Electrode assemblies used for electroporation of cultured cells.

Electroporation was initially developed for the introduction of DNA into cells which grow in suspension and was performed in a cuvette with two flat electrodes on opposite sides. Different configurations were subsequently developed for the electroporation of adherent cells in situ, while the cells were growing on nonconductive surfaces or a gold-coated, conductive support. We developed an assembly where the cells grow and are electroporated on optically transparent, electrically conductive indium-tin oxide (ITO). This material promotes excellent cell adhesion and growth, is inert and durable,

Cultured human muscle cells and respiratory chain deficiencies.

Cultured muscle cells are useful in the study of respiratory chain disorders. Muscle tissue is affected in most cases and muscle biopsies are often taken for diagnostic purposes. Small samples of the biopsies can provide large numbers of muscle cells. In contrast with most other cell types, the muscle cells can differentiate in culture. Upon differentiation, multinuclear myotubes are formed by fusion of myoblasts and muscle-specific proteins are synthesized in the myotubes. Muscle cells can be used for diagnostic purposes, e.g. by measurement of the lactate and pyruvate synthesis, although wit

Effects of prolactin on cultured human endometrial cells.

We report the effects of PRL on cultured human endometrial cells (epithelial cells and stromal cells) and discuss the role of PRL in implantation. Essentially similar responses to PRL in cell attachment and growth were obtained both in epithelial and stromal cells: a low concentration of PRL (3-30 ng/ml of PRL) enhanced cell attachment and growth. In contrast, the presence of a high concentration of PRL (over 100 ng/ml) resulted in a decrease in cell attachment and growth in normal culture conditions. Morphological analysis of epithelial glandular reformation on basement membrane extracts indi

Geometrical constraints on the shape of cultured cells.

Previous studies on cultured cells revealed that the values of certain shape descriptors were highly correlated despite the fact that there was little mathematical interdependence (Heckman, CA: In Advances in Cell Culture, Vol. 4, K. Maramorosch, ed, pp 85-156, 1985). The independence of such descriptors was tested by calculating the values of correlations among descriptors for a data set consisting of macroscopic objects. The descriptors were unlinked for this data set, confirming that high correlations reflected specific characteristics of cultured cells. These characteristics were identifie

Hypersensitivity of cultured ataxia-telangiectasia cells to etoposide.

Fibroblasts from patients with ataxia-telangiectasia (A-T) were found to be hypersensitive to killing by the antineoplastic agent etoposide. The A-T fibroblast strains GM5823, GM367, and GM2052 were twofold to threefold more sensitive to killing by etoposide than fibroblasts from normal controls (AG1521, AG1522, and IMR90). A simian virus 40 (SV40)-transformed, immortal human fibroblast line (GM5849) derived from the A-T cell line GM5823 was also studied. GM5849 retained the unusual sensitivity of nontransformed A-T fibroblast lines to x-irradiation, bleomycin, and neocarzinostatin (zinostatin

Alkaline phosphatase isozymes in cultured human cancer cells.

Alkaline phosphatase, an ubiquitous enzyme is known to exist in several isozymic forms. At least three different isozymes have now been identified in humans. Alkaline phosphatase isozymes are among the substances synthesized ectopically by a variety of human tumors and many continuous cell lines derived from different cancers have retained the capacity to produce these membrane-located glycoproteins. This paper reviews the identification of alkaline phosphatase isozymes in cultured tumor cells and relates these findings with recent developments concerning these cell membrane located glycoprote

Regenerating human epithelia with cultured stem cells: feeder cells, organoids and beyond.

More than 40 years ago, Howard Green's laboratory developed a method for long-term expansion of primary human epidermal keratinocytes by co-culture with 3T3 mouse embryonic fibroblasts. This was a breakthrough for in vitro cultivation of cells from human skin and later for other epithelia: it led to the first stem cell therapy using cultured cells and has vastly increased our understanding of epithelial stem cell biology. In recent years, new methods to expand epithelial cells as three-dimensional organoids have provided novel means to investigate the functions of these cells in health and dis

Beyond cultured cells: Frontiers in in situ structural biology.

Structural biology is no longer confined to isolated macromolecules. Correlative light and electron microscopy workflows at cryogenic temperatures have been developed and optimised in cultured cell systems, opening the door for cellular structural biology. The next frontier is expanding these approaches to primary cells, tissues, biopsies, and patient material. The complexity of these samples means each presents unique challenges. Here, we review recent advances that enable the study of primary cells and complex samples. These examples demonstrate the diversity in available workflows and indic

Nucleus Morphometry in Cultured Epithelial Cells Correlates with Phenotype.

Phenotype of cultured ocular epithelial transplants has been shown to affect clinical success rates following transplantation to the cornea. The purpose of this study was to evaluate the relationship between cell nucleus morphometry and phenotype in three types of cultured epithelial cells. This study provides knowledge for the development of a non-invasive method of determining the phenotype of cultured epithelium before transplantation. Cultured human conjunctival epithelial cells (HCjE), human epidermal keratinocytes (HEK), and human retinal pigment epithelial cells (HRPE) were analyzed by

Transcriptome analysis of primary bovine extra-embryonic cultured cells.

The dataset described in this article pertains to the article by Hue et al. (2015) entitled "Primary bovine extra-embryonic cultured cells: A new resource for the study of in vivo peri-implanting phenotypes and mesoderm formation" [1]. In mammals, extra-embryonic tissues are essential to support not only embryo patterning but also embryo survival, especially in late implanting species. These tissues are composed of three cell types: trophoblast (bTCs), endoderm (bXECs) and mesoderm (bXMCs). Until now, it is unclear how these cells interact. In this study, we have established primary cell cultu

[Karyotype analysis using cells cultured from fetal bladder centesis].

OBJECTIVE: To explore the feasibility of karyotype analysis using cells cultured from fetal bladder centesis samples. METHODS: Samples were derived from fetal bladder centesis for 3 fetuses featuring giant bladder and oligohydramnios. Following in vitro culture, cells were routinely processed and stained for chromosome analysis. RESULTS: For all 3 cases, cell culture has achieved success. Sufficient metaphase cells were obtained for chromosome counting and karyotype analysis. The karyotypes of the 3 fetuses were respectively 46, XY, 46, XX, t(1;5)(q22;q12)[7]/46, XX[4], and 46, XY. Riluzole increases glutamate uptake by cultured C6 astroglial cells.

Riluzole is a drug approved for the treatment of amyotrophic lateral sclerosis (ALS) and may be effective for the treatment of other neurodegenerative and neuropsychiatric disorders. Riluzole exerts diverse actions on the central nervous system, including altering glutamate release and uptake, and therefore act diminishing glutamate extracellular levels, but the underlying mechanism of these actions is still unknown. Here, we demonstrate that riluzole stimulated glutamate uptake and augmented the expression of the glutamate EAAC1 transporter in C6 astroglial cell cultures. The effect of riluzo

Radioactive labeling of mitochondrial translation products in cultured cells.

The mammalian mitochondrial genome contains 37 genes, 13 of which encode polypeptide subunits in the enzyme complexes of the oxidative phosphorylation system. The other genes encode the rRNAs and tRNAs necessary for their translation. The mitochondrial translation machinery is located in the mitochondrial matrix, and is exclusively dedicated to the synthesis of these 13 enzyme subunits. Mitochondrial disease in humans is often associated with defects in mitochondrial translation. This can manifest as a global decrease in the rate of mitochondrial protein synthesis, a decrease in the synthesis

Cytotoxicity of voriconazole on cultured human corneal endothelial cells.

The purpose of the present study was to evaluate the toxicity of voriconazole on cultured human corneal endothelial cells (HCECs). HCECs were cultured and exposed to various concentrations of voriconazole (5.0 to 1,000 μg/ml). Cell viability was measured using a Cell Counting Kit-8 (CCK-8) and live/dead viability/cytotoxicity assays. Cell damage was assessed using phase-contrast microscopy after 24 h of exposure to voriconazole. To analyze the effect of voriconazole on the intercellular barrier, immunolocalization of zonula occludens 1 (ZO1) was performed. A flow cytometric assay was performed

Radiation-induced bystander effects in cultured human stem cells.

BACKGROUND: The radiation-induced "bystander effect" (RIBE) was shown to occur in a number of experimental systems both in vitro and in vivo as a result of exposure to ionizing radiation (IR). RIBE manifests itself by intercellular communication from irradiated cells to non-irradiated cells which may cause DNA damage and eventual death in these bystander cells. It is known that human stem cells (hSC) are ultimately involved in numerous crucial biological processes such as embryologic development; maintenance of normal homeostasis; aging; and aging-related pathologies such as cancerogenesis and

Effects of vibration on differentiation of cultured PC12 cells.

Different types of physiological-mechanical stress, such as shear stress in vascular endothelial cells or hydrostatic pressure in chondrocytes are well known as regulators of cell function. In this study, the effects of vibration, a type of non-physiological mechanical stimulation, on differentiation of rat pheochromocytoma (PC12) cells are reported. A nano-vibration system was designed to produce nanometer-scale vibration. The frequency and amplitude of the nano-vibrations were monitored by a capacitance displacement sensor connected to an oscilloscope. When PC12 cells exposed to nerve growth

Membrane insertion by anthrax protective antigen in cultured cells.

The enzymatic moieties of anthrax toxin enter the cytosol of mammalian cells via a pore in the endosomal membrane formed by the protective antigen (PA) moiety. Pore formation involves an acidic pH-induced conformational rearrangement of a heptameric precursor (the prepore), in which the seven 2beta2-2beta3 loops interact to generate a 14-strand transmembrane beta-barrel. To investigate this model in vivo, we labeled PA with the fluorophore 7-nitrobenz-2-oxa-1,3-diazole (NBD) at cysteine residues introduced into the 2beta2-2beta3 loop. Each labeled PA was bound to CHO cells, and NBD fluorescenc
