Volumes of cochlear nucleus regions in rodents.

The cochlear nucleus receives all the coded information about sound from the cochlea and is the source of auditory information for the rest of the central auditory system. As such, it is a critical auditory nucleus. The sizes of the cochlear nucleus as a whole and its three major subdivisions - anteroventral cochlear nucleus (AVCN), posteroventral cochlear nucleus (PVCN), and dorsal cochlear nucleus (DCN) - have been measured in a large number of mammals, but measurements of its subregions at a more detailed level for a variety of species have not previously been made. Size measurements are re

The neurons of the medial geniculate body in the mustached bat (Pteronotus parnellii).

The neurons in the medial geniculate body were studied in Golgi preparations from adult mustached bats (Pteronotus parnellii). Their somatic and dendritic configurations were compared with those of cells in other, nonecholocating mammals. A second goal was to use the thalamic nuclear subdivisions derived from Golgi material to integrate the findings in parallel studies of cytoarchitecture, immunocytochemistry, and tectothalamic connections. Three primary divisions are defined. The ventral division is large and has a stereotyped neuronal organization. Medium-sized perikarya (about 10 microns in

Synaptic endfeet in the 'acoustic nerve nucleus' of the rat. An electron microscopic study.

The medial portion of the cochlear nerve of the rat contains astrocytes, oligodendrocytes and neurons. These neurons form what has been called the 'acoustic nerve nucleus'. This nucleus has been studied here at the electron microscopic level. Its neurons are large and round, showing an eccentric nucleus, fibrillary bodies and rough endoplasmic reticulum which is not arranged in stacks. The somata and dendrites receive synaptic endfeet which can be classified into three groups according to vesicle size and shape. In general, the ultrastructural characteristics of these cells are similar to thos

Heterogeneous Skeletal Muscle Cell and Nucleus Populations Identified by Single-Cell and Single-Nucleus Resolution Transcriptome Assays.

Single-cell RNA-seq (scRNA-seq) has revolutionized modern genomics, but the large size of myotubes and myofibers has restricted use of scRNA-seq in skeletal muscle. For the study of muscle, single-nucleus RNA-seq (snRNA-seq) has emerged not only as an alternative to scRNA-seq, but as a novel method providing valuable insights into multinucleated cells such as myofibers. Nuclei within myofibers specialize at junctions with other cell types such as motor neurons. Nuclear heterogeneity plays important roles in certain diseases such as muscular dystrophies. We survey current methods of high-throug

Breeding status affects motoneuron number and muscle size in naked mole-rats: recruitment of perineal motoneurons?

Naked mole-rats live in large colonies and exhibit a strict reproductive hierarchy. Each colony has one breeding female and one to three breeding males; all other individuals are nonreproductive subordinates. Subordinates show a remarkable lack of sex differences in behavior and anatomy, but can become reproductive if removed from the colony. We recently reported that the striated perineal muscles and their innervating motoneurons, which are sexually dimorphic in all other mammals examined to date, are not dimorphic in subordinate naked mole-rats. Here we asked whether sexual differentiation o

Role of nuclear ATPases in nuclear mechanics and cell migration through confined spaces: Opposite effects of BRG1 and cohesin.

Deformation of the nucleus often presents a barrier to cell migration through tight spaces, such as those encountered as cells move through tissues or across extracellular matrix barriers. Reorganization of the nucleus to allow its passage through spaces much smaller than its resting diameter requires forces generated by the cytoskeleton, as well as active reorganization within the nucleus driven by ATPases that cross-link or move chromatin. Here, we show that two different nuclear ATPases, the BRG1/BRM motor of the BAF or SWI/SNF complex and the bifunctional cross-linking and loop extruding c

Disordered development of macrophages in non-Hodgkin's lymphoma.

Macrophage development in 20 untreated patients with non-Hodgkin's lymphoma (NHL) has been studied and compared with that in 20 normal subjects. Morphometric measurements were carried out on ultrastructural features of cell, nucleus and mitochondria during 6 days suspension culture of blood monocytes in the presence of autologous serum and lymphocytes. The results were subjected to multivariate and univariate analysis of variance. Statistically significant differences were found between the subject groups with respect to the volumes and surface areas of cell, nucleus and mitochondria, to the e

CNS-endocrine pancreas system. I. The hypothalamus response to insulin deficiency.

The anterior, medial and posterior hypothalamic nuclei in alloxan-diabetic male rats were studied by karyometry. Selective responsiveness of separate nuclei of the medial basal hypothalamus (MBH) and the anterior hypothalamus was revealed. In the MBH cell nuclear-size changes were most prominent in the ventromedial nucleus and less pronounced, but significant, in the arcuate nucleus. In the anterior hypothalamus a significant response was produced by the supraoptic nucleus. The authors do not exclude the possibility that the reaction of the neurosecretory cells of the supraotic nucleus is indi

Development of the dorsal lateral geniculate nucleus in normal and visually deprived Siamese cats.

Neuronal cell bodies in the lateral geniculate nucleus of normal and of monocularly-deprived Siamese cats have been measured. Seventeen normally reared Siamese cats, ranging in age between 20 and 120 days, were used to determine rates of normal geniculate cell growth. A second group of five adult Siamese cats reared from bith with the lids of one eye closed were used to study the effects of monocular visual deprivation upon geniculate cell size. For each of the normal and visually deprived Siamese cats, the cross-sectional areas of 600 lateral geniculate cells were measured from camera lucida

Primary afferent projections to the central octavus nuclei in the elasmobranch, Rhinobatos sp., as demonstrated by nerve degeneration.

Nerve degeneration studies were performed to determine the central projections of the octavus nerve in the guitarfish. Rhinobatos sp. Most first-order octavus nerve afferents are confined to the ventral longitudinal column of the medulla, which consists of four octavus nuclei: the nucleus octavus anterior (NOA), nucleus octavus magnocellularis (NOM), nucleus octavus descendens (NOD); and nucleus octavus posterior (NOP). Central projections of the octavus nerve outside of the ventral octavus nuclear column are to the vestibulolateral lobe of the cerebellum, the large multipolar cells that lie d

Morphometric-interferometric investigations of neurons in the medial thalamic nucleus of newborn rats following maternal treatment with ethanol.

The nucleus and cell size, as well as the dry weight and protein contents of neurons in the medial thalamic nucleus of newborn rats, exposed to ethanol in utero, were examined on the first day of postnatal life. Comparison was made with a control group of animals, not exposed to ethanol. No significant changes in the protein content and dry weight values were observed. The perikarya of neurons from animals exposed to ethanol were significantly larger than those of the controls. The findings indicate that the nucleus and perikaryon are affected differently by alcohol, following intrauterine exp

Topology and Nuclear Size Determine Cell Packing on Growing Lung Spheroids.

Within multicellular living systems, cells coordinate their positions with spatiotemporal accuracy to form various tissue structures and control development. These arrangements can be regulated by tissue geometry, biochemical cues, as well as mechanical perturbations. However, how cells pack during dynamic three-dimensional multicellular architectures formation remains unclear. Here, examining a growing spherical multicellular system, human lung alveolospheres, we observe an emergence of hexagonal packing order and a structural transition of cells that comprise the spherical epithelium. The cytolinker plectin regulates nuclear mechanotransduction in keratinocytes.

The transmission of mechanical forces to the nucleus is important for intracellular positioning, mitosis and cell motility, yet the contribution of specific components of the cytoskeleton to nuclear mechanotransduction remains unclear. In this study, we examine how crosstalk between the cytolinker plectin and F-actin controls keratin network organisation and the 3D nuclear morphology of keratinocytes. Using micro-patterned surfaces to precisely manipulate cell shape, we find that cell adhesion and spreading regulate the size and shape of the nucleus. Disruption of the keratin cytoskeleton thro

Effects of prenatal alcohol exposure on the sexually dimorphic nucleus of the preoptic area of the hypothalamus in male and female rats.

Prenatal alcohol exposure can produce a variety of behavioral and physiological alterations, including changes in sexually dimorphic behaviors. It has been proposed that alcohol alters these behaviors by altering prenatal androgen and/or other steroid levels. This study was designed to examine the effects of prenatal alcohol exposure on a sexually dimorphic neuroanatomical structure, namely, the sexually dimorphic nucleus of the preoptic area of the hypothalamus (SDN-POA). This nucleus is larger in males than females and is sensitive to perinatal sex steroid exposure. The SDN-POA was examined

Measurement of cellular traction forces during confined migration.

To migrate efficiently through tissues, cells must transit through small constrictions within the extracellular matrix. However, in vivo environments are geometrically, mechanically, and chemically complex, and it has been difficult to understand how each of these parameters contribute to the propulsive strategy utilized by cells in different confining environments. To address this, we employed a sacrificial micromolding approach to generate polymer substrates with tunable stiffness, controlled adhesivity, and user-defined microscale geometries. We combined this together with live-cell imaging

Maintenance of neuronal size gradient in MNTB requires sound-evoked activity.

The medial nucleus of the trapezoid body (MNTB) is an important source of inhibition during the computation of sound location. It transmits fast and precisely timed action potentials at high frequencies; this requires an efficient calcium clearance mechanism, in which plasma membrane calcium ATPase 2 (PMCA2) is a key component. Deafwaddler (dfw2J ) mutant mice have a null mutation in PMCA2 causing deafness in homozygotes (dfw2J /dfw2J ) and high-frequency hearing loss in heterozygotes (+/dfw2J ). Despite the deafness phenotype, no significant differences in MNTB volume or cell number were obse

Histometric changes and cell death in the thalamus after neonatal neocortical injury in the rat.

Freezing injury to the developing cortical plate results in a neocortical malformation resembling four-layered microgyria. Previous work has demonstrated that following freezing injury to the somatosensory cortex, males (but not females) have more small and fewer large cells in the medial geniculate nucleus. In the first experiment, we examined the effects of induced microgyria to the somatosensory cortex on neuronal numbers, neuronal size, and nuclear volume of three sensory nuclei: ventrobasal complex, dorsal lateral geniculate nucleus, and medial geniculate nucleus. We found that there was

Apoptosis in auditory brainstem neurons after a severe noise trauma of the organ of Corti: intracochlear GDNF treatment reduces the number of apoptotic cells.

We have studied the morphological and cellular changes in the cochlear nucleus (CN) after cochlear nerve degeneration and whether these changes can be prevented by rescuing the primary cochlear neurons from degeneration with local glial cell line derived neurotrophic factor (GDNF) treatment. Degeneration of spiral ganglion neurons was seen to lead to a reduction of the volume of the anteroventral cochlear nucleus (AVCN); the size of the cell nuclei in the AVCN also was reduced. No differences were observed in cell density. After intrascalar GDNF treatment the volume of the AVCN was significant

The morphology of the diencephalon in the Prosimii. II. The Lemuroidea and Lorisoidea. Part I. Thalamus and metathalamus.

This study (Part I) covers the comparative structure of the thalamus and metathalamus in the prosimian superfamilies, Lorisoidea and Lemuroidea. Several lemuroid specimens, namely Lemus, Cheirogaleus, Microcebus, Avahi and Daubentonia, and lorisoid specimens such as Galago, Perodicticus and Loris have been used in this extensive study. The anterior nuclear group does not show much difference among these species, although attention is focussed on the differentiation of the principal anterior nucleus into anteromedial and anteroventral parts; the latter increases in structural complexity. Exfoliative cytopathology of alveolar rhabdomyosarcoma.

Exfoliative cytopathology of alveolar rhabdomyosarcoma is presented. A 34-year-old male was found to have alveolar rhabdomyosarcoma of the retroperitoneum at autopsy. Ascites was recognized in the course of this disease. Cytological features in the ascites were characterized by relatively small-sized ovoid cells observed singly or in loose clusters. The neoplastic cells possessed small amount of rather vesicular cytoplasm and an eccentric nucleus. The nucleus characteristically revealed multiple deep indentations in the nuclear membrane, a few prominent eosinophilic nucleoli and finely granula
