Synergistic movements of Ca(2+) and Bax in cells undergoing apoptosis.

Apoptosis is a physiological counterbalance to mitosis and plays important roles in tissue development and homeostasis. Cytosolic Ca(2+) has been implicated as a proapoptotic second messenger involved in both triggering apoptosis and regulating cell death-specific enzymes. A critical early event in apoptosis is associated with the redistribution of Bax from cytosol to mitochondria and endoplasmic reticulum (ER) membranes; however, the molecular mechanism of Bax translocation and its relationship to Ca(2+) is largely unknown. Here we provide functional evidence for a synergistic interaction bet

The alteration by ouabain of calcium movements in human red cell ghosts.

1. The influence of ouabain on net Ca movements was studied in human erythrocyte ghosts by atomic absorption spectrophotometry. 2. Ouabain (10(-5) M) showed a dual effect, altering both entry and exit of Ca from K-rich ghosts incubated in a high-Na medium in the presence of 10 mM-Ca. 3. Stimulation of Ca entry was observed in the first 15 min at 37 degrees C, whereas during the subsequent 15 min incubation ouabain elicited Ca extrusion. This latter effect was eliminated when the ouabain concentration was raised to 1 mM. 4. Ouabain-dependent Ca movements were abolished by replacing both interna

Retrograde movements determine effective stem cell numbers in the intestine.

The morphology and functionality of the epithelial lining differ along the intestinal tract, but tissue renewal at all sites is driven by stem cells at the base of crypts1-3. Whether stem cell numbers and behaviour vary at different sites is unknown. Here we show using intravital microscopy that, despite similarities in the number and distribution of proliferative cells with an Lgr5 signature in mice, small intestinal crypts contain twice as many effective stem cells as large intestinal crypts. We find that, although passively displaced by a conveyor-belt-like upward movement, small intestinal

Source of the eye-movement signal reaching real-motion cells.

The stability of visual perception despite eye movements suggests the existence in the visual system of neurons able to recognize whether the movement of a retinal image is due to the actual movement of an object or is self-induced by the ocular movement. We found neurons of this type in several areas of the monkey visual cortex and named them "real-motion" cells. Extracellular recordings were carried out from single neurons of the cortical prestriate area V3A of two awake macaque monkeys. Eighty-seven neurons were studied by comparing their responses during stimulus movement across the statio

Ret signaling in ureteric bud epithelial cells controls cell movements, cell clustering and bud formation.

Ret signaling promotes branching morphogenesis during kidney development, but the underlying cellular mechanisms remain unclear. While Ret-expressing progenitor cells proliferate at the ureteric bud tips, some of these cells exit the tips to generate the elongating collecting ducts, and in the process turn off Ret. Genetic ablation of Ret in tip cells promotes their exit, suggesting that Ret is required for cell rearrangements that maintain the tip compartments. Here, we examine the behaviors of ureteric bud cells that are genetically forced to maintain Ret expression. These cells move to the

Complementary cortical and thalamic contributions to cell type-specific striatal activity dynamics during movement.

Coordinated motor behavior emerges from information flow across brain regions. How long-range inputs drive cell type-specific activity within motor circuits remains unclear. Cytomegalovirus disrupts Lamin A/C to control microtubule-mediated nuclear movement and cell migration.

Human cytomegalovirus (HCMV) disrupts the inner nuclear Lamin A/C meshwork to facilitate virion egress. Here, we show that targeting Lamin A/C also plays a previously unrecognized role in HCMV-induced rewiring of cytoskeletal connections that control nuclear movement and cell migration at later stages of infection. Specifically, HCMV was found to employ distinct strategies to downregulate Lamin A/C and SUN2, which together regulate linker of nucleoskeleton and cytoskeleton (LINC) complexes that most often connect the nuclear membrane with cytoplasmic actin filaments. Unexpectedly, inhibition o

Mechanical Cues for Triggering and Regulating Cellular Movement Selectively at the Single-Cell Level.

Cell motility plays important roles in many biophysical and physiological processes ranging from in vitro biomechanics, wound healing, to cancer metastasis. This work introduces a new means to trigger and regulate motility individually using transient mechanical stimulus applied to designated cells. Using BV2 microglial cells, our investigations indicate that motility can be reproducibly and reliably initiated using mechanical compression of the cells. The location and magnitude of the applied force impact the movement of the cell. Based on observations from this investigation and current know

Splenomegaly induced by anemia impairs T cell movement in the spleen partially via EPO.

The spleen is an important secondary lymph organ. Splenomegaly induced by anemia could affect the function of spleen in immune responses. We observe that anemia induced in mice with reduced peripheral T cell trafficking to the spleen T cell zones as well as CCL21 and CCL19 expression. In accordance with previous research, we found that the production of EPO in the mice kidney was sharply increased post anemia. In addition, mice were injected with different doses of EPO. Our results show that with the increased dosage of EPO, the chemokine expression in the spleen is lowered with a decrease in

Effects of Stromal Cell-Derived Factor 1 on Osteoblast Activity During Orthodontic Tooth Movement.

OBJECTIVE: This study aimed to clarify the relationship between stromal cell‑derived factor 1 (SDF‑1) and osteoblast-related responses in periodontal ligament (PDL) during orthodontic tooth movement (OTM) after tooth extraction, based on the hypothesis that local inhibition of SDF‑1/C‑X‑C chemokine receptor type 4 (CXCR4) signaling with the CXCR4 antagonist AMD3100 would attenuate osteoblast differentiation and alveolar bone remodeling during OTM. METHODS: Thirty-six 6-week-old Wistar/Sprague-Dawley male rats were randomly divided into control and experimental groups. After the extraction of

Steroid hormone signaling synchronizes cell migration machinery, adhesion and polarity to direct collective movement.

Migratory cells - either individually or in cohesive groups - are critical for spatiotemporally regulated processes such as embryonic development and wound healing. Their dysregulation is the underlying cause of formidable health problems such as congenital abnormalities and metastatic cancers. Border cell behavior during Drosophila oogenesis provides an effective model to study temporally regulated, collective cell migration in vivo. Developmental timing in flies is primarily controlled by the steroid hormone ecdysone, which acts through a well-conserved, nuclear hormone receptor complex. Ecd

The Equine Movement Disorder "Shivers" Is Associated With Selective Cerebellar Purkinje Cell Axonal Degeneration.

"Shivers" is a progressive equine movement disorder of unknown etiology. Clinically, horses with shivers show difficulty walking backward, assume hyperflexed limb postures, and have hind limb tremors during backward movement that resembles shivering. At least initially, forward movements are normal. Given that neither the neurophysiologic nor the pathologic mechanisms of the disease is known, nor has a neuroanatomic locus been identified, we undertook a detailed neuroanatomic and neuropathologic analysis of the complete sensorimotor system in horses with shivers and clinically normal control h

Determination of pathways for sodium movement across corneal endothelial cell derived plasma membrane vesicles.

A bovine corneal endothelial cell plasma membrane vesicle preparation was used to investigate passive Na+ transport across the plasma membrane of these cells. Sodium accumulation rate into the vesicle was not dependent on the presence of HCO3- or a HCO3- gradient, but was stimulated by a trans-vesicle pH gradient. Amiloride, furosemide and DIDS all reduced the rate of Na+ accumulation. The data indicate the presence of at least two independent pathways for passive sodium movement across the vesicle: the first probably via a Na+/H+ exchanger and the second a furosemide inhibitable Na+ entry mec

Effect of deoxycholic acid on Ca2+ movement, cell viability and apoptosis in human gastric cancer cells.

Deoxycholic acid (DOA) is one of the secondary bile acids used as a mild detergent for the isolation of membrane associated proteins. This study examined whether the secondary bile acid, DOA, altered Ca(2+) movement, cell viability and apoptosis in SCM1 human gastric cancer cells. The Ca(2+)-sensitive fluorescent dye fura-2 was used to measure [Ca(2+)]i. DOA-evoked [Ca(2+)]i rises concentration dependently. The response was reduced by removing extracellular Ca(2+). DOA-evoked Ca(2+) entry was inhibited by store-operated Ca(2+) channel inhibitors (nifedipine, econazole and SKF96365), the protei

Chemokines in Leishmaniasis: Map of cell movements highlights the landscape of infection and pathogenesis.

Pathogen interactions with the host immune response components are critical for establishing protective immunity and pathological responses against Leishmania parasites. A predominant proinflammatory profile associated with enhanced phagocytosis trigger a cell-mediated immune response that is relevant to infection control. On the other hand, an anti-inflammatory phenotype, correlated with a predominant modulated/regulatory response, favors intracellular proliferation of Leishmania parasites and disease progression. In this context, chemokines play an important role in determining cellular comp

A marker of acid-secreting membrane movement in rat gastric parietal cells.

A monoclonal antibody (mab 146.14) marker of the movement of acid-secreting membranes in rat gastric parital cells has been produced and characterized. Mab 146.14 recognized a 95-kD major component of a purified membrane fraction of rat gastric mucosa, the protein composition of which was similar to that of well characterized porcine H+ -K+ ATPase-enriched membranes, and that presented the characteristic shift of density depending on whether it was purified from resting or stimulated tissues. Further biochemical analysis characterized the antigen as a membranous protein that might be in its na

Complex spikes perturb movements and reveal the sensorimotor map of Purkinje cells.

Computations that are performed by the cerebellar cortex are transmitted via simple spikes of Purkinje cells (P-cells) to downstream structures, but because P-cells are many synapses away from muscles, we do not know the relationship between modulation of simple spikes and control of behavior. Here, we recorded the spiking activities of hundreds of P-cells in the oculomotor vermis of marmosets during saccadic eye movements and found that following the presentation of a visual stimulus, the olivary input to a P-cell coarsely described the direction and amplitude of the visual stimulus as well a

Excitability of anterior horn cells after periodic or discrete repetitive movements.

INTRODUCTION/AIMS: It has been well established that spasticity interferes with smooth joint movements. Although the degree of spasticity is related to the excitability of anterior horn cells and is thought to improve after repetitive movements, the effect of the rhythm of repetitive movements on the excitability of anterior horn cells remains unknown. Therefore, we investigated the excitability of anterior horn cells after periodic and discrete repetitive movements using F waves. METHODS: Right-handed, healthy subjects were recruited for this study. Subjects then performed periodic or discre

Symmetry and fluctuation of cell movements in neural crest-derived facial mesenchyme.

In the face, symmetry is established when bilateral streams of neural crest cells leave the neural tube at the same time, follow identical migration routes and then give rise to the facial prominences. However, developmental instability exists, particularly surrounding the steps of lip fusion. The causes of instability are unknown but inability to cope with developmental fluctuations are a likely cause of congenital malformations, such as non-syndromic orofacial clefts. Here, we tracked cell movements over time in the frontonasal mass, which forms the facial midline and participates in lip fus
