a long history of use in the treatment of tumors and inflammatory disorders, pathologies often associated with dysregulated angiogenesis and immune-vascular interactions. However, their effects on endothelial cell functions relevant to angiogenesis and inflammation remain insufficiently characterized. AIM OF THE STUDY: The aim of this study was to identify the bioactive compounds from ethnopharmacologically relevant medicinal fungi by screening their ethanolic extracts for their effects on angiogenesis- and inflammation-related endothe

Homing of Mesenchymal Stem Cells.

SUMMARY: Mesenchymal stem cells (MSCs) are primarily fibroblast-like cells. Yet, once studied under conditions of shear stress when flowing along endothelial cells in vitro or in blood vessels, as well as in classic migration assays such as chemotaxis assays, MSCs have recently been found to function similarly to leukocytes in many ways. Firstly, MSCs express several homing receptors which are typically activated during extravasation of leukocytes. Secondly, some of these receptors are definitely functional, and required for their tissue localization in certain physiological or pathological co

Examination of patients suspected as having hypersensitivity to iodinated contrast media with leukocyte migration test.

In vivo tests may be used for the diagnosis of allergy to iodinated contrast media (ICM); however, the tests do not provide definitive diagnosis and are associated with risks for patients. Diagnoses based on in vitro tests are limited, and there are almost no relevant studies. Herein, the authors examined involvement of allergic reaction from a multilateral standpoint in 39 patients suspected of having ICM allergies using leukocyte migration test (LMT). The positive rate of LMT was 44%. A comparison with the positive rate of LMT in drugs other than ICM (74%) indicated 30% difference, which was

Inhibitory Effect of Punicalagin on Inflammatory and Angiogenic Activation of Human Umbilical Vein Endothelial Cells.

Punicalagin, a major ellagitannin isolated from pomegranate, is proved to have various pharmacological activities with an undefined therapy mechanism. The objective of this research was to demonstrate the effect of punicalagin on anti-inflammatory and angiogenic activation in human umbilical vein endothelial cells (HUVECs) and their potential mechanisms. Endothelial-leukocyte adhesion assay was applied to evaluate primary cultures of HUVECs activation following tumor necrosis factor alpha (TNF-α) treatment. The endothelial cell proliferation, migration, permeability and tube formation were ass

Netrin-1 suppresses CCL2-driven macrophage migration: implications for modulating vascular inflammation and aortic sinus enlargement in hyperlipidaemic mice.

BACKGROUND: Atherosclerosis is driven by sustained recruitment and accumulation of monocytes and macrophages within arterial walls through chemokine-mediated pathways. Netrin-1, an immunomodulatory molecule originally identified in neural development, has emerged as a potential regulator of vascular inflammation, though its precise effects on chemokine-driven leukocyte migration remain undefined. METHODS: We investigated netrin-1 effects on macrophage migration in vitro using real-time impedance-based assays and assessed systemic effects in vivo through intravital microscopy and aortic sinus

Circular RNA GLIS2 promotes colorectal cancer cell motility via activation of the NF-κB pathway.

Circular RNAs (circRNAs) are a newly discovered type of biological molecule that belongs to the noncoding RNA family. Abundant evidence has shown that circRNAs are involved in the progression of various cancers. However, the particular functions of circRNAs in colorectal cancer (CRC) remain elusive. In this study, we investigated the differentially expressed circRNAs in three pairs of cancer tissue and adjacent normal tissue of CRC. We revealed that circGLIS2 expression was higher in CRC tissue and cell lines. Gain-and-loss function assays showed that circGLIS2 was involved in the regulation o

Immune response to Japanese Encephalitis virus in mother mice and their congenitally infected offspring.

The immune response to Japanese encephalitis virus (JEV) was assessed in JEV-infected mice (mothers) and their offspring. The congenitally infected baby mice responded poorly in all assays for cell-mediated immunity. The total number of their splenic cells remained unaltered but the percentage of T cells was significantly reduced; a depressed delayed hypersensitivity response was seen against both homologous (JEV) and heterologous (sheep erythrocytes) antigens. In addition, significantly higher leukocyte migration inhibition (LMI) of spleen cells in the presence of specific antigen was observe

A new leukocyte hyperadhesion syndrome of delayed cord separation, skin infection, and nephrosis.

Leukocyte adhesion deficiency (LAD) I is a well-described genetic disorder in which leukocytes are unable to migrate to sites of inflammation due to mutations in the ITGB2 gene coding for the β subunit of β2 (CD18) leukocyte integrins. The classic symptoms of the disease present in the newborn period as failure of separation of the umbilical cord and recurrent bacterial infections, which continue throughout life. We report on a patient with these clinical manifestations but with normal ITGB2 gene sequencing excluding LAD-I, normal carbohydrate-deficient transferrin testing excluding LAD-II, an

Leukocyte procoagulant activity in man: an in vitro correlate of delayed-type hypersensitivity.

Human mononuclear leukocytes generate cell-bound procoagulant activity (LPCA) after incubation with an antigen (mumps or tuberculin) to which the donor was previously sensitized. An inhibitor of coagulation appears to be liberated into the extracellular culture fluid during incubation of leukocytes with the sensitizing antigen. Removal of this activity before measuring LPCA resulted in a reliable test that correlated directly with delayed skin reactivity. The assay was particularly sensitive in that cells from weakly sensitized donors who reacted only to high doses of tuberculin (100 TU) in th

Changes in leukocyte migration during carbonic anhydrase activity inhibition.

Experiment 1 evaluated changes in leukocyte migration during acetazolamide (AZ) inhibition of carbonic anhydrase activity in leukocytes. AZ induced changes in the intracellular calcium concentration, and extracellular calcium is thought to be a factor inducing an increase in leukocyte migration. Next, Experiment 2 determined whether extracellular calcium concentration was a primary factor influencing leukocyte migration in the absence of AZ. The distance of leukocyte migration increased in a dose-dependent manner with AZ despite the presence of IL-8 or LPS in Experiment 1. The extracellular ca

Pseudomonas aeruginosa Alters Critical Lung Epithelial Cell Functions through Activation of ADAM17.

Severe epithelial dysfunction is one major hallmark throughout the pathophysiological progress of bacterial pneumonia. Junctional and cellular adhesion molecules (e.g., JAMA-A, ICAM-1), cytokines (e.g., TNFα), and growth factors (e.g., TGFα), controlling proper lung barrier function and leukocyte recruitment, are proteolytically cleaved and released into the extracellular space through a disintegrin and metalloproteinase (ADAM) 17. In cell-based assays, we could show that the protein expression, maturation, and activation of ADAM17 is upregulated upon infection of lung epithelial cells with Ps

Single-Cell Sequencing Reveals That CD4+ T Cells Eliminate Senescent Prostate Epithelium to Delay Progression of Benign Prostatic Hyperplasia.

Benign prostatic hyperplasia (BPH) is an age-related condition characterized by progressive prostate enlargement driven in part by the accumulation of senescent epithelial cells and their pro-inflammatory secretome. Using human single-cell RNA sequencing and laser capture microdissection, we identified C-X-C Motif Chemokine Ligand 13 (CXCL13) as a key chemokine secreted by senescent prostate epithelial cells. CXCL13 recruits CD4+ T cells via the C-X-C Chemokine Receptor Type 5 (CXCR5) receptor, facilitating immune recognition through human leukocyte antigen-DR isotype (HLA-DR) and promoting se

CD44 Presence in inflamed pulp tissue.

Inflammation of pulp tissue appears as a consequence of caries progression. Its main characteristic is inflamed infiltrate whose cells contain lymphocytes. CD44 is a widely expressed adhesion molecule present in several body cells such as leukocytes and parenchymatous cells, including endothelial cells, epithelial cells, and unstriated muscle cells. It interacts with hyaluronic acid, collagen, laminin, and fibronectin, and there are data that indicate an important role in the migration of leukocytes from the bloodstream toward inflammation areas. This project, which applied the immunologic ass

Neutrophil Activities in Human Ocular Toxoplasmosis: An In Vitro Study With Human Cells.

PURPOSE: Retinal damage in ocular toxoplasmosis reflects Toxoplasma gondii-induced cell lysis and reactive inflammation. Human retinal histopathology demonstrates the presence of neutrophils, but activities of this leukocyte subset are unstudied. We conducted in vitro experiments to evaluate roles for neutrophils as retinal taxis for T. gondii and as contributors to the inflammation. METHODS: Human neutrophils were isolated from peripheral blood. Migration to disease-relevant chemokines was evaluated in transwells, seeded with human retinal endothelial cells for some assays, using neutrophils
