Regulation of cellulase production via calcium signaling in Trichoderma reesei under PEG8000 stress.

In this study, the effect of polyethylene glycol 8000 (PEG8000) stress on cellulase biosynthesis in Trichoderma reesei CICC2626 via calcium signaling was investigated, and a plausible mechanism by which intracellular Ca2+ regulates the transcription of cellulase genes was proposed. The results indicated that the total cellulase (filter paper-hydrolyzing activity [FPase]), endoglucanase (carboxymethyl cellulase activity [CMCase]), and β-glucosidase activities of the strain were 1.3-, 1.2-, and 1.3-fold higher than those of the control (no PEG8000 addition) at a final concentration of 1.5% (w/v)

Comparing deconstruction of sugarcane bagasse by a coculture of thermophilic bacteria and a fungal cellulase system.

BACKGROUND: Lignocellulosic biomass is a promising feedstock for sustainable bioethanol production due to its abundance, renewability, and high carbohydrate content. Among these feedstocks, sugarcane bagasse (SCB), a fibrous byproduct of sugar and ethanol mills, is particularly abundant in Brazil. However, the complex and recalcitrant structure of lignocellulose hinders its biological conversion into biofuels. There are different enzymatic architectures to overcome this challenge. Fungal enzyme systems rely on non-complexed cellulase components that act independently and are often used in comb

Mechanistic partitioning and functional synergy of bacterial cellulosome and fungal cellulases in cellulose saccharification.

Synergistic saccharification involving bacterial cellulosome and fungal cellulases holds significant promise for industrial applications; however, this integrated approach remains largely under-investigated. This study dissects the synergy from structural characterization, adsorption analysis, crystallinity evolution, and hydrolysis performance evaluation. The cellulosome exhibited superior enhancement of substrate accessibility after enzymatic hydrolysis of sugarcane bagasse compared with fungal cellulases. The distinct changes in crystallinity observed during the hydrolysis of microcrystalli

Cellulase adsorption and relationship to features of corn stover solids produced by leading pretreatments.

Although essential to enzymatic hydrolysis of cellulosic biomass to sugars for fermentation to ethanol or other products, enzyme adsorption and its relationship to substrate features has received limited attention, and little data and insight have been developed on cellulase adsorption for promising pretreatment options, with almost no data available to facilitate comparisons. Therefore, adsorption of cellulase on Avicel, and of cellulase and xylanase on corn stover solids resulting from ammonia fiber expansion (AFEX), ammonia recycled percolation (ARP), controlled pH, dilute acid, lime, and s

Alkaliphilic microbial cellulases: sources, structural insights, and bioengineering approaches for industrial applications.

As the biofuel/biorefinery/green economy advances quickly, the need for durable cellulases to operate under varying conditions in the industrial sector has increased. A specific type of cellulase that has emerged from this demand is alkaliphilic cellulase (ALKC). Due to their stability and high catalytic activity at elevated pH ranges (e.g., pH 8-11), ALKCs are being developed as biocatalysts for alkaline pretreatment and other harsh, high-pH industrial processes. The purpose of this review is to provide an updated overview of ALKCs focusing on their microbial origins, structural characteristic

Investigating the synergetic dispersing effect of hydrolyzed biomacromolecule Cellulase and SDS on CuPc pigment.

In this paper, the dispersion performance of biomacromolecule hydrolyzed cellulase from Trichoderma reesei on copper phthalocyanine (CuPc) pigment was first studied. The effect of hydrolysis time, cellulase concentration and environmental pH on the dispersion performance was investigated by particle size distribution and suspension transmittance measurement. The hydrolysis degree of cellulase was determined by FTIR, XRD, UV-vis and fluorescence spectra, potential and particle size analysis, respectively. Subsequently, the hydrolyzed cellulase was combined with sodium dodecyl sulfate (SDS) for

Early cellular events and potential regulators of cellulase induction in Penicillium janthinellum NCIM 1366.

Cellulase production by fungi is tightly regulated in response to environmental cues, and understanding this mechanism is a key pre-requisite in the efforts to improve cellulase secretion. Based on UniProt descriptions of secreted Carbohydrate Active enZymes (CAZymes), 13 proteins of the cellulase hyper-producer Penicillium janthinellum NCIM 1366 (PJ-1366) were annotated as cellulases- 4 cellobiohydrolases (CBH), 7 endoglucanases (EG) and 2 beta glucosidases (BGL). Cellulase, xylanase, BGL and peroxidase activities were higher for cultures grown on a combination of cellulose and wheat bran, wh

Cellulase production by Trichoderma reesei when cultured on xylose-based media supplemented with sorbose.

The ability of L-sorbose to stimulate cellulase production In shake flask culture of Trichoderma reesei was examined in mineral salts media (initial pH 5.0) containing either 1.0% D-xylose, 1.0% cellulose, and/or 0.1, 0.3, or 0.5% L-sorbose. When sorbose was the only carbon source, growth was limited, little substrate was utilized, pH increased, and cellulase activity was not apparent. The other carbon sources promoted good growth, pH dropped sharply to 2.5-3.0, substrate was utilized rapidly, and cellulase activity was detected. After three weeks of fermentation, twice as much cellulase activ

TSC2 is a positive master regulator of cellulase production by affecting protein secretion in Trichoderma reesei.

Understanding the molecular mechanism behind fungal cellulase production is key to enhancing cellulase production, but the relevant signaling pathways are yet to be explored. This study elucidates TSC2 as a positive master regulator of cellulase production in Trichoderma reesei. Genetic deletion of tsc2 severely impaired cellulase secretion, particularly under cellulose induction. The mTOR inhibitor Torin 1 significantly inhibited the production of CMC and BGL at the early stage. Torin 1-mediated mTORC1 inhibition partially restored CMC and BGL production in ΔTSC2, linking hyperactive mTORC1 t

Treatment of a Large Gastric Phytobezoar With Coca-Cola, Cellulase, and Endoscopic Fragmentation in a Community Hospital.

Bezoars are indigestible masses that can cause gastrointestinal complications such as obstructions and perforations. We report the case of a 60-year-old man with a history of type 2 diabetes mellitus and duodenal ulcer surgery. The patient developed a large gastric phytobezoar that caused persistent dyspepsia and epigastric pain. Upper endoscopy confirmed a 10-cm phytobezoar and a 1.5-cm active ulcer at the gastric angle. To soften the bezoar, Coca-Cola and cellulase were administered to the patient. This treatment significantly reduced the bezoar's hardness, permitting its successful endoscop

Optimization of a natural medium for cellulase by a marine Aspergillus niger using response surface methodology.

The components of a natural medium were optimized to produce cellulase from a marine Aspergillus niger under solid state fermentation conditions by response surface methodology. Eichhornia crassipes and natural seawater were used as a major substrate and a source of mineral salts, respectively. Mineral salts of natural seawater could increase cellulase production. Raw corn cob and raw rice straw showed a significant positive effect on cellulase production. The optimum natural medium consisted of 76.9 % E. crassipes (w/w), 8.9 % raw corn cob (w/w), 3.5 % raw rice straw (w/w), 10.7 % raw wheat b

Effective microwell plate-based screening method for microbes producing cellulase and xylanase and its application.

Cellulase and xylanase are main hydrolysis enzymes for the degradation of cellulosic and hemicellulosic biomass, respectively. In this study, our aim was to develop and test the efficacy of a rapid, high-throughput method to screen hydrolytic-enzyme-producing microbes. To accomplish this, we modified the 3,5-dinitrosalicylic acid (DNS) method for microwell plate-based screening. Targeted microbial samples were initially cultured on agar plates with both cellulose and xylan as substrates. Then, isolated colonies were subcultured in broth media containing yeast extract and either cellulose or xy

Transcriptome sequencing of Bacillus sp. HMM and effect of GH gene overexpression on cellulase activities.

Bacillus sp. HMM can efficiently yield cellulase. However, transcriptome data and sequences of important genes such as cellulase gene are indistinct now, which restricts genetic reconstruction of strain HMM. In current paper, GO, COG and KEGG classification of unigenes from transcriptome sequencing of strain HMM was analyzed in detail, and cellulase gene GH (TR1834) encoding glycosylhydrolase was selected. Moreover, transgenic Bacillus sp. HMM overexpressing GH (GHT) with recombinant plasmid PHY-300PLK-GH was constructed. Avicel activity, CMC-Na activity and filter paper activity of GH transge

Delivery of Hexaconazole by the Carboxymethylcellulose grafting and Cellulase/pH responsiveness hollow mesoporous silica nanoparticles.

Conventional pesticides are easily lost, and poorly targeted deposition causes pollution and economic losses because of their large particle sizes. Cellulase/pH Responsiveness Hollow Mesoporous Silica Nanoparticles (Hex/HMSN-CMC) was developed to respond rapidly to changes in the environment. The particle size of Hex/HMSN-CMC was 133.44 nm. The characterization results confirmed the introduction of CMC and a Hex loading capacity of 12 %. Controlled-release kinetics indicated that the drug-loaded system could exhibit specific responses to pH and cellulase, with the cumulative release rate excee

Modulating the pH-activity profile of cellulase A from Cellulomonas fimi by replacement of surface residues.

One industrial process for the production of cellulosic ethanol and or value-added products involves exposing the cellulose content of plant materials by steam explosion in the presence of strong acid, followed by its neutralization and subsequent digestion with a cocktail of cellulolytic enzymes. These enzymes typically have activity optima at slightly acidic or neutral pH and so generating enzymes that are more active and tolerant in more acidic conditions would help to reduce associated costs. Here, we describe the engineering of cellulase A from Cellulomonas fimi as a model to replace resi

Cellulase adsorption and an evaluation of enzyme recycle during hydrolysis of steam-exploded softwood residues.

The sugar yield and enzyme adsorption profile obtained during the hydrolysis of SO2-catalyzed steam-exploded Douglas-fir and posttreated steam-exploded Douglas-fir substrates were determined. After hot alkali peroxide posttreatment, the rates and yield of hydrolysis attained from the posttreated Douglas-fir were significantly higher, even at lower enzyme loadings, than those obtained with the corresponding steam-exploded Douglas-fir. The enzymatic adsorption profiles observed during hydrolysis of the two substrates were significantly different. Ultrafiltration was employed to recover enzyme in

Functional engineering of the ACE3 C-terminal residue S705 improves cellulase production in Trichoderma reesei.

Improving cellulase production in Trichoderma reesei remains essential for reducing enzyme costs in lignocellulosic biorefineries, whereas effective rational point engineering of ACE3, an essential transcription factor for cellulase production, remains largely unexplored. In this study, we identified serine 705 (S705) in the C-terminal region of ACE3 as an effective engineering site for enhancing cellulase production. Acidic substitutions at S705, such as S705E, severely reduced pNPCase and FPase activities and markedly repressed cellulase-related gene expression. Further analysis showed that
