Recent experimental evidence has led to the interpretation that "enamel-like" material is deposited along the forming mouse molar root surface by cells of Hertwig's epithelial root sheath (HERS cells) and that this material is integral to the developmental program for cementogenesis. The experimental strategy described in this study was to examine selected developmental stages of root formation for mouse first and second mandibular molars in order to localize the cellular sites of amelogenin gene transcripts using high resolution in situ hybridization. Amelogenin is the major structural protei

Periodontal regeneration is no longer viewed as the simple replacement of lost alveolar bone, but as the reprogramming of a diseased regenerative niche. Predictable repair requires coordinated reconstruction of cementum, periodontal ligament (PDL), and alveolar bone within a chronically infected, inflamed, and mechanically loaded microenvironment. Unlike regeneration in bone, skin or muscle, periodontal regeneration requires reconstruction of an integrated cementum-PDL-bone unit. Success therefore depends not on repair of a single tissue compartment, but on coordinated cementogenesis, PDL fibr

Contrary to previous reports, epithelial cell clusters with ultrastructural features similar to classically defined epithelial rests of Malassez were found, for the first time, in areas of repairing orthodontic root resorption. These observations were made on the buccal root surface of premolars extracted for orthodontic purposes from adolescent patients who had required rapid maxillary expansion. Ultrastructurally, the epithelial clusters ranged in size from 2-8 cells and were characterized by the presence of true desmosomes (macula adherens) and tonofilaments. Certain ultrastructural feature

Hertwig's epithelial root sheath (HERS) cells were isolated and recombined with ectomesenchymal cells in vitro utilizing extracellular matrix components as substrate. After 14 days in culture, HERS cells were differentiated and exhibited a stratified organization. These features resembled those observed in vivo as epithelial rests of Malassez. A mineralization process was also present in HERS cells, in which calcium salts were deposited. This mineralization was correlated with the strong immunoexpression of osteopontin by HERS. The results obtained add support to the possible role of HERS in t

Native and recombinant human bone morphogenetic/osteogenic proteins (BMPs/ OPs) singly initiate bone induction in vivo. The finding of synchronous but spatially different BMPs/OPs expression during periodontal tissue morphogenesis suggests novel therapeutic approaches using morphogen combinations based on recapitulation of embryonic development. Twelve furcation defects prepared in the first and second mandibular molars of three adult baboons (Papio ursinus) were used to assess whether qualitative histological aspects of periodontal tissue regeneration could be enhanced and tissue morphogenesi

Many types of localized reactive lesions may occur on the gingiva, including focal fibrous hyperplasia, pyogenic granuloma, peripheral giant cell granuloma and peripheral ossifying fibroma. Peripheral ossifying fibroma is a gingival growth, usually arising from interdental papilla and represents up to 2% of all lesions that are biopsied. Other terms used to describe this lesion include peripheral cementifying fibroma, peripheral fibroma with cementogenesis, peripheral fibroma with osteogenesis, peripheral fibroma with calcification, calcified or ossified fibrous epulis and calcified fibroblast

Periodontal furcation defects are usually addressed by the placement of a physical barrier which may limit the regenerative potential of periodontal wounds. This study morphometrically quantified the regenerative effect of brain-derived neurotrophic factor (BDNF) in furcation defects in a non-human primate model. Grade II furcation defects (with and without induced inflammation prior to surgery) were created on the first and second molars of eight non-human primates. Defects were treated with open flap debridement and subsequently filled with either: Group A; BDNF (500 µg mL-1 ) in high-molecu

1. The scleral grafts appeared to be well accepted as there were no signs of antigenicity or untoward reactions. 2. The gingival connective tissue, periodontal ligament and the periosteum were observed intertwined with sclera at the interface. 3. Sclera was invaded by host fibroblasts, capillaries, and appeared in some areas to be raplaced by a dense connective tissue. 4. Areas of cementogenesis could be observed in all specimens. 5. There were no signs of osteogenesis within the scleral grafts. 6. The alveolar crest appeared relatively nonreactive to sclera. 7. There were no signs of external

Cementum distribution was studied on transversely sectioned roots of 33 three-rooted maxillary and 33 two-rooted mandibular permanent molars. The roots were sectioned at a level midway between furcation and apical region. Cementum thickness was measured at the midpoint of each of the four root surfaces and at a site exhibiting maximal cementum thickness. A reflected light microscope was used. Midpoint cementum thickness ranged between 5 and 800 microm in maxillary molars and between 5 and 700 microm in mandibular molars. Maximal cementum thickness ranged between 25 and 1140 microm in maxillary

This report concerns a family showing both the lethal and mild form of hypophosphatasia in half-sibs. In addition, several other paternal family members with the mild form are documented. The lethal form is characterized by extremely low to absent alkaline phosphatase activity in serum with hypomineralization of the skeleton, whereas mildly affected individuals have enzyme levels intermediate between normal and lethal states. On the basis of this pedigree and because the mildly affected individuals have both biochemical abnormalities and the clinical phenotype of premature tooth loss, we prefe

The development and sequence of eruption of the maxillary cheek-teeth of rabbits were studied by histological methods. The presence of three deciduous molars which were replaced by correspondent premolars and of three permanent molars without predecessors was confirmed. The eruption of the maxillary deciduous molars was shown to begin at 4 days postnatally and that of the permanent molar at 9 days, while the eruption of the premolars occurs from 24 days on, replacing the deciduous molars which are exfoliated. The last tooth to erupt is the M3. At 32 days all the permanent cheek-teeth are erupt

Bone sialoprotein (BSP) is a multifunctional extracellular matrix protein found in mineralized tissues, including bone, cartilage, tooth root cementum (both acellular and cellular types), and dentin. In order to define the role BSP plays in the process of biomineralization of these tissues, we analyzed cementogenesis, dentinogenesis, and osteogenesis (intramembranous and endochondral) in craniofacial bone in Bsp null mice and wild-type (WT) controls over a developmental period (1-60 days post natal; dpn) by histology, immunohistochemistry, undecalcified histochemistry, microcomputed tomography

Invasive cervical resorption (ICR) occurs in the cervical area of the teeth due to the formation of a soft tissue that progressively resorbs dentin. The disease is asymptomatic unless the pulp is exposed. This article presents a case involving a mandibular canine that was treated with a calcium-enriched mixture (CEM) cement. After a full mucoperiosteal flap was performed, the soft tissue was curetted away and the cavity filled with CEM biomaterial. One week later, the supragingival surface of the CEM was polished and covered with composite resin. At a 1-year follow-up visit, the pulp was healt

Amelogenins, major components of developing enamel, are predominantly involved in the formation of tooth enamel. Although amelogenins are also implicated in cementogenesis, their precise spatial expression pattern and molecular role are not clearly understood. Here, we report for the first time the expression of two alternate splice forms of amelogenins, M180 and the leucine-rich amelogenin peptide (LRAP), in the periodontal region of mouse tooth roots. Lack of M180 and LRAP mRNA expression correlated with cementum defects observed in the amelogenin-null mice. The cementum defects were charact

Root resorption is an adverse outcome of orthodontic tooth movement. In addition to the iatrogenic response and compromising the crown-root ratio, root resorption has led to increased malpractice litigation against orthodontists. A clinically acceptable method of treating root resorption has not been validated in the literature to date. Previous research has shown that low-intensity pulsed ultrasound (LIPUS) can enhance healing of various types of traumatized connective tissues and stimulate dental tissue formation. The purpose of this study was to evaluate the effect of LIPUS on the healing p

Resistin-like molecule-β/found in inflammatory zone 2 (RELM-β/FIZZ2) is a cysteine-rich secretory protein that is localized in the epithelium of the gastrointestinal tract and lung alveoli. Previous reports have suggested that this protein regulates glucose metabolism and inflammation. In the present study, to analyze the involvement of RELM-β/FIZZ2 in tooth development, we immunohistochemically examined the localization of RELM-β/FIZZ2 in tooth germs of embryonic days (E) 15-20 and postnatal days (P) 7-42 rats. RELM-β/FIZZ2 was hardly detected in the tooth germ at the bud (E15) stage. However

Ectopic calcification within joints has been reported in humans and rodents exhibiting mutations in genes that regulate the level of extracellular pyrophosphate, e.g., ank and PC-1; however, periodontal effects of these mutations have not previously been examined. These initial studies using ank and PC-1 mutant mice were done to see if such mineral deposition and resulting ankylosis were occurring in the periodontium as well. Surprisingly, results indicated the absence of ankylosis; however, a marked increase in cementum formation on the root surfaces of fully developed teeth of these mutant m

Periodontitis is a chronic inflammatory disease that inflicts damage to periodontal tissues, leading to loss of teeth, and affects systemic health. Traditional treatments can delay inflammation, whereas regeneration of the periodontal complex (periodontal ligament, cementum, and alveolar bone) can better restore periodontal tissue function. In recent years, the regeneration of alveolar bone and the periodontal ligament has been widely reviewed although cementum has received less attention. As an avascular mineralised structure around the tooth, cementum can anchor periodontal ligament fibres t

4 vertical lesions at 3 teeth in 2 volunteer adults with severe periodontitis were treated by open surgical debridement, osseous allografts and barrier membranes. Roots were notched at both gingival margins and the deepest visible calculus and flaps were sutured coronally. Patients were placed on 0.12% chlorhexidine gluconate 2x daily post-surgery, and blocks were harvested 5 to 6 weeks after surgery. No abnormal clinical responses were noted during the observation period. In the 4 sites, the average preoperative pocket depth was 9.4 mm; the post-operative pocket depth averaged 4.9 mm, recessi
